탄닌화합물의 Tyrosinase 억제 활성 (Inhibitory Effects of Tannins on Tyrosinase Activity)

  • For the use of tannins in the whitening-effect cosmetics, inhibitory effect against tyrosinase activity was determined. Three condensed tannins including gallocatechin, gallocatechin 3',4'-di-O-gallate and epicatechin 3-O-gallate and three hydrolyzable tannins, 1,2,6-tri-O-galloyl-${\beta}$-D-glucose, 2,3-(S)-HHDP-D-glucose and pedunculagin showed 15-29% mild inhibitory effects against tyrosinase activity.

  • In the course of searching for BACE1 (${\beta}$-secretase) inhibitors from natural products, the ethyl acetate soluble fraction of green tea, which was suspected to be rich in catechin content, showed potent inhibitory activity. (-)-Epigallocatechin gallate, (-)-epicatechin gallate, and (-)-gallocatechin gallate ware isolated with IC$\_$50/ values of 1.6${\times}$10$\^$-6/ M, 4.5${\times}$10$\^$-6/ M, and 1.8${\times}$10$\^$-6/ M, respectively. Seven additional authentic catechins were tested for a fundamental structure-activity relationship. (-)-Catechin gallate, (-)-gallocatechin, and (-)-epigallocatechin significantly inhibited BACE1 activity with IC$\_$50/ values of 6.0${\times}$10$\^$-6/ M, 2.5${\times}$10$\^$-6/ M, and 2.4${\times}$10$\^$-6/ M, respectively. 비자 열수 추출물의 항산화 활성 및 뇌신경세포 보호효과 연구 (The Antioxidant Activities and Neuroprotective Effects of Hot Water Extracts from Torreyae Semen)

  • Objectives : This study was designed to estimate the antioxidative and neuroprotective effects of Torreyae Semen hot water extracts (TS). Methods : Torreyae Semen was extracted by hot water for 2 hours with a temperature of 105 degrees. Polyphenols and total flavonoid were measured and LC-MS/MS was used to certificate anticipated antioxidative compounds. The antioxidant activities of TS were measured as scavenging effects of 1,1-Diphenyl-2-picrylhydrazyl (DPPH) and Nitrite Oxides (NO). Cell viability and proliferation rate was measured MTT assay. The toxicities to thymocytes and splenocytes were evaluated by the proliferation rate of primary cultured cells of 7 weeks, male Balb/c mice. The antioxidant activities of TS on C6 mouse glioma cells were measured by the analysis of total glutathione contents variation. The neuroprotective effects against oxidative stresses were measured by MTT assay. Results : Polyphenols of TS was $92.00{\pm}1.24{\mu}g/mg$, and total flavonoids was $0.36{\pm}0.14{\mu}g/mg$. TS includes gallocatechin, epigallocatechin, gallocatechin gallate and epigallocatechin gallate. TS included gallocatechin, epigallocatechin, gallocatechin gallate, epigallocatechin gallate. TS showed DPPH and NO scavenging effects as dose-dependent manner at the concentrations of $0-10mg/m{\ell}$. In MTT assay, TS shows no significant toxicity to C6 cells, primary cultured thymocytes and splenocytes of Balb/c mice. TS increased the level of total glutathiones. TS increased cell viabilities of C6 cells against oxidative stresses such as $H_2O_2$, sodium nitroprusside (SNP), Rotenone at the concentrations of $0-0.063mg/m{\ell}$. Conclusions : TS shows the antioxidant and neuroprotecitive effects in these experiments.

갈로카테킨-3-갈레이트가 풍부한 열전환 카테킨의 피부 장벽 회복에 대한 개선 효과 (Effect of Heat-epimerized-catechin-mixture Rich in Gallocatechin-3-gallate on Skin Barrier Recovery)

