Effects of pruning intensity and diameter of bearing mother branches on the growth and berry quality in 'Kyoho' grapevines ('거봉' 포도의 전정강도와 결과모지 굵기가 생육 및 과실품질에 미치는 영향)

  • This study was carried out to investigate suitable pruning methods to avoid poor berry setting of 'Kyoho' grapevine in Suwon and Anseong areas. Light pruning increased percentage of bud bursts and number of shoots per bearing mother branches(BMB), cluster weight and percentage of berry sating but it decreased shoot length, number of leaves and, leaf area and leaf weight. There was little difference in soluble solids, titratable acidity, and anthocyanin contents of berries by the treatment of either light or heavy pruning. Shoot length became longer as diameter of BMB thickened, leaf area and chlorophyll content also became larger and diameter of BMB. Total carbohydrate and N, P, K, Ca, and Mg contents showed significantly little difference, but total carbohydrate and Ca contents showed a tendency of continuous increase as diameter of BMB thinned. Number of seeded berries per cluster became fewer as BMB thinned but percentage of berry setting showed an increasing tendency. Cluster weight, berry weight, berry number per cluster, soluble solids, anthocyanin content were higher in case of around 1cm of diameters of BMB.

Effects of Artificial Pollination using Pollen Suspension on Fruit Set and Quality Attributes of 'Fuji' Apples (인공수분시 꽃가루 현탁에 따른 '후지' 사과의 착과 및 과실품질에 미치는 영향)

  • This study investigated the effects of using a pollen suspension for artificial pollination on the labor costs, fruit set, and fruit quality attributes of 'Fuji' apples. The pollen germination rate was 20% in a 20% fructose solution after 6 hours, and the pollen remained stable for 6 hours in the same solution. The king fruit per flower cluster exhibited a normal fruit set, regardless of the treatment. Plus, none of the artificial pollination applications affected the fruit length/diameter ratio, flesh firmness, soluble solids concentration, or titratable acidity in the harvested 'Fuji' apple fruits. However, the labor savings were significantly higher with the pollen suspension treatment when compared with the cotton swab or love-touch. Therefore, the labor cost was four-fold less in use of pollen suspension than in use of cotton swab for artificial pollination.

  • To examine the sex morph, fruiting characteristics and seed viabilities of Acer palmatum var. matsumurae (Koidz.) Makino, the flowers and seed-sets of 101 trees were collected from early April to late October 2010, and floral structure and seed viability were studied. Among investigated 101 trees, 73.3% of trees were protandrous (PA: pollen is shed before stigmas are receptive) and 26.7% were protogynous (PG: stigmas are receptive before pollen is shed). Acer palmatum var. matsumurae has heterodichogamous sexual system. Number of flowers and seed-sets per inflorescence are significantly different between sex morphs, and PA has more flowers and seed-sets per inflorescence than PG. Rates of sound seeds and decayed seeds are significantly different between sex morphs. Rates of sound seeds were 46.4% in PA, but 65.6% in PG. and rates of decayed seeds were 49.6% in PA, but 23.6% in PG. The differences of decayed seeds' rates between sex morphs might be related with the timing of pistil flower blooming and insect pests' outbreak.

  • Peanut was cultured with varying planting density and sowing date with and without vinyl-film mulching. Vinyl-film mulching raised soil temperature 4.6$^{\circ}C$ higher than the non-mulched during April and early May, but later-on the soil temperature was similar between the two treatments. The ripening and slender grain ratio increased with closer spacing and earlier sowing. Compared with ordinary cultural practices of May 10th sowing without mulching the peanut yield increased by 9% by mulching sown May 10th, and by 49% by mulching and early sowing April 20th.

Cone and Seed Characteristics among Different Cone Abundance Classes in a Seed Orchard of Pinus koraiensis (잣나무 채종원에서 구과 착과수에 따른 구과 및 종자특성)

  • This study was conducted to examine the relationship between cone abundance and cone/seed characteristics to improve seed quality and seed orchard management of Pinus koraiensis. The data was collected from P. koraiensis seed orchards at Chungju in 2007. That year was a rich year of cone harvest and the average number of cone per tree was 2.5 times much than that of normal year. We collected the cones from all trees in the sample plot of the seed orchard and investigated morphological characters such as cone length, cone width and cone weight. According to cone abundance per tree, the samples were divided into eight classes and analysed by cone analysis method. There is a decreasing tendency of number of fertile scale, seed potential, percent of filled seeds, seed weight per cone and seed efficiency, when the cone number per tree is increased. Additionally, the implications of the results to seed orchards management of P. koraiensis were discussed.

  • This study was conducted to find the flowering and seed setting characteristics of Eleutherococcus spp. Plants of Eleutherococcus spp. were 7 years old trees reproducted by cutting method using seven species in Gongju. Initial flowering date was foremost by May 9 in E. sieboldianus and hindmost by August 23 in E. divaricatus among seven species. Flowering duration was most short by 20 days in E. sieboldianus and extremely long by 42 days in E. sieboldianus. Number of flowers per plant in E. sieboldianus and E. chilsanensis were higher by 4677 and 4634 than those in E. divaricatus and E. sieboldianus by 780, 84, respectively. The fruit setting percent was the highest in E. sieboldianus by 95% and numbers of fruit setting per cluster showed the highest value by 153 in E. sieboldianus. The seed setting percent was almost same by ${92{\sim}95%}$ in E. sessiliflorus, E. chiisanensis, E. koreanus, E. divaricatus and E. seoulensis, but lower by ${11{\sim}37%}$ in E. senticosus and E. sieboldianus. The dry weight per plant and 100-fruit weight showed the highest value by 270g, 6.7g in E. sessiliflorus, respectively.

