The Cytoprotective Action of Portulaca oleracea 70% EtOH Extracts via the Heme Oxygenase-1 on Hydrogen Peroxide-induced Oxidative Stress in Human Keratinocyte HaCaT Cells (마치현 70% 에탄올 추출물의 Heme Oxygenase-1 발현을 통한 산화적 스트레스에 대한 사람각질형성세포 보호 효과)

  • Keratinocytes are first barrier against outer challenges on skin. However, it is still largely unknown about effective protectors against ultraviolet B (UVB), and oxidative stress in human keratinocyte, HaCaT cells. Inducible heme oxygenase (HO)-1 acts against oxidants that are thought to play a role in the pathogenesis of skin disorders. Therefore, the purpose of this study was to evaluate the effect of Portulaca oleracea 70% EtOH extracts against hydrogen peroxide (H2O2)-induced oxidative stress in human keratinocytes, HaCaT cells. P. oleracea 70% EtOH extracts showed the potent protective effects on H2O2-induced toxicity by induced the expression of HO-1 in human keratinocyte, HaCaT cells. Furthermore, P. oleracea 70 % EtOH extracts caused the nuclear accumulation of nuclear factor E2-related factor 2 (Nrf2) in human keratinocytes, HaCaT cells. In addition, we found that treatment with c-Jun N-terminal kinase (JNK) inhibitor (SP600125) reduced P. oleracea 70% EtOH extracts-induced HO-1 expression, and JNK inhibitor (SP600125) also inhibited protective effects by P. oleracea 70% EtOH extracts. Therefore, these results suggest that P. oleracea 70 % EtOH extracts increases cellular resistance to H2O2-induced oxidative injury in human keratinocyte, HaCaT cells, presumably through JNK pathway-Nrf2-dependent HO-1 expression.

  • Bee venom (BV) therapy has been used as a traditional medicine to treat a variety of conditions, such as arthritis, back pain, cancerous tumors, and skin diseases. However, regulatory effects of BV on tumor necrosis factor (TNF)-${\alpha}$-induced HaCaT cell migration or anti-inflammatory have not been explored. In the present study, we investigated the effects of BV on HaCaT cell migration and anti-inflammation. HaCaT cell migration was evaluated by wound-healing assay. The pro-inflammatory cytokines such as TNF-${\alpha}$, interleukin (IL)-$1{\beta}$, and IL-8 were examined by ELISA or Western blotting. BV treatment led to an increase in migration of HaCaT cells for 24 and 48 h. Especially, 10 ng/ml of BV were significantly increased HaCaT cell migration. Also, BV suppressed the secretion of TNF-${\alpha}$, IL-$1{\beta}$, and IL-8 in culture medium with HaCaT cells. In addition, Western blot results demonstrate that BV suppressed the expression of TNF-${\alpha}$ and IL-$1{\beta}$, in HaCaT cells. Especially, 1 or 10 ng/ml of BV markedly decreased the expression of pro-inflammatory cytokines. These results demonstrate the potential of BV for the prevention of skin inflammation induced by TNF-${\alpha}$.

Relationship Between Egg Shell Quality and Shell Breakage and Factors that Affect Shell Breakage (란각질과 란각파열과의 관계 및 란각파열에 영향하는 제요인)

  • About 6 to 8% of the eggs annually produced are broken or cracked between the hen and the consumer's carton. The majority of this breakage is due to interrelationships between egg shell quality and the many biological, environmental and managerial factors which have been individually identified as affecting breakage. Some of the factors associated with shell damage discussed in this review include age of hens, temperature ana humidity of the laying house, design of the cage systems and of the cage floors, type of material used to manufacture the cages frequency of daily egg collection in the laying house and, Probably most important, the frequency and quality of handling equipment maintenance. Age and genetic constitution of the hens, environmental temperature, and design and quality of equipment maintenance are the major factors that influence shell breakage. There is a curvilinear relationship between shell quality and shell breakage which explains why small changes in shell quality may be associated with large changes in the incidence of breakage. Published data indicate the incidence of breakage may range from as low as 1 to 2% to 35% or more for other egg producers.

