Control of Methane Emission in Ruminants and Industrial Application of Biogas from Livestock Manure in Korea

  • Methane is known to be one of the major greenhouse gases. On a global scale, livestock farming may contribute 18% of total greenhouse gas emissions. Though methane contribution is less than 2% of all the factors leading to global warming, it plays an important role because it is 21 times more effective than carbon dioxide. Methane emission is a direct result of the fermentation process performed by ruminal microorganisms and, in particular, the archael methanogens. Reducing methane emission would benefit both ruminant production and the environment. Methane generation can be reduced by electron-sink metabolic pathways to dispose of the reducing moieties. An alternative way for methane control in the rumen is to apply inhibitors against methanogens. Generating methane from manure has considerable merit because it appears to offer at least a partial solution to two pressing problems-environmental crisis and energy shortage. An obvious benefit from methane production is the energy value of the gas itself. Control of methane emission by rumen microbes in Korea has mainly been focused on application of various chemicals, such as BES and PMDI, that inhibit the growth and activity of methanogens in the rumen. Alternatives were to apply long-chain polyunsaturated fatty acids and oils with or without organic acids (malate and fumarate). The results for trials with methane reducing agents and the situation of biogas production industries and a typical biogas plant in Korea will be introduced here.

Effects of Defaunation on Fermentation Characteristics and Methane Production by Rumen Microbes In vitro When Incubated with Starchy Feed Sources

  • An in vitro experiment was conducted to examine the effects of defaunation (removal of protozoa) on ruminal fermentation characteristics, $CH_4$ production and degradation by rumen microbes when incubated with cereal grains (corn, wheat and rye). Sodium lauryl sulfate as a defaunation reagent was added into the culture solution at a concentration of 0.000375 g/ml, and incubated anaerobically for up to 12 h at $39^{\circ}C$. Following defaunation, live protozoa in the culture solution were rarely observed by microscopic examination. A difference in pH was found among grains regardless of defaunation at all incubation times (p<0.01 to 0.001). Defaunation significantly decreased pH at 12 h (p<0.05) when rumen fluid was incubated with grains. Ammonia-N concentration was increased by defaunation for all grains at 6 h (p<0.05) and 12 h (p<0.05) incubation times. Total VFA concentration was increased by defaunation at 6 h (p<0.05) and 12 h (p<0.01) for all grains. Meanwhile, defaunation decreased acetate and butyrate proportions at 6 h (p<0.05, p<0.01) and 12 h (p<0.01, p<0.001), but increased the propionate proportion at 3 h, 6 h and 12 h incubation (p<0.01 to 0.001) for all grains. Defaunation increased in vitro effective degradability of DM (p<0.05). Production of total gas and $CO_2$ was decreased by defaunation for all grains at 1 h (p<0.05, p<0.05) and then increased at 6 h (p<0.05, p<0.05) and 12 h (p<0.05, p<0.05). $CH_4$ production was higher from faunation than from defaunation at all incubation times (p<0.05).

Survey on Nutritional Status of Pre-school Children in Asan Measured by Anthropometric and Nutrient Intake Analysis (아산시 거주 미취학 아동들의 신체계측 및 영양상태 조사)

  • The nutritional status of pre-school children is important for both physical growth and functional development. This study investigated the anthropometric, nutrient intakes and dietary quality of the pre-school children living in Asan for planning nutritional education program. The dietary intakes were measured by 2-day 24 hr recall recorded by children's mothers and the anthropometric data were collected by measuring children's weights, heights, and percent body fat at Asan Public Health Center. The children were 161 boys and 129 girls aged 5 and 6 years. When children's nutritional status was determined by their anthropometric status (underweight, normal weight and overweight) determined by z-score (normal range between -1.00 and 1.00), only 1.4% of the children belonged to underweight group while 55.7% and 42.9% belonged to normal and overweight groups, respectively. Significantly higher number of children belonged to overweight group for 5 year-olds. No differences were observed in nutrient intakes, nutrient adequacy ratio (NAR) and index of nutritional quality (INQ) values among nutritional status groups. Intakes of the most nutrients were adequate, but NAR and INQ of calcium and zinc were low. When nutrient intakes, NAR and INQ were compared by sex or age, all nutrient intakes were appropriate except calcium and zinc intakes of 6 year-olds. Nutrient intakes of 5 years were higher than those of 6 years. The mean of nutrient adequacy ratio (MAR) was 0.85 and mean INQ was 1.51 for all participants. The nutritional management program for pre-school children in Asan would be directed differently by age groups and education material should contain contents educating to consume moderate amount of food for 5 year-olds and to increase intake of foods with mineral nutrients such as calcium and zinc for 6 year-olds.

