Genotype distribution and gene frequency of angiotensin I-converting enzyme in Korean population

  • The angiotensin converting enzyme (ACE) is a key component of the renin-angiotensin system thought to be important in the pathogenesis of hypertension and cardiovascular diseases. Deletion polymorphism in the ACE gene may be a risk factor for myocardial infarction. The insertion/deletion (I/D) polymorphism of the ACE detected by PCR analysis appears to be associated with hypertension in Koreans and its nucleotide was subcloned into T-vector and its nucleotide sequences were determined. We also examined an association between hypertension and genetic variance of ACE. We identified the angiotensin I-converting enzyme genotype in 127 hypertensive and 189 normotensive Korean subjects. The distribution of ACE genotype II, ID, DD were 39.2%, 40.2%, 20.6% respectively and the frequency for ACE alleles I and D were 0.593 and 0.407, respectively in all subjects. The frequency of D allele in Korean males is higher than that of Korean females (male; 0.438 : female; 0.267), and the frequency of I allele in Korean females is higher than that of Korean males (female; 0.733 : male; 0.562). Genotype distributions of angiotensin I-converting enzyme genes in Korean normal adult population were different from that of Caucasians (P<0.001). There were no significant differences in genotype frequency between the hypertensive control group (n=127) and the normotensive group (n=189). We observed significant differences of ACE genotype distribution between the male group and the female group in total (P=0.001) and in hypertensive Korean subjects (P=0.013).

Structure-Antifungel Activity Relationships of Cecropin A Hybrid Peptides against Trichoderma sp.

  • The hybrid peptides, CA-ME, CA-MA and CA-BO, with the N-terminal sequence 1-8 of cecropin A and the N-terminal sequences 1-12 of melittin, magainin 2 and bombinin, respectively, have more improved antibacterial activities. CA-MA was found to have stronger antifungal activity against Trichoderma sp than other hybrid peptides and their parental peptides. In order to elucidate the relationships between the peptide structure and antifungal activity, several analogues of CA-MA or CA-BO were also designed and synthesized by the solid phase method. An tifungal activity was measured against T. reesei and T. viride, and hemolytic activity was measured by a solution method against human red blood cells. The residue 16 of CA-MA, Ser, was found to be important for antifungal activity. When the residue was substituted with Leu, showed powerful antifungal activity was dramatically decreased. CA-MA, P1, P4 and P5 designed in this study showed powerful antifungal activity against T. reesei and T. viride with low hemolytic activity against human red blood cells. These hybrid peptides will be potentially useful model to further design peptides with powerful antifungal activity for the effective therepy of fungal infection and understand the mechanisms of antifungal actions of hybrid peptides.

Some properties of Cucumber mosaic virus and a potfvirus isolated from Freesia

  • Freesia, a member of the Iridaceae family, has fragant, tubular shaped flowers and is very popular ornamental plants in the world. Diseased freesia plants showing systemic leaf streak mosaic symptoms were collected from a cultivated farm in Kyonggi province, Korea in 2003, and its causal agents were investigated. Two viruses, Cucumber mosaic virus (Fr-CMV) and a potyvirus, were identified from the leaf tissues of the diseased freesia based on sequence analysis and host range tests. CMV-Fr could infect systemically on Chenopodium quinoa, C. amaranticolor, N. glutinosa, and N. benthamiana, and this biological property is distinguishable from ordinary strains of CMV. A filamentous potyvirus-shaped virus could not infect general indicator plants by mechanical inoculation. Single RT-PCR products was successfully amplified with a set of degenerate primers specific to the Potyvirus genus and total nucleic acids from the infected tissues, and was cloned into the pGEMT-Easy vector. Nucleotide sequences confirmed it belongs to the Potyvirus genus with either a new species or an isolate of Freesia mosaic virus (no information is available for the FrMV). This is the first report of FrMV in Korea and more characterizations of the two viruses are in progress.

Molecular Characterization of AceB, a Gene Encoding Malate Synthase in Corynebacterium glutamicum

  • The aceB gene, encoding for malate synthase, one of the key enzymes of glyoxylate bypass, was isolated from a pMT1-based Corynebacterium glutamicum gene library via complementation of an Escherichia coli aceB mutant on an acetate minimal medium. The aceB gene was closely linked to aceA, separated by 598 base pairs, and transcribed in divergent direction. The aceB expressed a protein product of Mr 83, 000 in Corynebacterium glutamicum which was unusually large compared with those of other malate synthases. A DNA-sequence analysis of the cloned DNA identified an open-reading frame of 2, 217 base pairs which encodes a protein with the molecular weight of 82, 311 comprising 739 aminoo acids. The putative protein product showed only limited amino acid-sequence homology to its counteliparts in other organisms. The N-terminal region of the protein, which shows no apparent homology with the known sequences of other malate synthases, appeared to be responsible for the protein s unusually large size. A potential calciumbinding domain of EF-hand structure found among eukaryotes was detected in the N-terminal region of the deduced protein.

