Reduction of Oxidative Stress by Chondroitin Sulfate in the Ovariectomy-Induced Aging Rat (난소절제로 유도한 노화쥐에서 chondroitin sulfate에 의한 산화 스트레스의 감소효과)

  • The ovarian hormone-deficiency induced ovariectomy rat is widely used as an aging model due to its practicality, convenience, and cost effectiveness. The surgically ovariectomized rat induces reactive oxygen species (ROS) generation like aging phenomena. Free oxygen radicals have been proposed as important causative agents of aging. The purpose of this study was to investigate the effect of chondroitin sulfate (CS) to prevent ovariectomy (OVX)-induced oxidative stress. The OVX rats were given intraperitoneally CS at doses of 100 mg/kg and 200 mg/kg daily for fifteen weeks. Malondialdehyde (MDA) levels were determined as well as the activities of superoxide dismutase (SOD), catalase (CAT), reduced-glutathione (GSH), oxidized-glutathione (GSSG), glutathione peroxidase (GPx) in the liver. The liver antioxidative enzyme activity was elevated while MDA concentration decreased in all CS treated animals. The results demonstrated that CS reduced oxidative stress in a dose dependent manner. These results suggest that CS might be a useful candidate for antioxidative reagent.

  • 생체막의 주요 구성성분인 인지질의 2번 위치에 결합한 불포화지방산은 각종 전이금속이나 각종 활성산소들의 공격을 쉽게 받아 지질과산화반응이 일어나서 생체에 유독한 화합물을 생성하게 된다. 생체는 이러한 기구의 해독을 위하여 크게 2가지 방어기전을 갖고 있다. 즉 Vitamin- C, $\alpha$-tocopherol, flavonoid, SOD, catalase 등과 같이 생성된 활성산소를 제거시키는 기구와. 활성산소에 의해 생성된 과산화물을 제거시키는 기구로 glutathione peroxidase (GPX)가 알려졌으며 GPX에 의해 독성이 낮은 수산화물까지 환원시키는 기구가 보고되었다. 그러나 인지질의 과산화물 그대로는 GPX의 기질이 쥘수 없으므로, 산화된 지방산을 절단하는 효소에 대한 기구의 해석이 요구되고 있다. 최근 여러질병에 관련되어 있는 인지질 2번위치의 지방산을 분해하는 phospholipase $A_2$ (PLA$_2$)가 과산화지질의 분해에 관여한다는 주장이 제기되었다. 따라서 본 연구에서는 rat liver microsome에 $CCl_4$투여로 일어나는 과산화반응에 있어서 PLA$_2$의 역할을 규명하기 위하여 본 실험을 행하였다.

Activity of Superoxide dismutase(SOD) by fermented soybean (발효 대두 식품의 Superoxide dismutase(SOD) 활성)

  • This study was performed to evaluate the inhibition effects of fermented soybean on lipid perosidation and antioxidative relative enzyme activity. in vivo. Fermented soybean was induced the high SOD activity, while significantly inhibited on the peroxide value of linoleic acid and lipid perxidation from rat microsome induced by Fe$^{2+}$ ascorbate system, Sprague-Dawley(SD) male rats were fed basic diet, and experimental diets group added 200 or 500 mg/kg fermented soybean for 2 weeks. The effect of fermented soybean is also significantly increased catalase and glutathione peroxidase activities, while significantly inhibited the lipid peroxidation of rat liver microsome in a dose dependent manner. Therefore, these results suggest that fermented soybean has antioxidative activity which is related enzyme to prevention of oxidative stress.s.

A Study on the Mechanism of Calcium Binding Inhibition of Cardiac Sarcoplasmic Reticulum by Oxygen Free Radicals (산소대사물에 의한 심장근 Sarcoplasmic reticulum의 칼슘운반 억제 기전에 관한연구)

  • Mechanism of calcium transport inhibition of cardiac sarcoplasmic reticulum (SR) by oxygen free radicals was examined. Effects of oxygen free radicals generated by xanthine/xanthine oxidase (X/XO) system on isolated porcine ventricle SR were studied with respect to its calcium binding, lipid peroxidation, SH-group content and alteration of membrane protein components. The results are as follows. 1) Calcium binding of isolated SR was markedly inhibited by X/XO. 2) During the incubation of sarcoplasmic reticulum with xanthine/xanthine oxidase, there were marked inclose in lipid peroxidation and reduction of SH-group content. 3) An antioxidant, p-phenylenediamine effectively prevented the lipid peroxidation but partially prevented the calcium binding inhibition of X/XO treated SR. 4) The reduction of SH-group content of SR treated with X/XO was partially prevented by p-phenylendiamine. 5) When modifying SH-group of SR by treatment with DTNB, the inhibition of calcium binding activity was partially prevented. 6) On gel-permeation chromatography of X/XO-treated sarcoplasmic reticulum, there was an increase of small molecular weight products, probably protein degradation products. 7) Semicarbazide, which prevents the cross-linking reaction of protein components, did not affect the calcium binding inhibition of X/XO-treated SR. From these results, it is suggested that the inhibition of calcium binding of SR by oxygen free radicals results from the consequence of multiple changes of SR components, which are lipid peroxidation, SH-group oxidation and degradation of protein components.

