Neuroprotective Compounds Isolated from Lysimachia christinae

  • We previously reported that dried Lysimachia christinae whole plant extract exerted significant neuroprotective activity. In this study, we tried to isolate neuroprotective compounds of L. christinae. We evaluated the neuroprotective activity of the four fractions (hexane, chloroform, ethyl acetate, and n-butanol fractions) of methanol extract. Among them, ethyl acetate and n-butanol fractions showed most potent neuroprotective activity against glutamate excitotoxicity. Nine compounds were isolated from ethyl acetate and n-butanol fractions of L. christinae extract and identified as cynaroside (1), (3,4,5,6-tetrahydroxytetrahydro-2H-pyran-2-yl)methyl-3-hydroxy-2-octyldopentaconta-23,33-dienoate (2), androst-16-ene-3,6-diol (3), 2-hydroxy-24-propoxytetracos-4-enoic acid (4), 2-hydroxy-24-methoxytetracos-4-enoic acid (5), 12-(stearoyloxy)octadec-9-enoic acid (6), β-sitosterol (7), (E)-4-(3,4-dimethoxyphenyl)but-3-en-1-yl palmitate (8) and (1S,2S,3R,4R)-4-(((2S,3R,4R,5R,6S)-2-(((2R,3R,4S,5R,6R)-2-(3,4-dimethoxyphenethoxy)-3,5-dihydroxy-6-(hydroxymethyl) tetrahydro-2H-pyran-4-yl)oxy)-4,5-dihydroxy-6-methyltetrahydro-2H-pyran-3-yl)oxy)cyclohexane-1,2,3-triol (9) by spectroscopic data such as UV, IR, NMR, Mass spectroscopy. Their neuroprotective activity was evaluated by MTT assay. Cynaroside (1) and androst-16-ene-3,6-diol (3) had significant neuroprotective activity against glutamate-injured HT22 cells. The neuroprotective efficacy of cynaroside (1) and androst-16-ene-3,6-diol (3) was related to their anti-oxidative activity.

The Clinical Analysis of Ventricular Septal Defect Review of 97 cases (심실중격결손증의 임상적 고찰)

  • From 1977 through 1984, 97 patients of V.S.D. were treated surgically at Department of Thoracic and Cardiovascular Surgery, Korea University college of Medicine. Among 97 patients, 3 patients were treated by PDA ligation, 3 patients were treated by PDA ligation and subclavian flap aortoplasty, 1 patient were treated by pulmonary artery banding. All of the above patients were analyzed clinically. The results were as follows; 1. Of the patients, 52 patients were male [55.5%] and 45 patients were female [44.5%]. Their age ranged from 7 days to 32 year, and the mean age was 9 year of age and 28.9% of patients were between 4 and 8 year of age. 2. The most common clinical symptoms were frequent U.R.l. and D.O.E.. 3. The most common chest PA findings were cardiomegaly and increased pulmonary vascularity. 4. Associated anomaly was founded in 27 cases and PDA was most common associated anomaly and others were A.S.D., pulmonary stenosis, aortic regurgitation, D.C.R.V.. 5. Pulmonary hypertension was founded in 37 patients and it`s incidence was increased by patient age and shunt amount. 6. On Kirklin`s anatomical classification, type 11 defect was most common [45.5%], and type 1 was 35.5%, and type 111 was 4.4%, and type 1V was 4.4%. 7. Mean E.C.C. time was 69.1 min. and varied by closing method and associated anomaly as in case of simple closure; 47.8 min., in case of patch closure; 77.2 min., in cases with associated anomaly; 92.7 min.. 8. Mean postoperative ventilatory assisted time was 7.3 hour and varied by preoperative pulmonary artery pressure and E.C.C. time, as the group with pulmonary hypertension; 10.5 hour, the group without pulmonary hypertension; 5.5 hour, the group of short E.C.C time [within 1 hour]; 4.4 hour, the group of long E.C.C. time [over 1 hour]; 8.4 hour. 9. Overall operative mortality was 9.3% [9 cases].

