Postmortem Changes of Sterile Fish Muscle Inoculated with a Proteolytic Pseudomonas sp. (무균 어육에 단백질 분해세군 접종후의 저장중의 변화)

  • 사후 저장기간 중 어육내에 진행되는 단백질 분해에 어육 자체효소와 새균활동 중 어느 쪽이 기여하는 바 큰가를 분별하기 위하여 금방 죽인 볼낙류(Sebastodes)의 어육을 무균방법으로 잘라내어 4 OZ 유리용기에 넣고 플라스틱으로 된 나사뚜껑으로 밀봉한 후 감마선 조사하지 않는 군(무균육과 무군육에 단백질 분해성 Pseudomonas 를 접종한 것으로 세분)과 0.5과 2.0 Mrad 선량 조사한 군(무균육, 조사 전에 접종시킨 어육, 그리고 조사후에 접종시킨 어육으로 세분)으로 나누며 $0^{\circ}C$에서의 저장 중의 어육의 총균수, pH, 관능 상해, 그리고 0.5M KCl 및 $70\%$ ethanol 가용성 총질소와 Amino-N을 검측함으로써 어육 단백질 분해 과정을 조사하였다. 0.5 Mrad 감마선량은 무균 어육에 접종된 $10^6$ per gm Muscle Pseudomonas sp.를 완전 사멸시키는데 충분하였으므로 완전한 무균상태와 처음부터 단백질 분해성 세균활동이 활발한 상태에 놓여 있는 어육 내에 진행되는 단백질 분해과정을 비교할 수 있었다. 기름지지 않는 어류에 속하는 볼박 어육의 저장 중에 일어나는 pH의 상승, 관능상태의 저락, 그리고 단백질 분해에서 오는 총질소와 Amino-N의 축적은 모두 세균의 번식활동과 직접 관련되어 있으며, 완전 무균 어육의 단백질 분해는 전혀 검측되지 안했으므로 통상 저장중에 일어나는 어육 담백질 분해는 전부가 세균활동에 기인됨이 확실해 졌다. 감마선 조사 직후 단백질 분해성 세균을 접종시킨 어육을 조사치 않고 접종시킨 어육에 비교할 때, 전자의 경우 세균번식이 현저히 장해 받았으며, 이에 따라 PH 상승과 총질소 및 Amino-N 축적도 상대적으로 부진하였다. 이 방사선을 조사받은 어육에 접종된 단백질 분해성 세균활동의 장해는 조사선량에 비례함이 밝혀졌다. 관능상태로 판단할 때 0.5 Mrad선을 조사받은 어육은 조사 받지 않은 것보다 선도유지기간이 2배 이상으로 연장되었다. 그러나 2.0 Mrad 선량 조사받은 어육의 선도유지는 오히려 조사 받지 않은 것보다 못하였다. 이 고선량 조사로 인한 식품의 물리적 및 화학적 변화의 해독성은 접종시킨 세균 번식 장해현상으로 나타났으며, 이 현상은 조사식품의 안전성 검측을 위한 하나의 도구로 사용될 가능성을 말해준다.

미생물의 성 pheromone

  • pheromone은 생물의 체내에서 합성되고 체외로 분비되어 동종개체에 작용함으로써 특정의 생리적 변화를 유발하는 물질이다. 생물 중에서는 성장의 어느 단계에서 pheromone과 같은 물질이 관여하고 있다는 것이 알려져 있다. 또한, 단세포생물에서 세포 외로 분비하여 2개의 세포를 서로 접합시키는 물질이 있는데 이러한 물질을 성pheromone이라 부르고 있다. 특히, 효모 버섯, 조류, 섬모충류에서 연구되고 있다. 효모는 성접합형을 달리하는 두 균주가 상호pheromone을 분비, 수용하여 성분화를 행하여가고 있다. 그리고 세균에 있어서도 pheromone 양상의 물질이 세포 외로 분비되고 이 물질에 의하여 세균의 생리적 조건에 영향을 주는 예가 있다. 지금까지 알려져 있는 미생물의 성 pheromone에 대하여 크게 나누어 보면 지용성 성 pheromone과 수용성 성 pheromone으로 대별할 수 있다. 본 총설에서는 세균과 자낭균효모, 담자균효모유래의 성 pheromone 중 peptide(수용성) 성 pheromone에 대해서만 집약하였다.

