Ethephon 단용처리와 Ethephon 및 염화칼슘 혼합처리에 따른 사과 'Fuji'/M.26의 성숙기 과실특성 비교 (Comparison of Fruit Characteristics of 'Fuji'/M.26 in Response to Ethephon Treatment and Combined Treatment of Ethephon and CaCl2 During Maturing Stages)

  • 만생종 'Fuji' 사과(Malus × domestica)는 과피 착색에 따라 수확시기가 가변적인 품종이다. Ethephon은 ethylene 발생을 촉진하는 식물생장조절제로 사과나무를 포함한 호흡급등형 과수작물의 과실 성숙과 관련된 생리적 반응을 유도한다. 본 연구는 'Fuji'/M.26 사과의 경제적이고 안정적인 수확기 조절 방법을 제시하고자 과실 비대생장 종료 후 ethephon 단용처리와 ethephon 및 염화칼슘 혼합처리가 과실특성에 미치는 영향을 조사하였다. 과실 비대생장이 종료된 만개 후 145일에 100 mg/L ethephon과 100 mg/L ethephon 및 0.5% 염화칼슘 혼합용액을 분무처리 후 10일 주기로 과실특성을 조사하였다. Ethephon 단용처리구와 ethephon 및 염화칼슘 혼합처리구 모두 만개 후 155일부터 전분함량이 감소되어 과실성숙이 촉진되었으며, 수확 10일 전 전분함량은 대조구의 수확기 전분함량과 유사하였다. 과피 착색은 ethephon 및 염화칼슘 혼합처리구에서 ethephon 단용처리구보다 착색률이 낮았으나 대조구보다는 높았다. 과실 평균 경도는 ethephon 단용처리구가 가장 낮았으며, ethephon 및 염화칼슘 혼합처리구는 대조구와 유사하였다. 수확기 당산비는 세 처리구 간에 유의한 차이는 없었으나 수확 10일 전 ethephon 단용처리구와 ethephon 및 염화칼슘 혼합처리구의 적정산도가 대조구보다 유의적으로 낮았다. 특히 2021년 수확 10일 전 ethephon 단용처리구와 ethephon 및 염화칼슘 혼합처리구의 당산비는 수확기 당산비와 유사하였다. 따라서 'Fuji'/M.26의 과실 비대생장이 종료된 시점에 100 mg/L ethephon과 0.5% 염화칼슘 혼합처리를 통해 과실성숙과 과피 착색을 수확기보다 10일 촉진할 수 있을 것으로 생각된다.

  • The aims of this research were to investigate the preprocessing effect of spectrum data on prediction performance and to develop a robust model to predict SSC in intact apple. Spectrum data of 320 Fuji apples were measured with the on-line transmittance measurement system at the wavelength range of 550∼1100nm. Preprocess methods adopted for the tests were Savitzky Golay, MSC, SNV, first derivative and OSC. Several combinations of those methods were applied to the raw spectrum data set to investigate the relative effect of each method on the performance of the calibration model. PLS method was used to regress the preprocessed data set and the SSCs of samples, and the cross-validation was to select the optimal number of PLS factors. Smoothing and scattering corection were essential in increasing the prediction performance of PLS regression model and the OSC contributed to reduction of the number of PLS factors. The first derivative resulted in unfavorable effect on the prediction performance. MSC and SNV showed similar effect. A robust calibration model could be developed by the preprocessing combination of Savitzky Golay smoothing, MSC and OSC, which resulted in SEP= 0.507, bias=0.032 and R$^2$=0.8823.

Measurement of Dynamic Compressive Properties of Apples using the Oscillatory Test

  • Purpose: This study performed the oscillatory test using the texture analyzer to characterize the viscoelastic behavior of apples such as the storage modulus (E'), the loss modulus (E"), the complex modulus (${\mid}E^*{\mid}$) and the energy dissipated per cycle ($W_{diss}$). Methods: The sinusoidal deformation with the frequency of 1-10 Hz and the maximum displacement of 0.1 mm were applied to the flesh tissues of Fuji, Golden Delicious and Red Delicious apples. The Lissajous figure was used to measure the phase angle(${\delta}$) between stress and strain curve. Results: Trigger force was critical to the measurement of the phase angle. E', E", ${\mid}E^*{\mid}$ and Wdiss were measured using the Lissajous figure and the phase angle. The complex modulus of Golden Delicious apple was significantly lower than those of Fuji apple and Red Delicious apple. Conclusions: Apple flesh was exhibiting more elasticity at low frequency, and more viscosity at high frequency. Dynamic compressive properties of Fuji apple were similar to those of Red Delicious apple but significantly different from those of Golden Delicious apple.

