Improvement on the Quality and Functionality of Skipjack Tuna Cooking Drip Using Commercial Enzymes (효소분해에 의한 참치 자숙액의 품질 및 기능성 개선)

  • For the use of skipjack tuna cooking drip (STC) as a source of functional seasoning, the STC was hydrolyzed with various commercial enzymes, such as Alcalase, Flavourzyme, Neutrase and Protamex, and its hydrolysate was also investigated on the food component characteristics. The hydrolysate incubated with Alcalase for 30 min (HA30) showed 56.8% for angiotensin I converting enzyme (ACE) inhibitory activity and 1.18 for antioxidative activity, which were high or similar compared to the other enzymatic hydrolysates. There were no differences in ACE inhibitory activity and antioxidative activity among HA30, two-step enzymatic hydrolysates, and ultrafilterates (molecular weight cut off, 10 kDa). The HA30 was very stable on the digestive enzymes, such as chymotrypsin, pepsin, trypsin according to the TCA (trichloroacetic acid) soluble index. The results suggested that skipjack tuna cooking drip could be used as a source for preparing functional seasoning sauce.

Effects on Quality Characteristics of Extruded Meat Analog by Addition of Tuna Sawdust (참치 톱밥의 첨가가 압출성형 인조육의 품질 특성에 미치는 영향)

  • In this study, tuna sawdust was added to extruded meat analog in order to develop a meat analog with high quality. Addition of tuna sawdust has merit for utilizing a byproduct from poultry processing. Physicochemical characteristics were examined through the extrusion cooking process. The basic mixture of sample mixed with 65% deffated soy flour 25% isolated soy protein, and 10% corn starch was setup as the raw material. Three kinds of samples were made in total by addition of 15% and 30% tuna sawdust to this mixture. The extrusion process had a screw speed of 250 rpm, die temperature of $140^{\circ}C$, and moisture content of 50%. As addition of tuna sawdust increased, breaking strength and density decreased, specific length increased, and integrity and water holding capacity decreased. Likewise, nitrogen solubility index and protein digestibility decreased as addition of tuna sawdust increased. DPPH radical scavenging activity increased as addition of tuna sawdust addition, whereas it decreased as storage period increased to 30 or 60 days. The value of rancidity decreased as addition of tuna sawdust increased. However, 60 days later, radical scavenging activity increased more or less, and a significant difference was detected 150 days later. In conclusion, addition of tuna sawdust increased soft texture, and nutrition of the basic mixture sample. The process promoting functionality such as improvement of antioxidant function was confirmed through this study.

Development of Spaghetti Sauce with Adductor Muscle of Pearl Oyster (진주조개 패주 스파게티 소스의 개발)

  • This study was carried out to prepare spaghetti sauce with adductor muscle of pearl oyster (SSAM) and to compare with commercial spaghetti sauce (CSS). From the results of organic acid, pH, hunter color value and sensory evaluation, the optimal addition ratio of adductor muscle of pearl oyster was 11% based for preparing SSAM and reasonable $F_0$ value was about 4 min for keeping storage of SSAM. The proximate composition of SSAM was 69.8% for moisture, 3.7% for protein, 4.2% for crude lipid and 3.4% for crude ash. SSAM was superior in sensory flavor and texture to CSS. There was, however, no significant difference (p<0.05) in sensory color between CSS and SSAM. The total amino acid content (3,033.4 mg/100 g) of SSAM was higher than that (2,305.7 mg/100 g) of CSS and the major amino acids were aspartic acid, glutamic acid, leucine and lysine. Calcium and phosphorus contents in SSAM were 48.5 mg/100 g and 27.1 mg/100 g, respectively. Calcium level based on phosphorous was 1.78, which was a good ratio for absorbing calcium. The free amino acid content and taste value of SSAM were 989.4 mg/100 g and 141.04 mg/100 g, respectively These results suggested that the major taste active compounds among free amino acid were glutamic acid and aspartic acid.

