To understand the molecular structure of Korean garlic viruses, cDNA cloning of virus genomic RNA was attempted. Virus particles were isolated from virus-infected garlic leaves and a cDNA library was constructed from garlic virus RNA. One of these clones, S81, selected by random sequencing has been identified as a member of potexvirus group other than potyvirus and carlavirus. The clone is 873 bp long contains most of the coat protein (CP) coding region and 3'-noncoding region including poly(A) tail. A putative polyadenylation signal sequence (AAUAAA) and the hexanucleotide motif (ACUUAA), a replicational cis-acting element conserved in the 3'-noncoding region of potexvirus RNAs are noticed. The clone S81 shows about 30-40% identity in both nucleotide and amino acid sequences with CPs of potexviruses. The genome size of the virus was analysed to be 7.46 knt by Northern blot analysis, which was longer than those of other potexviruses. The open reading frame encoding CP was expressed as a fusion protein (S81CP) in Escherichia coli and the recombinant protein was purified by immobilized metal binding affinity chromatography. Polyclonal antibody was raised against S81CP in rabbit to examine the occurrence of garlic potexvirus in Korean garlic plants by immunoblot analysis. Two virus protein bands of Mr 27,000 and 29,000 from garlic leaf extract of various cultivars reacted with the antibody. It was shown that Mr 27,000 band might not be a degradation product of Mr 29,000 band, suggesting that two types of potexvirus different in size of coat protein could exist in Korean garlic plants.

A geophysical mapping was performed for Hwasan caldera which is located in Euisung Sub-basin of the southeastern part of the Korean Peninsula. In order to overcome the limitation of the previous studies, remote sensing technic was used and dense potential data were obtained and analyzed. First, we analyzed geological lineament for target area using geological map, digital elevation model (DEM) data and satellite imagery. The results were greatly consistent with the previous studies, and showed that N-S and NW-SE direction are the most dominant one in target area. Second, based on the lineament analysis, highly dense gravity data were acquired in Euisung Sub-basin and an integrated interpretation considering air-born magnetic data was made to investigate the regional structure of the target area. The results of power spectrum analysis for the acquired potential data revealed that the subsurface of Euisung Sub-basin have two density discontinuities at about 1 km and 3-5 km depth. A 1 km depth discontinuity is thought as the depth of pyroclastic sedimentary rocks or igneous rocks which were intruded at the ring vent of Hwasan caldera, while a 3-5 km depth discontinuity seems to be associated with the depth of the basin basement. In addition, three-dimensional gravity inversion for the total area of Euisung Sub-basin was carried out, and the inversion results indicated two followings; 1) Cretaceous Palgongsan granite and Bulguksa intrusion rocks, which are located in southeastern part and northeastern part of Euisung Sub-basin, show two major low density anomalies, 2) pyroclastic rocks around Hwasan caldera also have lower density when compared with those of neighborhood regions and are extended to 1.5 km depth. However, a poor vertical resolution of potential survey makes it difficult to accurately delineate the detailed structure caldera which has a vertically developed characteristic in general. To overcome this limitation, integrated analysis was carried out using the magnetotelluric data on the corresponding area with potential data and we could obtain more reasonable geologic structure.

To delineate geothermal water movement at the Pohang geothermal development site, Self-Potential (SP) survey and monitoring were carried out during pumping tests. Before drilling, background SP data have been gathered to figure out overall potential distribution of the site. The pumping test was performed in two separate periods: 24 hours in December 2003 and 72 hours in March 2004. SP monitoring started several days before the pumping tests with a 128-channel automatic recording system. The background SP survey showed a clear positive anomaly at the northern part of the boreholes, which may be interpreted as an up-flow Bone of the deep geothermal water due to electrokinetic potential generated by hydrothermal circulation. The first and second SP monitoring during the pumping tests performed to figure out the fluid flow in the geothermal reservoir but it was not easy to see clear variations of SP due to pumping and pumping stop. Since the area is covered by some 360 m-thick tertiary sediments with very low electrical resistivity (less than 10 ohm-m), the electrokinetic potential due to deep groundwater flow resulted in being seriously attenuated on the surface. However, when we compared the variation of SP with that of groundwater level and temperature of pumping water, we could identify some areas responsible to the pumping. Dominant SP changes are observed in the south-west part of the boreholes during both the preliminary and long-term pumping periods, where 3-D magnetotelluric survey showed low-resistivity anomaly at the depth of $600m\~1,000m$. Overall analysis suggests that there exist hydraulic connection through the southwestern part to the pumping well.

