A Survey on Diseases and Insect Pests in Sweet Persimmon Export Complexes and Fruit for Export in Korea (단감수출단지 과원과 수출단감 병해충 조사)

  • Between 2010 and 2012, diseases and insect pests of sweet persimmon were surveyed at sweet persimmon export complexes and non-export orchards in Suncheon, Jeonnam Province; Jinju, Changwon (Dongeup and Bukmyeon), and Gimhae, Gyeongnam Province; and Ulzu, Ulsan. The following diseases were found in the sweet persimmon orchards: angular leaf spot (Cercospora kaki), anthracnose (Colletotrichum gloeosporioides and Colletotrichum acutatum), circular leaf spot (Mycosphaerella nawae), powdery mildew (Phyllactinia kakicola), and gray mold (Botrytis cinerea). Circular leaf spot was the most frequent and serious disease, and C. gloeosporioides and C. acutatum were found on fruits. Thirty-three insect pest species that belonged to 32 genera of 20 families in 5 orders were found in the sweet persimmon orchards; the two-spotted spider mite, Tetranychus urticae, was also found in the surveyed orchards. Apolygus spinolae, Pseudaulacaspis cockerelli, and Adoxophyes orana were widely found in the surveyed orchards; Spodoptera litura and Homona magnanima were also recorded. Damage by insect pests was low, and the quarantine insect pests peach pyralid moth (Dichocrocis punctiferalis) and persimmon fruit moth (Stathmopoda masinissa) were rarely or not found in the sweet persimmon export complexes. In addition, other quarantine insect pests, such as persimmon false spider mite (Tenuipalpus zhizhilashviliae) and Japanese mealybug (Planococcus kraunhiae), were not detected. These quarantine insect pests were also not found in the sorting places, storage houses, and fruits for export; however, scale insects and two-spotted spider mites were found at a low rate. Although anthracnose (C. acutatum) infested fruit was found in the storage houses, only one in Jinju and Gimhae.

Quality Characteristics of Low-Dose X-Ray-Irradiated Imported Navel Oranges during Storage under Low Temperature (3℃) (저선량 X선 조사 수입 오렌지의 저온저장 중 품질 특성)

  • This study evaluated the effects of low-dose X-ray irradiation treatment on quality characteristics and sensory evaluation of imported navel oranges during storage at $3^{\circ}C$ for 45 days. The samples were irradiated at doses of 0.2, 0.4, 0.6, 0.8, and 1.0 kGy, and changes in their color value, hardness, Brix/acid ratio, total sugar content, reducing sugar content, vitamin C contents, and sensory evaluation were investigated. There was no significant increase or decrease in Brix/acid ratio, total sugar content, reducing sugar content, or vitamin C content between the non-irradiated and irradiated samples. Color value of orange peels decreased with increasing levels of irradiation treatment. Color b value of orange pulp increased with an increase in irradiation dose. Difference in hardness between the non-irradiated and irradiated samples decreased at the end of storage. For the sensory evaluation after 30 days, sweetness and overall acceptability of irradiated samples at more than 0.6 kGy were low. These results suggest that X-ray irradiation under 0.6 kGy does not affect quality characteristics and sensory evaluation.

Development and Evaluation of PCR-Based Detection for Pseudomonas syrinage pv. tomato in Tomato Seeds (토마토 종자로부터 PCR을 이용한 Pseudomonas syringae pv. tomato의 검출)

  • The bacterial speck of tomato caused by Pseudomonas syringae pv. tomato leads to serious economic losses especially on fruits of susceptible genotype. Thus, Pseudomonas syringae pv. tomato is a plant quarantine bacterium in many countries including Korea. In this study, we developed specific PCR assays for detection of the bacterium from tomato seeds. A specific primer set is designed from the hrpZ gene for specific detection of Pseudomonas syringae pv. tomato. A 501 bp PCR product corresponding to hrpZ gene was amplified only form Pseudomonas syringae pv. tomato strains, but no PCR product was amplified from other tomato bacterial pathogens, such as Pseudomonas syringae pv. glycinea, P. syringae pv. maculicola, P. syringae pv. atropurpurea, P. syringae pv. morsprunorum, and from other P. syringae pathovar strains. The nested-PCR primer set corresponding to an internal fragment of the 501 bp sequence (hrpZ) gine was used to specific detection of Pseudomonas syringae pv. tomato in tomato seed. A 119 bp PCR product using nested PCR primer was highly specific and sensitive to detect low level of Pseudomonas syrigae pv. tomato in tomato seeds. We believe that the PCR assays developed in this study is very useful to detect Pseudomonas syringae pv. tomato from the tomato seeds.

