Characterization of Entomopathogenic Fungus from Trialeurodes vaporariorum and Evaluation as Insecticide (온실가루이 병원성 곰팡이의 특성 및 살충제 개발을 위한 평가)

  • The greenhouse whitefly, Trialeurodes vaporariorum, is an economically important pest for greenhouse crops because they cause direct damage by feeding on plant nutrients and indirect damage as transmits many virus vectors. It has recently become a serious problem because of the continuous use of insecticide resulting in resistance among greenhouse whitefly population. To overcome these problems, in this study, the biological characteristics and virulence of an entomopathogenic fungus isolated from the cadaver of nymph greenhouse whitefly were investigated. Isolated fungus was identified as Isaria fumosorosea by morphological examinations and genetic identification using sequences of the ITS, ${\beta}$-tubulin, and EF1-${\alpha}$ regions. This fungus was named as I. fumosorosea SDTv and tested for the virulence against nymphs T. vaporariorum and the cold activity, the thermotolerance and the stability of UV-B irradiation on conidia. Mortality rate of greenhouse whitefly showed from 84 to 100% and the virulence increased with increasing conidial concentrations, $1{\times}10^5$ to $10^8$ conidia/ml. Conidia were stable at $35^{\circ}C$, 0.1 $J/cm^2$ of UV irradiation and germinated after 8 days at $4^{\circ}C$. Additionally, the activities of chitinases and proteases produced by I. fumosorosea SDTv were varied according to the medium. In conclusion, I. fumosorosea SDTv which showed high mortality rate against greenhouse whitefly will be used effectively in the integrated pest management programs against the greenhouse whitefly.

  • Several entomopathogenic fungi have been exploited to be developed into biological control agents in insect pest management. The beet armyworm, Spodoptera exigua, is a serious insect pest infesting various crops, but not effectively controlled by commercial chemical pesticides due to its high insecticide resistance. A fungal isolate was isolated from S. exigua larvae collected from cabbage field in Andong, Korea. The fungus could be cultured in potato dextrose agar. Larvae of S. exigua injected with the cultured conidia showed a potent entomopathogenicity. To identify the fungus isolate, its internal transcribed space (ITS) and surrounding partial 18S/28S regions were sequenced. The ITS sequence was highly matched (99%) to that of Nomuraea rileyi. Morphological characters of its hyphae and conidia were well fit to those of known N. rileyi. This study reports the first record of an entomopathogenic fungus, N. rileyi, in Korea.

Identification of Antiviral-related Genes Up-regulated in Response to Bombyx mori Nucleopolyhedrovirus (누에로부터 핵다각체병 바이러스 방어관련 유전자 정보 분석)

  • Silkworm larvae often suffer from viral infections causing heavy losses to the economy of silk industry. Insects exhibit both humoral and cellular immune responses that are effective against various pathohens like bacteria, fungi, protozoa, etc., but no insect immune responses is effective against viral infection. To obtain genes related to insect antiviral immunity from Bombyx mori, the cDNA library was constructed from B. mori nucleopolyhedrovirus (BmNPV)-infected B. mori. From the cDNA library, we selected 411 differentially expressed clones, and the 5' ends of the inserts were sequenced to generate ESTs. In this work, 135 unigenes were generated after the assembly of 411 differentially expressed clones ESTs. Of these 135 unigenes, we selected 109 antiviral response-related candidates except 26 clones that high similarity with genes derived from BmNPV. Among 109 unigenes, a total of 80% had significant matches to genes from other organisms in the database, wheres 20% of the unigenes had not matched in the database. Functional groups of these sequences with matches in database were constructed according to their putative biological function. Three largest categories were control of cellular oraganization (52%), metabolism (20%), and protein fate (10%). The genetic information reported in this study will provide more information about antiviral-related genes in silkworms.

