Structural and Functional Relationship of the Catalytical Subunit of Recombinant Pyruvate Dehydrogenase Phosphatase (rPDPc): Limited Proteolysis (Pyruvate dehydrogenase phosphatase의 catalytical subunit의 구조와 활성에 대한 연구)

  • Pyruvate dehydrogenase phosphatase (PDP)와 kinase는 당대사시 해당과정에서의 대사 산물인 pyruvate를 acetyl CoA로 만들어 구연산 회로로 진입시켜 주는 효소인 pyruvate dehydrogenase complex (PDC)의 활성을 조절하는 중요한 효소이다. PDP의 catalytic subunit는 PDC의 dihydrolipoamide acetyltransferase (E2), PDP regulatory subunit (PDPr), 그리고 칼슘 결합 도메인 등으로 구성되어 있는 것으로 추측되어지고 있다. 본 연구에서는 그 구조와 기능과의 상관관계를 알아보기 위해 PDPc를 E. coli JM101에서 발현시켜 순수 정제 후 단백분해 효소를 이용한 제한적 가수분해 방법을 이용해 그 구조와 기능과의 상관관계에 대해 연구하고자 하였다 정제된 PDPc는 trypsin, chymotrypsin, Arg-C 그리고 elastase를 이용하여 3$0^{\circ}C$ 그리고 pH 7.0에서 제한적으로 분해시켰으며 각 분해산물의 아미노 말단의 아미노산 배열을 분석하였다. 그 결과 PDPc는 trypsin, chymotrypsin, elastase에 의해 N-terminal의 50 kD과 C-terminal의 10 kD의 두개의 분해산물을 만들었으며, Arg-C에 의해 50kD의 분해산물은 약 35kD와 15kD으로 더 가수분해가 되었다. 이러한 결과로 볼 때 PDPc는 앞에서 추측한데로 세개의 주요한 기능적 도메인으로 이루어져 있음을 알 수 있었다 또한 C-terminal의 10kD은 PDPc의 활성에는 영향을 주지 않는 것으로 밝혀졌으나 다른 도메인의 기능은 더 연구가 되어져야 할 것으로 생각된다.

  • There are posters of a tourist and traffics as like railroads and ships among Cassandre's posters, and they are common in dealing with 'travelling'. This series of travelling posters which includes many of his representative works, are important in the point of showing his unique styles and the process how Hey have been changed. In this thesis, I tried to analyze, especially in the plastic and graphic aspects, how Cassandre catch the peoples' desire and dreams of the travelling and images on the mechanical property of railroads and ships in 1920's and 1930's and how he put them into his works. I think that has revealed sense and visual language which has revealed through this analysis still give some messages to advertising media of moving images based on electronic technology today.

  • The purpose of this thesis is to suggest the new orientalism fashion through the comprehensive approach to analyze the art of the Tang dynasty, China. Tang dynasty has high culture, economy, polity and art. Also the character of Tang's culture is very international. Therefore that point is coincide with fusion culture in the early 2000's. As a method of accomplishing this research, the documents and fashion magazines related to the art and fashion were examined. Through analyzing the art in the Tang's related to fusion culture, reach a conclusion that the modern fashion has four characters. 1. The cause of high culture and economy, the trend of fashion was decorative 2. The cause of realism, the trend of fashion were modern and simple. 3. The cause of rhythmical line, the trend of fashion had natural drape. 4. The cause of globalism, the trend of fashion mixed east with west This research keep going for showing new orientalism and coinciding with 21th century's trend.

The Effects of Cinnamomum cassia Blume, Aconitum carmichaeli Debx, and Pueraria lobata Benth on Glucose and Energy Metabolism in C2C12 Myotubes (C2C12 골격근 세포에서 육계, 부자, 갈근 물 추출물의 당대사 및 에너지 조절 효과)

  • Objectives: The prevalence of obesity and metabolic syndrome is increasing worldwide. Regulation of cellular energy metabolis has the potential to be manipulated therapeutically to serve as a target for obesity and insulin resistance. Skeletal muscle is regarded as a target for regulation of energy metabolism and insulin resistance. In this study, the authors investigated the regulatory effect of (Cinnamomum cassia Blume, CCB), Aconitum carmichaeli Debx (ACD), and Benth (Pueraria lobata Benth, PLB) on energy and glucose metabolism in C2C12 myotubes. Methods: The water extracts of CCB, ACD, and PLB (0.5 mg/ml) were treated in differentiated C2C12 myotubes. The expressions of adenosine monophosphate-activated protein kinase (AMPK) and phosphorylation AMPK were detected with western blotting. Glucose metabolism was investigated with glucose uptake assay and glucose consumption assay, total adenosine triphosphate (ATP) content was also analyzed. Results: CCB, ACD, and PLB activated the phophorylation of AMPK, they also increased the glucose metabolism and total ATP contents in C2C12 myotubes. Conclusions: This study suggests that CCB, ACD, and PLB have the potential to increase energy and glucose metabolism in skeletal muscle.

