Effect of Phospholipase D on the L-$\alpha$-Dimyristoyl-phosphatidyl Choline Liposome Containing Cholesterol, L-$\alpha$-Phosphatidylinositol and L-$\alpha$-Phosphatidylserine (Cholesterol, L-$\alpha$-Phosphatidylinositol, L-$\alpha$-Phosphatidylserine을 함유한 L-$\alpha$-Dimyristoyl-phosphatidyl Choline 리포솜에 대한 Phospholipase D의 작용에 관한 연구)

  • When the reaction rate constant k of phospholipase D on liposome was measured by the ANS fluorometry, k of phospholipase D on DMPC liposome which was made of L-$\alpha$-PI, cholesterol and L-$\alpha$-PS decreased than that of phospholipase D on DMPC liposome with cholesterol or with PI and cholesterol. Optimal $Ca^{2+}$ concentration, the most important factor on effect of phospholipase D, also decreased to 1mM, as compared with 10mM and 60mM respectively when cholesterol and PI were added, and cholesterol only was added. The change of cholesterol Mol% had a great influence on k value of phospholipase D. But in case of addition of L-$\alpha$-PS to cholesterol, the influence was relatively diminished.

Characterization of Thermal Products of Alpha-Tocopherol

  • Alpha-tocopherol was thermally oxidized and degraded at high temperatures and the resulting products were chromatographically separated and identified by LC-MS. Alpha-tocopherol dissolved in glycerol was heated at 200^{\circ}C for 30 min. The thermal products were separated by hexane extraction and analyzed by HPLC using a reversed phase $\mu$-Bondapak $C_{18}-column$ with two kinds of elution solvents: a mixture of acetonitrile and methanol (3:2), and of acetonitrile, methanol, 2-propanol, chloroform and methylene chloride (3:2:5:0.5:0.5) in a gradient mode. The isolated thermal. products of alpha-tocopherol were more viscous than alpha-tocopherol, and dark brown in color. Major thermal degradation products of alpha-tocopherol were identified by LC-MS, and the structures of thermal products were proposed. Alpha-tocopherol and its thermal degradation products were degraded into fragments, mainly at the non-aromatic parts. The degradation products of alpha-tocopherol. were combined with oxidized product (tocopherylquinone) to make thermal. products through dimerization.

  • The inhibitor had the inhibitory activities against hydrolysis of PNPG, sucrose and ONPG by $\alpha$-Dglucosidase, $\alpha$ - and $\beta$ -galactosidase, but it did not inhibit amylases and other carbohydrases. Kinetic studies exhibited that the inhibitory substance non-competitively inhibited the enzyme reaction with a Ki value of 118 $\mu$g/m$\ell$, and enzyme-inhibitor complex was formed slowly.

Carcinogenic Role of Tumor Necrosis Factor-α Inducing Protein of Helicobacter pylori in Human Stomach

  • Helicobacter pylori is the definitive carcinogen for stomach cancer and is known to induce proinflammatory cytokines, such as tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and interleukin-1(IL-1) in the stomach. Based on our findings that TNF-$\alpha$ is an endogenous tumor promoter, we identified the TNF-$\alpha$ inducing protein (Tip$\alpha$) gene family, and confirmed Tip$\alpha$ and HP-MP1 as new carcinogenic proteins of H. pylori. Tip$\alpha$ protein is unique to H. pylori, and this paper shows the strong tumor promoting activity of Tip$\alpha$ gene family, in cooperation with Ras protein and its mechanisms of action in relation to NF-${\kappa}B$ activation, and discusses the carcinogenic role of Tip$\alpha$ in stomach cancer. Our recent finding showing that penicillin-binding proteins of other bacteria are weak homologues of Tip$\alpha$ is also discussed.

  • ${\alpha}$-L-Arabinofuranosidase was purified from the culture supernatant of Bacillus sp. DSNC 101. The enzyme had a molecular weight of 56 kDa. Optimum temperature and pH for ${\alpha}$-L-arabinofuranosidase activity were 55$^{\circ}C$ and 7.0 respectively. The Michaelis constant(Km) and maximal reaction velo-city(Vmax) for p-nitrophenyl-${\alpha}$-L-arabinofuranoside were 1.0 mM and 113.6 U/mg protein, respe-ctively. ${\alpha}$-L-Arabinofuranosidase was completely inhibited by HgCl$_2$ and CuSO$_4$. The enzyme was spe-cific for the ${\alpha}$-linked arabinoside in the furanoside configuration. The enzyme was produced during growth on agricultural residue such as rice straw, but not during growth on spelt xylan, glucose or cellobiose.