Comparative analysis of AGPase proteins and conserved domains in sweetpotato (Ipomoea batatas (L.) Lam.) and its two wild relatives

  • Conserved domains are defined as recurring units in molecular evolution and are commonly used to interpret the molecular function and biochemical structure of proteins. Herein, the ADP-glucose pyrophosphorylase (AGPase) amino acid sequences of three species of the Ipomoea genus [Ipomoea trifida, I. triloba, and I. batatas (L.) Lam. (sweetpotato)] were identified to investigate their physicochemical and biochemical characteristics. The molecular weight, isoelectric point, instability index, and grand average of hyropathy markedly differed among the three species. The aliphatic index values of sweetpotato AGPase proteins were higher in the small subunit than in the large subunit. The AGPase proteins from sweetpotato were found to contain an LbH_G1P_AT_C domain in the C-terminal region and various domains (NTP_transferase, ADP_Glucose_PP, or Glyco_tranf_GTA) in the N-terminal region. Conversely, most of its two relatives (I. trifida and I. triloba) were found to only contain the NTP_transferase domain in the N-terminal region. These findings suggested that these conserved domains were species-specific and related to the subunit types of AGPase proteins. The study may enable research on the AGPase-related specific characteristics of sweetpotatoes that do not exist in the other two species, such as starch metabolism and tuberization mechanism.

Morphological and Phylogenetic Analysis of a New Record of Paraconiothyrium kelleni from Soil in Korea

  • A fungal strain designated KNUF-21-66Q1 was isolated from soil in Chungcheongbuk Province, Korea. Moderate growth of colonies was observed on potato dextrose agar, oatmeal agar (OA), malt extract agar, and cornmeal agar media at 25℃, and the detailed morphology was examined on OA medium. The colonies on OA medium were flat, had entire margin, hyaline, and yellow concentric rings in 3-4 weeks. Conidiomata were pycnidial, solitary or clustered, globose to subglobose, black-brown, and 300-500 ㎛ in diameter. Conidiogenous cells were smooth, hyaline, globose to ampulliform, and 6.0-9.0×3.0-6.0 ㎛ in size (n=15). Conidia were hyaline to pale brown, slightly golden, obovoid to slightly ellipsoidal, smooth, guttulate, and 3.0-4.7×2.1-3.3 ㎛ in size (n=100). The strain was confirmed based on phylogenetic analysis using internal transcribed spacer regions, the partial 28S rDNA of large subunit, and β-tubulin gene sequences. The morphological observations and phylogenetic analysis revealed that the strain KNUF-21-66Q1 was similar to the previously described Paraconiothyrium kelleni. To our knowledge, this is the first report of P. kelleni in Korea.

제주 연안에서 분리된 해양방선균의 이화학적 특성 및 다양성 (Physico-chemical Characteristics and Diversity of Marine Actinomycetes Isolated from the Coast of Jeju Island)

  • 제주도 연안해역 4개 지역(한림, 애월, 신촌, 함덕) 해수의 온도, 염분농도, 용존산소량(DO), 화학적 산소요구량(COD), 부유물질(SS), 암모니아성 질소($NH_3-N$), 질산성 질소($NO_3-N$) 및 아질산성 질소($NO_2-N$)와 같이 다양한 이화학적 특성을 확인하였다. 해수의 평균 온도는 $26.23{\sim}28.6^{\circ}C$, 염분농도는 31.4~32.88‰, pH는 8.15~8.35, COD는 0.48~0.91 mg $L^{-1}$, DO는 6.78~6.87 mg $L^{-1}$로 나타났다. 해수에서 분리된 해양방선균은 총 52종으로 제주시 동부지역(A, B)에서는 24균주, 서부지역(C, D)은 28균주가 분리되었다. 분리된 해양방선균은 16S rRNA 염기서열 분석을 이용하여 최종적으로 동정되었으며, 염기서열 정보를 기초로하여 유사도(similarity)를 조사하였다. 분리된 방선균의 16S rRNA 염기서열 분석을 통하여 계통 분류학적으로 어떤 division에 속하는지를 확인하여 본 결과 제주도 제주시 동부지역(Site A, B) 해양에서 분리된 방선균 24균주는 Gram positive bacteria (division)/Actinobacteria (class)/Actinomycetales (order)/Streptomy-cineae (suborder)/Streptomycataceae (family)/Streptomyces(genus)에 22 균주, Actinomycetales (order)/Streptosporangineae (suborder)/Nocardiopsaceae (family)/Nocardiopsis (genus)에 2 균주가 분리되었다. 제주시 서부지역(Site C, D) 해양에서 분리된 방선균 28균주는 Gram positive bacteria (division)/Actinobacteria (class)/Actinomycetales (order)/Streptomycineae (suborder)/Streptomycataceae (family)/Streptomyces (genus)에 27균주, Actinomycetales (order)/Streptosporangineae (suborder)/Nocardiopsaceae (family)/Nocardiopsis (genus)에 1균주가 분리되었다.

