Reducing Power and ${\alpha}-Glucosidase$ Inhibitory profiles of (-)-Catechin and Its glycoside ((-)-Catechin 및 배당체의 환원력 및 ${\alpha}-glucosidase$저해 활성)

  • From the EtOAc fraction of the MeOH extract of Ulmus davidiana, (-)-catechin (1), (-)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and (-)-catechin-7-O-${\beta}$-D-xylopyranoside (3) were isolated and characterized on the basis of $^1H-and\;^{13}C-NMR$, and FABMS spectral data. Compounds 1-3 showed more strong reducing power activities than ${\alpha}-tocopherol$, a positive control.

  • The bark of ash(Fraxinus rhynchophylla) and elm(Ulmus davidiana var. japonica) trees were collected, extracted with acetone-$H_2O$(7:3, v/v), fractionated with hexane, chloroform and ethylacetate, and freeze dried to give some dark brown powder. Each fraction of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. The ash bark contained a large amount of coumarin derivatives such as aesculetin and aesculin in addition to trace amount of ligstroside and oleuropein. Most of the elm bark extractive were (+)-catechin and its glycosides such as (+)-catechin-7-O-xylopyranose and (+)-catechin-7-O-apiofuranose in addition to a small amount of procyanidin B-3, a dimeric (+)-catechin. NMR and FAB-MS spectrometric analyses were performed to characterize the structures of isolated phenolic compounds.

Phenolic Compounds from Barks of Ulmus macrocarpa and Its Antioxidative Activities (왕느릅나무 수피의 페놀성 화합물 및 항산화 활성)

  • Phytochemical examination of Barks of Ulmus macrocarpa has led to the isolation and characterization of two fla-vanonol, taxifolin $7-O-{\beta}-D-glucopyranoside$ (1), taxifolin $3'-0-7-{\beta}-glucopyranoside$ (2), two flavanone eriodictyol $7-O-{\beta}-D-glucopyranoside$ (3), nalingenin $7-O-{\beta}-D-glucopyranoside$ (4), three flavan 3-ol, (+)-catechin (5), (-)-epicatechin (6), (+)-catechin 7-O-{\beta}-D-glucopyranoside (7) and one proanthocyanidin, procyanidin B-1 (8). Antioxidative activity of these compounds was determined by measuring the radical scavenging effect on 1,1-diphenyl-2- picrylhydrazyl (DPPH) radicals . (+)-Catechin (5) , (-)-epicatechin (6), (+)-catechin $7-O-{\beta}-D-glucopyranoside$ (7) and procyanidin B-1 (8) showed significant antioxidative activity.

Preparation of a Silk Fibroin Film Containing Catechin and Its Application

  • Silk fibroin (SF) film containing catechin was prepared and the antimicrobial activity as well as physical property of the film was examined. Tensile strength of the SF film decreased with increasing concentration of catechin, and water vapor permeability of the film decreased. The film's antimicrobial activity against Escherichia coli O157:H7 increased with increasing catechin concentration. Sausage samples were inoculated with E. coli 0157:H7 and Listeria monocytogenes, and the sausage packaged with the SF film containing catechin had a decrease in the populations of E. coli O157:H7 and L. monocytogenes by 0.83 and 0.85 log CFU/g after 12 days of storage, respectively, compared to the control. In addition, the sausage had a better quality than the control regarding lipid oxidation. Our results indicate that sausages can be packed with the SF film containing catechin to extend shelf life.

  • The MeOH extract of Sorbus commixta (Rosaceae) exhibited strong DPPH radical scavenging activity, and through activity-guided fractionation two antioxidant compounds were isolated and identified as $catechin-7-O-{\beta}-D-xylopyranoside$ (1) and $catechin-7-O-\;{\beta}-D-apiofuranoside$ (2) by physicochemical and spectrometric methods. To evaluate the antioxidant effect of these compounds, some in vitro tests, such as the DPPH radical scavenging activity test, the superoxide radical scavenging activity test and the lipid peroxidation inhibitory activity test, were performed. Compounds 1 and 2 showed stronger activities than both a-tocopherol and butylated hydroxy anisole (BHA) in each assay.

  • The bark of Ulmus parvifolia Jacq. (Ulmaceae) has been used for the treatment of gonorhea, edema, scabies and eczema marginatum. Previous investigations conducted with the heartwood and leaves have demonstrated it to contain sesquiterpenes as well as fat acids from the heartwood and flavonol glycosides from leaves. However, no phytochemical work has been done on the bark parts of this plant. Investigation of the phytochemical constituents in the barks of U. parvifolia has resulted in the isolation of sterols, sterol glucoside and a catechin glycoside, $(+)-catechin\;7-O-{\alpha}-{_L}-rhamnopyranoside$, all of which were isolated for the first time from this plant. Sterols were consisted of the three components, ${\beta}-sitosterol$, stigmasterol and campesterol in a ratio of 92.1:4.1:3.8, and sterol glucoside was identified as ${\beta}-sitosterol\;3-O-{\beta}-{_D}-glucoside$. The structure of the catechin $7-O-{\alpha}-{_L}-rhamnoside$ was established primarily by analysis of $^1H-and$ COSY-45 NMR, HMQC and HMBC and EI mass spectra of the heptaacetate. Especially, HMBC spectrum provides effective way for the determination of the point of attachment of the rhamnosyl group to catechin moiety.

