A gene encoding thermostable cellulase A (TmCelA) was isolated from Thermotoga maritima. The open reading frame of TmCelA gene was 774 bp long which predicted to encode 257 amino acid residues with a molecular weight of 29,732 Da. To examine the biochemical properties, the TmCelA was overexpressed in E. coli BL21, and expressed protein was purified. The optimum temperature of recombinant TmCelA was 90-95 ℃, and the optimum pH of recombinant TmCelA was approximately pH 5.0. Recombinant TmCelA was stable at temperature below 90 ℃.
Purpose: Urinary cytology and cystoscopic exam are effective methods for diagnosis of transitional cell carcinoma(TCC). But the former shows drawbacks such as the need for a well-trained examiner, and wide imprecision related to the variability of microscopic exam; the latter is an invasive method. $UBC^{TM}$ test detects the epitope on specific cytokeratin fragments released from epithelium of bladder cancer by immunoradiometric assay. We compared $UBC^{TM}$ test with urinary cytology for diagnosis of TCC to evaluate the utility of $UBC^{TM}$ test. Materials and Methods: Eighty-four patients with hematuria were included in our study. $UBC^{TM}$ tests (IDL Biotech, Sweden) were assayed in mid-stream urine according to the ordinary assay protocol. Nineteen patients were confirmed as TCC by cystoscopic examination and underwent transurethral resection (Group A). Other patients had various benign urinary tract conditions (Group B). Samples were considered positive as the $UBC^{TM}$ concentration was greater than $12{\mu}g/L$. Results: $UBC^{TM}$ levels were significantly different between group A ($95.9{\pm}166.4\;{\mu}g/L$) and group B ($19.2{\pm}85.6{\mu}g/L$) (P<0.001). Sensitivity for diagnosis of TCC was 89.5% (17/19) in UBC test and 47.4% (9/19) in cytology (p<0.05). Specificity for diagnosis of TCC was 81.5% (53/65) in $UBC^{TM}$ test and 100% (65/65) in cytology. $UBC^{TM}$ test was significantly more sensitive in stage Ta, $T_1$ tumors (84.6 vs 38.5%, p<0.05) and in grade I (83.3% vs 16.7%, p<0.05) than cytology. $UBC^{TM}$ test showed a tendency to be more sensitive as the grade was higher (83.3% in Grade I, 90% in Grade II and 100% in Grade III). Conclusion: $UBC^{TM}$ test could be a useful method in distinguishing TCC from other benign genitourinary diseases. Moreover, $UBC^{TM}$ test could be an especially valuable marker for diagnosis of TCC in patients with early TCC of low grade TCC compared to urinary cytology. Therefore, mbined use of $UBC^{TM}$ test in association with cytology is helpful to overcome the limited sensitivity of cytology.
$Ho^{3+}/Yb^{3+}/Tm^{3+}$ tri-doped $NaY_{1-x}(WO_4)_2$ phosphors with proper doping concentrations of $Ho^{3+}$, $Yb^{3+}$ and $Tm^{3+}$ ($x=Ho^{3+}+Yb^{3+}+Tm^{3+}$, $Ho^{3+}$=0.04, 0.03, 0.02, 0.01, $Yb^{3+}$=0.35, 0.40, 0.45, 0.50 and $Tm^{3+}$=0.01, 0.02, 0.03, 0.04) were successfully synthesized via the microwave sol-gel route, and their upconversion properties were investigated. Well-crystallized microcrystalline particles showed fine and homogeneous microcrystalline morphology with particle sizes of $1-2{\mu}m$. The optical properties were comparatively examined using photoluminescence emission and Raman spectroscopy. Under excitation at 980 nm, the doped particles exhibited white emissions based on blue, green and red emission bands, which correspond to the $^1G_4{\rightarrow}^3H_6$ transitions of $Tm^{3+}$ in the blue region, the $^5S_2/^5F_4{\rightarrow}^5I_8$ transitions of $Ho^{3+}$ in the green region, the $^5F_5{\rightarrow}^5I_8$ transitions of $Ho^{3+}$, and the $^1G_4{\rightarrow}^3F_4$ and $^3H_4{\rightarrow}^3H_6$ transitions of $Tm^{3+}$ in the red region. The pump power dependence of the upconversion emission intensity and the Commission Internationale de L'Eclairage chromaticity coordinates of the phosphors were evaluated in detail.