Characterization of Lecanicillium lecanii Btab0l Isolated with Bioactivities to Tabacco Whitefly (Bemisia tabaci) (담배가루이(Bemisia tabaci) 병원성곰팡이 Lecanicijjium lecanii Btab0l 균주의 특성)

  • Cultural characteristics Lecanicillium lecani Btab01 and its insecticidal activity against tobacco whitefly (Bemisia tabaci) were investigated. On potato dextrose agar, tryptic soy agar and SDA+Y media, mycelial growth of L. lecani Btab01 was best at $20{\sim}25^{\circ}C$ and suppressed above $28^{\circ}C$. Both solid culture and liquid culture of L. lecani Btab01 showed high insecticidal activity, 93.9 and 98.3% respectively, against nymph of tobacco whitefly, but there is no significant difference. When culture of L. lecani Btab01 was treated at the concentration of $10^5$, $10^6$, $10^7$ and $10^8$ cfu/ml, their insecticidal activity were 5.8%, 33.8%, 77.3% and 98.5% respectively, and $LT_{50}$ values were 16.1 days, 7.3 days, 5.1 days and 3.5 days respectively. When nymphs were treated by the cultures of L. lecani Btab01 and maintained under saturated condition for zero hour, 24 hours and 168 hours, their control activities were 0%, 20.3% and 100% respectively. Spore germination of L. lecani Btab01 was increased about two times by adding edible oil. When L. lecani Btab01 was treated to control nymph with 0.1% edible oil, it showed high control activity(98.6%) compared to single treatment of L. lecani Btab01 (79.9%).

  • Fifty three diseased root samples of Chinese cabbage and 22 soil samples were taken from six alpine areas in Kangwon province to examine the organisms causing root rot of Chinese cabbage. Among thirteen microorganisms detected from diseased roots samples, Pythium ultimum, Pythium echinocarpum and Rhizoctonia solani were detected frequently and were pathogenic to Chinese cabbage. Five to 100% of young Chinese cabbage plants grown in the sample soils taken from alpine cabbage fields were infected either alone or together with P. ultimum, P. echinocarpum and R. solani, depending upon the or gin of sample soils. When relative density of the three organsims in the sample soils was estimated based on the infection frequency on the plants grown in those sample soils, P. ultimum was prevalent only in Chahang-i-li area. P. echinocarpum was prevalent in Yongsan-il-li, Hoengwe-li, and Munmaek areas. R. solani was solely found in Maebongsan area where both P. ultimum and P. echinocarpum were not detected. In the three areas where both P. ultimum and P. echinocarpum were present, density of P. echinocarpum was generally greater than that of P. ultimum. Seedlings of thirty major Chinese cabbage varieties were inoculated independently with the three organisms in the greenhouse. Six varieties were resistant to P. ultimum. Three and two varieties were moderately resistant to P. ultimum and P. echinocarpum, respectively. All varieties were highly susceptible to R. solani.

  • Biological control of lepidopterous insect pests, Agrotis segetum, Artogeia rapae, Mamestra brassicae, Plutella xylostella, Spodoptera exigua, and S. litura with entomopathogenic fungi, Beauveria bassiana and Metarhizium anisopliae isolated from Gyeongbuk province were investigated. Mean lethal concentration ($LC_50$) values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of A. segetum larvae were $1.2\times10^7\;conidia/m\ell$ and $5.2\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of A, rapae larvae were $1.2\times10^7\;conidia/m\ell$ and $5.2\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of M. brassicae and P. xylostella, larvae were $1.5\times10^6\;conidia/m\ell$, $9.7\times10^5\;conidia/m\ell$, $3.0\times10^6\;conidia/m\ell$ and $1.4\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of S. exigua, and S. litura larvae were $6.3\times10^6 \;conidia/m\ell$, $2.6\times10^6\;conidia/m\ell$, $1.6\times10^7\;conidia/m\ell$ and $3.4\times10^6\;conidia/m\ell$ respectively.

Insect Pests and Natural Enemies of Hibiscus syriacus in Korea (무궁화 해충의 천적과 종류)

  • Insect pests and their natural enemies of Hibiscus Linne (Malvaceae) were investigated from March 2002 to November 2004. Fourteen insect pest species of 9 families in 5 orders were collected from Hibiscus syriacus: 5 species in Homoptera, 3 species in Lepidoptera, 2 species in Coleoptera, 1 species in Orthoprera, 1species in Hemiptera, 1 spedies in Acarina, and 1 species in Stylommatophora. Especially, Aphis gossypii Glover (Aphididae), Anomis megogona Walker(Noctuidae) and Tetranychus urticae Koch (Tetranychidae) were very important species because of their increasing daminge. The highest donsities were observed from May to June in August in Tetranychus urticae. As the enemies and ento-mopathogens of insect pests on Hibiscus syriacus, 1 species of bacteria, 3 species of fungi, 1 species of fungi, 1 species of Hemiptera, 1 species of Coleoptera, 2 species of Hymenopetera, 2 species of Diptera, and 1 species of Acarina were investigated. As the predators and parasitoids of Aphis gossypii, Aphidoletes aphidoletes aphidimyza Rondani (Cecidomyiidae), Meliscaeva cinctella Zetterstedt (Syrphidae), Harmonia axyridis Pallas (Coccinellidae), and Aphidius gifuensis Ashmead (Braconidae), entomopathogenic fungi, Vericillium lecani naturalis strain (Moniliaceae) and Beauveria bassiana naturalis strain strain (Hypocreaceae) were observed and Bacillus thuringiensis naturalis strain (Bacillaceae), B. bassiana, Metarhizium anisopliae naturalis strain (Hypocreaceae), predators of Tetranychus urticae, Amblyseius sp. (Phytoseiidae), and Orius sp. (Anthocoridae) were observed.