Quality Characteristics of Low-Dose Electron Beam Irradiated-Imported Navel Orange during Storage at Room Temperature (20℃) (저선량 전자선 조사 수입 오렌지의 상온저장 중 품질 특성)

  • This study investigated the effects of low-dose electron beam irradiation treatment on the physicochemical and sensorial properties of imported navel oranges during storage at $20^{\circ}C$ for 12 days. The samples were irradiated at doses of 0.2, 0.4, 0.6, 0.8, and 1.0 kGy, after which changes in color value, hardness, Brix/acid ratio, total sugar contents, reducing sugar contents, vitamin C contents, and sensory evaluation were investigated. There were no significant differences between non-irradiated and irradiated samples in terms of color value, Brix/acid ratio, total sugar contents, total reducing sugar contents, and vitamin C contents. Hardness of irradiated samples significantly decreased in the early storage period in an irradiation dose-independent manner, and the difference between non-irradiated and irradiated samples decreased again at the end of storage. For the sensory evaluation, there was no significant difference between non-irradiated and irradiated samples up to 0.6 kGy, and all sensory item scores decreased at the end of the storage period regardless of irradiation. These results suggest that electron beam irradiation below 0.6 kGy does not affect physicochemical and sensory properties during storage at $20^{\circ}C$. Thus, electron beam irradiation up to 0.6 kGy applied to imported navel oranges is the optimum dose for minimizing quality changes and disinfestation treatment.

인터넷 기반 원스톱서비스 시스템 개발에 관한 연구 -수출컨테이너화물 원스톱서비스 시스템 개발-

  • 오늘날 우리 나라가 당면한 최우선 경제과제중 하나는 물류부문의 혁신을 통한 국가경쟁력 강화라고 할 수 있으며, 이를 위해 정부도 1993년 물류체계 개선을 위한 장기구상으로 ‘화물유통체계 개선 10개년 기본계획’을 수립하여 적극 추진 중에 있다. 그러나 이러한 노력에도 불구하고 PORT-MIS사용자를 상대로 한 설문조사에서는 선박입출항 업무 관련 서류의 40%, 항만시설 사용 업무와 관련된 서류의 31%, 하역업무 관련 서류의 10%만이 EDI를 활용하고 있었다. EDI 활용이 저조한 사유로는 전송시간이 많이 걸리며, EDI 소프트웨어가 작동되지 않으며, 수신확인이 되지 않기 때문이라 응답을 하였다. 이처럼 오늘날 항만물류산업이 겪고 있는 물류 데이타 흐름의 단절적 현상은 시간이 흐를수록 해결될 기미가 보이고 있지 않다. 따라서 본 논문에서는 우리 나라가 겪고 있는 물류관련 업무를 한번의 데이터 입력으로 해결할 수 있는 원스톱 서비스 시스템개발을 목표로 우선 PORT-MIS EDI 업무를 처리할 수 있는 시스템을 구축하였다. 이는 향후 화주, 운송사, 선사, 포워더, 창고업자, 하역회사, 철도청, 화물터미널, 컨테이너 터미널, 해양수산청, 관세청, 출입국관리사무소, 검역소 사이에 서로 교환되는 적하목록, Booking List, 컨테이너 Pick up정보, 위험물 정보, COPINO 정보를 비롯하여 대 관세청 신고 등 수출컨테이너 화물업무의 전반적인 영역으로까지 쉽게 확대할 수 있을 것이다. 본 연구결과 구축된 시스템은 원천정보를 중앙의 통합데이터베이스에 저장하여 이를 사용자의 요구에 의해 인터넷을 통해 전달하는 FTP와 웹 EDI 방식을 결합한 하이브리드 형태이다.인터넷으로 주문처리하고, 신속 안전한 배달을 기대한다. 더불어 고객은 현재 자신의 물건이 배달되는 경로를 알고싶어 한다. 웹을 통해 물건을 주문한 고객이 자신이 물건의 배달 상황을 웹에서 모니터링 한다면 기업은 고객으로 공간적인 제약으로 인한 불신을 불식시키는 신뢰감을 주게 된다. 이러한 고객서비스 향상과 물류비용 절감은 사이버 쇼핑몰이 전국 어디서나 우리의 안방에서 자연스럽게 점할 수 있는 상황을 만들 것이다.SP가 도입되어, 설계업무를 지원하기위한 기본적인 시스템 구조를 구상하게 된다. 이와 함께 IT Model을 구성하게 되는데, 객체지향적 접근 방법으로 Model을 생성하고 UML(Unified Modeling Language)을 Tool로 사용한다. 단계 4)는 Software Engineering 관점으로 접근한다. 이는 최종산물이라고 볼 수 있는 설계업무 지원 시스템을 Design하는 과정으로, 시스템에 사용될 데이터를 Design하는 과정과, 데이터를 기반으로 한 기능을 Design하는 과정으로 나눈다. 이를 통해 생성된 Model에 따라 최종적으로 Coding을 통하여 실제 시스템을 구축하게 된다.the making. program and policy decision making, The objectives of the study are to develop the methodology of modeling the socioeconomic evaluation, and build up the practical socioeconomic evaluation model of the HAN projects including scientific and technologica

