Effect of Initial Solidification Condition During Directional Solidification on the Grain Growth and the Tensile Properties of Superalloy CM247LC (초내열합금 CM247LC의 조직 및 인장특성에 미치는 초기 일방향응고 조건의 영향)

  • Initial solidification condition during directional solidification of superalloy CM247LC was controlled with various cooling methods such as insertion of alumina disc or Ni foil or inoculant, and direct pouring of melt onto chill plate. Rapid cooling with direct pouring of melt onto chill plate resulted in generation of many fine grains and precipitation of fine γ' particles, as well as small dendrite arm spacing. Tensile properties of directionally solidified superalloy CM247LC were closely related to microstructure which was governed by initial solidification conditions. Directionally solidified CM247LC with small dendrite arm spacing and fine precipitates showed good tensile properties.

Importance of Strain Improvement and Control of Fungal cells Morphology for Enhanced Production of Protein-bound Polysaccharides(β-D-glucan) in Suspended Cultures of Phellinus linteus Mycelia (Phellinus linteus의 균사체 액상배양에서 단백다당체(β-D-glucan)의 생산성 향상을 위한 균주 개량과 배양형태 조절의 중요성)

  • Strain improvement and morphology investigation in bioreactor cultures were undertaken in suspended cultures of Phellinus linteus mycelia for mass production of protein-bound polysaccharides(soluble ${\beta}$-D-glucan), a powerful immuno-stimulating agent. Phellineus sp. screened for this research was identified as Phellinus linteues through ITS rDNA sequencing method and blast search, demonstrating 99.7% similarity to other Phellinus linteus strains. Intensive strain improvement program was carried out by obtaining large amounts of protoplasts for the isolation of single cell colonies. Rapid and large screening of high-yielding producers was possible because large numbers of protoplasts ($1{\times}10^5{\sim}10^6\;protoplasts/ml$) formed using the banding filtration method with the cell wall-disrupting enzymes could be regenerated in relatively high regeneration frequency($10^{-2}{\sim}10^{-3}$) in the newly developed regeneration medium. It was demonstrated that the strains showing high performances in the protoplast regeneration and solid growth medium were able to produce 5.8~6.4%(w/w) of ${\beta}$-D-glucan and 13~15 g/L of biomass in stable manners in suspended shake-flask cultures of P. linteus mycelia. In addition, cell mass increase was observed to be the most important in order to enhance ${\beta}$-D-glucan productivity during the course of strain improvement program, since the amount of ${\beta}$-D-glucan extracted from the cell wall of P. linteus mycelia was almost constant on the unit biomass basis. Therefore we fully investigated the fungal cell morphology, generally known as one of the key factors affecting cell growth extent in the bioreactor cultures of mycelial fungal cells. It was found that, in order to obtain as high cell mass as possible in the final production bioreactor cultures, the producing cells should be proliferated in condensed filamentous forms in the growth cultures, and optimum amounts of these filamentous cells should be transferred as active inoculums to the production bioreactor. In this case, ideal morphologies consisting of compacted pellets less than 0.5mm in diameter were successfully induced in the production cultures, resulting in shorter period of lag phase, 1.5 fold higher specific cell growth rate and 3.3 fold increase in the final biomass production as compared to the parallel bioreactor cultures of different morphological forms. It was concluded that not only the high-yielding but also the good morphological characteristics led to the significantly higher biomass production and ${\beta}$-D-glucan productivity in the final production cultures.

