Expression patterns of innate immunity-related genes in response to polyinosinic:polycytidylic acid (poly[I:C]) stimulation in DF-1 chicken fibroblast cells

  • Polyinosinic:polycytidylic acid (poly[I:C]) can stimulate Toll-like receptor 3 (TLR3) signaling pathways. In this study, DF-1 cells were treated with poly(I:C) at various concentrations and time points to examine the comparative expression patterns of innate immune response genes. The viability of DF-1 cells decreased from 77.41% to 38.68% when cells were treated different dose of poly(I:C) from 0.1 ㎍/mL to 100 ㎍/mL for 24 h respectively. The expressions of TLR3, TLR4, TLR7, TLR15, TLR21, IL1B, and IL10 were increased in dose- and time-dependent manners by poly(I:C) treatment. On the contrary, the expression patterns of interferon regulatory factors 7 (IRF7), Jun proto-oncogene, AP-1 transcription factor subunit (JUN), Nuclear Factor Kappa B Subunit 1 (NF-κB1), and IL8L2 were varied; IRF7 and IL8L2 were increasingly expressed whereas the expressions of JUN and NF-κB1 were decreased in a dose-dependent manner after they were early induced. In time-dependent analysis, IRF7 expression was significantly upregulated from 3 h to 24 h, whereas JUN and NF-κB1 expressions settled down from 6 h to 24 h after poly(I:C) treatment although they were induced at early time from 1 h to 3 h. Poly(I:C) treatment rapidly increased the expression of IL8L2 from 3 h to 6 h with a plateau at 6 h and then the expression of IL8L2 was dramatically decreased until 24 h after poly(I:C) treatment although the expression level was still higher than the non-treated control. These results may provide the basis for understanding host response to viral infection and its mimicry system in chickens.

24 hours' Exposed Temperature and Thermal Comfort in Summer (여름철 도시의 일상생활에서 경험하는 환경온도와 온냉감 평가)

  • All the outdoor and indoor spaces are connected with each other. The human being moves toward those spaces with temperature fluctuation between the natural and artificial temperature. We conducted an experiment which subjects were wearing the data logger in urban life, and measured 24 hours' exposed temperature and thermal comfort in summer. Results were as follows. 1. Subjects controls their micro climate like this. Most of them(84.6%) get weather information. Fashion(46.2%) and weather(30.8%) are the reasons to select clothes. They spend their time in indoor environment for 84.92% hours of a day and have an air-conditioner(61.5%) in their houses. 2. Temperature fluctuation which subjects were exposed for 24 hours were from 15.6$^{\circ}C$ to 33.8$^{\circ}C$ and average fluctuation was 9.02$^{\circ}C$. The median value of experienced temperature were 26-26.5$^{\circ}C$ and average temperature was 26.18$^{\circ}C$. They experienced cold shock of 3.96 times in a day.

Quality Characteristics of a Korean Rice Cake (Karedduk) with Mixture of Trehalose and Modified Starch by Using Response Surface Methodology (반응표면분석을 이용한 트레할로스와 변성전분 혼합사용 떡의 품질 특성)

  • Texture and descriptive sensory characteristics of a Korean rice cake (Karedduk) added with mixture of trehalose and Sun-Tender were investigated after 2, 24, and 48 hrs of storage at $5^{\circ}C$. A central composite design was used for treatment arrangement. The trehalose were added in 0, 3, 6, 9 and 12% levels and Sun-Tender added in 0, 0.3, 0.6, 0.9, and 1.2% levels to dry rice flour. The mixture amounts of trehalose and Sun-Tender on texture and descriptive sensory characteristics of a Korean rice cake (Karedduk) were optimized using response surface methodology (RSM). The two independent variables selected for the RSM were amounts of trehalose ($X_1$) and amounts of Sun-Tender ($X_2$). The predicted value at stationary point or a minimum for texture hardness by RSM was found as 591.5440 after 24 hrs of storage at $5^{\circ}C$. Also the adhesiveness, hardness, cohesiveness and sweetness by sensory characteristics were significantly different as compared to the control. The results suggested that a Korean rice cake (Karedduk) added with mixture of trehalose and Sun-Tender can be made from the mixture amounts of trehalose 9.5826% and Sun-Tender 0.2216% in retarding retrogradation.