Shoot yield and growth characteristics of Pteridium spp. according to the dickness of sowed seed root (고사리의 종근 규격에 따른 파종 2년차 신초 출현양상 및 수량성)

  • 고사리(Ferns)는 고사리 속(Pteiidium spp.)에 속하는 양치식물의 총칭으로서 우리나라에는 22과 70속 272종이 분포되어 있는 것으로 보고되었다. 고사리의 어린 순에는 가식부 100g 당 칼슘 15.0mg, 칼륨 185.0mg 등이 함유되어 있으며 골다공증, 심혈관질환 등에 효과가 있고 식이섬유로 인해 변비 예방에도 유용한 것으로 알려져 있다. 2017년 기준 우리나라의 고사리 재배면적은 3,280ha 수준으로 그중 전라북도의 고사리 재배는 재배면적 285ha이며 연간 생산량 1,985톤으로 전국 생산량의 21%를 차지하고 있어 전라북도의 산채 재배작목 중 중요한 비중을 차지하고 있다. 고사리 재배는 정식 후 수확기간이 길고 제초노력 등의 노동력 의존도가 상대적으로 낮아 최근 재배면적이 늘어나고 있는 추세이나, 종근의 적정 규격 등에 대한 기준이 제시되지 않아 경영비 부담이 큰 실정이다. 본 연구는 고사리 종근 규격을 종근 두께 기준 A등급($14.1{\pm}3.31mm$), B등급($8.3{\pm}1.88mm$), C등급($3.3{\pm}2.31mm$)로 구분하여 표고 500m의 허브시험장 연구포장에 3월 28일 정식한 후 2018년 4월부터 신초 출현양상 및 수량성을 조사하였다. 파종 2년차 고사리 신초의 출현양상은 4월 11일부터 출현을 시작하여 6월 11일 까지 빠르게 증가하여 7월 9일 이후 고온기이후 증가세가 완만해지는 경향을 보였고 9월 10일까지 누적 신초수는 A 등급 파종구에서 80.0개를 보였다. 신초 출현 후 포자엽이 전개되어 상품성을 상실하기까지의 포자엽 전개일수는 주 수확시기인 4~5월에는 5.3~6.7일이 소요되었으나, 포자엽 전개이후 울폐가 이루어지는 8~9월에는 9.7~11.7일이 소요되어 생육 초기 차광에 의한 전개일수 연장효과가 기대되었다. 생육기간 동안 조사시기별 신초의 두께는 6월 11일과 9월 11일 조사한 신초에서 굵어지는 경향을 보였고, 상품성 있는 고사리 신초의 규격을 신초장 20cm로 포자엽 전개 직전으로 한정하여 수확을 실시한 결과 파종 2년차 수확은 4월18일 이후 4회 이루어졌으며 신초의 생체중은 5.2~6.0g/개의 범위로 종근 규격 간에는 차이가 인정 되지 않았다. 4월 18일에서 5월 8일까지의 수확기간 동안 누적 신초 수확량은 A 등급과 B 등급의 종근을 파종한 처리에서 각 32.2개/$m^2$, 30.3개/$m^2$로 C 등급 파종구에 비해 유의하게 수확 신초수가 증가하였다. 단위면적당 신초 수량성은 4월 30일 3회차 수확시부터 종근 규격간 차이가 있었으며 누적 신초수량은 A 등급과 B 등급 파종구에서 각 183.3kg/10a, 169.9kg/10a로 유의하게 높았는데 이는 파종된 종근의 두께에 따라 상품성 있는 신초 출현수가 유의하게 증가된 때문으로 판단되었다.

