Antioxidative, antidiabetes, antibacterial, anticancer and angiotensin-converting enzyme (ACE) inhibitory activities of methanol extracts of dried persimmon, fresh persimmon and persimmon leaves were investigated. Total polyphenol content of dried persimmon, fresh persimmon and persimmon leaves were 147.79, 301.45 and $315.90\;{\mu}g/mg$, respectively, of which fresh persimmon and persimmon leaves had significantly higher total polyphenol than dried persimmon. Activities of DPPH radical scavenging, lipid peroxidation inhibition and salivary $\alpha$-amylase inhibition were increased in persimmon leaves related to total polyphenol contents. Anticancer activities against AGS of fresh persimmon and persimmon leaves were $65{\sim}70%$; however, there were no significant differences between dried persimmon and fresh persimmon on free radical scavenging activity and inhibitory activity of salivary $\alpha$-amylase. Also, extracts of dried persimmon, fresh persimmon and persimmon leaves showed good ACE inhibitory activities. Dried persimmon and fresh persimmon showed antibacterial activities on E.coli O157:H7. Therefore, there are many difference activities by dried and parts of persimmon. From this result, it is suggested that persimmon leaves is believed to have possible antioxidative, antidiabetes and anticancer capacities by polyphenol, but further studies on the identification of the active compound(s) as antioxidant, antidiabetic, antihypertensive and antibacterial materials will be needed to develop a better understanding of its potency on persimmons.
This study was worked out to investigate the compounds of antioxidant constituents extracted from Stachys Sieboldii MiQ. and their effects on antioxidant activity by DPPH method, ferric thiocyanate method, and nitrite scavenging ability. Solvents such as methanol, hexane, chloroform, ethyl acetate, butanol, and water were used for this purpose. Total concentrations of polyphenols and flavonoids were measured in the methanol fraction. The ethyl acetate fraction showed the strongest activity by DPPH method, ferric thiocyanate method, and nitrite scavenging ability. The ethyl acetate extract was fractionated on a silica gel column using elution solvent (chloroform: methanol: water = 70 : 30 : 5 lower phase) at a flow rate of 1.0 mL/min. UV-VIS spectral data of each fraction showed adsorption maxima in the range of 284~330 nm. Among fractions, the fraction 1 that has λ$\_$max/ (nm) of 284 nm showed the strongest activity by DPPH method. The UV-VIS spectral data of phenolic compounds were known to lie in the range of 210~290 nm and 300~550 nm. Therefore, the results of our study suggested that Stachys sieboldii MIQ. contains phenolic compounds showing natural antioxidant activity.
For the screening of anti-inflammation and antioxidative activities, ethanolic extract of 40 species of traditional herbal medicines were examined their hyaluronidase inhibitory effect and radical scavenging activity in vitro. From the result of the hyaluronidase inhibitory activity using a Morgan-Elson assay, Astragali Radix, Eucommia Cortex, Schizandrae Fructus, Scutellaria Radix, Acanthopanacis Cortex, Chaenomelis Fructus, Amomum xanthioides Wallich and Moutan Radicis Cortex showed more than 50% hyaluronidase inhibitory effects at the concentration of 10 mg/mL. In the various solvent fractions (n-hexane, chloroform, ethyl acetate, water) prepared from ethanolic extracts, the ethyl acetate fraction of all extracts tested showed strong activity. Antioxidative activity was evaluated by assaying electron-donating ability to DPPH free radical and scavenging of hydroxyl radical (ㆍOH) generated through Fenton reaction, respectively. Rubus coreanus Miq, Moutan Radicis Cortex, Paeoniae Radix Alba, Plantaginis Semen and Sorbus commixta Hedl. showed high activity more than 90%, yet similar activity to $\alpha$-tocopherol and BHA at the concentration of 1 mg/mL in electron donating activity. The scavenging effects of ethanolic extracts on hydroxyl radical were investigated using a 2-deoxyribose oxidation method and tested all extracts showed significant radical scavenging activity. The experiment was also performed to examine whether herbal medicines having significant lipid peroxidation inhibitory activity, Schizandrae-Fructus is the strongest inhibitory activity in both linoleic acid and liposome peroxidation.