Utilization of Steam-treated Oil Palm Fronds in Growing Saanen Goats: II. Supplementation with Energy and Urea

  • The objective of this study was to evaluate the effect of protein and energy on goats fed oil palm fronds (OPF) as roughages. Twenty-four male Saanen goats aged between 7 and 8 months and weighing $23.4{\pm}1.6kg$ were used in a $2{\times}3$ factorial design. Factors were three levels of urea (3%, 4% or 5%) and two levels of energy (low energy (LE) or high energy (HE)). On average, all parameters measured, including dry matter intake (DMI), nutrient digestibility, digestible nutrient intakes, ruminal ammonia-N ($NH_3$-N), ruminal total volatile fatty acid (total VFA) and individual VFA concentrations (mM/L), microbial N supply, P/E ratio and N retention were higher for HE compared to LE diets. Significant (p<0.05) interactions were found between levels of urea and energy for nonstructural carbohydrate (NSC) and energy (DE) digestibilities, ruminal $NH_3$-N and total VFA concentrations. HE diets had higher N absorption and retention than LE diets. Interactions between urea and energy for plasma urea nitrogen (PUN), heat production (HP), and urine and faeces N excretion were significantly lower (p<0.05) for the HE diets than those recorded for the LE diets. The results indicated that supplementation of energy enhanced utilization of urea and resulted in higher animal performance as a consequence of improved ruminal fermentation, microbial yield and N balance. However, the optimal level of urea supplementation remained at 3% in the HE diet.

기체 흐름 기술을 이용한 원거리 대기압 질량분석 이미징 기술

  • 분석 방법의 간편함과 용이함의 장점은 물론, 시료 전처리 과정이 적어 시료물질의 임의 파괴나 훼손을 방지한다는 이유에서 최근 10년 간 많은 연구가 이루어지고 있는 대기압 질량분석 기술은 기압차이가 없는 대기압 분위기에서 질량분석이 이루어지기 때문에 시료를 질량분석기 입구 바로 앞에 스테이지를 설치하고서 시료를 이온화하는 경우가 대부분이다. 이 때문에 균질하지 않은 시료의 관심 영역을 모니터링하면서 질량분석을 하기에는 어려움이 있으며, 공간 정보를 추가한 질량분석 이미징에 한계가 있었다. 이에 본 연구팀은 질량분석기 입구에 챔버와 보조 펌프를 장착하여 강제로 기체 흐름 일으켜 시료로부터 발생한 이온을 질량분석기 입구로 유도하여, 원거리에서 시료를 이온화해도 질량분석기 입구까지 이온을 성공적으로 전달시키는 방법을 제안한다. 이를 이용하면 분석하고자 하는 시료를 현미경 스테이지 위에 위치시켜 분석하고자하는 부분을 현미경으로 확인하면서 질량분석을 할 수 있으며, 나아가 대기압 질량 분석 이미징 기술을 구현할 수 있다. 대기압 탈착/이온화원은시료에 열적 손상이 없는 조건으로 시편의 이온화 및 탈착 과정이 이루어지게 하기 위해 저온 대기압 헬륨 플라즈마 젯과 펨토초 레이저를 결합하여 대기압 이온화원을 제작하였다. 이온 전달관은 1/4" (6.35 mm) 외경의 60 cm 길이의 스테인리스 스틸관을 사용하여 질량분석기에서 약 60 cm 떨어진 현미경 위의 시료의 질량분석이 가능하게 했다. 보조 펌프의 계기압과 저온 대기압 헬륨 플라즈마 젯의 헬륨 기체의 유속을 변화시키면서 시료인 PDMS (polydimethylsiloxane) 의 질량 스펙트럼 (m/z 270.314) 세기를 관찰하여 최적의 이온 전달 조건을 찾았다. 추가로 현미경 스테이지에 정밀 2-D 자동 스캐닝 스테이지를 장착하여 질량분석 정보에 공간 정보를 더할 수 있는 질량분석 이미징 기술 방법을 개발하여 생체 시편의 질량분석 이미징을 얻었다.