  • 우리나라의 고사리 재배면적은 2018년 기준 5,406ha 수준을 보이고 있으며 전체 산채류 재배면적에서 가장 높은 비중을 차지하고 있다. 고사리(Ferns)는 고사리 속(Pteridium spp.)에 속하는 양치식물의 총칭으로서 우리나라에는 22과 70속 272종이 분포되어 있는 것으로 보고되었다. 고사리의 어린 순에는 가식부 100g 당 칼슘 15.0mg, 칼륨 185.0mg 등이 함유되어 있으며 골다공증, 심혈관 질환 등에 효과가 있고 식이섬유로 인해 변비 예방에도 유용한 것으로 알려져 있다. 고사리 뿌리에서 추출한 전분은 중국 및 일본 등지에서 면류나 제과용으로 이용되고 있어 고사리 재배의 부가가치를 향상시킬 수 있는 방안으로 기대되고 있다. 본 연구에서는 고사리의 재배년차간에 뿌리 전분의 생산성과 이화학적 특성을 조사하기 위해 2020년 3월 26일 각 재배년차별로 굴취한 뿌리를 세척 후 고무망치로 파쇄하고 수침 방법을 사용해 추출한 전분의 이화학적 특성을 조사하였다. 재배년차간 고사리 뿌리의 생산성은 5년생과 7년생에서 3,000 kg/10a 이상으로 유의하게 높은 수량을 보였다. 세척한 고사리 뿌리를 24시간 sodiun metasulfite 4% V/W 용액에 수침하여 전분을 추출한 후 24시간 간격으로 3회에 걸쳐 진탕, 침전과 세척의 과정을 거친 후 조사한 전분 추출 수율은 뿌리 생체중 대비 6.8~7.6% 수준이었으며 재배년차간 차이는 없었다. 추출한 전분의 색도분석에서 L 값은 재배년차가 높아질수록 낮아지는 경향을 보였으나 a 값은 3.7~4.8, b 값은 13.5~15.9로서 뚜렷한 차이를 보이지 않았다. 고사리 뿌리전분의 호화특성에서 최고점도는 3년생 고사리 뿌리에서 추출한 전분이 557.4 RVU로 높은 값을 보였으나 재배년차간 통계적 유의성을 인정할 수는 없었으며, 최저검도에서는 5년생에서 추출한 전분이 269.9RVUfh 낮은 경향을 보였다. 또한 최대 점도 시간은 5.0~5.2hr로 재배년차간 차이는 없었으며 호화 저지온도는 67.0~68.9℃ 범위로 유의한 차이를 보이지 않았다. 고사리 뿌리에서 채취한 전분의 무기물 함량을 조사한 결과 황의 함량은 1년생에서 0.081%로 유의하게 높았으며, 인산 함량은 3년생에서 72mg/kg으로 가장 높았고 1년생에서 31mg/kg으로 가장 낮은 값을 나타냈다. 칼륨의 함량은 7년생에서 추출한 전분에서 376.3mg/kg으로 높았으나 재배년차 간 뚜렷한 경향을 보이지 않았으며 칼슘, 마그네슙 및 철분 함량에서는 5년생 전분에서 유의하게 높은 경향을 보였다.

  • This study was performed to determine the optimal hydrolysis conditions for the production of a flavoring from the precipitation of snow crab cooker effluent (PSCCE) with commercial proteases. Based on cost-per-enzyme activity and sensory evaluations, Flavourzyme$^{(R)}$ 500 MG plus Protamex$^{(R)}$ (1:1 ratio, w/w) were selected as suitable enzymes. Three independent variables consisting of the substrate concentration (S), enzyme-to-substrate ratio (E/S), and hydrolysis time (T) were examined using response surface methodology (RSM). A model equation obtained from RSM was used to predict the degree of hydrolysis (DH) as follows: % DH = 52.285 - 6.371[S] + 5.469[E/S] + 7.599[T] - $5.818[S]^2$ - $5.633[E/S]^2$ - $6.528[T]^2$ - 3.265[E/S][S] - 5.415[T][S] + 4.315[T][E/S]. From the ridge analysis, the conditions favoring the highest degree of hydrolysis were pH 7.45, $55^{\circ}C$, a S of 21.82%, an E/S of 0.50%, and a T of 3.74 h.