Optimum Fermentation Conditions and Fermentation Characteristics of Mulberry (Morus alba) Wine (오디(Morus alba) 와인의 최적 발효조건 및 발효 특성)

  • For the development of mulberry wine, we investigated its optimum fermentation conditions as well as quality changes during fermentation. The physicochemical characteristics of the mulberry fruit used in the study were pH 4.56, 0.50% titratable acidity, and 13.0 $^{circ}Brix$ soluble solids. The mulberry wine fermented with Saccharomyces cerevisiae KCCM 12224 (Sc-24) at 24 $^{circ}Brix$ soluble solids and $26^{circ}C$ showed excellent characteristics in terms of ethanol production, titratable acidity, and redness. The sucrose, fructose, and glucose contents of the mulberry wine drastically decreased with fermentation time. The citric acid content was maintained during the fermentation period, and malic acid decreased, but lactic and succinic acids increased. The cyanidin-3-glucoside content, a major anthocyanin pigment, of the mulberry wine drastically decreased from 195.5 mg% at the initial stage of fermentation to 15.37 mg% at 2 days of fermentation. However, cyanidin-3-rutinoside decreased gradually. In summary, a mulberry wine of high quality was made by fermentation for 8 days at $26^{\circ}C$ using mashed mulberry fruit containing $24^{\circ}Brix$ soluble solids, after adding 200 ppm $K_2S_2O_5$ and inoculating with 3%(v/v) Sc-24.

Solids Loss with Water Uptake during Soaking of Soybeans (대두의 침지과정중 침출액의 성분변화)

  • Dried soybeans (varieties: Saeal, Kwanggyo, Tanyob) took up water rapidly for first 3hr followed by a slower rate of uptake. The beans took up an equal weight of water (100% hydration)after approximately 3.5hr at $50^{\circ}C$, 5hr at $30^{\circ}C$ and 7.5hr at $20^{\circ}C$ respectively. pH of the soaking solutions decreased during the soaking period. This was undoubtedly caused by the ionization of the cellular components resulting in increased levels of hydrogen ions in the liquor. Soluble solids were leached out of the beans at fairly steady rate throughout the hydration and the amount was greater with higher temperature. This amounted to 0.4-0.7g at $20^{\circ}C$ and 10.2-15.0g at $50^{\circ}C$ per 100g soybeans. Temperature was the most important factor in determining the rate of water absorption and of solid losses. Of the total solids lost, 12-25% was protein. The proportion of protein loss increased as the soaking time and temperature increase. Amount of protein loss was 80-200mg at $20^{\circ}C$ and 440-480mg at $50^{\circ}C$ after 24hr soaking per 100g soybeans. About 5% of soluble sugars, including fructose, sucrose, raffinose, and stachyose, was removed from the beans after 24hr soaking at $20^{\circ}C$.

Studies on Lipids of Urechis unicintus -on the Composition of Lipids, Fatty acid and Sterol- (개불의 지질에 관한 연구 -지질, 지방산 및 Sterol 조성에 관하여-)

  • This work was done in order to clarify the chemical characteristics, composition, fatty acid components and sterol components of the lipids from Urechis unicintus. The results obtained are summarized as follows ; 1. The lipid content, iodine value and unsaponifiable matter of the total lipids are 1.89, 111, and $14.3\%$, respectively. 2. The main components of the total lipids are phospholipids $39.8\%$, free sterol $27.7\%$ and triglyceride $21.4\%$, and two unidentified fractions are detected. 3. The main fatty acids of total lipids are $C_{20:4}(19.4\%),\;C_{16:0}(13.7\%),\;C_{20:1}(11.3\%)\;and\;C_{18:1}(10.4\%)$ 4. Sterols found are Cholesterol($57.6\%$), Brassicasterol(?)($20.3\%$), 24-methylenecholesterol ($17.7\%$),22-dehydrocholesterol($3.0\%$)and 22.trans-24-norcholesta-5,22-dien-$3{\beta}$-ol(?)$1.4\%$.