Treatment of Partial Thickness Skin Defect with Cultured Allogenic Keratinocytes (Kaloderm®) (동종유래각질세포(Cultured Allogenic Keratinocytes, Kaloderm®)를 이용한 부분층 피부 결손의 치료)

  • Purpose: Grafting with autograft skin remains the most effective method for treating skin defects. When insufficient donor sites are present or patients are afraid of the operation, a skin graft is impossible. Cultured allogenic keratinocytes speed wound healing by providing cover and by producing growth factors and extracellular matrix protein. We report an application of cultured allogenic keratinocytes ($Kaloderm^{(R)}$, Tegoscience, Seoul, Korea) in the treatment of an acute partial thickness skin defect. Methods: From March 2005 to January 2006, 20 patients with a partial thickness skin defect were treated with cultured allogenic keratinocytes. The wound was covered with a sheet of cultured allogenic keratinocytes and ointment with $Bactigras^{(R)}$ gauze. The wound was inspected every two or three days. We regarded completion of epithelialization as wound healing. Results: The mean period between time of injury and time of $Kaloderm^{(R)}$ application was 7.5 days. The time taken from application of $Kaloderm^{(R)}$ to complete closure of the wounds was 7.2 days. Conclusion: In view of the favorable outcome, cultured allogenic keratinocytes are safe and effective biologic dressing materials for use in the treatment of open wounds.

A Study on the Comparison of Explosive Lower Limit Concentration & Thermal Specific of Wheat Powder Dust & Salicylic Acid Dust (밀가루분진 및 살리실산분진의 폭발하한농도 및 열적특성 비교에 관한 연구)

  • We have examined In order to compare each other from explosion and combustion characteristics about the dusts which collects from manufacturing process of wheat flour and cosmetics manufacturing process of functional Keratin removal soap at the small and medium enterprise style. We measured explosive pressure and explosive lower limit which follows in change of concentration change at the time of talc addition uses Hartman dust explosion apparatus, also measured weight loss and endothermic quantity uses DSC and TGA. The explosion test results show that increased explosive lower limit concentration and explosive pressure decreased by the increased ratio of the talc dust. And the DSC results show that heat flux and temperature decreased by the increased ratio of the talc dust. Also increased in raising temperature causes initial smoldering temperature to move towards low temperature section and the endothermic quantity increased on a large scale. Together the TGA results show that weight loss decreased by the increased ratio of the talc dust. From this research we have assured the successive dust explosion mechanism study will play a key role as a significant safety securing guideline against the dust explosion.

Effects of Catechin-rich Green Tea Extract on the MMP-1 Activity of HaCaT Keratinocyte Cells and on UVB-induced Skin Damage in Hairless Mice (각질형성세포에서 MMP-1 활성 및 자외선 유도 무모쥐 피부손상에 대한 카테킨 고함유 녹차추출물의 영향)

  • Background: Skin is an organ that protects the human body from various environmental stimuli that can induce immune system activation. Skin aging can be largely divided into two categories: physiological aging, which is caused by the a decreased physiological function of the skin and structural changes with aging, and photoaging, which is caused by the chemical stress induced by external stimuli such as ultraviolet (UV) radiation. Methods and Results: The objective of this study was to investigate the anti-wrinkle and UV protective effect of catechin-rich green tea extract (CGTE) in activated keratinocyte (HaCaT cells) and UV-induced skin damage in hairless mice. The results showed that CGTE inhibits the tumor necrosis factor-alpha interferon-gamma ($TNF-{\alpha}+IFN-{\gamma}$)-induced expression of matrix metalloproteinase (MMP)-1 in HaCaT cells. In addition, the CGTE treatment significantly reduced wrinkle formation, epidermal thickness, collagen deposition, and transepidermal water loss in dorsal skin irradiated with UVB. However, the ${\beta}$-glucosidase activity was significantly increased. The CGTE treatment inhibits mRNA expression and enzyme activity of MMP-2 and MMP-9 in the dorsal skin irradiated with UVB. Conclusions: It is expected that CGTE can be effectively used as a functional food and cosmetic ingredient to improve skin moisture retention and reduce wrinkle formation.

