Effects of Houttuynia cordata Thunb on Lipidperoxide and Cholesterol in 2,3,7,8-TCDD-damaged Rats (2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD)로 처치된 흰쥐에서 어성초가 과산화지질 및 콜레스테롤에 미치는 영향)

  • TCDD, one of the notorious toxic environmental pollutants, damages various organs including liver and is regarded as an endocrine disrupter. To investigate the effects of Houttuynia cordata Thunb (HCT) on the biochemical parameters of function, liver and serum of TCDD-treated rats were used. Seven days after the injection of TCDD (1 $\mu\textrm{g}$/kg), HCT (200 mg/kg) was administered to rats on every other day for four weeks. The lipidperoxide activity was examined by measuring the level of total cholesterol, HDL-cholesterol, LDL-cholesterol, total lipid and triglyceride (TG) in serum, and malondialdehyde (MDA) in liver tissue of rats. Result showed that lipidperoxidation was inhibited In the significant level when 2,3,7,8-TCDD-damaged rats were treated with HCT.

  • Ibotonic acid와 FeCl$_3$ 투여군은 phosphate buffered saline을 투여한 대조군에 비해 흰쥐의 능동적 회피학습 습득율을 감소시켰고 해마영역의 과산화지질 형성도를 유의하게 증가시켰으며 acetylcholinesterase의 활성을 감소시켰으나 acetylcholine의 농도에는 별영향을 주지 못하였다. 이들 약물은 신경세포 지질막등에 작용하여 노인성 치매에서와 유사한 cholinergic신경계통에 neuronal degeneration을 유발하여 학습능력 감소를 유발한 것으로 보인다. 4,결론 중추 Cholinergic성 신경계활성의 변화와 뇌내 과산화지질의 과다 형성이 학습능력 감소 및 노인성치매의 원인이라고 말할수는 없지만 이러한 뇌내의 변화률이 치매과정과 연관성을 갖는 것으로 보여진다.

Effect of Korean Red Ginseng Powder on the Lipid Concentrations and Tissue Lipid Peroxidation in the Rats Fed High Fat Diet (고지방급여 흰쥐의 혈청과 간의 지질 농도 및 조직 과산화지질 농도에 미치는 홍삼분말의 영향)

  • Effect of Korean red ginseng (KRG) on the level of serum and liver lipids and lipid peroxidation was investigated in the rats fed high fat diet. Content of serum total cholesterol was significantly decreased (P<0.05) in KRG I group and KRG II group. Content of HDL-cholesterol was significantly increased by 69.75% and 39.15% in KRG I and KRG II group compared to control group, respectively. Atherogenic index (hi) was also significantly decreased by 74.76% and 37.38% in KRG I and KRG II groups compared to control group, respectively. Serum triglyceride content was significantly decreased (p<0.05) in only KRG II group. Antioxidative activity of KRG on the lipid peroxidation of serum and tissues in rats was also studied in vivo by measuring the formation of thiobarbituric acid reactive substances (TBARS). Contents of TBARS in the serum of both KRG groups were significantly decreased (p<0.05) and that of nonheme iron in serum was significantly increased (p<0.05) in a dose-dependent manner, which suggested that lipid peroxidation contents are inversely correlated with serum nonheme iron content. Content of TBARS in liver was significantly decreased (p<0.05) in KRG I and KRG II groups, without any influence in other tissues. Content of TBIARS in liver microsomal fractions stimulated by Fe$^{2+}$/ascorbate was significantly decreased (p<0.05) in KRG I and KRG II groups, whereas this observation did not occur in liver mitochondrial fractions. When the effect of KRG on TBARS content in the liver fractions of homogenates, microsomes, and mitochodria stimulated by Fe$^{2+}$/ascorbate was tested in vitro experimental model, TBARS of liver three fractions was significantly decreased at 6 mg/mL KRG compared with those of control. These results suggested that KRG powder have hypocholesterolemic effect as well as antioxidative effect in the serum and liver of the rats fed high fat diet.