Investigation of Conservative Genes in 711 Prokaryotes (원핵생물 711종의 보존적 유전자 탐색)

  • A COG (Cluster of Orthologous Groups of proteins) algorithm was applied to detect conserved genes in 711 prokaryotes. Only COG0080 (ribosomal protein L11) was common among all the 711 prokaryotes analyzed and 58 COGs were common in more than 700 prokaryotes. Nine COGs among 58, including COG0197 (endonuclease III) and COG0088 (ribosomal protein L4), were conserved in a form of one gene per one organism. COG0008 represented 1356 genes in 709 of the prokaryotes and this was the highest number of genes among 58 COGs. Twenty-two COGs were conserved in more than 708 prokaryotes. Of these, two were transcription related, four were tRNA synthetases, eight were large ribosomal subunits, seven were small ribosomal subunits, and one was translation elongation factor. Among 58 conserved COGs in more than 700 prokaryotes, 50 (86.2%) were translation related, and four (6.9%) were transcription related, pointing to the importance of protein-synthesis in prokaryotes. Among these 58 COGs, the most conserved COG was COG0060 (isoleucyl tRNA synthetase), and the least conserved was COG0143 (methionyl tRNA synthetase). Archaea and eubacteria were discriminated in the genomic analysis by the average distance and variation in distance of common COGs. The identification of these conserved genes could be useful in basic and applied research, such as antibiotic development and cancer therapeutics.

Studies on Sanitary Conditions in a Special Grade Chicken Processing Plant in Korea I. Storage Period of Packed Chicken Meat in relation to the Bacterial Contamination of Each Processing Part (특급도계장의 위생실태에 관한 연구 I. 작업부분별 세균오염도 측정)

  • In order to investigate the storage period of packed chicken meat in relation to the bacterial contamination of each processing part, a special grade poultry processing plant located at Dongdoochun-City, Gyeonggi-Do, was surveyed mainly in point of sanitary conditions. The results are summarized as follows. 1. processing room near reception and packing room next to shipping area showed the highest bacterial counts in air. 2. Equipments and instruments for slaughtering also showed a high degree of bacterial contamination even before the operation. This finding suggested that the possibility of the bacterial contamination of carcasses through equipments and instruments would be high, if the continuous sanitary control measures are not properly taken. 3. The bacterial count of scalding water was 3${\times}$10$\^$7/ for standard plate count and 4${\times}$10$\^$5/ for most probable number of coliform bacteria at 1 hour after the start of operation. These values were maintained until the completion of processing. 4. At the ending of chilling process, the bacterial counts of chilling water were 3.3 ${\times}$10$^4$ and 3.0${\times}$10$^3$for standard plate count, when the volume of water used per bird were 21 and 51, respectively. Bacterial contamination was further decreased about 10$\^$-3/-10$\^$-4/ times as the effective chlorine concentration in chilling water was maintained about 50 ppm. 5. An average standard plate count of the residual water In abdominal cavity of carcasses was 3.9${\times}$10$^4$immediate after chilling, however, it increased to 4.7${\times}$10$\^$7/ when the carcasses were packed and stored for 10 days at 4$^{\circ}C$. off-odour development was observed as the bacterial count was not less than 10$\^$6//$m\ell$ of the residual water of the packed carcasses. Data indicated that if the standard plate count of the residual water in abdominal cavity of carcasses was not less than l0$^4$immediate after chilling, the packed chicken meat would not be stored longer than 5 days at 4$^{\circ}C$.

  • The gene encoding a extremely thermostable 4-$\alpha$-glucanotransferase from a hyperthermophilic archaeon, Thermococcus litoralis, was cloned, sequenced and expressed in Escherichia coli. The amino acid sequence of the enzyme was distantly related to other functionally-related ones, such as D-enzymes. The enzyme is of industrial interest because of a novel activity of producing cycloamylose and is also important for fundamental studies of protein, sugar-metabolizing enzymes. In this paper, the overexpression of 4-$\alpha$-glucanotransferase in E. coli was carried out expression vector system with lac and T7 promoters. The enzyme was successfully overexpressed, and purified by the heat treatment of a cell-free extract, successive Butyl-Toyopearl and Mono Q chromatographies. The purified recombinant enzyme showed the same specific activity and the same mobility in SDS-PAGE as natural enzyme.

Microbial Contamination in Kitchens and Refrigerators of Korea Households

  • The objectives of this study were to investigate the microbial contamination level of domestic kitchen environments and to provide information to improve food safety in 50 domestic house kitchens located in Seoul, Incheon, and Gyeonggi-do. Dishcloth, chopping board, and refrigerator swabs were examined for the presence of coliforms, Salmonella spp., Campylobacter jejuni/coli, Escherichia coli, Listeria monocytogenes, and Staphylococcus aureus. The means and standard deviations of coliform counts for dishcloths was $4.8{\pm}1.84log\;CFU/100g$, chopping boards, and refrigerator drawers were $4.04{\pm}1.53$, $4.11{\pm}1.65log\;CFU/100cm^2$, respectively. Salmonella spp. and Campylobacter jejuni/coli were not detected in all samples. E. coli were detected in 3 on the dishcloths and 1 of 50 samples in the refrigerator drawer. Listeria monocytogenes was detected in the drawer of the refrigerator in 2 of 50 samples. In the case of Staphylococcus aureus, the detection on dishcloths, chopping boards, and drawers in refrigerators was 21, 12, and 14 of 50 samples, respectively. The results of microbiological tests of domestic kitchen utensils can be used to emphasize the importance of the sanitary conditions in domestic kitchen environments.