The Development of a Natural Seasoning Using the Enzymatic Hydrolysate of Fish Skin (어피의 효소적 가수분해물을 이용한 천연조미료의 개발)

  • A study on the optimum hydrolysis conditions of fish skin through the aid of enzymes and the development of a natural seasoning using the hydrolysate has been carried out for the effective utilization of fish skin. Using the "pH-drop" techniques the collagenase and pronase were identified as most suitable for this purpose. The $K_m$ and $V_{max}$ values of pronase were 1.82 mgN/ml and 0.06 mgN/mL/min, respectively. The hydrolysis conditions of the cod skin for the pronase were as follows: reaction temperature, $50^{\circ}C$; reaction time, 3hrs; pH 6; enzyme concentration, 0.03%. The degree of hydrolysis at these conditions was 76.8%. But after hydrolyzing cod skin with collagenase for 1hr, when the pronase was treated, the degree of hydrolysis was 83.13%. The molecular weight of the hydrolysate was 8,000 daltons. Among the amino acids in the hydrolysate, glycine(27.95%), glutamic acid(10.94%), proline(7.39%), aspartic acid(9.47%) and serine(7.39%) were responsible for 64.23% of the total amino acids. But valine, methionine, isoleucine, leucine, phenylalanine and histidine having a bitter taste were only 13.05%. From the results of the sensory evaluation, the imitation sauce which was made of 20% fermented soy sauce prepared from the hydrolysate was at least similar to the traditional soybean sauce in product quality. The complex seasoning containing 31.7% of the hydrolysate was nearly equal to complex seasonings on the market, too.

Proteolytic Conditions for the Hydrolysate of Flounder Skin Gelatin (효소에 의한 가자미피 젤라틴 가수분해물의 제조 조건)

  • In order to develop a new flavourant using the fish skin gelatin, the proteolytic renditions for the gelatin hydrolysate of the alkali (B-type) and Alcalase (E-type) pretreated flounder (Limanda aspera) skin gelatin were investigated, and some physical properties, molecular weight and amino acid compositions of the hydrolysates were, also, compared with each other. The proteolytic conditions of the gelatins (B-type and E-type) by trypsin were as follows : reaction temperature, 55$^{\circ}C$ : pH, 9.0 : enzyme concentration, 0.1% : re-action time, 4hrs for B-type and 1 hr for E-type. The degrees of hydrolysis of the B-type and E-type gelatin un-der the renditions stated above were 63% and 82%, respectively. The rnajor molecular weights of the hydrolysates were 15,000 dalton for B-type and 12,400 dalton for E-type. Among the amino acids in the hydrolysates, glycine, alanine, proline, hydroxyproline and serine having a sweet taste were responsible for 57% of the total amino acid. But valine, leucine, phenylalanine, tyrosine, methionine, arginine and histidine having a bitter taste were only 18%.

Monitoring on Benzo(a)pyrene Content in Oriental medicine (유통 한약재 중 벤조피렌 함유량에 관한 모니터링)

  • Benzo(a)pyrene is a polycyclic aromatic hydrocarbons (PAHs) whose metabolites are mutagenic and highly carcinogenic and is listed as a Group 1 carcinogen by the IARC. It has been found at variable concentrations in several foods and is associated with several factors during the process including contaminated raw materials, exposure of environment, and procedure of process or cooking. In this study, benzo(a)pyrene in 45 oriental medicines were determined by HPLC/FLD. The calibration curves of benzo(a)pyrene was linear over the concentration range of 0.5~40 ng/mL with correlation coefficient of above 0.999. The limit of detection (LOD) and limit of quantitation (LOQ) of benzo(a)pyrene were 0.04 and 0.10 ${\mu}g/kg$. Benzo(a)pyrene in 3 samples out of 45 samples was not detected. The level of benzo(a)pyrene in 26 (57.7%), 8 (17.8%) and 7 (15.6%) samples was 0.1~0.5, 0.5~1.0 and 1.0~5.0 ${\mu}g/kg$, respectively. Especially, content of benzo(a)pyrene in Coptis Rhizome is the highest (5.97 ${\mu}g/kg$). In conclusion, these results suggest that could be applied to fundamental study and guideline on drying condition to decrease content of benzo(a)pyrene in oriental medicine.

