Tocopherol Content and Composition in Peanut

  • The content and composition of tocopherol were analyzed in peanuts produced in Korea. The tocopherol assay was consisted of an extraction with n-hex-ane:isopropanol mixture followed by high pressure liquid chromatography with fluorescence detection. Tocopherol content was higher in leaves and seeds but lower in seed coats and shells. $\alpha$-/${\gamma}$- Tocopherol ratio was as low as 0.53 in seeds and as high as 22.5 in shells. Tocopherol content in seeds of nine varieties ranged from 142 to 220 ng/mg dry weight, and the $\alpha$-/${\gamma}$-tocopherol ratio from 0.40 to 0.75. Tocopherol content decreased by 18% but $\alpha$-/${\gamma}$-tocopherol ratio increased by 44% in roasted seeds. The results indicate that the level of tocopherol in Korean peanut varieties is moderate and the ratio of $\alpha$- to ${\gamma}$-tocopherol is low.

Vitamin E status of 20- to 59-year-old adults living in the Seoul metropolitan area of South Korea

  • BACKGROUND/OBJECTIVES: Vitamin E is a fat-soluble vitamin and functions primarily as a lipid antioxidant. Inadequate vitamin E status may increase risk of several chronic diseases. Thus, the objectives of this study were to estimate intake and plasma concentration of each tocopherol and to evaluate vitamin E status of Korean adults. SUBJECTS/METHODS: Three consecutive 24-h food recalls and fasting blood samples were collected from healthy 20- to 59-y-old adults (33 males and 73 females) living in the Seoul metropolitan area, South Korea. ${\alpha}$-, ${\beta}$-, ${\delta}$-, and ${\gamma}$-tocopherol intakes and plasma concentrations of tocopherols (${\alpha}$-, ${\delta}$-, and ${\gamma}$- tocopherol) were analyzed by gender. RESULTS: Dietary vitamin E and total vitamin E intake (dietary plus supplemental vitamin E) was $17.68{\pm}14.34$ and $19.55{\pm}15.78mg$ ${\alpha}$-tocopherol equivalents, respectively. The mean daily ${\alpha}$-tocopherol, and ${\gamma}$-tocopherol intakes were $3.07{\pm}2.27mg$ and $5.98{\pm}3.74mg$, respectively. Intakes of total vitamin E and each tocopherol of males were significantly higher than those of females (P < 0.05). Plasma ${\alpha}$-tocopherol concentration was $15.45{\pm}10.16$ of males and $15.00{\pm}4.54{\mu}mol/L$ of females, respectively. There were no significant differences in plasma tocopherol concentrations by gender ($P{\geq}0.05$). Plasma ${\alpha}$-tocopherol was negatively correlated with ${\gamma}$-tocopherol intake (P < 0.05). Twenty-three percent of the subjects had plasma ${\alpha}$-tocopherol concentrations < $12{\mu}mol/L$ indicating a biochemical deficiency of vitamin E. Approximately 8% and 9% of these participants had plasma ${\alpha}$-tocopherol:total lipid ratio less than $1.59{\mu}mol/mmol$ and plasma ${\alpha}$-tocopherol:total cholesterol ratio less than $2.22{\mu}mol/mmol$, respectively, which are also indicative of vitamin E deficiency. CONCLUSIONS: Vitamin E intakes of Korean adults were generally adequate with the Korean Dietary Reference Intakes for vitamin E. However, ${\alpha}$-tocopherol intake was lower than that reported in other countries, and 23% of the subjects in the current study were vitamin E deficient based on plasma ${\alpha}$-tocopherol concentrations.

  • This study was conducted to investigate the effects of $\alpha$-, ${\gamma}$-, $\delta$-Tocopherols on oxidative stability of lipid in hamburger patties during frozen storage. Hamburger patty samples were prepared with the addition of tocopherol by fat basis: control, treat 1 (control+0.02% $\alpha$-tocopherol), treat 2 (control+ 0.02% ${\gamma}$-tocopherol were cooked at 85$^{\circ}C$ for 3 minutes and the stored at -1$0^{\circ}C$ or 3$0^{\circ}C$). The fatty acids composition of the hamburger samples were analyzed during the storage periods (1, 20, 40, 60 and 80 days). there was no difference in the fatty acids composition among control and treatment groups in early storage time; however, content of unsaturated fatty acid was decreased in control group and slightly decreased or almost not changed in treatment groups as the storage period passed. No difference in the ratio of saturated fatty acid : unsaturated fatty acid (SFA : USFA) and saturated fatty acid : monounsaturated fatty acid : unsaturated fatty acid (SFA : MUFA : PUFA) between control and treatment groups at early storage time. However, SFA : USFA and SFA : MUFA : PUFA was decreased in control and slightly decreased or not changed in all treatment groups as the storage period passed.