Antioxidant and Anti-inflammatory Activity of Stem Bark Extracts from Ulmus davidiana var. japonica

  • Ulmus davidiana var. japonica is a deciduous tree used in traditional medicine. In this study, we examined the antioxidant and anti-inflammatory activity of extracts and compounds isolated from U. davidiana var. japonica stem barks for development of cosmetic phyto-materials. Phytochemical examination of the stem bark led to the isolation and characterization of three known compounds, (+)-catechin (1), (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and procyanidin B3 ((+)-catechin-($4{\alpha}{\to}8$)-(+)-catechin) (3). Their bioactivities including antioxidant (DPPH ree radical scavenging assay) activity, anti-inflammatory (nitric oxide nhibition assay) were evaluated. Most of the crude extracts and isolates indicated significant antioxidant potential compared with BHT and ${\alpha}$-tocopherol as controls. Futhermore, all compounds showed higher inhibitory activities for NO production in Raw 264.7 cells than the L-NMMA using the positive control. Eespecially, (+)-catechin (1) and (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2) which could inhibit more than 90% of the NO production at a concentration of 100 ${\mu}g/m{\ell}$, implying excellent anti-inflammatory activity.

  • One flavonoid, lignan glycosides and two diarylheptanoids were isolated from the sapwood of Betula maximowicziana. The structures were determined as : catechin 7-O-𝛽-D-xylopyranoside, lyoniresinol 9'-O-𝛽-D-glucopyranoside, 11-oxo-3, 8, 12, 17-tetrahydroxy-9-ene[7, 0]-metacyclophane and 11-oxo-3, 8, 9, 10, 12, 17-hexahydroxy[7, 0]-metacyclophane, respectively, on the basis of spectrosopic data and chemical correlations.

  • The barks of oak trees (Quercus acutissima and Quercus variabilis) were collected, extracted with acetone-$H_2O$ (7:3, v/v), fractionated with hexane, $CH^2C1^2$ EtOAc and -$H_2O$, then freeze dried to give dark brown powder. The EtOAc soluble mixtures of the trees were chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. The structures of isolated compounds were characterized by $^1H$, $^13C$ and 2D-NMR spectroscopy and molecular weights were determined by FAB-MS spectra. The isolated compounds from Quercus acutissima were (+)-catechin, (+)-gallocatechin, gallic acid and taxifolin-3-O-$\beta$-D-glucopyranoside and the compounds from Quercus variahilis (+)-catechin, caffeic acid and taxifolin-3-O-$\beta$-D-glucopyranoside.

Validation of Analytical Methods for Unriped Rubus crataegifolius, Gardenia jasminoides and Ulmus macrocarpa Marker Compounds for Standardization of Natural Complex LS-RUG-com Preparation as Functional Ingredient (천연복합소재 LS-RUG-com의 기능성원료 표준화를 위한 산딸기 미숙과, 치자 및 유백피 지표성분의 분석법 검증)

  • LS-RUG-com preparation in a complex extract from mixture of three natural plants. Rubus crataegifolius/unriped, Gardenia jasminoides and Ulmus marcrocarpa that have been widely used in traditional functional health food. This study was conducted to establish the HPLC analysis methods that can be used to establish quantitative analysis of R. crataegifolius, G. jasminoides and U. macrocarpa for standardization of LS-RUG-com preparations. HPLC analysis methods for simultaneous determination of ellagic acid and geniposide and single determination of catechin-7-O-β-D-apiofuranoside were established for the quality control of natural plants complex (LS-RUG-com). Validation of HPLC analysis were performed by checking specificity, accuracy, precision, limit of detection and quantitation, and linearity following ICH (International Council for Harmonisation) guideline. As the result of quantitative analysis, the contents of ellagic acid, geniposide and catechin-7-O-β-D-apiofuranoside in each plant extracts were 11.2 mg/g (ellagic acid) and 72 mg/g (geniposide) and 10.2 mg/g (catechin-7-O-β-D-apiofuranoside). The contents of ellagic acid, geniposide and catechin-7-O-β-D-apiofuranoside in LS-RUG-com were 4.62~6.82 mg/g (ellagic acid), 19.2~28.8 mg/g (geniposide) and 1.36~2.04 mg/g (catechin-7-O-β-D-apiofuranoside) respectively.