Objectives: The aim of this study was to assess the physicochemical properties, cytotoxicity and penetration into dentinal tubules of ChlorCidTM Surf (3% sodium hypochlorite [NaOCl] with surfactant) in comparison to ChlorCidTM (3% NaOCl without surfactant). Materials and Methods: The physicochemical properties evaluated were pH, surface tension, free available chlorine (FAC) and contact angle. Cytotoxicity was evaluated in L929 fibroblasts exposed to the solutions by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide and neutral red assays. Assessment of penetration into dentinal tubules was performed by staining single-rooted permanent human teeth with crystal violet (n = 9), which were irrigated with the solutions and analyzed in cervical, middle and apical segments. Data were analyzed by one-way analysis of variance (ANOVA) and Tukey's post-test, 2-way ANOVA and Bonferroni's post-test or t-test (α = 0.05). Results: ChlorCidTM Surf and ChlorCidTM FAC values were close to those indicated by the manufacturer. ChlorCidTM Surf showed lower surface tension and contact angle on dentin, and higher pH than ChlorCidTM (p < 0.05). The penetration of ChlorCidTM Surf was higher in cervical and middle segments, compared with ChlorCidTM (p < 0.05). There was no difference in irrigant cytotoxicity (p > 0.05). Conclusions: ChlorCidTM Surf showed lower surface tension, lower contact angle on root canal dentin, higher penetration into dentinal tubules and more alkaline pH, compared with ChlorCidTM. However, both solutions showed similar cytotoxicity and FAC content.

This study was performed to compare the shear bond strength of primary enamel & dentin treated by AQ Bond $Plus^{TM}$ and G $Bond^{TM}$, recently developed 6th generation dentin bonding system, to that of Single $Bond^{TM}$ being widely used. Also by observing the resin tag under scanning electron microscope, Resin tags of each material were also observed under scanning electron microscope and compared to one another. The possibility of clinical application of All-in-One system which has an advantage to reduce chair-time for children with difficult behavior pattern was evaluated. The results obtained are as follows: 1. No statistically significant difference between groups was found in shear bond strength of primary enamel. 2. In primary dentin, the shear bond strength of AQ Bond $Plus^{TM}$ was $1.15\;{\pm}\;0.37\;MPa$, G $Bond^{TM}$ was $1.69\;{\pm}\;0.74\;MPa$ and Single $Bond^{TM}$ was $0.56\;{\pm}\;0.11\;MPa$. There were no statistical difference between AQ Bond $Plus^{TM}$ and G $Bond^{TM}$ and between G $Bond^{TM}$ and Single $Bond^{TM}$, whereas statistically significant difference was found between AQ Bond $Plus^{TM}$ and Single $Bond^{TM}$. 3. Under scanning electron microscope, resin tags observed in AQ Bond $Plus^{TM}$ and G $Bond^{TM}$ were very weak and tangled while strong and thick tags were shown with many lateral branches in Single $Bond^{TM}$. The result of the present study coupled with the advantages of less working time over the previous generation suggests that All-in-One system might be effectively used in adhesive dental procedures for primary teeth.

This paper describes the design overview of shared file system $SANique^{TM}$ and analyzes the performance evaluation results of I/O intensive stress test based on various cluster file system architectures. Especially, we illustrate the performance analysis for the comparison results between the $SANique^{TM}$ and the Linux file system EXT3 system that is used to generally in Unix world. In order to perform our evaluation, Oracle 10g database system is operated on the top of cluster file system, and we developed the various kinds of testing tools which are compiled by ESQL/C from Oracle. Three types of architectures are used in this performance evaluation. Those are the cluster file system $SANique^{TM}$, EXT3 and the combined architecture of $SANique^{TM}$ and EXT3. In this paper, we present that the results of $SANique^{TM}$ outperforms other cluster file systems in the overhead for providing the true sharing over the connecting server nodes.
The Landsat Thematic Mapper (TM) has suggested that spatial and spectral characteristics would be suited to evaluate water quality of lake. But, TM has not been commonly used for the analysis of in-land water quality, such as surface water temperature, chlorophyll-a, suspended sediments, and Secchi depth in domestic research. This paper summarizes the analysis of Landsat 5 - TM image collected on 22 Feb 1996 for evaluation of chlorophyll-a and surface temperature in the Lake Soyang. And, field measurements collected in the Lake Soyang were used to obtain water optical algorithms for calibration of satellite data. It is concluded that we can assess chlorophyll-a with remote sensing reflectance and surface temperature with thermal band in lake Soyang. However, surface temperature calculated with thermal band of Landsat TM are underestimated. Relationship between remote sensing reflectance and chlorophyll-a using the ratio of TM band 1 and band 3 is as follows; Y = 17.206 - 6.4711 * (Rrs(band1) / Rrs(band3)) $R^2$=0.8762 and, using the ratio of TM band 1 and band 2 as follows; Y = 57.77 - 35.771 * (Rrs(band1) / Rrs(band2)) $R^2$=0.8317.

