한국 재래닭 경제형질 관련 QTL 탐색 및 표지유전자 개발

  • The analyses of relationship between specific genes and economic traits were performed on the basis of the genetic characterization of Korean native chicken in this study. Three generation reference pedigree was established from the several lines of Korean native chicken and cornish breed for the linkage analysis and many economic traits were collected for QTL(quantitative trait loci) detection. The experimental design were planed to detect QTLs of meat quality and laying performance from those two distinctive breeds.

  • In order to make the high quality Korean cattle, it has been identified the gene which influence to various economic characters. In this paper, we introduce Restricted Partition Method for gene-gene interaction analysis. Further, economic traits, longissimus muscle dorsi area (LMA), carcass cold weight (CWT) and average daily gain (ADG) are applied with Restricted Partition Method (RPM). The SNP (19_1)$^*$SNP (28_2) was selected and was best marker on Single nucleotide polymorphisms (SNPs). It also influenced SNP (19_1)$^*$SNP (28_2) was an very important marker for economic character and to make the thing know it became.

Current Research Status for Economically Important Candidate Genes and Microarray Studies in Cattle (소의 경제형질 관련 후보 유전자 및 Microarray 연구현황)

  • Researches in livestock are currently actively progressing to improve economically important traits using DNA markers. In cattle, the candidate genes have been selected based on their known functions in the target QTL (quantitative trait locus) region in order to identify QTN (quantitative trait nucleotide) for improving productivities. In this review, molecular genetic studies for the meat related traits, one of the major determinant of market prices, have been fully described. Also recent emerging microarray technique for identifying candidate genes in cattle has been discussed. In case of microarray, cDNA microarrays have been replaced to oligoarrays in order to minimize the experimental errors in cattle. Since the first draft of bovine genome sequences was appeared in the public domain, more markers in relation to the quantitative traits will be discovered in a short period of time and genes affecting difficult-to-measure traits, such as disease resistance, can also be selected for marker assisted selection in near future.

  • Kim and Lee (2015) identified a superior FABP4 gene that improves the grade and fatty acid of Korean cattle. This study selects a superior genotype by expanding genes that influence the economic traits of Korean cattle. Expanded genes are FABP4, SCD, FASN and SREBPs that are related to grade and fatty acid (Oh, 2014). We use the adjusted economic-trait values with environmental factors excluded. We also applied multifactor dimensionality reduction(MDR) method to data of the adjusted economic-trait values. As a result, we identified superior genes and genotypes which improved the grade and fatty acid of Korean cattle.

  • The propose of the present study was to evaluate the relative economic importance of the major Production traits in laying hens. Based on the results of this study, it was showed that feed requirement, average egg weight and hen-housed e99 Production influenced greatly on the income over feed and chick cost.

  • 외래 유전자를 이식하여 형질전환동물을 생산하는 방법은 현재 약 3가지 정도가 실제 이용되고 있다. 현재 가장 많이 사용되고 있는 방법으로는 DNA 미세주입법인데 생쥐와 비교하여 다른 동물에서의 형질전환효율은 상당히 낮은 것으로 나타나 형질전환가축의 생산을 위해서는 많은 비용과 시설 및 노동이 필요하여 실용성에 문제를 갖고 있다. 각 가축에 적합한 DNA 미세주입 조건을 확립시키고 형질전환동물의 생산효율을 높이는 연구가 필요하다. 가축에서 형질전환기술이 실제로 응용되고 있는 분야로는 대량생산이 어려운 의약품을 형질전환가축의 젖으로 합성 분비시키게 함으로써 생리적으로 활성이 있는 의약품의 대량생산이 가능할 것으로 예상된다. Growth Hormone이나 Growth Factor들을 이용한 성장과 관련된 형질전환가축의 기술은 예상했던 것보다 큰 성과는 없었는데 이식 유전자의 과잉발현으로 인한 부작용으로 실용화될 수 없었다. 그러므로 형질전환동물의 실용화를 위해서는 효율적인 유전자 이식방법의 개발과 이식 유전자의 발현으로 인한 부작용을 최소화하면서 좋은 표현형을 얻을 수 있도록 이식유전자의 발현을 인위적으로 조절할 수 있는 regulatory system의 개발과 가축의 경제형질에 관여하는 유전자의 식별이 필요하다.