Isolation and Morphological Characterization of Monilinia sp. KV-27 Associated with Apple Anthracnose of Fuji Apples in Korea

  • This study was undertaken to isolate and to identify a fungal pathogen Monilinia sp. KV-27 associated with apple anthracnose. Rotted Fuji apples were used for the isolation of the fungus. The infected tissues were sterilized with 70% ethanol, washed with sterilized distilled water and were transferred to 50 ml containing potato dextrose broth (PDB) flasks. The peripheral hyphae of the fungal colony which developed from the infected tissues were isolated on to potato dextrose agar (PDA). On PDA plates the fungus grew well at $25^{\circ}C$ and occupied more than half of a 9 cm petri dish within 5 days. The fungal cultures on PDA were used for morphological observation and identification of the fungus. Conidiophores were produced on the gray to whitish sporodochial structures scattered on PDA plates. These conidiophores gave rise to chains of conidia, which were branched and easily detached in water. These structures were dark brown to black and consisted of hyphal masses. Conidia produced on PDA plates were hyline or light colored, lemon shaped or ellipsoidal ($10-13{\times}8.5-11{\mu}m$) in size.

  • In this study, the wear characteristics of five different dental composite resins cured by conventional halogen light and LED light sources were investigated. Five different dental composite resins of Surefil, Z100, Dyract AP, Fuji II LC and Compoglass were worn against a zirconia ceramic ball using a pin-on-disk type wear tester with 15 N contact force in a reciprocal sliding motion with sliding distance of 10 mm/cycle at 1Hz under the room temperature dry condition. The wear variations of dental composite resins were linearly increased as the number of cycles increased. It was observed that the wear resistances of these specimens were in the order of Dyract AP > Surefil > Compoglass > Z100 > Fuji II LC. On the morphological observations by SEM, the large crack formation on the sliding track of Fuji II LC specimen was the greatest among all resin composites. Dyract AP showed the least wear with few surface damage. There is no significant difference in wear performance between conventional halogen light curing and light emitting diodes curing sources. It indicates that a light emitting diodes (LED) source can replace a halogen light source as a curing unit for composite resin restorations.

사과의 성숙에 따른 Pectin질의 변화 (Changes in the Pectic Substance during Maturity of Apple)

  • Fuji 과실의 성숙에 따른 pectin 질의 변화를 조사하기 위하여 LMP (low methoxy pectin), HMP (high methoxy pectin) IAP (ionically associated pectin), CBP (covalently bounded pectin), methoxyl 및 구성당의 함량변화와 DE(degree of esterification)를 조사하였다. 그 결과 LMP+HMP와 IAP+CBP는 AIS(alcohol insoluble solid)의 각각 70%, 22%를 차지하였고 이들 성분은 성숙에 따라 다소 증가하였다. Pectose 질을 구성하는 중성당중 hexose는 LMP, HMP, IAP, CBP 다같이 $20{\sim}50%$가 감소하였는데 특히 Gal의 감소율은 63%이었다. Pentose는 $1{\sim}9%$가 증가하였고 uronic acid는 $2{\sim}4%$감소하였다. 그러나 HMP, CBP에서는 uronic acid의 상대적 함유비율은$3{\sim}5%$ 증가되었다.

사과의 품종별 휘발성 향기성분 (volatile Flavor Components in Various Varieties of Apple (Molus pumila M.))

  • 사 과의 휘발성 향기성분을 분석하기 위해 국내에서 생산되는 후지, 쓰가루, 홍로, 홍옥(조나단) 등 4가지 품목 휘발성 향기성분을 SDE 방법으로 추출하고, GC-FID와 GC/MS로 분석, 동 정하였다. 후지, 쓰가루, 홍로, 홍옥(조나단)에서 각각 100종, 68종, 85종, 94종의 화합물의 동 정되었으며, alcohol류, aldehyde류 , esterfb가 사과의 전체적 향기에 영향ㅇ르 미치는 것으 로 여겨진다. 주요 휘발성 향기에 영향을 미치는 것으로 여겨진다. 주요 위발성 향기성분으 로 hexanal, butanol, (E)-2-hexenal, 3-methyl-1-butanol, hexanol이 모든 품종에서 확인되 었으며, 그외 품종별로 약간의 차이는 있지만 ethy1 acetyl2-methylbutyate,hexyl, aceteate, (E,E)-$\alpha$-facetate도 다량 함유되어 있었다. 사과 품종별로 분리, 동정된 휘발성 향기성분을 정량한 결과, 후지 17.18mg/kg, 쓰가루 7.74mg/kg, 홍로 16.71 mg/kg, 홍옥(조나단) 15.81 mg/kg이 함유되어 있었다.