Expression of γ-Tocopherol Methyltransferase Transgene Improves Tocopherol Composition in Lettuce (Latuca sativa L.)

  • A cDNA encoding ${\gamma}-tocopherol$ methyltransferase (${\gamma}-TMT$) from Arabidopsis thaliana was overexpressed in lettuce (Latuca sativa L.) to improve the tocopherol composition. Seven lines of lettuce ($T_0$) containing the ${\gamma}-TMT$ transgene were produced by Agrobacterium-mediated transformation. The inheritance and expression of the transgene were confirmed by DNA and RNA gel blot analyses as well as quantification of tocopherols and ${\gamma}-TMT$ activities. The ratio of ${\alpha}-/{\gamma}-tocopherol$ content (TR) varied from 0.6 to 1.2 in non-transformed plants, while the $T_0$ plants had ratios of 0.8 to 320. The ratio ranged from 0.4 to 544 in 41 $T_1$ progenies of the $T_0$ transgenic line gTM3, and the phenotypic segregation indicated monogenic inheritance of the transgene (i.e., 3:1 = dominant:wild-type classes). There was a tight relationship between the TR phenotype and ${\gamma}-TMT$ activity, and enzyme activities were affected by the copy number and transcript levels of the transgene. The TR phenotype was stably expressed in $T_2$ progenies of $T_1$ plants. The results from this study indicated that a stable inheritance and expression of Arabidopsis ${\gamma}-TMT$ transgene in lettuce results in a higher enzyme activity and the conversion of the ${\gamma}-tocopherol$ pool to ${\alpha}-tocopherol$ in transgenic lettuce.

토코페롤류의 항산화작용과 Linoleic Acid Methylester에서 생성된 cis/trans-, trans/trans-Hydroperoxide Isomer (Antioxidant Effect of Tocopherols and Tocotrienols and cis/trans-, trans/trans-Hydroperoxide Isomer from Linoleic Acid Methylester)

  • Linoleic acid methylester와 토코페놀류$({\alpha}-,\;{\beta}-,\;{\gamma}-,\;{\delta}-tocopherol$${\alpha}-,\;{\gamma}-,\;{\delta}-tocotrienol)$를 사용한 model system에서의 자동산화과정에서 온도$(40,\;60,\;80^{\circ}C)$와 산소의 양 $(0,\;10,\;20%\;O_2)$에 따르는 항산화효과를 고찰하였다. Linoleic acid methylester에서 생성된 주요 산화생성물인 13-cis/trans-, 13-trans/trans-, 9-trans/cis-, 9-trans/trans-hydroperoxide를 HPLC를 이용하여 분석하였으며, 그 양적 변화로 항산화정도를 비교하였다. 모든 토코페롤류$({\alpha}-tocotrienol$제외)는 $60^{\circ}C$ 범위에서 항산화효과가 있었다. ${\alpha}-tocopherol$, ${\alpha}-tocotrienol$ and ${\delta}-tocotrienol$의 경우 $80^{\circ}C$ 범위에서 산화촉진효과가 관찰되었다. $40^{\circ}C$ 범위에서는 주로 cis/trans-hydroperoxide가 많이 생성되었고 $80^{\circ}C$ 범위에서는 trans/trans-hydroperoxide가 많이 생성되었다. $80^{\circ}C$ 범위의 hydroperoxide 생성량간의 오차가 심한 구간을 제외한 $40^{\circ}C/10%,\;O_2$에서부터 $60^{\circ}C/20%,\;O_2$까지의 산화구간에서 cis/trans-: trans/trans-hydroperoxides의 비율은 ${\alpha}-T>{\alpha}-T_3>{\gamma}-T>{\beta}-T>{\gamma}-T_3>{\delta}-T>{\delta}-T_3$의 순으로 고찰되어, 토코페롤류 중 ${\alpha}-tocopherol$이 가장 낮은 항산화효과가 있는 것으로 나타났다.

애기장대 gamma-Tocopherol Methyltransferase 유전자를 이용한 상추의 형질전환 (Transformation of Arabidopsis gamma-Tocopherol Methyltransferase into Lettuce (Lactuca sativa L.))

