Prevalence and molecular characterization of novel recombinant enterovirus G isolates in Jeju Province of South Korea

  • Enterovirus species G (EV-G) is highly diverse, and is ubiquitous in pig populations, usually without diarrhea. The present study aimed to investigate the presence of novel EV-G recombinants with the torovirus papain-like cysteine protease (PLCP) in Jeju pig herds. EV-G1-PLCP mono-infections were most prevalent in diarrheic weaned piglets. The PLCP genes of the Jeju isolates varied in size and junction sequences, and were greatly heterogeneous, with 77.0-90.7% homology amongst all recombinants. Our results suggest that the exogenous PLCP gene has undergone continuous rapid mutation in the individual EV-G genomes following cross-order recombination, thereby causing clinical disease in swine.

  • The green algae Scenedesmus acuminatus was cultured in different media: animal wastewater and an artificial culture medium in order to evaluate potential use for tertiary treatment. The experiments were conducted with air flowrate 1~2 L/min at $28{\sim}30^{\circ}C$. The nitrogen and phosphorus showed very similar removal efficiencies (68~77 % and 69~80 % for nitrogen and phosphorus respectively). The optimal fed period was estimated as three days in the semi-continuous experiment. The effects of $CO_2$ (4.5 %) injection on nutrient uptake from animal wastewater (biological treatment effluent) were compared to an air injection under the same conditions of light and photoperiod. The uptake rates of nutrient with air injection were observed 0.009 gN/gChl-a/day, 0.028 gN/gChl-a/day and T-P 0.003 gP/gChl-a/day for nitrate, total nitrogen and phosphorus respectively. The rates were enhanced by addition of $CO_2$ to 0.026 gN/gChl-a/day, 0.076 gN/gChl-a/day and T-P 0.018 gP/gChl-a/day. This study establishes that $CO_2$ addition during nutrient deprivation of microalgal cells may accelerate tertiary wastewater treatment.

Effects of Two Halophytic Plants (Kochia and Atriplex) on Digestibility, Fermentation and Protein Synthesis by Ruminal Microbes Maintained in Continuous Culture

  • Eight continuous culture fermenters were used in a completely randomized design to evaluate various nutritional values of Kochia (Kochia scoparia) compared with Atriplex (Atriplex dimorphostegia). Dried and pelleted samples (leaves and stems) provided substrate for metabolism by ruminal microbes maintained in a continuous culture fermentation system. Results indicated that there were no differences (p>0.05) in dry matter (DM) and crude protein (CP) digestibility between the two halophytic plants. Atriplex had higher (p<0.05) organic matter (OM) digestibility compared with Kochia. Neutral detergent fiber (aNDF) digestibility of Atriplex (411 g/kg) was higher (p<0.05) than that of Kochia (348 g/kg), however acid detergent fiber (ADF) digestibility was higher (p<0.05) in Kochia compared with Atriplex (406 vs. 234 g/kg). There were no differences (p>0.05) between the two halophytic plants in molar proportion of acetate and propionate, but the concentration of butyrate and valerate in Kochia were about two fold of Atriplex (p<0.05). When Kochia provided substrate to the microbes, protein synthesis was higher (p<0.05) compared with feeding Atriplex (5.96 vs. 4.85 g N/kg of OM truly digested). It was concluded that Kochia scoparia and Atriplex dimorphostegia had similar digestibility of DM and CP. It appears that these halophytic plants may not have enough digestible energy for high producing ruminants.

