Effect of Young Phragmites communis Leaves Powder on Lipid Metabolism and Erythrocyte Antioxidant Enzyme Activities in High-Fat Diet Fed Mice (갈대순분말이 고지방을 급여한 마우스의 지질대사 및 적혈구 항산화방어계에 미치는 영향)

  • This study investigated the effect of young Phragmites communis (Pc) leaves on lipid profiles, lipid metabolism and erythrocyte antioxidant defense system in high-fat diet fed mice. Three groups of mice were fed different diets for 8 weeks: normal diet (Normal), high-fat diet (High-fat; 37% calories from fat) and high-fat diet supplemented with 1% Pc (wt/wt, HF-Pc). Body weight, daily food intake and energy intake tended to decrease by Pc supplement in high-fat fed mice. Pc supplementation significantly lowered plasma triglyceride and total cholesterol concentrations compared to the high-fat control group. Pc also lowered hepatic and heart cholesterol contents, whereas it significantly increased fecal excretion of triglyceride and cholesterol compared to the high-fat control group. Pc significantly inhibited fatty acid synthase, 3-hydroxy-3-methylglutaryl CoA reductase and acyl-CoA:cholesterol acyltransferase activities compared to the high-fat control group. Erythrocyte superoxide dismutase and catalase activities were also significantly higher in the high-fat group than in the normal group, however Pc supplementation reversed these changes. The Pc supplementation significantly lowered erythrocyte lipid peroxidation level compared to the high-fat control group. Accordingly, these results suggest that Pc improves lipid metabolism and erythrocyte antioxidant defense system in high-fat diet fed mice.

Effects of an aqueous extract of purple sweet potato on nonalcoholic fatty liver in high fat/cholesterol-fed mice (고지방/고콜레스테롤 식이를 섭취한 마우스에서 자색고구마 열수추출물 보충이 지방간 저항성에 미치는 영향)

  • Purpose: Anthocyanins from purple sweet potato (PSP) have been investigated in vitro and in animals and found to have a protective effect against oxidative hepatic damage. In this study, we investigated that aqueous extract of PSP can ameliorate the dysfunction of lipid metabolism in mice fed a high fat/cholesterol diet. Methods: Forty C57BL/6J mice were randomly divided into 5 groups (n = 8) and fed one of the following diets for 8 weeks; normal fat (NF) diet; high fat/cholesterol (HFC) diet; HFC with 1.25% PSP (HFPL) diet; HFC with 2.5% PSP (HFPM) diet; HFC with 5% PSP (HFPH) diet. Results: Non-alcoholic fatty liver was manifested in the HFC group by showing increased levels in plasma alanine aminotransferase (ALT) activity, total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C), increased level of TC and presence of many large lipid droplets in the liver, and increased fat cell size in the HFC group compared with the NF group. However, administration of HFC induced a significant decrease in food intake, resulting in decrease in fat mass. Co-administration of PSP did not lead to reversal of body weight changes, ALT activity, and lipid levels in plasma and the liver, but suppressed excess enlargement of the fat cell size through increasing carnitine palmitoyltransferase-1 (CPT-1) gene expression in the liver. Accordingly, the number of fat droplets in the liver was reduced in PSP administered groups. Conclusion: Taken together, these results suggest that PSP may have a protective effect on the dysfunction of lipid metabolism. Conduct of further studies on the coordinated regulation of PSP for lipid metabolic homeostasis at the liver-adipose tissue axis is needed.

Effects of Cheonggukjang Added Phellinus linteus myceria on Lipid Metalbolism in Adult Female Rats (상황버섯균사체청국장이 고지혈증을 유도한 암쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of Cheonggukjang (traditional soybean food, CK) and Cheonggukjang added with Phellinus linteus myceria (CKP) on the lipid metabolism, growth, food intake, and food efficiency ratio(FER) in adult female rats. Sprague-Dawley rats were fed a high fat diet (control diet: 0.1% cholesterol, 10% Lard) for 4 weeks. The rats were randomly assigned to each treatment group: control, and two kinds of CK and CKP (powders of CK and CKP as dietary protein sources). After 4 weeks of experimental diet consumption, the body weights and the uterine fat pad weights of the CK and CKP diet groups were more significantly decreased than those of the control diet group. The hepatic cholesterol, triglyceride and total lipid levels were significantly lower in both the CK and CKP diet fed groups than those in the control group. The concentrations in serum triglyceride and LDL-cholesterol were significantly decreased in the CK and CKP diet groups compared to those in the control group. Fecal total lipid, moisture and wet weight excretion in the CK and CKP diet groups were more increased than those in the control group. These results showed that feeding of cheonggukjang and cheonggukjang added with Phellinus linteus myceria both the triglyceride and LDL-cholesterol in serum, as well as the triglyceride and cholesterol in the livers of the rats.