  • 겨울 상추 품종인 청치마 상추의 자엽조직을 ${\gamma}$-TMT유전자가 도입된 A. tumefaciens LBA 4404와 공동배양한 후, 50 mg/L kanamycin, 500 mg/L carbenicillin, 0.1 mg/L NAA, 0.5mg/L BA가 첨가된 MS재분화 배지에 옮겨 약4주 배양하여 재분화된 신초를 얻었다. 재분화된 신초를 50 mg/L kanamycin, 250mg/L carbenicillin이 포함된 MS 기본 배지에 옮겨 발근된 소식물체를 얻었다. 선발된 형질전환 식물체의 genomic DNA에 대한 PCR분석과 Southern분석을 수행하여 애기장대 ${\gamma}$-TMT유전자 특이적 DNA단편이 상추의 genomic DNA내에 삽입되었음을 확인하였다. 선발된 상추 형질전환체 잎의 $\alpha$-tocopherol/${\gamma}$-tocopherol 함량 비율이 대조 식물체에 비하여 약 4배 증가하여 도입된 ${\gamma}$-TMT유전자가 안정적으로 발현함을 나타내었다.

흰쥐의 Testosterone 함량(含量), Protein 대사(代謝) 및 스테미너 증진(增進)에 미치는 인삼(人蔘)과 Tocopherol의 영향(影響) (Effects of Korean Ginseng and Tocopherol on Testosterone Content, Protein Metabolism and Stamina Promotion in Rats)

  • 인삼(人蔘)과 tocopherol이 흰쥐의 체중증가(體重增加), 혈청(血情)의 testosterone 함량(含量)의 변화(變化), Protein 대사(代謝) 및 스테미너 증진(增進)에 미치는 영향을 비교(比較)검토(檢討)하기 위하여 Srague-Dawley 계(系) 수컷 흰쥐에게 분말형태의 인삼(人蔘)(2.5 g/kg)과 ${\alpha}-tocopherol$(250mg/kg)을 일반사료에 섞어 6주간 투여(投與)하여 실험(實驗)한 결과(結果) 다음과 같은 결과(結果)를 얻었다. 1. 체중증가량(體重重增加量)은 인삼투여군(人蓼投輿群)과 tocopherol 투여군(投與群)이 모두 대조군(對照群)에 비하여 증가(增加)하였으나 통계적(統計的)인 유의성(有意性)은 보이지 않았다. 2. 혈청중(血淸中)의 testosterone 함량변화(合量變化)에 있어서는 인삼투여군(人蔘投與群)은 대조군(對照群)에 비하여 별다른 차이가 없었으나 tocopherol 투여군(投與群)에서는 유의적(有意的)인 증가(增加)를 나타내었다. (p<0.05). 3. 인삼투여군(人蔘投與群)은 대조군(對照群)에 비하여 albumin, ${\alpha}_1-globulin$, ${\alpha}_2$, ${\gamma}-globulin$ 및 A/G ratio에 있어서는 유의적(有意的)인 차이(差異)가 없었으나 총단백질(總蛋白質)과 ${\beta}-globulin$에 있어서는 유의적(有意的)인 증가(增加)를 나타내었다(p<0.005). 그리고 tocopherol 투여군(投與群)은 총단백질함량(總蛋白質含量)이 대조군(對照群)에 비하여 증가(增加)하였고 (P<0.005) albumin치(値)는 감소(減少)하였으나 통계적(統計的)인 유의성(有意性)은 없었다. 또한 ${\alpha}_1-$, ${\alpha}_2-$, ${\beta}-$, 및 ${\gamma}-globulin$ 함량(合量)은 모두 대조군(對照群)에 비하여 유의적(有意的)으로 증가(增加)하였으며(P<0.001) 이로 인하여 A/G ratio치(値)는 현저하게 감소(滅少)하였다(P<0.001). 4. 스테미너 증진효과를 보기 위하여 인삼(人蓼) 및tocopherol을 투여(投與)한 흰쥐를 동일(同一)한 조건(條件)에서 계속 수영시킨 후 지쳐서 죽을 때까지의 시간(時間)을 측정(測定)한 결과(結果) 모두 대조군(對照群)에 비하여 별다른 변화(變化)를 나타내지 않았고 개체(個騷)간의 차이가 매우 큰 것으로 나타났다.