  • We reported that amorphous silicon (a-Si) thin film provide sample plate exhibiting a multimodality to measure biomolecules by secondary ion mass spectrometry (SIMS) and laser desorption/ionization mass spectrometry (LDI-MS). Kim et al.1 reported that a-Si thin film were suitable to detect small molecules such as drugs and peptides by SIMS and LDI-MS. Recently, bacterial identification has been required in many fields such as food analysis, veterinary science, ecology, agriculture, and so on.2 Mass spectrometry is emerging for identifying and profiling microbiology samples from its advantageous characters of label-free and shot-time analysis. Five species of bacteria - S. aureus, G. glutamicum, B. kurstaki, B. sphaericus, and B. licheniformis - were sampled for MS analysis without lipid extraction in sample preparation steps. The samples were loaded onto the a-Si thin film with a thickness of 100 nm which did not only considered laser-beam penetration but also surface homogeneity. Mass spectra were recorded in both positive and negative ionization modes for more analytical information. High reproducibility and sensitivity of mass spectra were demonstrated in a mass range up to mass-to-charge ratio(m/z) 1200 by applying the a-Si thin film in mentioned above MS. Principle component analysis (PCA) - a popular statistical analysis widely used in data processing was employed to differentiate between five bacterial species. The PCA results verified that each bacterial species were readily distinguished and differentiated effectively from our MS approach. It shows a new opportunity to rapid bacterial profiling and identification in clinical microbiology. More details will be discussed in the presentation.

Shape optimization for partial double-layer spherical reticulated shells of pyramidal system

  • Triangular pyramid and Quadrangular pyramid elements for partial double-layer spherical reticulated shells of pyramidal system are investigated in the present study. Macro programs for six typical partial double-layer spherical reticulated shells of pyramidal system are compiled by using the ANSYS Parametric Design Language (APDL). Internal force analysis of six spherical reticulated shells is carried out. Distribution regularity of the stress and displacement are studied. A shape optimization program is proposed by adopting the sequence two-stage algorithm (RDQA) in FORTRAN environment based on the characteristics of partial double-layer spherical reticulated shells of pyramidal system and the ideas of discrete variable optimization design. Shape optimization is achieved by considering the objective function of the minimum total steel consumption, global and locality constraints. The shape optimization of six spherical reticulated shells is calculated with the span of 30m~120m and rise to span ratio of 1/7~1/3. The variations of the total steel consumption along with the span and rise to span ratio are discussed with contrast to the results of shape optimization. The optimal combination of main design parameters for six spherical reticulated shells is investigated, i.e., the number of the optimal grids. The results show that: (1) The Kiewitt and Geodesic partial double-layer spherical reticulated shells of triangular pyramidal system should be preferentially adopted in large and medium-span structures. The range of rise to span ratio is from 1/6 to 1/5. (2) The Ribbed and Schwedler partial double-layer spherical reticulated shells of quadrangular pyramidal system should be preferentially adopted in small-span structures. The rise to span ratio should be 1/4. (3) Grids of the six spherical reticulated shells can be optimized after shape optimization and the total steel consumption is optimized to be the least.

Determination of Optimal Dietary Sulfur Amino Acids Ratio Relative to Lysine for Growing Barrows and Gilts

  • This experiment was conducted to investigate the effects of dietary SAA (sulfur-containing amino acids) on growth performance, nutrient digestibility and blood urea nitrogen (BUN) content, and to determine the optimal SAA:lysine ratio for growing barrows and gilts. A total of 150 pigs (75 barrows and 75 gilts, Landrace${\times}$Yorkshire${\times}$Duroc) were assigned to 6 treatments with 5 replicates of 5 pigs per pen. All pigs were fed diets containing either 1.12 (for barrows) or 1.33% (for gilts) dietary lysine with increasing SAA levels (50, 55 and 60% of dietary lysine) in a $2{\times}3$ factorial design. Throughout the whole experimental period (15 to 54 kg body weight), there was no interaction between sexes and SAA:lysine ratios on ADG, ADFI and FCR. However, increasing the SAA:lysine ratio from 50 to 60% in a diet showed a trend to increase ADG and ADFI of barrows. None of differences in nutrient digestibilities except for calcium and phosphorus were observed and gilts showed higher digestibility of calcium and phosphorus (p<0.05). Among dietary SAA:lysine ratios, there were no differences in apparent nutrient digestibility. Mean values of the essential amino acids (EAA), non-essential amino acids (NEAA) and total amino acids (TAA) digestibilities were higher in gilts than barrows (p<0.01). However, no differences in mean value of EAA, NEAA and TAA digestibilities were observed among dietary SAA:lysine ratios. Between sexes and among SAA:lysine ratios, no significant difference in BUN concentration was observed. This study demonstrated that the optimal inclusion ratio of SAA:lysine was 55% and below 50% in barrows and gilts, respectively.