  • 지금까지 (-)-epigallocatechin-3-gallate(EGCG)는 인간의 피부에 유용한 녹차 카테킨 중에서 가장 강력한 항산화 성분으로 알려져 왔다. 본 연구팀은 용매, 온도, 압력 등 다양한 조건을 변화시키며, 멸균과정(autoclaving) 중에 발생하는 이성질체화(epimerization) 과정을 연구하여, gallocatechin-3-gallate(GCG) 함량이 크게 증가된 열전환-EGCG-복합체(heat-epimerzied-EGCG-mixture, HE-EGCG-mix)를 순수한 EGCG로부터 조제 하였다. 이러한 열전환-EGCG-복합체는 무모쥐 SKH-1을 이용한 실험에서, 손상된 피부 장벽의 회복 시에 인보루크린 7(involucrin 7) 단백질의 발현량을 EGCG 처리 시보다 증가시킴을 확인하였다. 또한, in vitro 실험을 통하여 GCG는 $PPAR-{\alpha}$에 대한 전이활성(transactivation) 효과가 EGCG보다 뛰어남을 확인하였다. 이러한 결과는 열전환-EGCG-복합체에 함유된 고함량의 GCG 성분에 의해서, 피부 장벽 손상 회복 시 PPAR에 의해 매개된 각질형성세포(keratinocyte)의 분화가 더욱 촉진될 수 있음을 암시한다. 따라서, EGCG의 C-2 에피머(epimer)인 GCG는 녹차 카테킨을 이용한 피부 장벽 개선 용도의 화장품과 건강식품 개발 시 주요 소재로 활용될 수 있다.

Gallocatechin Gallate Inhibits Platelet Aggregation by Arachidonic Acid Liberation and $TxA_2$ Synthase Activity

  • We have previously reported that green tea catechins (GTC) displayed anti-thrombotic activity, and that this might be due to anti-platelet rather than anti-coagulation effects. In the present study, we have studied the anti-platelet activity and mechanism of gallocatechin gallate (GCG), which is a component of GTC. GCG inhibited the collagen- and U46619-induced aggregation of rabbit platelets, with IC$\^$50/ values of 63.0 and 48.3 ${\mu}$M, respectively. GCG also inhibited collagen-induced serotonin release and TxB$_2$ formation in a similar manner of platelets aggregation. (omitted)

Prolyl Endopeptidase Inhibitors from Green Tea

  • Three prolyl endopeptidase (PEP) inhibitors were isolated from the methanolic extract of green tea leaves. They were identified as (-)-epigallocatechin gallate, (-)-epicatechin gallate, and (+)-gallucatechin gallate with the $IC_{50}$ values of 1.42${\times}$$10^{-4}$mM, $1.02{\times}10^{-2}$mM, and $1.09{\times}10^{-4}$mM, respectively. They were non-competitive with a substrate in Dixon plots and did not show any significant effects against other serine proteases such as elastase, trypsin, and chymotrypsin, suggesting that they were relatively specific inhibitors against PER The isolated compounds are expected to be useful for preventing and curing of Alzheimer's disease.

녹차 페놀류가 corn oil-in-water emulsion의 산화 중 hydroperoxide 생성에 미치는 영향 (Effect of Polyphenolic Compounds from Green Tea Leaves on Production of Hydroperoxide for Lipid Oxidation in Corn Oil-in-Water Emulsion)

  • 5% corn oil과 17 mM Brij 700으로 제조한 corn oil-in-water emulsion(O/W)의 droplet의 크기는 첨가되는 phenol의 양이 많아질수록 지방구가 다소 커지는 경향을 보였으며, continuous phase의 surfactant 양은 대조구에 비해 다소 낮아졌다. O/W에 녹차로부터 분리한 phenol류를 100-200 ppm의 농도로 첨가하고 30일간 저장하며 hydroperoxide의 생성량을 측정한 결과 대조구에 비해 낮은 hydroperoxide 함량을 보여 유도기간이 길어지는 것으로 나타났으며, phenol 종류별 hydroperoxide 생성억제 효과는 BHT > procyanidin B3-3-O-gallate>(+)-gallocatechin>(+)-catechin의 순서로 나타났다. 지방 산화의 결과 발생하는 aldehyde의 함량을 측정함으로서 산화정도를 측정하기 위하여 headspace hexanal 양을 측정한 결과, 대조구는 15일까지 hexanal이 검출되지 않았으나 phenol 첨가구는 20-30일로 headspace aldehyde의 불검출기간이 길어졌으며, 농도가 증가할수록 낮은 함량을 나타내었다. Phenol 종류별 headspacealdehyde 생성 억제 효과는 BHT > procyanidin B3-3-O-gallate > (+)-gallocatechin >(+)-catechin의 순서로 나타났다.