Oxidative stress can play a key role in cadmium (Cd)-induced dysfunction. The present study examined the effect of pine needle water extract (PN) on Cd-induced oxidative stress in rats. Sprague-Dawley male rats were divided into three groups: normal group, Cd control group (Cd) and PN-administered Cd group (Cd-PN). $CdCl_2$ in 0.9% NaCl was administered orally with a dose of 5mg/kg of body weight/week, while the PN was administered orally with a dose of 1.26g/kg of body weight/day. Body weight gain was not different between groups, whereas food intakes were significantly lower in the Cd-PN group than in normal or Cd group. Relative liver weight was significantly increased by cadmium administration compared to the normal group. Hepatic cytochrome P450 was significantly lower in Cd and Cd-PN groups than in normal group, while xanthine oxidase and alcohol dehydrogenase activities were significantly higher in the Cd-PN group than in normal or Cd group. Increased hepatic superoxide dismutase, monoamine oxidase, catalase, and glutathione peroxidase activities by cadmium administration were significantly decreased by PN supplement. PN did not affect the hepatic glutathione content in cadmium-administered rats; however, PN significantly lowered the hepatic lipid peroxide level and plasma alanine transferase activity compared to the Cd control group. These results suggest that the PN may alleviate Cd-induced oxidative stress without hepatotoxicity.

The objective of this study was to investigate the effect of hot water extract from Cornus walteri Wanger (CWE) on tert-butyl hydroperoxide (TBHP)-induced oxidative stress in HepG2 cells. Generation of reactive oxygen species (ROS), concentrations of cellular lipid peroxidation products and reduced glutathione, and antioxidant enzyme activity were used as biomakers of cellular oxidative status. Cells pretreated with CWE (25~200 ${\mu}g/mL$) showed an increased resistance to oxidative stress in a dose-dependent manner, as revealed by a higher percentage of surviving cells compared to control cells. ROS generation induced by TBHP was significantly reduced when cells were pretreated with 200 ${\mu}g/mL$ CWE for 4 h. Pretreatment with CWE (5~50 ${\mu}g/mL$) prevented the decrease in reduced glutathione and the increase in malondialdehyde and ROS evoked by TBHP in HepG2 cells. Finally, CWE pretreatments prevented the significant increase of glutathione peroxidase, catalase, glutathione reductase, and superoxide dismutase activities induced by TBHP. These results show that CWE has significant protective ability against a TBHP-induced oxidative insult and that the modulation of antioxidant enzymes by CWE may have an important antioxidant effect on TBHP-induced oxidative stress in HepG2 cells.
The purpose of the present investigation was to evaluate the effects of swimming training on response of lipid peroxide (MDA) and superoxide dismutase (SOD) enzyme activity of hyperlipidemic rats. Twenty-five male SD rats (6 weeks old) were randomly divided into a control group and 4 swimming groups after hyperlipidemia induction for 4 weeks through a 1% cholesterol diet. Swimming groups were then divided into unloaded swimming group, low-loaded swimming group, moderate-loaded swimming group and high-loaded swimming group by swimming intensity, and made to swim for 6 weeks (6 days/week). The loaded swimming group rats among the swimming groups swam a lead weight equivalent to 0%, 3%, 5% and 7% of body weight attached to the base of the tail. All data were expressed as mean and standard deviation by using an SPSS/$PC^+$ program, and to evaluate the differences between groups, data were analyzed by one-way analysis of variance and Duncan multiple range test (${\alpha}$=0.05) was performed to test the significant levels of differences between groups. The conclusions obtained from this study were as follows: 1) all swimming groups had significantly lower levels of MDA than the control group (p<0.001). Among the swimming groups, the moderate-loaded group had a significantly lower level than the unloaded group, low-loaded group and high-loaded group (p<0.001). 2) all swimming groups had significantly higher levels of SOD than the control group (p<0.01). Among swimming groups, the unloaded group, moderate-loaded group and high-loaded group had significantly higher levels than the low-loaded group (p<0.01).
We determined oxidative stress caused by thermal stress in olive flounder Paralichthys olivaceus based on the altered-mRNA expression and enzymatic activity of two key antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT), along with monitoring of several other biomarkers. When the fish were exposed to acute thermal change (from $20^{\circ}C$ to $25^{\circ}C$ and $30^{\circ}C$), the expression and activity of both enzymes were significantly higher at elevated temperatures ($25^{\circ}C$ and $30^{\circ}C$) than at $20^{\circ}C$. Lipid peroxidation (LPO) was also higher at $25^{\circ}C$ and $30^{\circ}C$ than at $20^{\circ}C$. In addition, the plasma $H_2O_2$ concentration was significantly increased by thermal stress. Furthermore, we investigated changes due to thermal stress by measuring levels of plasma alanine aminotransferase (AlaAT) and aspartate aminotrasferase (AspAT). Both were significantly increased by thermal stress. As an immune indicator, the lysozyme concentration was lower at $30^{\circ}C$ than at $20^{\circ}C$, indicating that thermal stress decreases immune function. Therefore, thermal stress could induce oxidative stress and suppress immune function and can cause physiological stress.