Production Performances and Heterosis Effects of Korean Native Chicken Breed Combinations by Diallel Crossing Test (토종 종계 이면교배조합 시험에 따른 생산능력 및 잡종강세효과)

  • This study was conducted to establish new synthetic lines of Korean Native Chicken. We performed $5{\times}5$ diallel crossings with GPS lines of Korean Native Chicken for the selection of parent stock. The production traits including viability, body weight, age at first egg laying, egg weight, hen-day egg production, and hen-housed egg production were measured and analyzed for 25 crosses with 1,157 hens. The heterosis effects of these traits were also estimated. The results showed that the average survival rate during laying periods was 67.7% in the pure lines and 77.1% in the crosses. The 25 cross combinations were shown to be distinctly divided into three groups according to body weight; nine crosses in the high-weight group, 12 crosses in the medium-weight group and four crosses in the light-weight group. The average body weight at 12 weeks of age was $1,873.8{\pm}43.0g$ in the high group, $1,595.4{\pm}56.6g$ in the medium group and $1,152.7{\pm}24.7g$ in the light group, and $1,560.2{\pm}339.3g$ in the pure lines and $1,640.9{\pm}213.7g$ in the crosses. In terms of egg production performance, the age at first egg laying was $139.7{\pm}4.9$ days in the pure lines and $135.8{\pm}5.1$ days in the crosses. The average egg weight was $52.3{\pm}1.6g$ in the pure line and $53.0{\pm}1.6g$ in the crosses. The average hen-housed egg production from 20 to 40 weeks was $58.3{\pm}12.9$ eggs in the pure line and $69.2{\pm}10.4$ eggs in the crosses. The average heterosis effect was 16.9% for survival rate, 5.4% for body weight at 12 weeks, -2.7% for age at first egg laying, 1.3% for egg weight, and 14.3% for hen-day egg production. Generally, the heterosis effect for viability and egg production traits was higher than that for growth trait. On the basis of these results, it is expected that the selection of the HF combination, which had excellent growth performance, is the most desirable as the paternal strain, and the selection of GW and FW combinations, which had excellent laying performance, are preferable as the maternal strain.

가스산업시설에서의 위험성 평가분야에서 지리정보시스템의 적용

  • 우리나라는 그동안 가스산업에서 고속성장을 이룩해 왔으나, 근래에는 중대가스산업사고가 빈번하게 일어나고 있다. 그건 이유로 1996년도에는 중대산업사고 예방제도가 전면적으로 실시되고 공정안전보고서를 작성하여 제출하도록 함으로서 위험성 평가의 중요성이 점차 높아가고 있으며, 위험성 평가 기법들에 대하여 여러 연구 단체에서 연구ㆍ시도되고 있다. 또한 이러한 연구ㆍ보고된 위험성 평가 기법들을 프로그램화하고 적용하려는 시도가 많이 있어 왔다. 본 연구에서는 기존에 연구ㆍ개발된 위험성 평가 기법들에 최근 컴퓨터 산업의 발달에 힘입어 각광을 받고 있는 지리정보시스템을 적용하고자 한다. 이러한 가스산업시설의 위험성 평가 시스템은 위험성을 평가하기 위해서 필요한 여러 가지 정보들을 지리정보시스템이 속성 데이터로서 저장하고 있으나, 가스산업시설에 관련된 주변의 도면들을 공간 데이터로서 저장하고 있다. 그리고 위험성 평가 시스템의 세부적인 기능을 모듈화하였다. 우선 위에 언급한 속성 데이터와 공간 데이터를 관리하는 모듈과 이러한 데이터를 가지고 사고영향 범위를 산출해내기 위한 모듈, 그리고 산출된 사고 영향 범위를 도면에 나타내는 모듈로 나뉘어져 있다. 이렇게 지리정보시스템에 구축되어 있는 도면에 위험성을 평가한 결과치를 나타냄으로서 위험성 평가 숙련자가 아니더라도 위험성 평가를 할 수 있고 결과를 분석하도록 도와 줄 수 있도록 할 수 있다. 또한 향후 재난관리시스템에서는 도면상의 도로에 교통량 가중치와 인근 소방서와 경찰서등의 위치를 관리하도록 지리정보시스템을 적용할 수 있으면, 가스시설물 관리시스템에서는 최근 대형가스사고의 대부분이 타공사에 의한 것임을 고려하여 가스배관망을 포함하여 기타 다른 지하배관망을 관리하도록 지리정보시스템을 적용할 수 있다. DIAION WA30을 라세미화 촉매로 사용하여 아이소옥탄 내에서 라세믹 나프록센 2,2,2-트리플로로에틸 씨오에스터의 효소적 DKR 반응을 수행해 보았다. 그 결과 DIAION WA30을 사용하지 않은 경우에 비해 반응 전환율과 생성물의 광학 순도는 급격히 향상되었다. 전통적 광학분할 반응의 최대 50%라는 전환율의 제한이 본 연구에서 찾은 DIAION WA30을 첨가함으로써 성공적으로 극복되었다. 또한 고체 염기촉매인 DIAION WA30의 사용은 라세미화 촉매의 회수 및 재사용이 가능하게 해준다.해준다.다. Studies on Protoplast Formation and Regeneration of Lyophyllum decastes (Lyophyllum decastes의 원형질체 분리와 재생에 관한 연구)