  • This study examined the physicochemical properties of rice embryo in order to investigate the antioxidative effect of black rice bran pigment fraction on rice embryo lipid oxidation. Color stability of rice bran pigment fraction and acid value of added rice embryo were determined at various conditions of pH, temperature, and storage time. Rice embryo contained more crude protein (22.1%) compared to the rice bran (9.4%). However, rice embryo had a higher lipid content than rice bran, causing lipid oxidation during storage. Pigment fraction were relatively stable in acidic pH of 3.0 stored at $4^{\circ}C$ for 9 days. Rice embryo showed increased acid value with increased storage time and temperature. Furthermore, rice embryo with pigment fraction resulted in lower acid value compared to the control group. Therefore, addition of black rice bran pigment fractions to rice embryo improved storage capacity.

Physicochemical Characteristics and Biological Activity of Irradiated Pectin Solution (감마선 조사 펙틴 용액의 이화학적 특성 및 생리활성 변화)

  • Pectin was dissolved in HCl, citric acid, and deionized distilled water (DW, 2%, v/v) and irradiated at different irradiation doses (2.5-50 kGy) by gamma ray to investigate its physicochemical characteristics and biological activity. Viscosity of pectin solution was significantly decreased by irradiation up to 10 kGy, then remained constant thereafter. Gamma-irradiation increased monosaccharide and polysaccharide levels up to 30-40 kDa. Electron donating ability of pectin solution was highest when DW was added and was increased by increasing irradiation dose (p<0.05). ${\beta}-Carotene$ bleaching assay revealed irradiation resulted in development of antioxidantive activity in pectin solution. Growth inhibition of cancer cell lines was observed in irradiated pectin solution in dose-dependent manner, with G36l showing the highest. Results suggested irradiation of pectin solution could be effective for preparation of functional pectin oligomer.

Manufacturing and Physical and Chemical Characteristics of Fruit Leathers Using Flesh and Pomace of Japanese Apricots (Prunus mume Sieb. et Zucc) (매실과육과 매실착즙박을 이용한 Fruit leather의 제조와 그 특성)

  • Fruit leathers were manufactured from flesh and pomace of Japanese apricots and evaluated for their physical, chemical, microbiological and sensory properties. The contents of total dietary fiber(TDF) in Japanese apricot leathers(JAL) and Japanese apricot pomace leathers(JAPL) were 4.06% and 7.82%, respectively. One hundred grams of leather contained 368kcal in JAL and 352kcal in JAPL. Water activities of fruit leathers were 0.36 in JAL and 0.48 in JAPL. None of the factors had an effect on the microbiological counts of any of the organisms. The L, a and b values of the fruit leathers were higher in those made of JAF than those made of JAP. The fruit leathers made of JAF were harder than those made of JAP. Sensory panelists preferred fruit leathers made of JAP to those made of JAF in all attributes, except for their color.

  • The chemical characteristics of flesh and pomace of Japanese apricot were investigated. The moisture contents of flesh and pomace of Japanese apricot were 89.94% and 91.39%. Free sugars and sugar-derivatives of Japanese apricot flesh (JAF), were 0.77% glucose, 0.47% fructose, 0.35% mannitol and 0.47% sorbitol, and of Japanese apricot pomace (JAP) were 0.01% glucose, 0.09% fructose, 0.38% mannitol and 0.06% sorbitol, respectively. The organic acids of flesh and pomace of Japanese apricot were citric acid, malic acid and oxalic acid. The predominant minerals in flesh and pomace of Japanese apricot were K, P, Ca and Al. The contents of dietary fiber (DF) in JAF were 2.94% of insoluble dietary fiber (IDF), 1.07% of soluble dietary fiber (SDF) and 4.01% of total dietary fiber (TDF). IDF of JAP were 6.25%, SDF 0.51% and TDF, 6.76%.