Variation of the Number of Empty Seeds per Cone in Pinus koraiensis Sieb. et Zucc (잣나무결실(結實)에 있어서 구과중(毬果中)의 쭉정이량(量)의 변이(變異))

  • The average number of empty seeds per cone on the mature tree was $10.03{\pm}0.61$. The ratios of empty seeds to total seeds and to full seeds were $7.38{\pm}0.35%$ and $8.42{\pm}0.49%$, respectively in average. The number of empty seeds per cone and the ratios were varied with the site condition; fruited year, and individual tree. The contribution rates of these sources of variance to the number of empty seeds per cone were respectively 3.50%, 8.06%, and 55.41%. No correlationships were found between the amount of empty seeds in number or ratio and the cone production, and the D.B.H. of mother tree. However, both the number of empty seeds and the ratio of empty seeds to total seeds showed significant positive correlations with the number of 1-year-old cones per tree.

Proper Tree Vigor and Crop Load in High Density Planting System for 'Fuji'/M.9 Apple Trees (사과 '후지'/M.9 밀식 사과원의 성과기 적정 결실 및 수세 기준)

  • Proper tree vigor and crop load were determined for 'Fuji'/M.9 apple trees in high density planting system from 2001 to 2003. Leaf/fruit ratio was highly correlated to mean fruit weight (y=1.715x+205.02, $R^2=0.66^{**}$) and yield (y=-35.l56x+5963.7, $R^2=0.44^{**}$). In addition, there was a significant correlation between the number of leaves per tree and mean fruit weight. However, crop load did not affect tree growth, soluble solids content of fruit, and Hunter a value. To harvest the fruits heavier than 300 g without biennial bearing, it was appropriate to crop 55 to 64 fruits in a tree with 55 leaves per fruit of adult tree. The good indices for proper tree vigor could be 20 to 25 cm of mean shoot length and above 95% of shoot termination rate. Moreover, no secondary growth and 20 to 30% of spur formation could be the indices for highly productive tree vigor.

  • To examine the sex morph, heterodichogamy, fruiting characteristics and seed viabilities of Acer palmatum, sixty three planted trees were monitered in Wonju-si for two years in terms of their sex morph, flowering and fruiting habit. Most flowers of A. palmatum are pollinated by Apis mellifera, Helophilus virgatus, a few (3.57%) are pollinated by wind. Twenty four trees (42.9%) in 2010, and twenty trees (31.7%) in 2011 were proved protandrous (PA: pollen is shed before stigmas are receptive), thirty two (50.8%) in 2010, thirty trees (47.6%) in 2011 were proved protogynous (PG: stigmas are receptive before pollen is shed). This result means that Acer palmatum has heterodichogamous sexual system. Five trees (7.9%) have changed their sex morphs; 4 trees change the sex morp PA to PG, and one does the sex morph PG to PA. Rates of sound seeds and decayed seeds were significantly different between sex morphs. Rates of sound seeds were 51.6% (2010) and 64.0% (2011) in PA, but 30.4% (2010) and 40.4% (2011) in PG. and rates of decayed seeds did 40.0% (2010) and 31.2% (2011) in PA, but 65.6% (2010) and 56.8% (2011) in PG. The differences of decayed seeds' rates between sex morphs might be related with the timing of insect pests' outbreak.

  • DNA 클로닝과 조작기술의 발전은 어떤 유전자의 특정한 위치에 선택적으로 돌연변이를 도입할 수 있는 site-specific mutagenesis 기술을 창출해 내었다. 이 기술로 DAN 염기의 치환, 결실, 삽입등을 클론된 유전자에 직접 도입할 수가 있게 되어 생체의 유전자 조작이나 유전자의 산물인 단백질의 구조와 기능을 의도적으로 변화시키는 protein engineering에 광범위하게 이용되고 있다. Protein engineering은 주로 단백질의 촉매 및 생리활성의 증가, 효소의 특성및 기질 특이성의 변화, 단백질 구조의 안정화 및 내염성 증가, 분자량의 감소, 효소및 생리활성 단백질의 구조의 안정화및 내열성 증가 등에 활용되고 있으며 산업적 유용성이 큰새로운 단백질의 창조에도 기여할 것으로 기대를 모으고 있다. Site-specific mutagenesis 기술로 현재 가장 널리 이용되는 것이 in vitro상에서 수행하는 oligonucleotide-directed site specific mutagenesis이다. 이 방법은 생화학적으로 합성한 특정한 염기서열을 가진 oligonucleotide들을 일종의 mutagen으로 사용하거나 효소적 DNA 합성을 위한 primer로 사용하여 클론된 DNA의 염기서열을 선택적으로 개조하거나 혹은 다른 조작을 하는 것이다. 여기서는 돌연변이율을 높이는 여러가지 개량된 방법들이 나왔으며 그중의 몇가지를 소개하였다.