Multispectral Wavelength Selection to Detect 'Fuji' Apple Surface Defects with Pixel-sampling Analysis

  • Purpose: In this study, we focused on the image processing method to determine the external quality of Fuji apples by identifying surface defects such as scabs and bruises. Method: A CCD camera was used to capture filter images with 24 different wavelengths ranging between 530 nm and 1050 nm. Image subtraction and division operations were performed to distinguish the defect area from the normal areas including calyx, stem, and glaring on the apple surface image. All threshold values of the image were examined to reveal the defect area of pretreated filter images. Results: The developed operation methods were [image (720 nm) - image (900 nm)]/image (700 nm) for bruise detection and [image (740 nm) - image (900 nm)]/image (590 nm) for scab detection, which revealed 81% and 90% recognition ratios, respectively. Conclusions: Our results showed several optimal wavelengths and image processing methods to detect Fuji apple surface defects such as bruises and scabs.

Fluoride penetration from three orthodontic adhesives: an experimental study

  • Objective: To examine the prophylactic potential of 3 orthodontic bonding adhesives: Fuji Ortho SC, Illuminate, and Resilience. Methods: Thirty-six Wistar Wag rats were randomly divided into 4 groups consisting of 9 rats each. One of the groups received no treatment and was used as a control. In the other groups, individual bands coated with one of the 3 adhesives were cemented to the lower incisors. Enamel samples were obtained after 6 and 12 weeks and analyzed using scanning electron microscopy in combination with energy dispersive spectrometry. Results: Six weeks after band cementation, no fluoride was found in the enamel of the lower incisors. After 12 weeks, there was no fluoride in the enamel of teeth coated with the Resilience composite. However, in the case of the Illuminate composite and the resin-modified glass ionomer Fuji Ortho SC cement, the depth of fluoride penetration reached $2{\mu}m$ and $4.8-5.7{\mu}m$, respectively. Conclusions: Fluoride ions from orthodontic adhesives can be incorporated into the surface layer of the enamel. Orthodontists may apply orthodontic adhesives, such as the Fuji Ortho SC, to reduce the occurrence of caries during orthodontic treatment with fixed appliances.

Bonding of a resin-modified glass ionomer cement to dentin using universal adhesives

  • Objectives: This study aims to assess the effect of universal adhesives pretreatment on the bond strength of resin-modified glass ionomer cement to dentin. Materials and Methods: Fifty caries-free human third molars were employed. The teeth were randomly assigned into five groups (n = 10) based on dentin surface pretreatments: Single Bond Universal (3M Oral Care), Gluma Bond Universal (Heraeus Kulzer), Prime&Bond Elect (Dentsply), Cavity Conditioner (GC) and control (no surface treatment). After Fuji II LC (GC) was bonded to the dentin surfaces, the specimens were stored for 7 days at 37℃. The specimens were segmented into microspecimens, and the microspecimens were subjugated to microtensile bond strength testing (1.0 mm/min). The modes of failure analyzed using a stereomicroscope and scanning electron microscopy. Data were statistically analyzed with one-way analysis of variance and Duncan tests (p = 0.05). Results: The surface pretreatments with the universal adhesives and conditioner increased the bond strength of Fuji II LC to dentin (p < 0.05). Single Bond Universal and Gluma Bond Universal provided higher bond strength to Fuji II LC than Cavity Conditioner (p < 0.05). The bond strengths obtained from Prime&Bond Elect and Cavity Conditioner were not statistically different (p > 0.05). Conclusions: The universal adhesives and polyacrylic acid conditioner could increase the bond strength of resin-modified glass ionomer cement (RMGIC) to dentin. The use of universal adhesives before the application of RMGIC may be more beneficial in improving bond strength.
