This study evaluated the characteristics of rice cookies with varying amounts (0, 250, 500, and 750 mg) of dandelion complex powder. Dandelion powder is considered a functional food with skin-moisturizing and atopic skin improvement effects by KFDA. Quality characteristics of AF343 and rice cookies were measured by determining antimicrobial activities, physical characteristics, sensory evaluation values, and antioxidant activities. An antibiotic susceptibility test of the powder showed positive activities in Escherichia coli (O157:H7), Staphylococcus aureus, Bacillus cereus, and Listeria monocytogenes. Rice cookie containing 250-500 mg powder showed significantly increased antioxidant activities compared to controls (p<0.05). However, sensory intensities between the two sample groups were not significantly different. Thus, rice cookies containing 250-500 mg dandelion complex powder were successfully developed to improve antioxidant and antimicrobial qualities. These products may attract the attention of health-conscious consumers in the highly competitive cookie industry.
We tried to analyze the HPV prevalence and HPV genotypes of sexually high-risk women living in Busan, the biggest seaport of South Korea. Six hundred sixty women engaging in high-risk occupations participated in this study. The prevalence of HPV infection and HPV genotyping were determined with $MyGene^{(R)}$ HPVDNA chip, which consisted of 16 high-risk HPV genotypes (oncogenic genotypes) and 8 low-risk HPV genotypes. The overall prevalence of HPV infection in this study population was 39.1% (258/660) and the 20's showed the highest prevalence of HPV infection (51.5%). The dominant HPV genotypes including single or multiple HPV-infected women were resulted in HPV-16 (15.9%), -53 (10.2%), -58 (7.7%), -18 (5.2%) in case of high-risk HPV genotype and HPV-70 (10.4%), -6 (4.1%), -11 (2.0%) in case of low-risk HPV genotypes. Remarkably, the proportion of women infected with high-risk HPV genotypes (62.0%) was almost four times higher than those of women infected with low-risk HPV genotypes (14.7%) and high/low-risk HPV genotypes (12.0%). Among the 258 HPV-infected women, single infection was 175, double infection 66, triple infection 12, quadruple infection 4, quintuple infection 1, respectively. Our finding suggests that the introduction and development of effective HPV vaccines should consider the current status of HPV genotypic infection in South Korean women.
Cancer is a complex disease heterogeneously composed of various types of cells including cancer stem-like cells responsible for relapse and chemoresistance in the tumor microenvironment. The conventional two-dimensional cell culture-based platform has critical limitations for representing the heterogeneity of cancer cells in the three-dimensional tumor niche in vivo. To overcome this insufficiency, three-dimensional cell culture methods in a scaffold-dependent or -free physical environment have been developed. In this study, we improved and simplified the HCT-8 colon cancer cell-based spheroid culture protocol and evaluated the relationship between cancer stemness and responses of chemosensitivity to 5- Fluorouracil (5-FU), a representative anticancer agent against colon cancer. Supplementation with defined growth factors in the medium and the culture dish of the regular surface with low attachment were required for the formation of constant-sized spheroids containing $CD44^+$ and $CD133^+$ colon cancer stem cells. The chemo-sensitivities of $CD44^+$ cancer stem cells in the spheroids were much lower than those of $CD44^-$ non-stem-like cancer cells, indicating that the chemoresistance to 5-FU is due to the stemness of colon cancer cells. Taken together, the inflammation and oncogenic gut environment-sensitive HCT-8 cell-based colon cancer spheroid culture and comparative evaluation using the simplified model would be an efficient and applicable way to estimate colon cancer stemness and pharmaceutical response to anticancer drugs in the realistic tumor niche.
This study was conducted to determine the optimum nutrient solution, pH, irrigation interval, light intensity and planting density to growth of common ice plant (Mesembryanthemum crystallinum L.) in a closed-type plant production system. Three-band radiation type fluorescent lamps with a 12-h photoperiod were used. Nutrient film technique systems with three layers were used for the plant growth system. Environmental conditions, such as air temperature, relative humidity and $CO_2$ concentration were controlled by an ON/OFF operation. Treatments were comparison of the nutrient solution of Horticultural Experiment Station in Japan (NHES) and the nutrient solution of Jeju National University (NJNU), pH 6.0 and 7.0, irrigation interval 5 min and 10 min, light intensity 90 and $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, and within-row spacing 10 cm, 15 cm, 20 cm and 25 cm with between-row spacing 15 cm. Optimum macronutrients were composed N 7.65, P 0.65, K 4.0, Ca 1.6 and Mg $1.0mM{\cdot}L^{-1}$. There were no significant interactions between pH 6.0 and 7.0 about shoot fresh weight and shoot dry weight of common ice plant. Irrigation interval 5 min and 10 min was also the same result. Shoot fresh weight and shoot dry weight were highest at $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. Shoot fresh weight and shoot dry weight were decreased according to increasing the planting density. From the above results, we concluded that optimum nutrient solution, optimum levels of pH, irrigation interval, light intensity and planting density were 6.0-7.0 and 10 min, $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ and $15{\times}15cm$, respectively for growth of common ice plant in a closed-type plant production system.