This study examined the antioxidant activity and neuroprotective effects of ethanol extracts obtained from Diospyros kaki core (DCE). The total polyphenol and flavonoid contents in DCE was 786.47±15.27 and 31.14±0.82 mg/g, respectively. In addition, DCE exhibited a dose-dependent induction of radical scavenging activity, determined by 1,1-diphenyl-picrylhydrazyl (DPPH), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonicacid) (ABTS), ferric reducing antioxidant power (FRAP), and reducing power assays. The viability of HT22 hippocampal cells was examined to investigate the neuroprotective effect of DCE. DCE treatment did not induce cytotoxicity at concentrations below 1,000 ㎍/mL. Additionally, DCE treatment in the background of H2O2 induce oxidative stress revealed a significant increase in the survival rat, indicated by increased SOD activity and decreased levels of MDA, a lipid peroxidation product. Therefore, the results suggest that DCE can be used as a source of natural antioxidants source and a therapeutic agent for the treatment of brain disorders induced by oxidative stress and neuronal damage.

The purpose of this study was to investigate the mechanism and effects of different types of ginseng on memory improvement in an experimental rat model. In this study, SD rats were induced for memory deficits through scopolamine treatment (1 mg/kg, i.p.) then administrated with ginseng extract for 7 weeks. The rats were divided into five groups: saline (1 mL/kg, NC: negative control), white ginseng (300 mg/kg, WG), red ginseng (300 mg/kg, RG), black ginseng (300 mg/kg, BG), and scopolamine (1 mg/kg, PC: positive control). The step through latency of the BG and RG groups was significantly longer than the PC group in the retention trial of multiple trial passive avoidance test. In the spatial reference memory triads of the Morris water maze test, the latency time of BG and RG was significantly lower than the PC group. In addition, in the prove test, the time spent in the platform quadrant of BG and RG groups were significantly longer than the PC group. Brain choline acetyltransferase (ChAT) activities BG and RG groups significantly increased compared to other groups. On the other hand, the levels of malondialdehyde (MDA) were significantly lower in the BG and RG groups compared to other groups. These result suggested that black ginseng could be useful to enhance learning memory and cognitive function by regulation of cholinergic enzymes.

The present study was undertaken to investigate the inhibition effects of sancho(Zanthoxylum schinifolium) on lipid peroxidation and liver damage in carbon tetrachloride tetracloride-treated mice. Mice aged 3 weeks old were fed diets containing either dry sancho powder, methanol extract or hyperoside isolated from sancho at the various levels for 2 weeks(sancho, methanol extract from sancho) or 3 days (hyperoside) before $CCl_{4}$ treatments. Seed coats of sancho added to diet with the levels from 1.25 to 5.0% significantly reduced(p<0.001) thiobarbituric acid reactive substances(TBARS) formation in liver and serum of mice treated with carbon tetrachloride. $CCl_{4}$ treatments significantly increased (p<0.001) in liver TBARS comparing with those of the untreated control, while methanol extract from the seed coat of sancho added to diet with the levels from 0.5 to 1.0% significantly reduced (p<0.01) to 20mg per kg of body weight showed significant reduction(p<0.01, p<0.001) of liver TBARS in mice treated with $CCl_{4}$. $CCl_{4}$ treatments significantly increased(p<0.05) in serum TBARS comparing with those the levels from 0.5 to 1.0% significantly reduced(p<0.01) serum TBARS in mice treated with $CCl_{4}$, and administration of hyperoside with the levels from 10 to 20mg per kg of body weight showed significant reduction(p<0.01, p<0.001) of serum TBARS in mice treated with $CCl_{4}$. $CCl_{4}$ significantly increased(p<0.001) in serum AST activities comparing with those of the untreated control, while either addition(0.5, 1.0%) of methanol extract from sancho or administration(10, 20mg/kg) of hyperoside significantly reduced(p<0.001, p<0.05) serum AST activities in $CCl_{4}$-treated mice. $CCl_{4}$ treatments showed significant increases(p<0.001) in serum ALT activities comparing with those of the untreated control, while either addition(0.5, 1.0%) of methanol extract from sancho or administration (10, 20mg/kg) of hyperoside showed moderate decreases(p<0.01, p<0.001) serum ALT activities in $CCl_{4}$-treated mice.