In this study, effective multispectral Landsat TM band combinations for a merging with the high resolution IRS-1C PAN data using the IHS method to improve landuse accuracy is discussed. From the pre-classified image using the merged images with TM all six band images(with the exception of band 6 image) and PAN image, a sample data which has ten classes was generated. An evaluation of the overall classification accuracy for the representative seven merged images which were merged using each TM three-band images and IRS-1C PAN image by IHS method for the sample area. The increase in classification accuracy is most significant with the inclusion of two of TM4, TM5 and TM7 infrared band images. Especially, the largest increase(11.8 percent) in landuse classification accuracy were investigated when Landsat TM247 bands were merged with IRS-1C PAN data. The classification accuracy when TM three band image and PAN image were used without merging is higher than result of the case of using the merged images.

Phytase increases the availability of phosphate and trace elements by hydrolyzing the phosphomonoester bond in phytate present in animal feed. It is also an important enzyme from an environmental perspective because it not only promotes the growth of livestocks but also prevents phosphorus contamination released into the environment. Here we present a novel phytase derived from Turicimonas muris, TmPhy, which has distinctive structure and properties compared to other previously known phytases. TmPhy gene expressed in the Pichia system was confirmed to be 41 kDa in size and was used in purified form to evaluate optimal conditions for maximum activity. TmPhy has a dual optimum pH at pH3 and pH6.8 and exhibited the highest activity at 70℃. However, the heat tolerance of the wildtype was not satisfactory for feed application. Therefore, random mutation, disulfide bond introduction, and N-terminal mutation were performed to improve the thermostability of the TmPhy. Random mutation resulted in TmPhyM with about 45% improvement in stability at 60℃. Through further improvements, a total of three mutants were screened and their heat tolerance was evaluated. As a result, we obtained TmPhyMD1 with 46.5% residual activity, TmPhyMD2 with 74.1%, and TmPhyMD3 with 66.8% at 80℃ heat treatment without significant loss of or with increased activity.
Dryfilm method by using 3M Petrifilm$^{TM}$ has been examined to replace conventional agar method for isolation of microorganisms from foods. The objectives of the present study were to evaluate suitability of dryfilm method as a microbial isolation method and to determine the effect of antimicrobial agent on dryfilm for isolation of microorganisms from foods. Five different foods, milk, ground beef, fishery surimi, Takju and wheat flour were used to isolate the natural microflora in foods and the inoculated Escheri chia coli. Standard method agar (SMA, Difco) and Petrifilm$^{TM}$ aerobic count (PAC, 3M) were used to isolate total microorganisms from foods. Violet red bile agar (VRBA), brilliant green lactose bile (BGLB) broth and Petrifilm$^{TM}$ coliform count (PCC, 3M) were used to isolate coliforms from foods. E. coli broth (EC broth) and Petrifilm$^{TM}$ E. coli count (PEC, 3M) were used to isolate E. coli from foods. Acidified potato dextrose agar (APDA) and Petrifilm$^{TM}$ yeast & mold count (PYMC, 3M) were used to isolate yeasts and molds from foods. Total aerobic plate counts isolated from five different foods by SMA and PAC (3M) were riot significantly different each other at P<0.05 level and were highly correlated each other ($\geq$0.96). Mugwort extract as an antimicrobial agent did not affect microbial enumeratiion of Dryfilm. Significantly higher number of coliform colonies were formed on VRBA than PCC (3M) from ground beef, but they were not significantly different in coliform colonies from milk samples. PCC (3M) and BGLB were not significantly different for enumeration of coliforms in milk and beef samples. Significantly higher number of E. coli were isolated by EC broth than PEC from ground beef, but these were not significontly different for enumeration of E. coli from milk. Yeast and mold counts isolated from Takju and wheat flour by APDA and PYMC (3M) were not significantly different at P<0.05 level. These data indicate that dryfilm method by using 3M Petrifilm$^{TM}$ can be successively used as an alternative to conventional agar method for enumeration of microorganisms in various foods.