Intratypic Variants of HPV-16 E6jE7 Oncogene Isolated from Sexually High-Risk Women in Busan. (부산지역 유흥업소 종사여성으로부터 분리된 HPV16형의 발암유전자(E6/E7) 돌연변이 유형 분석)

  • Recent studies have reported that the distribution of HPV-16 sequence variation differs geographically, and more specifically that HPV-16 E6/E7 intratypic variants might carry a high risk for development of ICC (invasive cervical cancer) and CIN (cervical intraepithelial neoplasia) in a given population. To investigate the genetic diversities of HPV-16 E6/E7 oncogene by region, we collected nineteen HPV-16 isolates from sexually high-risk women in Busan, and analyzed the HPV-16 E6/E7 coding regions (nt 34 to 880) with HPV-16 E6/E7 specific PCR amplification. At the nucleotide levet eleven variants of the E6 genes and nine variants of the E7 genes were identified as follows: E6 T178G (n=l1), E6 T178A (n=l), E6 T350G (n=3), E6 A442C (n=2), E6 AI04T, E6 All1G, E6 C116T, E6 G145T, E6 T183G, E6 C335T, E6 G522C and E7 A647G (n=12), E7 A645C, E7 A777C, E7 G663A, E7 T732C, E7 T760C, E7 A775T, E7 T789C and E7 T795G, respectively. At the amino acid levet the isolated HPV-16 E6 and E7 genes showed eleven E6 variants: E6 D25E (n=12), E6 L83V (n=4), E6 E113D (n=2), E6 MIL, E6 Q3R, E6 P5S, E6 Q14H, E6 D25N, E6 127R, E6 H78Y, E6 C140S and three E7 variants: N29S (n=12), L28F, T72S. HPV16 E6 L83V, the dominant variant in the Caucasian population, showed relatively low frequencies in our study population. We elucidated that the dominant HPV-16 E6/E7 variants were HPV-16 E6 D25E (63.2%) and HPV-16 E7 N29S (63.2%), which were phylogenetically included in Asian lineage. Further study is needed to evaluate the risk of cervical cancer related HPV-16 E6/E7 intratypic variants in the Korean population.

Treatment of Bunionette Deformity with S.E.R.I. (simple, effective, rapid, inexpensive) Operation (S.E.R.I. 수술법을 이용한 소건막류의 치료)

  • Purpose: The purpose of this study was to evaluate the clinical and radiological outcomes of the S.E.R.I. (simple, effective, rapid, inexpensive) operation for the bunionette deformity. Materials and Methods: Between March 2005 and February 2009, 22 patients (26 feet) who had been treated for the bunionette deformity with minimally invasive osteotomy were reviewed retrospectively. Clinically, Visual Analogue Scale (VAS), American Orthopaedic Foot and Ankle Society (AOFAS) score, shoes selectivity, disappearance of callus and patient's satisfaction level by Coughlin scoring system were evaluated. Radiologically, the bunionette was classified as four types according to the Fallat classification. The 4-5$^{th}$ intermetatarsal angle (4-5$^{th}$ IMA), the 5$^{th}$ metatarsophalangeal angle (5$^{th}$ MPA) and the length of 5th metatarsal bone (5$^{th}$ MTL) were analyzed at preoperatively and at final follow up visit. Results: VAS improved from $6.8{\pm}1.8$ points to $2.2{\pm}1.8$ points (p<0.05). AOFAS score improved from $54.0{\pm}14.2$ points to $90.0{\pm}4.8$ points (p<0.05). There was no change in shoes selectivity. 9 feet (34.6%) were satisfied with excellent results, 16 feet (61.5%) with good results and 1 foot (3.9%) with fair results. The average 4-5$^{th}$ IMA was corrected from $10.1{\pm}2.3^{\circ}$ to $4.4{\pm}1.7^{\circ}$ (p<0.05). The average 5$^{th}$ MPA was corrected from $11.5{\pm}8.6^{\circ}$ to $-0.1{\pm}4.1^{\circ}$ (p<0.05). The average 5$^{th}$ MTL was changed from $66.1{\pm}4.3$ millimeters to $64.1{\pm}4.4$ millimeters (p=0.069). There was no malunion, nonunion or delayed union and other perioperative complications. Conclusion: S.E.R.I. operation is less invasive and easy technique. This procedure is recommendable for the treatment of the bunionette deformity.