  • For an endomorphism ${\alpha}$ of a ring R, the endomorphism ${\alpha}$ is called semicommutative if ab=0 implies $aR{\alpha}(b)$=0 for a ${\in}$ R. A ring R is called ${\alpha}$-semicommutative if there exists a semicommutative endomorphism ${\alpha}$ of R. In this paper, various results of semicommutative rings are extended to ${\alpha}$-semicommutative rings. In addition, we introduce the notion of an ${\alpha}$-skew power series Armendariz ring which is an extension of Armendariz property in a ring R by considering the polynomials in the skew power series ring $R[[x;\;{\alpha}]]$. We show that a number of interesting properties of a ring R transfer to its the skew power series ring $R[[x;\;{\alpha}]]$ and vice-versa such as the Baer property and the p.p.-property, when R is ${\alpha}$-skew power series Armendariz. Several known results relating to ${\alpha}$-rigid rings can be obtained as corollaries of our results.

  • The concentrations of $\alpha$-tocopherol in the brain, liver, and serum were studied with and without saponification process between control and vitamin E supplemented rats. Young rats, 80-120g body weight, were fed control and vitamin E supplemented diets, ad libitum, for four weeks. $\alpha$-Tocopherol concentrations were determined by high pressure liquid chromatography. The $\alpha$-tocopherol concentration per wet weight base in the brain tissue was significantly lower than that in the liver. Vitamin E supplementation had no effect on brain $\alpha$-tocopherol levels in contrast to the significant increase in lover $\alpha$-tocopherol concentration with and without saponification is significantly greater in the brain than in the liver or serum. Further study is needed to clarify the nature of interaction or /and binding between $\alpha$-tocopherol and the complex membrane system in brain tissue. It can be speculated from this and other studies that the metabolism and the nature of interaction of $\alpha$-tocopherol with the complex membrane system in brain tissue rich in polyunsaturated fatty acids seems different from that in liver tissue or serum.

  • The effects of prostaglandin $F_2{\alpha}$ were investigated on the uterine smooth muscle motility in the pig. The results were summarized as follows : 1. Prostaglandin $F_2{\alpha}$ caused the contraction of the porcine uterine smooth muscle and the contractile responses increased between the concentration of prostaglandine $F_2{\alpha}$ $10^{-9}$ M and $5{\times}10^{-8}$ M with a dose-dependent manner. 2. The contractile response induced by prostaglandine $F_2{\alpha}$($10^{-8}$ M) was not blocked by pre-treatment with cholinergic receptor blocker, atropine ($10^{-6}$ M). 3. The contractile response induced by prostaglandine $F_2{\alpha}$(10$^{-8}$ M) was not blocked by pretreament with ${\alpha}$-adrenergic receptor blocker, phentolamine($10^{-6}$ M) and ${\beta}$-adrenergic receptor blocker, propranolol($10^{-6}$ M). From these results, it was concluded that the effects of uterine smooth muscle by prostaglandine $F_2{\alpha}$ were only the contraction mediated by prostaglandine TEX>$F_2{\alpha}$ receptor in pig, and that it may not be related to the cholinergic and adrenergic receptor.

Expression of Chemokine and Tumor Necrosis Factor Alpha Genes in Murine Peritoneal Macrophages Infected with Orientia tsutsugamushi

  • Scrub typhus, caused by Orientia tsutsugamushi infection, is clinically and histopathologically characterized by local as well as systemic inflammatory reactions, indicating that orientiae induce mechanisms that amplify the inflammatory response. To reveal underlying mechanisms of chemoattraction and activation of responding leukocytes, expression of chemokine and tumor necrosis factor alpha (TNF-$\alpha$) genes in murine peritoneal macrophages after infection with the obligate intracellular bacterium Ο.tsutsugamushi was investigated. The genes that were unregulated included macrophage inflammatory proteins l$\alpha$/$\beta$(MIP-l$\alpha$/$\beta$), MIP-2, monocyte chemoattractant protein 1(MCP-1), RANTES (regulated upon activation, normal T-cell expressed and secreted), gamma-interferon-inducible protein 10(IP-10) and TNF-$\alpha$. Peak expression of these chemokines and TNF-$\alpha$ was observed between 1 and 3 h after infection. These responses returned to or approached baseline preinfection levels 6 h after challenge. Semiquantitative reverse transcription (RT)-PCR analysis revealed dramatic Increases during infection in the steady-state levels of mRNA ceding for the inhibitory subunit of NF-kB (IkB$\alpha$), whose transcription is enhanced by binding of NF-kB within the IkB$\alpha$promoter region. Thus, Ο. tsutsugamushi appears to be a stung inducer of chemokines and TNF-$\alpha$ which may significantly contribute to inflammation and tissue damage observed in scrub typhus by attracting and activating phagocytic leukocytes.