Generation of Discrete $G^1$ Continuous B-spline Ship Hullform Surfaces from Curve Network Using Virtual Iso-parametric Curves

  • Ship hullform is usually designed with a curve network, and smooth hullform surfaces are supposed to be generated by filling in (or interpolating) the curve network with appropriate surface patches. Tensor-product surfaces such as B-spline and $B\'{e}zier$ patches are typical representations to this interpolating problem. However, they have difficulties in representing the surfaces of irregular topological type which are frequently appeared in the fore- and after-body of ship hullform curve network. In this paper, we proposed a method that can automatically generate discrete $G^1$ continuous B-spline surfaces interpolating given curve network of ship hullform. This method consists of three steps. In the first step, given curve network is reorganized to be of two types: boundary curves and reference curves of surface patches. Especially, the boundary curves are specified for their surface patches to be rectangular or triangular topological type that can be represented with tensor-product (or degenerate) B-spline surface patches. In the second step, surface fitting points and cross boundary derivatives are estimated by constructing virtual iso-parametric curves at discrete parameters. In the last step, discrete $G^1$ continuous B-spline surfaces are generated by surface fitting algorithm. Finally, several examples of resulting smooth hullform surfaces generated from the curve network data of actual ship hullform are included to demonstrate the quality of the proposed method.

Continuous Production of Natural Colorant, Betacyanin, by Beta vulgaris L. Hairy Root

  • It has been known that continuous cultivation of hairy root is difficult to maintain for a long period of time compared to the microbial and callus cultures. Chemostat cultivation was successfully carried out in order to economically produce a plant-based colorant, betacyanin, from red beet hairy root for more than 85 days in a 14-1 fermentor. The result from the chemostat cultivation was compared to those of the batch and fed-batch cultivations of red beet hairy roots. It was shown that hairy root reached its steady state within 50 days of the cultivation, and then maintained for about 25-30 days in a wide range of dilution rates. Total betacyanin production from the continuous process was also calculated to be 2.65g at 0.28(l/d) of dilution rate, compared to 0.196g from fed-batch cultivation. It was found that betacyanin production was a partially growth related process, yielding 0.376 mg/g-fresh wt. cell and $1.89{\times}10^{-5}$ mg/g-fresh wt. cell/d, with 0.92 of correlation factor in a partial growth-product model. It was also shown that the cell growth required was relatively large for maintenance amount of energy at a low dilution rate. The growth of hairy root was inhibited by high light intensity in following a photo-inhibition model. The growth parameters were estimated to be 0.3(l/d), $10.56kcal/\textrm{m}^2/h$,{\;}and{\;}35.81kcal/\textrm{m}^2/h$ for the maximum specific growth rate, half saturation light intensity, and inhibition light intensity, respectively.

The Disruption of Saccharomyces cerevisiae Cells and Release of Glucose 6-Phosphate Dehydrogenase (G6PDH) in a Horizontal Dyno Bead Mill Operated in Continuous Recycling Mode

  • Baker's yeast was disrupted in a 1.4-L stainless steel horizontal bead mill under a continuous recycle mode using 0.3 mm diameter zirconia beads as abrasive. A single pass in continuous mode bead mill operation liberates half of the maximally released protein. The maximum total protein release can only be achieved after passaging the cells 5 times through the disruption chamber. The degree of cell disruption was increased with the increase in feeding rate, but the total protein release was highest at the middle range of feeding rate (45 L/h). The total protein release was increased with an increase in biomass concentration from 10 to $50\%$(w/v). However, higher heat dissipation as a result of high viscosity of concentrated biomass led to the denaturation of labile protein such as glucose 6-phosphate dehydrogenase (G6PDH). As a result the highest specific activity of G6PDH was achieved at biomass concentration of $20\%$(ww/v). Generally, the degree of cell disruption and total protein released were increased with an increase in impeller tip speed, but the specific activity of G6PDH was decreased substantially at higher impeller tip speed (14 m/s). Both the degree of cell disruption and total protein release increased, as the bead loading increased from 75 to $85\% (v/v)$. Hence, in order to obtain a higher yield of labile protein such as G6PDH, the yeast cell should not be disrupted at biomass concentration and impeller tip speed higher than $20\%(w/v)$ and 10 m/s, respectively.