History of Symptom Discrimination based on Theory of Triple Energizers(三焦) before Tang Dynasty(唐代) (당대(唐代) 이전(以前)의 '삼초이론(三焦理論)에 기초한 변증(辨證)'의 연혁(沿革))

  • Objectives : I intended to find out the origin and development of symptom discrimination based on theory of triple energizers before Tang Dynasty(唐代). Methods : I extracted the topic-related contents from books in this era. After arraying them in a chronological order, I analyzed the materials. Results : The author in Zhong Zang Jing(中藏經) first mentioned that symptom of Huo Luan(霍亂) was deeply related with triple energizers. Ge Hong(葛洪) did not only come out with some symptoms and mechanisms for Shang Han(傷寒) and Za Bing(雜病), but also applied them to their usages for herb medicine. Chao Yuan Fang(巢元方) suggested its symptoms were more closely related with Shang Han and Za Bing than before. For the first time, Chao subdivided its symptoms to both cold-heat and weakness-strength sides. Sun Sai Miao(孫思邈) supplemented its discrimination invented by Chao(巢元方), adding more symptoms and prescriptions for triple energizers. Wang Tao(王燾) showed more prescriptions for triple energizers than before, and initially designed the discrimination for Huo Luan, diabetes(消渴), and epidemic Wen Bing(溫病). Conclusions : Before Tang dynasty, the symptom discrimination based on theory of triple energizers had been originated and developed for the purpose of repairing Za Bing, not just treating Wen Bing.

  • The purpose of this article is to review the historical media and reenactment activity which is used in history education. Especially, teachers are very interested in how to develop the interest of the students in class. In this article, we review the characters of the reenactment activity as well as the drama and film. The definition of the reenactment activity is to act like a historical persons. 'Reenactment activity' is not familiar to us in Korea but it is very famous to the students and the teachers who want to re-act the historical life. Reenactment actvitiy is more efficient than movie because doing reenactment is much cheeper than making film.

  • 조선조 세종대는 천문과학기술의 전성기였다. 세종 자신도 천문학에 밝았던 데다 유교적 정치이념을 구현하기 위한 국책 사업의 성격이 강했기 때문에 조선조의 천문학은 세계적 수준으로 발돋움하였다. 이것은 고려시대부터 축적된 천문과학기술과 창조적 재능을 지닌 많은 과학기술자들이 있었기 때문에 가능했다. 1432년 세종이 천문의기 제작을 명한 지 6년만인 1438년에 각종 관측기기를 완비한 천문대인 간의대가 완성되었다. 당대로서는 세계 최대 규모로 인정받고 있는 거대한 종합 천문대인 간의대의 주변에는 혼천의, 혼상, 규표 등 다양한 천문 관측기기를 설치하였다. 규표로는 24기를 정밀하게 측정할 수 있었고, 각종 해시계로는 한양의 정확한 시간을 잴 수 있었다. 해와 별을 관측하여 낮과 밤의 시간을 측정하는 일성정시의와 천체 관측기기인 소간의 등 다양한 관측의기들이 독창적으로 창제되었다. 아쉽게도 세종대에 제작한 천문의기들 가운데 현존하는 유물은 한 점도 남아 있지 않다. 당시 천문유물은 사라져버렸지만 문헌을 통해 세종시대 각종 천문 관측의기의 복원 연구를 통해 설계와 복원을 수행하였다. 천문의기 복원(復元)은 모형(模型)을 만드는 것이 아니므로 자재와 공법들은 전통적인 방식으로 제작하여야 한다. 각 부품들이 계시기(計時器)로서 정확히 작동하도록 복원하려면 정교하게 제작하여야만 한다. 뿐만 아니라 천문의기들은 왕궁(王宮)에서 사용한 것으로 외형적으로 왕권을 상징하는 용(龍)의 형상과 화려한 문양으로 장식되어 있기 때문에 복원 과정에서 예술적인 면도 신중히 고려하여야 한다. 여기서는 지금까지 복원한 조선의 각종 천문의기의 구조와 기능 및 복원 과정의 주요 내용을 소개하고자 한다.

Ethanol Effect of Nelumbo nucifera Root on Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨 흰쥐에 대한 연근 에탄올 추출물의 당대사 효소활성과 항산화 작용에 미치는 영향)

  • This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

The Mixed Herbal Extract, CAPA, Prevents Obesity and Glucose Intolerance in Obese Mice (비만형 동물모델에서 복합 한약 추출물 CAPA의 비만 및 당대사 개선 효능)

  • Objectives: This study investigated the effects of the mixed herbal extract from Cinnamomum cassia Blume, Atractylodes macrocephala Koidzumi, Pueraria lobata Benth, and Aconitum carmichaeli Debx (CAPA) on obesity and glucose tolerance in obese mice. Methods: Animals were divided in 6 groups, normal diet, high fat diet (HFD), HFD with CAPA 100 mg/kg (CAPA 100), CAPA 300 mg/kg (CAPA 300), and metformin 200 mg/kg or lorcaserin 10 mg/kg as positive controls, and treated for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after treatment. The oral glucose tolerance test was also carried out after treatment. Results: Compared to HFD, CAPA extract treated mice showed significant decreases in body weight, adipose tissue weight, lipid accumulations in liver and serum lipid levels without a reduction of food intake. And fasting glucose and glucose tolerance were all improved in the CAPA treated mice. Conclusions: Our results suggest that CAPA extract can prevent diet induced obesity and glucose intolerance without a reduction of energy intake in obese mice.

  • This paper was investigated by comparing "Sok-Mong-Gu Bun-Ju"(續蒙求分註) and "Mong-Gu"(蒙求). As a result, "Mong-Gu" was organized around the person anecdotes, "Sok-Mong-Gu Bun-Ju" is annotated with the person anecdotes. So the text show an differences were found on the configuration. Thus It reveal that "Sok-Mong-Gu Bun-Ju" is a tome and convenient to use the report has been made by scholars can be clarified.