  • This experiment was carried out to investigate proper conditions for protoplast isolation and regeneration from mycelia of Lyophyllum decastes. Novozym 234(10 mg/ml) with 0.6 M $MgSO_4$ in phosphate buffer(pH 4.0) was proper for protoplast isolation. The optimal reaction time of the mycelium with the lytic enzyme was four hours in shaking condition at 120 strokes per min. When the mycelium of L. decastes was cultured at $24^{\circ}C$ for 5 days, the formation of protoplasts was effective. The liquid medium was more effective for protoplast isolation than the solid medium. In the liquid medium, high yields of protoplasts were obtained from 0.6 M $MgSO_4$ osmotic stabilizer. Protoplasts of L. decastes were regenerated to normal hyphal growth and the regeneration frequency of the protoplasts in the complete agar medium containing Triton X-100(0.0025%) was $5.94{\sim}8.32%$. The regeneration medium stabilized with 0.6 M sucrose was the best for regeneration of the protoplasts. In contrast to protoplast formation, regeneration was inhibited by the inorganic salts used as osmotic stabilizer.

Effect of RuO$_2$ Thin Film Microstructure on Characteristics of Thin Film Micro-supercapacitor ($RuO_2$박막의 미세 구조가 박막형 마이크로 슈퍼캐패시터의 특성에 미치는 영향)

  • All solid-state thin film micro supercapacitor, which consists of $RuO_2$/LiPON/$RuO_2$ multi layer structure, was fabricated on Pt/Ti/Si substrate using a $RuO_2$ electrode. Bottom $RuO_2$ electrode was grown by dc reactive sputtering system with increasing $O_2/[Ar+O_2]$ ratio at room temperature, and a LiPON electrolyte film was subsequently deposited on the bottom $RuO_2$ electrode at pure nitrogen ambient by rf reactive sputtering system. Room temperature charge-discharge measurements based on a symmetric $RuO_2$/LiPON/$RuO_2$ structure clearly demonstrates the cyclibility dependence on the microstructure of the $RuO_2$ electrode. Using both glancing angle x-ray diffraction (GXRD) and transmission electron microscopy (TEM) analysis, it was found that the microstructure of the $RuO_2$ electrode was dependent on the oxygen flow ratio. In addition, x- ray photoelectron spectroscopy(XPS) examination shows that the Ru-O binding energy is affected by increasing oxygen flow ratio. Furthermore, TEM and AES depth profile analysis after cycling demonstrates that the interface layer formed by interfacial reaction between LiPON and $RuO_2$ act as a main factor in the degradation of the cyclibility of the thin film micro-supercapacitor.

  • $La_2NiO_{4+{\delta}}$ based oxides, a mixed electronic-ionic conductors (MIECs) with $K_2NiF_4$ type structure, have been considerably investigated in recent decades as electrode materials for advanced solid oxide fuel cells (SOFCs) due to their high electrical conductivity, and oxidation reduction reaction (ORR). In this study, structure properties of $La(Ca)_2Ni(Cu)O_{4+{\delta}}$ were studied as a potential cathode for intermediate temperature SOFCs (IT-SOFCs).