  • Chemotaxis is the movement of an organism toward chemical attractant and away from chemical repellents. Several bacteria are known to cometabolically degrade some pollutants and attracted to the pollutants. The chemotactic responses to these compounds influence the efficiency of bioremediation because the first precondition of pollutant degradation is definitely confrontation between microorganisms and pollutants. In this review, we summarize present knowledge of about the chemotactic mechanism to environmental pollutants of Pseudomonas species.

Optimal Conditions for Treatment of Swine Wastewater using Rhodopseudomonas palustris KK14 (Rhodopseudomonas palustris KK14를 이용한 돈분폐수처리의 최적조건 검토)

  • For the development of biological wastewater treatment process using photosynthetic bacteria (PSB), photosynthetic sludge process consisted of anaerobic digestion and PSB reactor were designed for the treatment of swine wastewater and the optimal operating conditions in flask-scale were examined. Photosynthetic bacteria from soil, pond, rice field, ditch etc. were isolated in synthetic medium containing high amount of organic acids and finally isolated one strain KK14 which showed the most degradating ability of organic acids was selected for the treatment of swine wastewater. It was identified as Rhodopseudomonas palustris. In the anaerobic digestion stage, the maximum organic acid productivity was obtained at pH 5.0, $37^{\circ}C$, HRT 2 day and under anaerobic standing condition. The optimal operating conditions of PSB reactor for the treatment of swine wastewater were pH 7.0, $30^{\circ}C$ under 4,000 lux illumination, and optimal initial COD loading (kg COD/kg D. C. W of PSB) was 2 (20% v/v seeding) in the main purification stage. Maximum removal rate of COD reached 92% under the above optimal conditions for 5 days.

Recombinant Expression of Agarases: Origin, Optimal Condition, Secretory Signal, and Genome Analysis (한천분해효소의 재조합발현 : 기원, 활성조건, 분비신호와 게놈분석 등)

  • Agarase can be used in the field of basic science, as well as for production of agar-derived high-functional oligosaccharides and bioenergy production using algae. In 2012, we summarized the classification, origin, production, and applications of agar. In this paper, we briefly review the literature on the recombinant expression of agarases from 2012 to the present. Agarase genes originated from 19 genera, including Agarivorans, Flammeovirga, Pseudoalteromonas, Gayadomonas, Catenovulum, Microbulbifer, Cellulophaga, Saccharophagus, Simiduia, and Vibrio. Of the 47 recombinant agarases, there were only two α-agarases, while the rest were β-agarases. All α-agarases produced agarotetraose, while β-agarases yielded many neoagarooligosaccharides ranging from neoagarobiose to neoagarododecaose. The optimum temperature ranged between 25 and 60℃, and the optimum pH ranged from 3.0 to 8.5. There were 14 agarases with an optimum temperature of 50℃ or higher, where agar is in sol state after melting. Artificial mutations, including manipulation of carbohydrate-binding modules (CBM), increased thermostability and simultaneously raised the optimum temperature and activity. Many hosts and secretion signals or riboswitches have been used for recombinant expression. In addition to gene recombination based on the amino acid sequence after agarase purification, recombinant expression of the putative agarase genes after genome sequencing and metagenome-derived agarases have been studied. This study is expected to be actively used in the application fields of agarase and agarase itself.

Antifungal Activity of Bacterial Strains isolated from Tidal Mudflat and Salted Seafood (traditional Jeotgal) Against Six Major Plant Pathogens (갯벌 및 젓갈에서 분리한 세균의 작물 주요 병원균에 대한 항균활성 효과 검정)

  • Many bacterial strains inhabit strong saline condition, such as tidal mudflat and salted seafoods, were identified and reported for the proposed protease activities and salt resistance; however antifungal activities against plant fungal pathogen have not well been studied until now. In this study, primary screening was performed for the isolation of promising strains against major plant pathogens like Sclerotinia sclerotiorum, Fusarium oxysporum, Phytophthora capsici, Botrytis cineria, Collectotrichum acutatum and Pythium ultimum. Totally 423 bacterial strain were isolated from laboratory media which was based on different morphological characteristics and all the strains were dual cultured against major fungal pathogens on PDA, finally 40 strains were selected as antifungal bacterial strain and identified by fatty acid phylogenic difference analysis from MIDI shorlock gas chromatography system. As a result, antifungal strains from tidal mudflat were 10 species of 6 genus. Paenibacillus macerans was dominant species; 5 strains among the 17 isolates from tidal mudflat. Antifungal strains from salted seafoods were 7 species of 3 genus and Bacillus atrophaeus was dominant species; 12 strains among the 23 isolates from salted fishes.