  • DC accelerated aging characteristics of $Pr_{6}O_{11}$-based ZnO varistors, which are composed of $ZnO+Pr_{6}O_{11}+CoO+Cr_{2}O_{3}+Dy_{2}O_{3}$ ceramics were investigated with CoO content in the range of 0.5 - 5.0 mol%. The varistors doped with 1.0 mol% revealing maximum value(66.61) in the nonlinear exponent exhibited excellent stability, in which the variation rates of the varistor voltage, the nonlinear exponent and leakage current are -1.93%, -10.48%, and +288.79%, respectively, under DC accelerated aging stress, such as $(0.85V_{lmA}/115^{\circ}C/24h)+(0.90V_{lmA}120^{\circ}C/24h)+(0.95V_{lmA}/125^{\circ}C/24h)+(0.95V_{lmA}/150^{\circ}C/24h)$. Next the varistors doped with 2.0 mol% exhibiting the nonlinear exponent of 47.39 showed high stability.

  • This paper describes the fabrication and characteristics of a Au/Ni/Ti/3C-SiC Schottky diode with field plate (FP) edge termination. The Schottky contacts were annealed for 30 min at temperatures ranging from 0 to $800^{\circ}C$. At annealing temperature of $600^{\circ}C$, it showed an inhomogeneous Schottky barrier and had the best electrical characteristics. However, the annealing of $800^{\circ}C$ replaced it with ohmic behaviors because of the formation of many different types of nickel silicides. The fabricated Schottky diode had a breakdown voltage of 200 V, Schottky barrier height of 1.19 eV and worked normally even at $200^{\circ}C$.

A Cellular Physiological Study on the Effects of Korean Ginseng -Part I. Effects on the Division of Saccharomyces- (인삼의 효과에 관한 세포생리학적 연구 -제 I 편 Saccharomyces의 분열에 미치는 영향-)

  • The effects of water extracts and powder of Korean ginseng on the division of Saccharomyces cerevisiae were studied. 1. The addition of several doses of water extracts and powder of ginseng to the yeast medium of Moyer and Coghill showed various promoted division of Saccharomyces. 2. The optimal dose of ginseng on tile division of Saccharomyces (0.08% dry ginseng medium solution per $10\;cells/mm^3$) could be recognized. 3. On the culture for 24 hours at $18^{\circ}C$, the cell number of control group was $13.25{\times}10^3\;cells/mm^3$ and that of the optimal dose group of water extracts of ginseng was $23.20{\times}10^3\;cells/mm^3$. On the culture, for 24 hours at $25^{\circ}C$, the cell number of control group was $16.85{\times}10^3\;cells/mm^3$ and that of the optimal dose group was $30.20{\times}10^3\;cells/mm^3$. The increasing rate of cell divison by the ginseng was about twice than that of control group. The optimal dose treatment of ginseng at $18^{\circ}C$ was more effective than control group at $25^{\circ}C$. 4. On the culture for 24 hours at $18^{\circ}C$, the increasing rate of water extracts of ginseng was 75.1%, and the rate of ginseng powder was 7.6%. On the culture for 24 hours at $25^{\circ}C$, the rate of water extracts of ginseng was 79.8%, and the rate of ginseng powder was 57.2%. Therefore water extracts of ginseng was more effective than ginseng powder of same dry weight, and the promoted effect of ginseng powder at $25^{\circ}C$ was more effective than at $18^{\circ}C$.