This study was carried out to establish of environment-friendly control system and to survey the occurrence status of Ricania shanthungensis in Jeonnam province from 2016 to 2017. R. shanthungensis occurred at 1,344 ha in 61 towns of 15 cities in Gwangju and Jeonnam province. At four sites in the stationary area, the densities of eggmass and pupae were lower in 2017 than in 2016, the hatching rate decreased, and the first hatching time, emergence of adults, and the start of spawning tended to be slower. Telenomus sp., an egg parasitoid of R. shanthungensis, was found in most areas. Sunchon had the highest rate of 46.1%, followed by Muan 40.6, Goksung 29.2, Gurye 25.8, and Gwangju 17, respectively. The optimum spraying time for the control of R. shanthungensis was early March. The spraying material and the dilution multiples are 10 times of the machine oil at environmental friendly cultivation and 500 times of the chlorpriphosphate wp. at normal cultivation. When sprayed these materials, 95% of egg prohibited to hatch. There was more than 80% of the insecticidal effects on the organic materials of the machine oil emulsion, the Sophora root extrect + microbial extract agent, and the castor emulsion. For adults, Sophora root extrect + microbial extract, neem extract, and machine oil were better. Four kinds of chemical pesticides such as dinotefuran wp were effective for nymph and adult control. We have developed an adult catching device using the most preferred daylight color and behavioral habits of R. shanthungensis. The capture device consisted of two daylight compact lamps (30W and 20W), a yellow plate, and a catcher using water, and caught about 700 individuals a day. Based on the above results, we have established a system for controlling and life cycle of R. shanthungensis in Jeonnam province.
PraziquantEI ($Distocide^{\circledR}$), the KcrEan product, was tEstEd for its safety and Efficacy in treatmEnt of Clonorchis sinensiJ infccticn during the period from April to SeptembEr, 1983 in Korea. A total of 55 egg positive cases were selected and treated with the regimen of 25 mg/kg t.i.d. for 1 day (total 75 mg/kg). The follow-up stool examination was done in 47 cases by cellophane thir;k smear and Stoll's egg counting techniques. The 8 uncured cases were treated again with the same regimen. The laboratory tests for blood picture and liver function were done in 27 cases and compared before and after the treatment. The results obtainEd are as follows: 1. After single course treatment, the cure and egg reduction rates were 83.0 and 99.1% respectively. With the second treatment, excellent results of 100% in both rates were obtained. 2. Several kinds of side effects such as dizziness, headache, etc. were complained by 29 cases (61.7 %), however, those were so mild and transient that no special treatment was necessary. 3. No significant change in laboratory findings was recognizable before and after the treatment. From the above results, it is concluded that $Distocide^{\circledR}$ is as effective and safe as $Biltricide^{\circledR}$ and highly recommendable in treatment of C. sinensis infection.
Objective of this study was to assess rice growth and percolation water salinity under the irrigation of the discharge waters from the municipal wastewater treatment plant and from the industrial wastewater treatment plant as alternative water resources during transplanting season. Three kinds of waters were irrigated; the discharge water from an industrial wastewater treatment plant (DIWT), the discharge water from the municipal wastewater treatment plant (DMWT), and groundwater. Concentrations of $COD_{er}$, $NH_4{^+}_-N$, $Mn^{2+}$, and $Ni^+$ in DIWT, SS content and $PO_4-P$ concentrations in DMWT were higher than those of reuse water criteria of other country for agricultural irrigation. The plant height in the irrigation of DMWT was shorter by 2 cm than the groundwater irrigation except for 10 days irrigation. However, the number of tillerings was not significantly different between DMWT and the groundwater. For the harvest index, there were no significant difference between DMWT and DIWT for 20 days irrigation, but slightly higher in DIWT than that of DMWT for 30 days irrigation regardless of soil types. The salinity of percolation water in the rhizosphere with irrigation of DIWT had more twofold than DMWT, but SAR value from DMWT had no significantly different from the groundwater irrigation. The average $EC_i$ values in the rooting zone irrigated with DIWT and DMWT for 30 days after rice transplanting were 4.7 and $3.4dS\;m^{-1}$ in clay loam soil, and were 3.5 and $2.5dS\;m^{-1}$ in sandy loam soil, respectively. There was dramatic decrease in $EC_i$ value at 30 days after rice transplanting even though $EC_i$ of DIWT had more twofold than DMWT. However, $EC_i$ from DMWT had no significant difference from the groundwater. Therefore, it might be considered that there was limited possibility to irrigate DMWT to overcome drought injury of rice transplanting season in paddy field.