국내산 참나무속 수종 잎의 추출성분 (Chemical Constituents of Domestic Quercus spp. Leaves)

  • 국내산 주요 참나무류인 신갈나무, 상수리나무, 떡갈나무, 졸참나무, 갈참나무 및 상수리나무 잎의 추출성분의 구조를 규명하고 수종 상호간의 성분의 특성 및 상호 연관성 등을 조사하였다. 신갈나무에서는 gallic acid, (+)-catechin, (-)-epicatechin, (+)-gallocatechin, kaempferol, a stragalin, astragalin-6"-O-gallate, isoquercirin, isoquercitrin-6"-O-gallate 및 myricetin 등 10종의 화합물이 단리되었고 상수리나무에서는 gallic acid, kaempferol과 quercetin이 떡갈나무에서는 gallic acid, (+)-catechin, (-)-epicatechin, (+)-galocatechin, (-)-epigallocatechin, kaempferol, quercetin, guajaverin 및 tamarixin 등 9종의 화합물이 단리되었으며 갈참나무에서는 gallic acid, caffeic acid, astragalin, quercetin과 isoquercitrin이 졸참나무에서는 gallic acid, (+)-catechin, (-)-epicatechin, kaempferol, quercitrin, isoquercitrin 및 myricetin 등 7종의 화합물이 단리되었고 굴참나무에서는 gallic acid, (+)-catechin, astragalin, astagalin-6"-O-gallate와 isoquercitrin 등 5종의 화합물이 단리되었다. Gallic aicd는 모든 수종에서 단리되어 참나무류 잎 추출성분의 지표 화합물로 이용될 수 있다.

친환경 버섯배지에 녹차 잔류물의 처리가 새송이버섯의 생장 및 Catechin류 함량에 미치는 영향 (Effects of Green Tea Residue Treatment in Eco-Friendly Medium on Growth and Catechin Content of Pleurotus eryngii)

  • The present study was conducted to investigate the translocation of polyphenols, especially catechin derivatives, from mushroom medium mixed with green tea residues into fruiting body of Pleurotus eryngii. Pleurotus eryngii was grown on the media incorporated by mixing or surface-treated with dry materials including leaf petioles and young stems or leaves of green tea. The dry materials treated in medium did not affect plant height and fresh weight of Pleurotus eryngii body. From the samples of Pleurotus eryngii, the eight main catechin derivatives (-)-gallocatechin(GC), (+)-catechin (C), (-)-epicatechin (EC), (-)-epigallocatechin (EGC), (-)-epigallocatechin gallate (EGCG), (-)-gallocatechin gallate (GCG), (-)-epicatechin gallate (ECG), and (-)-catechin gallate (EGCG), and caffeine were analyzed quantitatively by HPLC. The results showed that EGC in Pleurotus eryngii was 45% more detected, when incorporated with the dry materials, than untreated control. Especially, content of EGCG was increased in surface-treated Pleurotus eryngii up to 3.2 ppm, while it was not detected or reduced in control and other treatments. Caffeine content was greatly increased regardless of treatment method, compared with control (0.1ppm), showing 44 fold-amount in Pleurotus eryngii at early growth stage when incorporated with the dry materials into medium. The results indicates that functional catechin derivatives of green tea would be partly translocated into Pleurotus eryngii throught incorporation and surface treatment with residues of green tea plants.

보성산 녹차의 채엽시기에 따른 화학 성분의 변화 (Changes of Some Chemical Compounds of Korean (Posong) Green Tea according to Harvest Periods)

  • 국내산 녹차의 채엽시기가 녹차 중의 카테킨, 카페인, 아미노산 및 무기질 함량과 조성에 미치는 영향에 대하여 조사하였다. 녹차의 총 카테킨 함량은 40.61-52.04mg/g으로 채엽시기가 늦을수록 증가하였으며, 카테킨 조성은 채엽시기에 관계없이 EGC, EGCg, EC, CC, ECg, C, GCg 및 Cg의 순으로 함유되어 있음을 알 수 있었다. 카테킨류 중 기능성 작용이 우수한 EGC 및 EGCg는 전체의 50% 이상을 차지하였고 온화한 떫은맛으로 후미에 감미를 남기는 유리형 카테킨인 EC와 EGC는 전체의 53.21%에서 57.09%를 차지하였다. 카페인 함량은 4월차 17.56mg/g, 5월차 15.71mg/g, 6월차 14.92mg/g 및 7월차 14.61mg/g으로 채엽시기가 늦을수록 감소하는 경향을 나타냈다. 총 아미노산 함량은 4월차 45.84mg/g, 5월차 22.64mg/g, 6월차 12.22mg/g 및 7월차 10.45mg/g으로 채엽시기가 늦을수록 감소하였으며 특히, 중요한 생리적 기능을 갖는 theanine의 감소 폭이 큰 것으로 타났다. 무기질 함량은 채엽시기가 늦어질수록 Fe, Mn 및 Mg는 증가하였으며, Cu는 감소함을 알 수 있었다.