Inhibitory Actions of Quercetin and Rutin on $Fe^{2+}-induced$ Lipid Peroxidation ($Fe^{2+}$에 의한 지질 과산화에 있어서 Quercetin과 Rutin의 억제 작용)

  • Inhibitory effects of quercetin and rutin on lipid peroxidation of microsomes caused by iron(II) were investigated with respect to the scavenging action for oxygen radicals produced during oxidation of iron and the chelating action for iron. Lipid peroxidation by $Fe^{2+}$ alone was markedly inhibited by quercetin or rutin in a dose dependent fashion. Lipid peroxidation by ascorbate or NADPH in the presence of $Fe^{2+}$ was almost completely inhibited by both quercetin and rutin. The peroxidative action of $Fe^{2+}$ was inhibited by SOD and DABCO and slightly inhibited by catalase, DMSO and mannitol. Quercetin and rutin inhibited oxidation of $Fe^{2+}$ which is responsible for DETAPAC and they showed a significant initial chelating effect. Quercetin and rutin effectively inhibited lipid peroxidation by $H_{2}O_{2}$ and decomposed $H_{2}O_{2}$. Both $OH{\cdot}$ production in the presence of $Fe^{2+}$ and $^1O_2$ production by U.V. irradiation were inhibited by quercetin and rutin. Lipid peroxidations by $Cd^{2+},\;Cu^{2+},\;Ni^{2+},\;Pb^{2+}$ and $Zn^{2+}$ were almost completely inhibited by quercetin. Quercetin and rutin significantly prevented the loss of sulfhydryl groups by $Fe^{2+}$. These results suggest that inhibitory effects of quercetin and rutin on the peroxidative action of $Fe^{2+}$ in the presence or absence of ascorbate and NADPH may be attributable to their scavenging action on reactive oxygen species and chelating action on iron.

The Effects of Selenium on Cadmium-Induced Toxicity and Lipid Peroxidation in Rat Hepatocyte Primary Culture (랫드 간세포 일차배양에서 셀레늄이 카드뮴에 의해 유도된 독성 및 지질과산화에 미치는 영향)

  • The objective of present study was to investigate the antioxidative and hepatoprotective effects of selenium on cadmium-induced toxicity and lipid peroxidation in rat hepatocyte primary culture. To do this, two separate experiments were conducted. In Experiment 1, primary cultures of rat hepatocytes were incubated for 6 hr in the presence of various concentrations (1, 10, 50, 100, and $500\;{\mu}M$) of cadmium chloride. Cytotoxicity and lipid peroxidation were evaluated using the MTT assay and TBARS assay, respectively. Antioxidative and hepatoprotective effects were determined by measuring the activity of GOT and GSH-Px, respectively. Cell viability was reduced and lipid peroxidation was increased by cadmium in dose-dependent manners. There was significantly negative correlation (r=-0.943, p<0.01) between cell viability and lipid peroxidation GOT activity was increased and GSH-Px activity was decreased by cadmium at the concentration of $50\;{\mu}M$. In Experiment 2, primary cultures of rat hepatocytes were incubated for 6hr in the presence of 100\;{\mu}M$ of cadmium chloride and various concentrations (0.01, 0.1 and 1 ppm) of sodium selenite to assess the effect of selenium on cadmium-induced toxicity and lipid peroxidation. Cell viability and GSH-Px activity were increased by sodium selenite at the concentration of 1 ppm Whereas, lipid peroxidation and GOT activity were reduced by 0.1 ppm of sodium selenite. These results demonstrate that selenium has an antioxidative and hepatoprotective potentials against cadmium.

Studies on Antioxidative Substances of Ganoderma lucidum (영지의 항산화성 물질에 관한 연구)

  • To study antioxidative activities of Ganoderma lucidum, its extracts were fractionated by various organic solvents with different polarity the extracts were purified by thin chromatography, silicagel column chromatography and preparative liquid chromatography. In antioxidative activity tests using thiocyanate method, TBA method and weighing method, fraction 5 from the hexane extract and fraction II from the methanol extract showed antioxidative activity. When the antioxidative activities were expressed as TBA value using a homogeneous liver extracte of rats, the relative antioxidative activities of fraction 5 and fraction II were increased by 13.0% and 54.6%, respectively.