For the sensory evaluation, scores of color, sweetness, flavor, and overall acceptability decreased as irradiation dose and storage period increased. Samples irradiated at over 0.8 kGy showed low preference in all scores except color. These results suggest that electron beam irradiation below 0.6 kGy does not affect physicochemical and sensory properties; thus, electron beam irradiation up to 0.6 kGy in imported navel oranges is optimum for minimizing quality changes and disinfestation treatment simultaneously.

Epidemiological Aspects and Trends of the Extensive Eradication Programs for Bovine Brucellosis Outbreaks-Associated Human Brucellosis in Korea, 2002~2013

  • Over the last decade, human brucellosis (HB) has become a principal zoonosis in Korea. The objective of this study was observing the epidemiological trends of the extensive eradication programs for bovine brucellosis (BB) outbreaks-associated HB in Korea from 2002 to 2013. The raw data analysis in this study was obtained from the website of brucellosis outbreaks by Korea Center for Disease Control and Prevention (KCDC) and Animal and Plant Quarantine Agency (QIA), Korea, 2002~2013. The number of cases of BB outbreaks-associated HB in Korea was increased after 2003 and peaked in 2006 before decreasing thereafter. There were a total of 89,240 infected BB in domestic cattle with a cumulative incidence rate (CIR) of 611.8 per 100,000 cattle; during the same period there were a total of 703 cases of HB with a CIR of 0.12 per 100,000 persons. Moreover, the correlation coefficient of brucellosis outbreaks between cattle and human was highly significant (r=+0.985). The attack ratio of HB was 7.88 per 1,000 BB cases. All of the control measures of HB were applied in the extensive eradication programs for brucellosis outbreaks of infection source in domestic animals and contributed significantly to the reduction in the outbreaks of HB in Korea.

Degradation of Poultry Feathers by Bacillus amyloliquefaciens Y10 With Plant Growth-promoting Activity and Biological Activity of Feather Hydrolyzates (식물 성장 촉진 활성을 가진 Bacillus amyloliquefaciens Y10에 의한 가금 우모의 분해 및 생산된 우모 분해산물의 생리활성)

  • This study was conducted to characterize strain Y10, isolated from discarded chicken feathers. Strain Y10 was identified as Bacillus amyloliquefaciens through phenotypic and 16S rRNA gene analysis. B. amyloliquefaciens Y10 exhibited plant growth-promoting activities, including the production of fungal cell-degrading enzymes (cellulase, lipase, protease, and pectinase), siderophores, ammonia, and indoleacetic acid. Furthermore, strain Y10 was able to inhibit the mycelial growth of several phytopathogenic fungi. When 0.1% sucrose as a carbon source and 0.05% casein as a nitrogen source were added to the basal medium, adjusted to pH 10, and cultured at 35℃, the degradation rate of chicken feathers by strain Y10 was about two times higher than that of the basal medium, with the feathers almost completely degraded in four days. Strain Y10 also degraded various keratin substrates, including duck feathers, wool, and human nails. It was confirmed that the feather hydrolyzates prepared using strain Y10 exhibited antioxidant activities, such as 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity (EC50 = 0.38 mg/ml) and superoxide dismutase-like activity (EC50 = 183.7 mg/ml). These results suggest that B. amyloliquefaciens Y10 is a potential candidate for the development of bioinoculants and feed additives applicable to the agricultural and livestock industries, as well as the microbiological treatment of keratin waste.