  • Conductivity of LSGMC materials were affected by secondary phase segregation, composition and synthetic route. $La_{0.8}Sr_{0.2}Ga_{0.8}Mg_{0.1}Co_{0.1}O_{3-{\delta}}$ (LSGMC) powders were prepared using the glycine nitrate process to produce high surface area and compositionally homogeneous powders. The powders were synthesized with different 0.5, 1, 1.5, 2, 2.5 of glycine/cation molar ratios. A single perovskite phase from the synthesized powders was characterized with X-ray diffraction patterns. The obtained sintered pellets showed the dense grain microstructure. In case of 1.5 molar ratio, its density was higher than the others. The electrical conductivity measured at $800^{\circ}C$ was observed to be 0.131 $Scm^{-1}$. In addition, the linear thermal expansion behavior was indicated between $25^{\circ}C$ and $800^{\circ}C$.

Studios on the Thermal Properties of Silane Crosslinked Polyethylene Prepared by Various Crosslinking Conditions (Silane 가교 PE의 가교조건에 따른 열적특성 변화에 관한 연구)

  • The silane crosslinking method was applied for the crosslinking of polyethylene (PE). Crosslinking of PE was performed by, first grafting vinyltrimethoxysilane(VTMOS) to the main chain of PE using an extruder at $200{\sim}210^{\circ}C$, followed by exposure to three different silane crosslinking conditions (1. immersed in $80^{\circ}C$ water, 2. at $80^{\circ}C$ air forced convection oven, 3. exposed to air at room temperature ). The thermal characteristic changes of PE resins with respect to the silane crosslinking conditions were studied by measuring the crystalline melting temperature, density and crosslinking reaction rate. Because silane crosslinking was carried out at solid state, crystalline melting temperature, crystallinity, crystal growth rate, crosslinking reaction rate and the change in the density of silane crosslinked PE were affected by crosslinking condition and the type of base resin. The properties of silane crosslinked PE were different from those of Peroxide crosslinked PE which was crosslinked at the molten state. It was found, from the result of DSC analysis, that silane crosslinked linear low density polyethylene(LLDPE) crosslinked at room temperature had no secondary melting peak because the crosslinking reaction proceeds slowly as the crystalline grows. After crystallization, the melting point of PE was lowered by crystalline interruption of crosslinked site.

The comparison of characteristics of Li$_2$O-2SiO$_2$--xCuO conduction glasses prepared by microwave and conventional energies (고체 전지용 Li$_2$O-2SiO$_2$-xCuO 계 전도성 유리의 제조에 마이크로파 에너지의 이용 및 특성 비교)

  • Effect of microwave heat-treatment processing on the electrical conductivity and crystallization behavior for the $Li_2O-2SiO_2$-xCuO glasses with various CuO contents was compared with that of conventional heat-treatment processing. The electrical conductivities of samples heat-treated at $500^{\circ}C$ by different heat-treatment processing were increased with increasing CuO content and higher electrical conductivities were obtained from microwave heat-treated samples. From the result of XRD analyses, microwave heat-treatment processing enhanced the degree of crystallization in the formation of $Li_2Si_2O_5, Li_2Cu_5$($Si_2O_7)_2$, and $Li_2Cu_2O_3$ crystalline phases. The electrical conductivities of $Li_2O-2SiO_2$-1.3CuO (30 mol% CuO) glass heat-treated at $500^{\circ}C$ for 30 min under conventional and microwave heat-treatment processing were $0.11{\times}10^{-4}(\Omega \textrm {cm})^{-1}$ and $0.68{\times}10^{-4}(\Omega \textrm {cm})^{-1}$ at room temperature, respectively. It was speculated that microwave energy enhanced the degree of crystallization and increased electrical conductivity in the samples.