Evolution of a dextransucrase gene for constitutive and hyper-production and for synthesis of new structure dextran

  • After irradiation of a cloned dextransucrase gene (dsrB742) with ultrasoft X-ray, an E. coli transformant (pDSRB742CK) was first developed for the expression of an extracellular dextransucrase, having increased activity and the synthesis of a highly branched dextran. Seven nucleotides of the parent gene (dsrB742) were changed in the nucleotide sequences of dsrB742ck. Among them, four nucleotides were changed at the ORF of dsrB742, resulting in a 30 amino acids deletion in the N-terminal of DSRB742 dextransucrase. The activity of DSRB742CK dextransucrase in culture supernatant was approximately 2.6 times higher (0.035 IU/ml) than that of the DSRB742 clone. The pDSRB742CK clone produced DSRB742CK dextransucrase when grown both on a sucrose medium (inducibly) and on a glucose medium (constitutively). The DSRB742 clone did not produce dextran constitutively on a glucose medium. DSRB742CK dextran had 15.6% branching and 2.7-times higher resistance to dextranase hydrolysis compared to DSRB742 dextran. $^{13}C-NMR$ showed that DSRB742CK dextran contained ${\alpha}-(1{\rightarrow}3)$ branch linkages that were not present in DSRB742 dextran.

Biodegradative Activities of Fungal Strains Isolated from Terrestrial Environments in Korea

  • Polylactic acid (PLA) and polycaprolactone (PCL) are commercially available bioplastics that are exploited worldwide, and both are biodegradable. The PLA and PCL polymer-degrading activity of 30 fungal strains that were isolated from terrestrial environments were screened based on the formation of a clear zone around fungal colonies on agar plates containing emulsified PLA or PCL. Among them, five strains yielded positive results of biodegradation. Strains Korean Agricultural Culture Collection (KACC) 83034BP and KNUF-20-PPH03 exhibited PCL degradation; two other strains, KACC 83035BP and KNUF-20-PDG05, degraded PLA; and the fifth strain, KACC 83036BP, biodegraded both tested plastics. Based on phylogenetic analyses using various combinations of the sequences of internal transcribed spacer (ITS) regions, RPB2, LSU, CAL, and b-TUB genes, the above-mentioned strains were identified as Apiotrichum porosum, Penicillium samsonianum, Talaromyces pinophilus, Purpureocillium lilacinum, and Fusicolla acetilerea, respectively. Based on our knowledge, this is the first report on (i) plastic biodegraders among Apiotrichum and Fusicolla species, (ii) the capability of T. pinophilus to degrade biodegradable plastics, (iii) the biodegradative activity of P. samsonianum against PCL, and (iv) the accurate identification of P. lilacinum as a PLA biodegrader. Further studies should be conducted to determine how the fungal species can be utilized in Korea.

First Report of Soft Rot Caused by Pectobacterium brasiliense on Cucumber in Korea

  • Wilted and water-soaked lesion symptoms were observed on cucumbers in greenhouses located in Daejeon, Chungcheongnam-do, Korea, in June 2021. A bacterial strain, designated KNUB-04-21, was isolated from the cucumbers, which was subsequently identified as Pectobacterium brasiliense through a phylogenetic analysis based on sequences of the 16S rRNA region, dnaX, leuS, and recA genes. The biochemical characteristics of KNUB-04-21 were also similar to those of P. brasiliense through investigation using the API ID 32 GN system. The pathogenicity of KNUB-04-21 was confirmed by inoculating it into healthy cucumber plants. The reisolated strains were also found to be same to the original strain. To our knowledge, this is the first report of P. brasiliense being identified as the causative agent of cucumber soft rot in Korea.