Effect of Grape Seed Water Extract on Lipid Metabolism and Erythrocyte Antioxidant Defense System in High-Fat Diet-Induced Obese C57BL/6 Mice (포도씨열수추출물이 고지방식이로 유도한 비만마우스의 지질대사와 적혈구 항산화 방어계에 미치는 영향)

  • This study was investigate the effect of grape seed water extract (GSW) on lipid profiles, lipid metabolism and erythrocyte antioxidant defense system in high-fat diet-induced obese mice. Three groups of male C57BL/6 mice were fed different diets for 6 weeks: normal diet (Normal), high-fat diet (HF control; 37% calorie from fat) and high-fat diet supplemented with GSW (HF-GSW; 1% wt/wt). Supplementation of GSW did not affect the body weight, food intake, daily energy intake, white adipose tissue weights and plasma leptin level in high-fat fed mice. Plasma and hepatic cholesterol and triglyceride contents were significantly higher in the HF control group than in the Normal group; however, GSW supplement significantly lowered plasma triglyceride and hepatic cholesterol concentrations compared to the HF control group. GSW supplement significantly increased fecal excretion of triglyceride in high-fat fed mice. Hepatic carnitine palmitoyl transferase activity was significantly higher in the HF-GSW group than in the HF control group, while fatty acid ${\beta}$-oxidation tended to be lowered by GSW supplement. Erythrocyte superoxide dismutase activity was also significantly higher in the HF-GSW group than in the HF control group and glutathione peroxidase activity tended to be lowered in HF-GSW group. The GSW supplement significantly lowered erythrocyte lipid peroxidation level compared to the HF control group. Accordingly, these results suggest that GSW can be considered as a lipid-lowering agent and as being effective for enhancing erythrocyte antioxidant defense system in high-fat diet-induced obese mice.

Anti-obesity Effects of Ishige okamurae Extract in C57BL/6J mice Fed High-fat Diet (패 추출물이 고지방식이로 비만이 유도된 C57BL/6J 마우스의 항비만 효과에 미치는 영향)

  • We investigated the anti-obesity effects of Ishige okamurae extract (IOE) on body weight, epididymal adipose tissue weight, plasma lipid levels, and leptin levels in high-fat diet (HFD)-induced obese mice. After inducing obesity by feeding 45% HFD for 4 weeks, C57BL/6J mice were randomly assigned to HFD or HFD containing 5% IOE, and then fed for 6 weeks. The body weight and epididymal adipose tissue weight were increased by HFD, but they were significantly less in animals fed HFD containing 5% IOE than in those fed HFD. Levels of plasma triglyceride, total cholesterol, and LDL-cholesterol were significantly lower, but the HDL-cholesterol level was significantly higher in animals fed HFD containing 5% IOE compared with the HFD group. The plasma leptin level was significantly lower in animals fed HFD containing 5% IOE. The diet containing 5% IOE did not show any adverse effects on liver and kidney functions. These results suggest that IOE has a potential as anti-obesity agent by reducing body weight and lowering the levels of obesity-related factors in plasma.

Anti-obesity and Anti-inflammation Effects of Cheonggukjang in C57Bl/6 mice with High Fat Diet Induced Obesity (고지방식이로 유도된 비만 마우스에서 청국장의 항비만 및 항염증 효과)

  • The purpose of this study was to investigate the anti-obesity and anti-inflammation effect of the cheonggukjang (a soybean paste fermented for only a few days) in diet induced obesity mice. Weight gain was significantly decreased in the mice fed cheonggukjang compared High Fat Diets (HFD). The HFD plus cheonggukjang (CGJ) were also effective in improving the lipid metabolism. The levels of plasma triglyceride, cholesterol, ALT, AST, leptin, glucose, and insulin were significantly lower in CGJ than HFD group (p<0.05). The adiponectin level of CGJ group was significantly increased compared to the HFD group (p<0.05). In the CGJ group, the mRNA expression of adipogenic genes in the liver and adipose tissues, which are transcription factors crucial for adipogenesis, were significantly suppressed (p<0.05). The number of $CD11b^+F4/80^+$ T cells, $Gr-1^{int}CD11b^{high}$ cells, and $Gr-1^{int}CD11b^{high}$ cells were significantly higher in HFD group than CGJ group (p<0.05). The size of adipocyte was significantly reduced in CGJ group compared to HFD group. In addition, the contents of liver lipid droplets were significantly downregulated in the CGJ mice than HFD mice (p<0.05). Collectively, these data suggest the novel function of cheonggukjang in modulating adipogenesis through an immune function-alteration involving downregulation of adipogenic transcription factors and macrophage activation.

Effect of Ligusticum chuonxiong Hort Extracts on the Bioactivity in High-fat diet-fed Obese Rats (천궁 추출물이 고지방식이로 유도된 비만흰쥐의 생체활성에 미치는 영향)

  • This study was performed to investigate the antioxidative effect of Ligusticum chuanxiong Hort extracts (LCE) against the hyperlipidemia of high-fat diet-fed obese rats. The rats were divided into the three groups (normal group, control group and sample group) to perform the experimental research. 1.5 ml/kg of LCE was intraperitoneally administered into the sample group for 21 days. The equal dose of 0.9% saline was intraperitoneally administered into the normal group and the control group. On day 22, they were anesthetized with ether and dissected. The levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT) were examined in serum of rats. Superoxide dismutase (SOD) was measured in mitochondrial fraction. Malondialdehyde (MDA), catalase (CAT), and glutamate peroxidase (GPx) were determined in liver homogenate. High-fat diet markedly increased the levels of AST, ALT and MDA, significantly decreasing those of SOD, CAT and GPx. But Ligusticum chuanxiong Hort-pretreatment decreased the levels of AST, ALT, and MDA. increasing those of SOD, CAT and GPx. These results demonstrated the antioxidative effects, suggesting that LCE could be the candidate for the functional material.