Identification and Analysis of PIT1 Polymorphisms and Its Association with Growth and Carcass Traits in Korean Cattles (Hanwoo) (한우에서 Pituitary-specific Transcription Factor (PIT1) 유전자와 경제 형질과의 연관성 분석)

  • Pituitary-specific transcription factor (PIT1) 유전자는 동물의 성장을 조절하고 근육 형성에 관여하는 유전자로서 최근에는 단일염기다형성 변이가 한우를 비롯한 동물에서 관찰되었으며, 한우의 경제 형질과 연관성이 보고되었다. 본 연구는 PIT1 유전자의 단일염기다형성 변이가 한우에서 성장 인자에 미치는 영향과 경제 형질에 대한 유전자형간 육종가와의 상관성에 대해 알아보고자 하였다. 도체 성적을 보유하고 있는 한우 후보종모우 집단 268두를 대상으로 PIT1 유전자 A1256G 다형성을 조사하여 유전자형의 빈도를 분석하였고 각각의 유전형에 따른 기본적인 검정 성적을 바탕으로 경제 형질과의 연관성을 비교 분석하였다. 268두의 한우에서 PIT1 유전자의 A1256G 유전자형 빈도는 MseI 제한 효소를 사용했을 때 A 유전자 빈도(0.37)보다 G 유전자 빈도(0.62)가 높게 나타났다. 통계적 분석을 통하여 각 유전자형에 대한 경제 형질과의 관련성을 분석한 결과, 각 유전자형 간에 12개월령 체중 (body weight 12, BW12)에서 유의한 차이를 보였고, 등지방 두께 육종가 (Backfat thickness-estimated breeding value, BF-EBV)와도 유의한 차이가 있었지만 (p<0.05), marbling score (MS), carcass weight (CW), M. longissimus dorsi area (LDA) 등 다른 경제 형질과는 통계학적으로 유의한 차이가 없었다. PIT1 유전자의 A1256G 다형성은 한우의 성장과 도체체중에 관여하는 인자로 작용하는 것으로 보여진다.

Estimation of Environmental Effect and Maternal Effect for Swine Economic Traits (돼지의 경제형질에 대한 환경효과 및 모체효과의 추정)

  • This study looked into how much maternal genetic effect influenced on economic traits through estimation of genetic parameter and heritability over swine's economic traits by maternal animal model using GGP farm examination data of total 31,455 swine of Duroc species, Landrace species and Yorkshire species that were born between 2000 and 2008. As a result of significance test over each factor in surveyed all traits, high significance was approved in the effect of breed, gender, the date of swine's birth, the season swine's born, and difference in delivery in every trait (p<0.01). It is considered that it would be possible to get more efficient improvement effect provided correlation between additive genetic effect and maternal genetic effect as well as maternal genetic effect according to breed, traits, and improvement direction are properly considered as negative covariance existed between additive genetic distribution and maternal genetic distribution presumed for traits by each breed and their genetic relation also showed mostly strong negative correlation.

  • Developing economic traits with a high growth rate, robustness, and disease resistance in livestock is an important challenge. RFLP and AFLP are the classical methods used to develop economic traits. Whole-genome-based economic traits have recently been detected with the advent of next-generation sequencing (NGS) technologies. However, NGS technologies are rather costly for use in studies, and RNA-seq, RAD-Seq, RRL, MSG, and GBS have been used to overcome the issue of high costs. In this study, recent NGS-based studies were reviewed, particularly those that focused on minimum costs and maximum effects. Then, we presented further prospects on how to apply for selection of high economic-trait livestock.