Age and Growth of the Yellow Goosefish, Lophius litulon (한국근해 황아귀, Lophius litulon의 연령과 성장)

  • Age and growth of the yellow goosefish, Lophius litulon, were studied using samples collected from the southwestern waters of Korea. Vertebrae of the fish had relatively clear annuli on their surface. The opaque zone of vertebrae was formed once a year between March and April. The oldest fish observed in this study was 8 years old for females and 5 years old for males. The relationship between the radius (R) of vertebral centrum and total length (L) was as follows: L=12.7+4.8R for females and L=9.8+5.6R for males. The relationship between total length and body weight (W) was as follows : $W=0.0089L^{3.0311}$ for females and $W=0.0329L^{2.7752}$ for males. Growth in length of the fish was expressed by the von Bertalanffy's equation as $L_t=127.60(1-e^{-0.1228(t+0.3851)})$ for females and $L_t=82.23(1-e^{-0.1832(t+0.6431)})$ for males.

Smile Rearrangement of Herbicidal Flazasulfuron (제초성 Flazasulfuron의 Smile 자리옮김 반응)

  • A series of the herbicidal pyridylsulfonyl areas, none substitutent, 1-(4,6-dimethoxypyrimidine-2-yl)-3-(2-pyridylsulfonyl) urea, 3 and 3-trifluoromethyl substitutent, 1-(4,6-dimethoxypyrimidine-2-yl)-3-(3-trifluoromethyl-2-pyridylsulfonyl) urea, 5(Flazasulfuron) were synthesizied and the rate of hydrolysis of their has been studied in 25%(v/v) aqueous dioxane at $45^{\circ}C$. From the results of solvent effect($m{\ll}1,\;n{\ll}3\;&\;{\mid}m{\mid}{\ll}{\mid}{\ell}{\mid}$), thermodynamic parameter (${\Delta}S^{\neq}=0.54{\sim}\;-2.19\;e.u.\;&\;{\Delta}H^{\neq}=0.025\;Kcal.mol.^{-1}$), hydrolysis product analysis, $pK_a$ constant(3: 4.9 & 5: lit.4.6) and the rate equation, a marked difference in the kinetics of the reaction of 3 and 5(Flazasulfuron) was observed. It may be concluded that the hydrolysis of 5 proceeds through the $A-S_N2Ar$ reaction via conjugate acid$(5H^+)$ below pH 7.0, whereas, above pH 9.0, the hydrolysis proceeds through irreversibly $(E_1)_{anion}$ and reversibly $(E_1CB)_R$ mechanism via conjugate base(CB), respectively. But in case of 5, $A-S_N2Ar,\;(E_1)anion\;and\;(E_1CB)_R$ mechanism involved Smile rearrangement. The mate of rearrangement of 5 to a 3-trifluoromethyl-2-pyridylpyrimidinyl urea(PPU) in acid and 3-trifluoromethyl-2-pyridyl-4.6-dimethoxypyridinyl amine (PPA) in base was increased about 3.5 times by the introduction of trifluoromethyl group in the 3-position on the 2-pyridyl ring. From the basis of these findings, a possible mechanism for the hydrolysis of 5 was proposed and discussed.