Evaluation of L-Lactic Acid Production in Batch, Fed-batch, and Continuous Cultures of Rhizopus sp. MK-96-1196 Using an Airlift Bioreactor

  • Various processes which produce L-lactic acid using ammonia-tolerant mutant strain, Rhizopus sp. MK-96-1196, in a 3L airlift bioreactor were evaluated. When the fed-batch culture was carried out by keeping the glucose concentration at 30g/L, more than 140 g/L of L-lactic acid was produced with a product yield of 83%. In the case of the batch culture with 200g/L of initial glucose concentration, 121g/L of L-lactic acid was obtained but the low product yield based on the amount of glucose consumed. In the case of a continuous culture, 1.5g/L/h of the volumetric productivity with a product yield of 71% was achieved at dilution rate of $0.024\;h^{-1}$. Basis on these results three processes were evaluated by simple variable cost estimation including carbon source, steam, and waste treatment costs. The total variable costs of the fed-batch and continuous cultures were 88% and 140%, respectively, compared to that of batch culture. The fed-batch culture with high L-lactic acid concentration and high product yield decreased variable costs, and was the best-suited for the industrial production of L-lactic acid.

  • The paper is devoted to the study of fractional integration and differentiation on a finite interval [a, b] of the real axis in the frame of Hadamard setting. The constructions under consideration generalize the modified integration $\int_{a}^{x}(t/x)^{\mu}f(t)dt/t$ and the modified differentiation ${\delta}+{\mu}({\delta}=xD,D=d/dx)$ with real $\mu$, being taken n times. Conditions are given for such a Hadamard-type fractional integration operator to be bounded in the space $X^{p}_{c}$(a, b) of Lebesgue measurable functions f on $R_{+}=(0,{\infty})$ such that for c${\in}R=(-{\infty}{\infty})$, in particular in the space $L^{p}(0,{\infty})\;(1{\le}{\le}{\infty})$. The existence almost every where is established for the coorresponding Hadamard-type fractional derivative for a function g(x) such that $x^{p}$g(x) have $\delta$ derivatives up to order n-1 on [a, b] and ${\delta}^{n-1}[x^{\mu}$g(x)] is absolutely continuous on [a, b]. Semigroup and reciprocal properties for the above operators are proved.

고정화균체 반응기에서 L-Sorbose 연속생산

  • The conversion of D-sorbitol to L-sorbose by Gluconobater suboxydans was analyzed, and continuous production of L-sorbose was carried out in immobilized cell reactors. L-Sorbose production by high densities of resting cells was more effective than by conventional batch fermentations. Sorbitol dehydrogenase, an enzyme converting D-sorbitol to L-sorbose, did not suffer from substrate inhibition, but from product inhibition. When L-sorbose production was carried out with Ca-alginate-immobilized cells, about 60 g/l of L-sorbose was obtained. On the other hand, when the corn steep liquor (CSL) concentration of medium was reduced to 0.08%, 80 g/l of L-sorbose was obtained. Outgrowth inside the immobilized carriers was thought to block the pores of the carriers so that substrate could not easily diffuse through the carriers. Continuous production of L-sorbose was well accomplished in a bubble column reactor, and 6. 5 g/l.h of productivity and 81.2% of yield were obtained at a substrate feeding rate of 0.08h$^{-1}$ under the optimum conditions with carrier volume of 55% and aeration rate of 3 vvm.

Production of pullulan by Aureobasidium pullulans HP-2001 with continuous culture

  • In batch culture of A. pullulans HP-2001, maximal production of pullulan was 57.90 g/l at 96 hr. The maximal production of pullulan with 76.55 g/l occurred when substituted solution was 10% (w/v) sucrose, 0.25% (w/v) yeast extract and mineral salts. The maximal production of pullulan was 75.00 g/l with a dilution rate of $0.015\;h^{-1}$. Productivity of pullulan in the continuous culture fed with the fresh medium was higher than that fed with the fresh medium without nitrogen source.