Electrical resistivity surveys were conducted at areas of abandoned landfills in Cheonan and Wonju. Geology and extent of leachate migration around the landfills were evaluated with collected resistivity data by 2-D and 3-D resistivity inverse modeling. The Cheonan landfill is located above the paddy fields and the resistivity survey lines were crossed to examine possible pollution at the paddy fields by leakage of the landfill leachate. In Wonju, the landfill and the downgradient paddy fields are divided by a concrete barrier wall. At the bottom of the landfill, there is a leachate settlement system, which has not been in operation. To evaluate leachate leakage into the paddy fields, a total of 4 survey lines were used. According to the resistivity survey results, the landfill leachate in Cheonan appeared to be restricted only within the interior of the landfill, not to migrate into the subsurface of the paddy fields. These results are well consistent with electrical conductivity values of groundwaters obtained from a periodic analysis of water qualities. In Wonju, however, it was inferred that the leachate emanating from the landfill migrated beneath the abandoned leachate settlement system and the leachate would reach the downgradient paddy fields. Low resistivity area was observed in the old reservoir area and it appeared to be derived from convergence of groundwater flows from the surrounding valley and the moist wet land. In addition, groundwater flow into the paddy fields occurs beneath the old reservoir embankment at depths of $7{\sim}8m$. This paper reports details of the resistivity surveys for the uncontrolled landfills.
This research examined prostanozol and its metabolites in urine of women who took the medicine (prostanozol). Prostanozol and its metabolites were successfully separated and detected by using LC/ESI/MS and GC/TOF-MS. Mass spectrum of LC/ESI/MS estimated molecular weight of Prostanozol and its metabolites and that of GC/TOF-MS verified them. For M1, carbon number 17 of Prostanozol substituted to a keto group and it is called 17-keto-Prostanozol. M2 turned out to be hydroxy-17-keto-Prostanozol. It came from substitution of one hydroxyl group of pyrazole nucleus and A-ring of M1. Substitution of one hydroxyl group of B-ring or C-ring became M3, hydroxy-17-keto-Prostanozol. M4 was found to be a hydroxy-17-keto-Prostsnozol transposed from one hydroxyl group to a D-ring. M5 has a hydroxyl group of carbon number 17. One hydroxyl group is substituted from B-ring or C-ring and it is assumed to be hydroxy-17-hydroxy-Prostanozol. M6 was turned out to be dihydroxy-17-keto-Prostanozol transposed from one hydroxyl group to pyrazole nucleus or A-ring and to B-ring or C-ring. Like M6, M7 has a keto group at carbon number 17 and was identified as dihydroxy-17-keto-Prostanozol. M7 has one hydroxyl group at pyrazole nucleus or A-ring and also at D-ring. At last M8 was found to be dihydroxy-17-hydroxy-Prostanozol. Pyrazole nucleus or A-ring has got one hydroxyl group and other rings were substituted to another hydroxyl group. From above, M5, M7 and M8 were verified as new metabolites that were not discovered yet. Prostanozol and all of the 8 metabolites formed glucuronic conjugates as a result of conjugation reaction test in human body. Some of 8 metabolites were excreted without forming conjugates. Particularly M6 and M7 were excreted as sulfate conjugates.
In this study, we examine how land use zoning affects the land price controlling other variables such as road-facing condition of the land, land form, land age after its development and land size. We employ geographically weighted regression analysis which reflects spatial dependency as methodology with a data sample of land transaction price data of Jangyu, a new town, in Korea. The results of our empirical analysis show that the respective coefficients of traditional regression and geographically weighted regression are not significantly different. However, after calculating Moran's Index with residuals of both OLS and GWR models, we find that Moran's Index of GWR decreases around 26% compared to that of OLS model, thus improving the problem of spatial autoregression of residuals considerably. Unlike our expectation, though, in both traditional regression and geographically weighted regression where residential exclusive area is used as a reference variable, the dummy variable of the residential land for both housing and shops shows a negative sign. This may be because the residential land for both housing and shops is usually located in the level area while the residential exclusive area is located at the foot of a mountain or on a gentle hill where the residents can have good quality air and scenery. Although the utility of the residential land for both housing and shops is higher than its counterpart's since it has higher floor area ratio, amenity which can be explained as high quality of air and scenery in this study seems to have higher impact in purchase of land for housing. On the other hand, land for neighbourhood living facility seems to be valued higher than any other land zonings used in this research since it has much higher floor area ratio than the two land zonings above and can have a building with up to 5 stories constructed on it. With regard to road-facing condition, land buyers seem to prefer land which faces a medium-width road as expected. Land facing a wide-width road may have some disadvantage in that it can be exposed to noise and exhaust gas from cars and that entrance may not be easy due to the high speed traffic of the road. In contrast, land facing a narrow road can be free of noise or fume from cars and have privacy protected while it has some inconvenience in that entrance may be blocked by cars parked in both sides of the narrow road. Finally, land age variable shows a negative sign, which means that the price of land declines over time. This may be because decline of the land price of Jangyu was bigger than that of other regions in Gimhae where Jangyu, a new town, also belong, during the global financial crisis of 2008.