Recent Advanced Toxicological Methods for Environmental Hazardous Chemicals (환경 오염물질의 진보된 독성 평가 기법)

  • Recently, several new methods for the detection of genetic damages in vitro and in vivo based on molecular biological techniques were introduced according to the rapid progress in toxicology combined with cellular and molecular biology. Among these methods, mouse lymphoma thymidine kanase (tk) gene forward mutation assay, single cell gel electrophoresis (comet assay) and transgenic animal and cell line model as a target gene of lac I (Big Blue) and lac Z (Muta Mouse) gene mutation are newly introduced based on molecular toxicological approaches. The mouse lymphoma tk$\^$+/-/ gene assay (MOLY) using L5178Y tk$\^$+/-/ mouse lymphoma cell line is one of the mammalian forward mutation assays, and has many advantages and more sensitive than hprt assay. The target gene of MOLY is a heterozygous tk$\^$+/-/ gene located in 11 chromosome, so it is able to detect the wide range of genetic changes like point mutation, deletion, rearrangement, and mitotic recombination within tk gene or deletion of entire chromosome 11. The comet assay is a rapid, simple, visual and sensitive technique for measuring and analysing DNA breakages in mammalian cells, Also, transgenic animal and cell line models, which have exogenous DNA incorporated into their genome, carry recoverable shuttle vector containing reporter genes to assess endogenous effects or alteration in specific genes related to disease process, are powerful tools to study the mechanism of mutation in vivo and in vitro, respectively. Also in vivo acridine orange supravital staining micronucleus assay by using mouse peripheral reticulocytes was introduced as an alternative of bone marrow micronucleus assay. In this respect, there was an International workshop on genotoxicity procedure (IWGTP) supported by OECD and EMS (Environmental Mutagen Society) at Washington D. C. in March 25-26, 1999. The objective of IWGTP is to harmonize the testing procedures internationally, and to extend to finalization of OECD guideline, and to the agreement of new guidelines under the International Conference of Harmonization (ICH) for these methods mentioned above. Therefore, we introduce and review the principle, detailed procedure, and application of MOLY, comet assay, transgenic mutagenesis assay and supravital staining micronucleus assay.

Characterization of the nar Promoter of Escherichia coli to use as an inducible promoter in Wild-type host Agrobacterium.tumefaciens

  • In this study, the nar promoter of E. coli was characterized to see whether the nar promoter cloned onto pBBR122 can be used as an expression promoter of gram negative microbes. For this purpose, a plasmid with lacZ gene expressing ${\beta}-galactosidase$ instead of the structural genes of nar operon in a gram negative host strain(Agrobacterium.tumefaciens) was used to simplify an assay of induction of the nar promoter. The following effects were investigated to find optimal conditions: methods of inducing the nar promoter, optimal nitrate concentration, maximally inducing the nar promoter, the amount of expressed ${\beta}-galactosidase$ and induction ratio(specific ${\beta}-galactosidase$ activity after maximal induction/specific ${\beta}-galactosidase$ activity before induction). The following results were obtained from the experiments: the growth of Agrobacterium with E.coli nar promoter was not much affected by nitrate concentration in the shake-flask; induction of nar promoter was optimal when Agrobacterium was grown in the presence of 1% nitrate ion at the beginning of culture and when overnight culture was completely grown in the shake-flask before being transferred to other shake-flask; the amount of ${\beta}-galactosidase$ per cell and per medium volume was maximal when Agrobacterium was grown under aerobic condition to $OD_{600}$ of 1.7; then the nar promoter was induced under microaerobic and anaerobic condition made by lowering dissolved oxygen level(DO). After 2-3h of induction in the YEP medium selected as a main culture medium, the specific ${\beta}-galactosidase$ activity became about 17,000 Miller units in the fermentor cluture.