  • We have established a transformation system for entomopathogenic fungus, Metarhizium anisopliae, in order to develop mycoinsecticide by recombinant DNA techniques. Protoplasts of M. anisopliae would be transformed to a benomyl-resistant by introducing pBRG-4 plasmid DNA, which contains a ${\beta}-tubulin$ gene of Aspergillus flavus conferring resistance to benomyl and a pyr4 gene of Neurospora crassa, in the presence of 5% polyethylene glycol and 10 mM calcium chloride. Transformants occuring at a frequency of 10 colonies per $50\;{\mu}g$ pBRG-4 DNA grew on the $5\;{\mu}g/ml$ concentrations of benamyl, while the wild type was inhibited by $2.5\;{\mu}g/ml$. From the Southern analysis using genomic DNAs isolated from M. anisopliae transformants, the positive signals suggested that the ${\beta}-tubulin$ gene had integrated in the M. anisopliae genome by homologous recombination.

  • During the autumn-rearing season of 1971, at the silkworm rearing house at the college of agriculture, Seoul national university in Korea, the authors carried out a series of experiments (1) on the susceptibility of four important silkworm pathogens (Aspergillus flavus, Bacillus thuringiensis, Aspergillus oryzae, and Isaria farinosa) to Halamid (Tosylchloramide Sodium), (2) on the acute toxicity of Halamid to silkworm larvae at every instar, (3) on the inhibiting effect of Halamid to the attack of two silkworm infections, fungal muscardine and viral jundice, and the chronic toxicity of Halamid to silkworm larvae, and (4) on the yield and quality of the raw silk harvested from the Halamid treated silkworms. As the results of the experiments the authors found that Halamid could be applied usefully to silk worm rearing industry as an effective disinfectant for both silkworms themselves and their envir onmental articles, rearing houses and tools, also that Halamid exerts its disinfectant effect most efficiently as a disinfectant for silkworms when sprayed in 2% or 3% aqueous solution every second day throughout all tile instars of larval stage.

Influence of Pre-treated Eco-friendly Agricultural Materials on Control Efficacy of Isaria javanica Isolate against Sweet Potato Whitefly (Bemisia tabaci) (친환경 농자재와 곤충병원성 곰팡이 Isaria javanica의 처리 간격이 담배가루이(Bemisia tabaci) 방제에 미치는 영향)

  • For effective control of insect pest which can outbreak in a field during crop cultivation, various control agents such as eco-friendly agricultural materials (EFAM) including microbial control agents and chemical pesticides have been applied at fields and these control agents may be treated simultaneous or sequential in the same field to suppress diverse pests and diseases. The agents may influence each other and control efficacy may also differ from interactions. Therefore we need to test compatibility of microbial control agents with other agricultural agents. In this study, we investigated influence of pre-treated EFAMs, which are registered for whitefly control in greenhouse, on germination, mycelial growth and control efficacy of Isaria javanica isolate against sweet potato whitefly. The results showed that a mixture of paraffin oil+cinnamon oil among 4 EFAMs highly reduced germination ($8.9{\pm}1.3%{\sim}24.5{\pm}0.9%$) and mycelial growth ($0.81{\pm}0.01cm{\sim}0cm$) of I. javanica. To investigate the effects of the treatment interval between EFAMs and I. javanica on sweet potato whitefly control, four different EFAMs were pre-treated 0, 1, 4, and 7 days before applying I. javanica. Pre-treatment of four EFAMs inhibited insecticidal activity of I. javanica against sweet potato whitefly. Therefore when EFAMs and a mycopesticide using I. javanica spray simultaneous or sequential, application of EFAMs need more than 7 days interval after treatment of mycopesticide at field.