Effects of major phenolic acids identified from barley residues (straw, root) on the germination of rice and 3 paddy weeds such as Echinochloa crusgalli, Cyperus serotinus, and Potamogeton distintus were evaluated to obtain the basic informations on the development of naturally occurring herbicides. The predominant phenolic compounds extracted from barley residues in both straw and root were identified as p-coumaric, p-hydroxybenzoic, ferulic, vanillic, and salicylic acids by means of paper chromatography. Total phenol content of barley straw and root at the harvesting stage was 0.169% and 0.127% per dry weight, respectively. During the decomposing process, total phenol content slightly increased and then decreased. The germination of test plants was inhibited by treatments of 4 major authentic phenolic acids identified, most significantly on rice, and less on E. crusgalli, and C. serotinus. P. distintus, however, was markedly stimulated by them as the concentration increased, and then sprouted buds of pondweeds were changed to dark brownish color, resulting in the death as the treatment prolonged. The greater inhibitory effect appeared on shoot growth rather than germination. The aqueous extracts of barley residues showed the similar inhibitory effect on the germination and shoot growth of rice and three paddy weeds as the treatments of 4 authentic phenolic acids.
The behaviour of mixtures of herbicides was determined to obtain the basic informations about effective herbicide use, enhancing herbicidal efficacy and reducing the chemical cost. Fourteen herbicides with 91 mixed combinations were evaluated by Limpel et al method at the Echinochloa crus galli Beauv-Monochuria vaginalis Presl.-Scirpus hotarui Ohwi (importance values of these weeds were 63%, 16% and 10%, respectively) community type. Thirty eight mixed combinations showed the antagonistic response. Among these 14 mixed combinations including chlormethoxynil + naproanilide mixture were greater than 11% in antagonistic effect. On the other hand, 40 mixed combinations including chlormethoxynil + SW751 mixture showed additive response (${\pm}2%$). For synergistic response, 13 mixed combinations were belonged to this group. Particularly, 3 mixed combinations, chlormethoxynil + butachlor, chlormethoxynil + bifenox and nitrofen + ACN/MCPB/nitrofen mixtures were greater than 11% in synergistic effects. The mixture of thiobencarb + oxyfluorfen was analyzed by isobole technique. This mixture showed the synergistic response and the interaction index was approximately 2. The most optimum mixtur for inducing 90%n weed suppression was 0.012 kg ai/ha for oxyfluorfen and 0.45 kg ai/ha for thiobencarb.
To investigate the toxicological effects of ${\beta}$-glucan, we performed basic studying on ${\beta}$-glucan in Sprague-Dawley (SD) rats. Standard endpoints in this study included mortality, clinical observations, changes of body weights, analysis on food consumption, ophthalmoscopic examination, hematologic examination, serum biochemistry, analysis of organ weights, gross anatomic pathology and histopathology. No clinical signs and mortality were observed in animals treated with beta-glucan throughout the experimental period. The average body weight of each treatment groups showed similar levels at end of experiment. There were no treatment-related changes in mortality, body weights changes, food consumption, ophthalmoscopic examination. Although there were statistically significant differences between the control and treated groups in some relative and absolute organ weights, and hematological and biochemical analysis, the data were in biologically normal ranges and did not show a dose-dependent manner. In the morphological change, hepatic tissue were not showed ballooning degeneration and irregular arrangement of hepatic cell in ${\beta}$-glucan treatment groups with control group. Also, organs weights (liver, heart, kidney and stomach) and hematological indices (WBC, RBC, Hb, Hct and Platelet) did not show statistically significant differences among the experimental groups. In summary of these results, there were no changes in mortality, mean body weight, clinical signs, food consumption. There were no changes in ophthalmological examination, hematology, blood chemistry, necropsy and histopathology. In conclusion, although further investigation of glucan should be performed in the functions registered in many ancient literatures, ${\beta}$-glucan is physiologically safe and may have potential as candidate food for human health.