Effect of Glutathione-Enriched Saccharomyces cerevisiae FF-8 on Tissues Lipid Peroxidation in Orotic Acid-Induced Fatty Liver Model Rats (지방간의 과산화지질에 미치는 글루타티온 고함유 효모 Saccharomyces cerevisiae FF-8 균주 급여의 영향)

  • Glutathione is a well-known chemotherapeutic agent and a popular nutritional supplement for liver disease and oxidative stress. Our previous studies reported the suppressive effects of the glutathione-enriched Saccharomyces cerevisiae FF-8 (FF-8) strain on carbon tetrachloride- and alcohol-induced oxidative stress in rats. The purpose of the current study was to investigate the effect of the FF-8 strain on lipid peroxidation in tissues of rats with orotic acid (OA)-induced fatty liver. OA treatment showed a significant decrease in body weight gain compared to the normal diet, and simultaneous addition of FF-8 and OA had the same effect. OA treatment produced an increase in liver weight, however, this also increased with simultaneous addition of FF-8 and OA. Liver lipid peroxidation was significantly increased by OA, but was significantly decreased by FF-8 strain treatment. This same tendency was found in the kidney and heart. Concentration levels of hepatic glutathione and zinc are known to be closely associated with the antioxidant system, and OA treatment led to reductions in liver glutathione and zinc concentrations, whereas these were significantly increased by FF-8 strain treatment in OA feeding rats. These results suggest that the glutathione-enriched S. cerevisiae FF-8 strain may positively mediate orotic acid-induced oxidative stress by enhancing glutathione and zinc levels in rat livers.

Effects of Long-Term Ozone Exposure and Dietary Factors on Lipid Peroxidation in Lung & Liver Tissues of Mice (장기간의 오존조사와 식이요인이 생쥐의 폐와 간조직의 치질과산화에 미치는 영향)

  • The chronic effects of long-term ozone exposure and dietary factors on the lipid peroxidation were investigated in mouse lung and liver tissues. Eighteen groups of mice were exposed to ozone(0.25 or 0.50 ppm) or ambient air over an 18-month period. Within each esposure regimen. animals were fed diets containing different levels of antioxidants and unsaturated fat. Ozone exposure did not have an effect on the production of thiobarbituric acid-reactive substances in lung and liver or free malondialdehyde in the liver at all levels of dietary vitamin E. An inverse relationship between the level of vitamin I supplementation and the concentration of lipid peroxidation products was observed. Results indicate the possible adaptation of animals to long-term continuous ozone exposure by unknown mechanism and the effectiveness of dietary vitamin I at sufficient level(30ppm) to protect against tissue lipid peroxidation regardless of the degree of unsaturation of the dietary fat.

Effect of Antioxidation and Inhibition of Matrix Metalloproteinase-1 from Ligularia fischeri (곰취의 항산화와 UVA에 의한 MMP-1 발현 저해효과)

  • In this study, we examined the effects of antioxidant, lipid peroxidation inhibition and suppression of UVA-induced MMP-1 expression in human dermal fibroblasts. Ligularia fischeri extract showed free radical scavenging effect by 82.3% at 5 mg/mL and superoxide radical scavenging effect by 79.3% at 5 mg/mL in the kanthine/xanthine oxidase system, respectively. At the concentration of $500{\mu}g/mL$, L. fischeri extract showed 97% inhibition on lipid peroxidation of linoleic acid. UVA induced MMP expression in human dermal fibroblasts was reduced 35% by treatment with $100{\mu}g/mL$ of L. fischeri extract. These results suggest that L. fischeri extract can be used for an anti-aging agent by antioxidation, lipid peroxidation inhibition and reducing UVA-induced MMP-1 production.