Fingerroot (Boesenbergia pandurata) Extract Inhibits the Accumulation of Visceral Fat in C57BL/6J Mice (핑거루트(Boesenbergia pandurata) 추출물의 고지방 식이를 섭취한 마우스의 내장 지방 축적에 대한 효능)

  • Boesenbergia pandurata (Roxb.) Schltr. has been reported to possess anti-oxidative and anti-inflammatory properties. The aim of this study was to investigate the effect of Boesenbergia pandurata extract (BPE) in a high-fat diet (HFD)-induced obese mice model. C57BL/6J mice were fed with either the high-fat diet or a 0.5% BPE-supplemented HFD for 8 weeks. The BPE-containing HFD significantly reduced body weight gain and the accumulation of visceral fat mass in mice model without altering the amount of food intake. Moreover, mice fed with BPE-containing HFD had lower concentrations of lipids in their blood, lower hepatic lipid accumulation, and lower serum leptin levels compared with the HFD-fed mice. RT-PCR analysis showed that the expression levels of peroxisome proliferators-activated receptor ${\gamma}2$ ($PPAR{\gamma}2$) and CCAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$) genes in the epididymal fat tissue of mice fed the BPE-containing HFD increased 1.16- and 1.30-fold, respectively, compared to mice fed HFD only. In conclusion, BPE attenuated visceral fat accumulation and improved dyslipidemia in a mice model with HFD-induced obesity.

Effect of Dietary Grape Pomace on Lipid Oxidation and Related Enzyme Activities in Rats Fed High Fat Diet (포도박이 고지방식이를 섭취한 흰쥐의 지질 산화와 항산화 효소 활성에 미치는 영향)

  • The present study was conducted to investigate the effect of dietary supplementation of grape pomace on lipid peroxidation and related enzyme activities of rats fed high fat diet. Male Sprague-Dawley rats weighing about 90 g were assigned to 4 experimental groups of 8 rats on the basis of their body weight. The high fat diet contained additional 15% lard to AIN 93-based diet. Rats were fed experimental diets containing 5% grape pomace for 4 weeks. Dietary supplementation of grape pomace reduced serum concentration of lipid peroxide in rats fed high fat diet. Hepatic concentration of lipid peroxide tended to be lower by feeding grape pomace. Hepatic total glutathione content and GSH/GSSG ratio were increased by grape pomace feeding in normal or high fat diet groups. Hepatic superoxide dismutase activity of grape pomace group with high fat diet was induced significantly compared with high fat diet group without grape pomace. Hepatic catalase activity of high fat fed rats was induced by feeding grape pomace. Grape pomace diet increased glutathione-S-transferase and glutathione peroxidase activities in rat liver fed high fat. Hepatic glucose-6-phosphatase activity was not affected by dietary supplementation of grape pomace in rats fed high fat. These results suggest that dietary supplementation of grape pomace may alleviate lipid peroxidation through antioxidant effect in rats fed high fat.

Effects of Cheonggukjane on Lipid Metabolism in Hyperlipidemic Female Rats (고지방 섭취 암쥐에 청국장이 지질대사에 미치는 영향)

  • The effects of cheonggukjang(traditional fermented soybean food, CK) and cheonggukjang added Agaricus blazei (CKA) on lipid metabolism were investigated in female Sprague-Dawley rats (20 weeks old). The rats were fed a purified hyperlipidemic diet (control diet: 0.1% cholesterol, 10% fat, 18% casein) for 4 weeks. The rats were randomly assigned to each treatment group: control, two kinds of CK or CKA (powders of CK or CKA as dietary protein sources). After 8 weeks of experimental diets consumption, the body weights, and the uterine fat pad weights of CK and CKA diets groups were more significantly decreased than those of the control diet group. The liver cholesterol and triglyceride levels were significantly lower in both the CK and CKA diets groups than those in the control group. The concentrations in serum triglyceride, total cholesterol, LDL-cholesterol, and atherogenic index ratios were significantly decreased in the CK and CKA diets group s compared with those in the control group. The HDL-cholesterol/total-cholesterol ratios were significantly increased in the CK and CKA diets groups compared with those in the control group. The fecal cholesterol and triglyceride excretion in the CK and CKA diets groups were more increased than those in the control group. These results showed that both the feeding the rats with cheonggukjang and cheonggukjang added Agaricus blazei decreased the triglyceride, total cholesterol and LDL-cholesterol in serum as well as the triglyceride and cholesterol in liver, and increased the HDL-cholesterol/total-cholesterol ratio in serum.