The gene encoding $\beta$-agarase (agaB) which hydrolyzes $\beta$-1,4 linkages of agarose from Zobellia galactanivorans was cloned and fused to Saccharomyces cerevisiae mating factor alpha-1 secretion signal ($MF{\alpha}1$), in which the transcription of $MF{\alpha}1$-AgaB was under the control of AOX1 (alcohol oxidase 1, methanol inducible) promoter. The constructed plasmid pPIC-AgaB (9 kb) was integrated into HIS4 gene locus of Pichia pastoris genome. Successful integration was confirmed by performing colony PCR. The transformed cells showed red halos around its colonies in methanol agar plate by adding iodine solution, indicating the active expression of agaB in P.pastoris. By SDS-PAGE and zymographic analysis, the molecular weight of $\beta$-agarase was estimated to be a 53 kDa and about 15% N-linked glycosylation was occurred. The activity of extracellular $\beta$-agarase reached 1.34, 1.42 and 1.53 units/mL by inducing 0.1, 0.5, and 1% methanol, respectively, at baffled flask culture of P.pastoris GS115/pPIC-AgaB for 48 hr. Most of the enzyme activity was found in the extacellular fraction and the secretion efficiency showed 98%. Thermostability of recombinant $\beta$-agarase was also increased by glycosylation.
In order to reduce leakage from a reservoir, a large amount of cement milk (grout) was injected from boreholes drilled around the shores of the reservoir, and monitored to establish the infiltration of cement milk into the bedrock under the reservoir. From laboratory tests using rock core samples, it was revealed that the resistivity of cement milk is much lower than that of the groundwater at this location. Therefore, it was expected that the resistivity of the zones filled with cement milk would be significantly reduced. Geophysical surveys are expected to be suitable methods to check the effectiveness of grouting in improving the water-retaining performance of a reservoir. DC electrical surveys (seven in total) and two Controlled Source Audio-frequency Magneto-Telluric (CSAMT) surveys were conducted along survey lines in the reservoir to monitor the infiltration of cement milk during the grouting. Extremely low resistivity zones ($10\;{\Omega}m$ or less) were observed in resistivity sections obtained by 2D inversion. The zones are inferred to be fractured zones filled with cement milk. In sections showing the rate of change of resistivity, three zones that showed significant change showed gradual expansion to deeper parts as the grouting progressed. These zones correspond to highly permeable zones detected by Lugeon tests at grout boreholes. We have confirmed that it is possible to measure the resistivity change by DC electrical and CSAMT surveys from the surface of the reservoir. It seems that such monitoring results could be reflected in future grouting plans.

In this study the analysis is performed for influencing factors on the behavior of rigid piles (short pile) by research papers and case study. The results indicated that the point of virtual fixity should be calculated considering the relative stiffness of soil and pile, and Chang (1937) and P-Y method estimated the similar fixity. The values of ultimate resistances of a vertical pile to a lateral load are different for laboratory and field tests in cohesive soils and its ultimate values in laboratory tests are underestimated and in field tests are under or overestimated. The estimated resistance by Hansen (1961)'s method is similar to the value of field tests. The horizontal resistances to laterally loaded pile in cohesionless soils are overestimated in laboratory tests and generally overestimated in field tests. The ultimate resistances by Zhang (2005)'s method, used to the empirical distribution of the resistance, are similar to the test results. In the paper the calculating method and distribution of the ultimate resistance in cohesive soils are proposed. The estimated value by the proposed method is closer to the test results than any other method of calculating ultimate resistance of the piles embedded into cohesive soils.
This paper analyzes the displacements of CFRD which was completed by field measurement. It is to understand the deformation behavior of the dam body according to the water level change from the impounding time. And it was compared with numerical analysis results. As a result of measuring the behavior of the dam crest and downstream slope according to impounding, horizontal displacements in axis direction of the dam, upstream and downstream displacements and settlements occurred mostly when the water level reaches about half of the dam height. The displacements continued until the water level reached its maximum. After that, it showed a constant convergence regardless of the water level. Horizontal displacements of the face slab which is the most important in CFRD were similar at all locations. The Horizontal displacements of the face slab showed the trends of increasing in winter and decreasing in summer due to the effect of the outside temperature before impounding. Also, the displacements increased until the water level reached about half of the dam height. After that, they decreased with rising in water level. As a result, the face slab behaviors according to seasonal change after impounding as well as water level condition. It is judged because of the material characteristics of the concrete slab. Numerical analysis showed slightly different maximum settlement and depth of occurrence from the measuring data after construction of the dam. It is considered that this is due to various design and construction differences such as the estimation of input parameters in analysis, construction period, and the layer thickness of construction. For the overall period of the dam, the settlements were mostly completed during the construction period and some settlements occurred in the early days of impounding and then converged.