Antioxidant Potential of Enzymatic Extracts from Blueberry (Vaccinium corymbosum L.) (블루베리 (Vaccinium corymbosum L.) 유래 효소 추출물의 항산화성)

  • Enzymatic extracts were prepared from the blueberry (Vaccinium corymbosum L.) collected in Jeju, Korea. Five carbohydrases namely AMG, Celluclast, Termamyl, Ultraflo and Viscozyme, and five proteases namely Alcalase, Flavourzyme, Kojizyme, Neutrase and Protamex were used to prepare the enzymatic extracts. Antioxidant properties of each extracts were studied using stable 1,1-diphenyl 2-picrylhydrazyl (DPPH), reactive oxygen species (ROS), nitric oxide (NO) scavenging, metal chelating assays and lipid peroxidation inhibitory activity in hemoglobin-induced linoleic acid system. The phenolic content of all enzymatic extracts was in the range of 517.85-597.96 mg/100 g dried sample. DPPH and NO${\cdot}$scavenging, and metal chelating assays exhibited prominent activities. Viscozyme showed the highest DPPH activity $(0.046{\pm}0.002\;mg/mL)$ while AMG Showed the highest activity in NO${\cdot}$scavenging $(0.339{\pm}0.011\;mg/mL)$. All the extracts exhibited strong metal chelating activities. Blueberry enzymatic extracts also showed relatively good activity in hydrogen peroxide scavenging. AMG showed the highest lipid peroxidation inhibitory activity $(0.28{\pm}0.01\;mg/mL)$ in hemoglobin-induced linoleic acid system. In this results, the blueberry, which has potential antioxidant components, may be a good candidate as a natural antioxidant source.

Pretreatment of Used Newspaper to Increase Enzymatic Digestibility (효소 당화율을 높이기 위해 폐 신문지의 전처리)

  • A pretreatment method to increase enzymatic digestibility for waste paper such as newspaper was investigated. Ash content, substrate size and printed ink were considered to be factors that affect on enzymatic hydrolysis. The effect on enzymatic digestibility of varying these factor were measured. Printed ink had the highest effect of the three factors, so a method was developed to remove the ink during pretreatment. Fist, a pretreatment process using a percolation reactor was tried. The digestibility of the substrate pretreated at 170$\^{C}$, however, was less than that of the untreated substrate because only small portion of ink was removed. Therefore, a batch type process at less than 100$\^{C}$ was devised. Of several schemes, a method using amonia-hydrogen peroxide mixture on a shaking bath proved most effective. The digestibility obtained from this method was about 85%--approximately 20% greater than the untreated substrate. This proves the pretreatment method was very effective in treating waste paper. The high digestibility obtained from this pretreatment is probably due to the effects of the hydrogen peroxide that can enhance ink removal and substrate swelling.

Development of a cholesterol biosensor modified with carbon nanotube (탄소나노튜브를 이용하여 개조한 콜레스테롤 바이오 센서 개발)

  • A cholesterol biosensor was developed using a modified carbon electrode with carbon nanotubes. The disposable cholesterol biosensor was modified with carbon nanotubes to enhance electron transfer during the enzymatic reaction of cholesterol. Cholesterol oxidase and peroxidase, with potassium ferrocyanide as a mediator, were immobilized on a screen-printed carbon nanotube electrode. The electrochemical cholesterol biosensor developed using carbon nanotubes showed a rapid and reliable signal for measuring total cholesterol. The cholesterol sensor showed a linear response in 5 seconds with a small volume (0.5 μL) in the range of 100~400 mg/dL, with a coefficient of variation of 4.0%.

Effect of Peroxidized Lipid on the Protein Isolate and Protease Activity of Rice Bran (미강의 산화 지질이 단백질과 효소의 활성에 미치는 영향에 관한 연구)

  • The destructive effect of peroxidized lipid on the amino acid in protein isolate and the proteolytic activity of protease were studied in the model system of rice bran. The content of amino acids in the protein isolate decreased significantly when they reacted with peroxidized lipid (pov. 1200 meq/kg). Most of amino acids were lost by more than 90% in salt soluble protein isolates when analyzed by the method of enzyme hydrolysis. Formaldehyde reduced the activity most severely among peroxidized products. Formic acid, peroxidized lipid and hydroperoxide were also found to reduce the protease activity. The damaging effect of the secondary products on the protease activity was more serious than that of the primary products of lipid peroxidation. The destruction of amino acids in the total protein and Inhibition of protease activity by the peroxidized lipid were apparent.

  • To compare antioxidant action of u 3 polyunsaturated fatty acid (PUFA) on lipid peroxidation in rats, the formation of malondialdehyde (MDA) and membranes of liver and brain and activities of antioxidant-related enzymes such as catalase, glutathione peroxidase, and superoxide dismutase (SOD) in blood, were studied. Malondialdehyde contents of $\omega3$ PUFA and sardine oil groups were significantly decreased compared with lard group as control (p<0.05). Catalase and superoxide dismutase showed higher activities in $\omega3$ PUFA and sardine oil groups than those of lard control group. These findings suggest that fish oil has a inhibitory effect on formation of lipid peroxides in blood and membranes of rats.