Modulatory effects of $\alpha$- and $\gamma$-tocopherols on 4-hydroxyestradiol induced oxidative stresses in MCF-10A breast epithelial cells

  • The elevated level of circulating estradiol increases the risk of breast tumor development. To gain further insight into mechanisms involved in their actions, we investigated the molecular mechanisms of 4-hydroxyestradiol (4-$OHE_2$) to initiate and/or promote abnormal cell growth, and of $\alpha$- or $\gamma$-tocopherol to inhibit this process. MCF-10A, human breast epithelial cells were incubated with $0.1{\mu}M$ 4-$OHE_2$, either with or without $30{\mu}M$ tocopherols for 96 h. 4-$OHE_2$ caused the accumulation of intracellular ROS, while cellular GSH/GSSG ratio and MnSOD protein levels were decreased, indicating that there was an oxidative burden. 4-$OHE_2$ treatment also changed the levels of DNA repair proteins, BRCA1 and PARP-1. $\gamma$-Tocopherol suppressed the 4-$OHE_2$-induced increases in ROS, GSH/GSSG ratio, and MnSOD protein expression, while $\alpha$-tocopherol up-regulated BRCA1 and PARP-1 protein expression. In conclusion, 4-$OHE_2$ increases oxidative stress reducing the level of proteins related to DNA repair. Tocopherols suppressed oxidative stress by scavenging ROS or up-regulating DNA repair elements.

Contrasting rice sub-populations to tocols ratio associated with seed longevity

  • Understanding the mechanism(s) to overcome or prevent seed ageing deterioration during storage is of fundamental interest to seed physiologists. Vitamin E (tocols) is known as a key metabolite to efficiently scavenge lipid peroxy radicals which cause membrane breakdown resulting in seed ageing. However, in rice research this hypothesis has been tested for very few lines only without considering intraspecific variation in genomic structure. Here, we present a correlation study between tocols and seed longevity using a diverse rice panel. Seeds of 20 rice accessions held in the International Rice Genebank at the International Rice Research Institute, representing aus, indica, temperate japonica and tropical japonica subpopulations, were used for tocols analysis (quantification of ${\alpha}$-, ${\beta}$-, ${\gamma}$-, ${\delta}$-tocopherol/tocotrienol by ultra performance liquid chromatography) and storage experiments at $45^{\circ}C$ and 10.9% seed moisture content (sample taken for germination testing every 3 days up to 60 days). To examine interactions between DNA sequences and phenotype, the 700k high-density single-nucleotide polymorphism marker data-set was utilized. Both seed longevity (time for viability to fall to 50%; $p_{50}$) and tocols content varied across subpopulations due to heterogeneity in the genetic architecture. Among eight types of tocol homologues, ${\alpha}$-tocopherol and ${\gamma}$-tocotrienol were significantly correlated with $p_{50}$ (negatively and positively, respectively). While temperate japonica varieties were most abundant in ${\alpha}$-tocopherol, indica varieties recorded 1.3 to 1.7-fold higher ${\gamma}$-tocotrienol than those of other subpopulations. It was highlighted that specific ratio of tocol homologues rather than total tocols content plays an important role in the seed longevity mechanism.

어유와 대두유로부터 기능성 유지의 효소적 합성 (Enzymatic Synthesis and Characterization of Structured Lipids from Docosahexaenoic Acid - Enriched Fish oil and Soybean oil)

  • 본 연구에서는 DHA-enriched fish oil 과 대두유를 비율(w/w)을 달리하여 반응시키고 그 특성을 알아보았다. 어유의 비율이 증가할수록 trilinolein(LLL) peak는 감소하였고, 새로운 피크의 형성은 점차 증가하였다. 재구성 지질(어유 : 대두유 = 5 : 5, w/w, $55^{\circ}C$, 240, 180 rpm)의 지방산 조성 분석 결과 대두유에서 볼 수 없었던 DHA가 약 13% 증가한 반면, linoleic acid는 24.17% 감소하였으며 포화 지방산의 함량은 약 23%, 불포화 지방산의 함량은 약 77%이었다. 위치별 지방산 함량으로 sn-2 position은 약 linoleic acid 28%, oleic acid 22%, DHA 18% 그리고 palmitic acid 17% 이었다 재구성 지질의 tocopherol 분석 결과 $\alpha$, $\gamma$, and $\delta$-tocopherol 함량은 각각 23.58 mg/100 g, 32.28 mg/100 g, 17.94 mg/100 g이었다. 대두유와 비교하여 재구성 지질의 $\alpha$-tocopherol은 2.1배 증가하였으며, $\gamma$-tocopherol은 1.6배 감소하였다. 유지의 산패 정도를 알아보기 위하여 유도기간(induction period)을 측정한 결과 기질로 사용한 어유 보다는 대두유와 어유를 반응시킨 재구성 지질의 유도기간이 길었으며, 재구성 지질에 항산화제를 첨가한 군 간의 유도기간은 단일 항산화제를 첨가한 군이 최대 2.1배 길었다. 그리고 항산화제 첨가에 따른 향기 패턴을 알아본 결과 항산화제를 첨가하지 않은 그룹과 항산화제를 첨가한 그룹간에 향기 패턴에 차이가 있음을 확인하였다.