Investigation of the effect of Hibiscus sabdariffa L. extracts on tight-junction related genes in human keratinocyte HaCaT cells (히비스커스 추출물이 인간 각질 형성 세포의 밀착 연접 관련 유전자 발현에 미치는 영향 연구)

  • Objectives : Hibiscus (Hibiscus sabdariffa L.) is rich in antioxidants such as flavonoids and anthocyanins and is known to have anti-inflammatory activity and anti-aging function of the skin, but there is no study on its effect on the skin barrier. This study aim to investigate the positive effect on the skin barrier by confirming the effect of water extracts of Hibiscus sabdariffa L. (WEHS) on the tight junction-related gene expression. Methods : The antioxidant efficacy of WEHS was investigated through ABTS and DPPH assays, and 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium was performed to examine the effect on cell viability. quantitative Reverse transcription polymerase chain reaction and immunoblot analysis were performed to confirm the effect of WEHS on the expression of tight junction-related genes, and immunofluorescence microscopy was used to confirm the movement of Claudin 1 protein into tight junctions. Results : WEHS showed strong antioxidant activity and induced an increase in both mRNA and protein expression levels of Claudin 1 among tight junction-related genes. The strong localization of Claudine 1 protein increased by WEHS to the tight junction was confirmed by immunofluorescence microscopy. Conclusions : Hibiscus was confirmed through this study to show antioxidant activity and the function of promoting the expression of the tight junction Claudin 1 gene, suggesting that Hibiscus can be used as a material for the prevention and treatment of skin moisturizing and atopy, which have an important influence on tight junction.

Development of Anti-aging from Natural Materials by Inhibition of UV Stimulating (자외선 자극에 의한 피부노화 억제 천연물 소재 개발)

  • ln this study, natural extracts extracted from cypress sapiens, a natural material, were investigated as materials that could protect skin aging caused by ultraviolet rays, and experiments were conducted on the synthesis of filaggrins that make up the natural moisturizing factor of the skin, the synthesis of pro-colagen, a fibrous protein, which plays an important role in moisturizing the dermis, and elastin, which is an enzyme that decomposes collagen. As a result, cypress ethanol extract (COE) was a dependent inhibitor to collagenase and elastase, inhibiting the synthesis of filaggrin and the expression of MMP-1 for exfoliated cells damaged by ultraviolet rays. Therefore, it is estimated that ethanol extract will have the effect of delaying wrinkles and as a functional cosmetic material that inhibits skin aging convergence. Based on this study, we would like to further study the mechanism of the synthesis of filaggrin on the suppression of expression of MMP, which is the anti-wrinkle effect.

Using similarity based image caption to aid visual question answering (유사도 기반 이미지 캡션을 이용한 시각질의응답 연구)

  • Visual Question Answering (VQA) and image captioning are tasks that require understanding of the features of images and linguistic features of text. Therefore, co-attention may be the key to both tasks, which can connect image and text. In this paper, we propose a model to achieve high performance for VQA by image caption generated using a pretrained standard transformer model based on MSCOCO dataset. Captions unrelated to the question can rather interfere with answering, so some captions similar to the question were selected to use based on a similarity to the question. In addition, stopwords in the caption could not affect or interfere with answering, so the experiment was conducted after removing stopwords. Experiments were conducted on VQA-v2 data to compare the proposed model with the deep modular co-attention network (MCAN) model, which showed good performance by using co-attention between images and text. As a result, the proposed model outperformed the MCAN model.

Physiological Study of the Extract of Junglans nigra Shells for the Cosmeceutical Application (코스메슈티컬 적용을 위한 흑호두 과피의 생리학적 연구)

  • Among natural products, the shells of black walnut(Juglans nigra), which are not used commercially and mostly discarded, were examined to investigate the physiological activity and the efficacy for the cosmetic application. DPPH radical scavenging activity of hot water extract of black walnut shells was 76.06% at 300 ㎍/mL. ABTS radical scavenging activity of the extract was 61% at 1000 ㎍/mL, showing excellent antioxidant activity. When the black walnut shell extract was applied to HaCaT cells, a skin keratinocyte, the viability of the cells was 92.6% at 250 ㎍/mL, showing a remarkably low effect on cell viability. At the concentration of 500 ㎍/mL, 67.35% of nitric oxide(NO) production was inhibited. It also showed an inhibitory effect on Hyaluronidase 31 times higher than that of Vitamin C at 100 ㎍/mL concentration. In conclusion, the black walnut shell extract showed high potentials for the cosmeceutical applications, suggesting the possibility of using it as a high value-added natural material in various industries such as food, fragrance, healthcare, and pharmaceuticals.
