답전윤환(畓田輪換) 체계(體系)에 따른 토양(土壤)의 층위별(層位別) 양분분포(養分分布) 및 양분수지(養分收支) (Effects of Paddy-Upland Rotation Systems on Nutrient Balance and Distribution in Soil Profile)

  • 1989~'93년(年)까지 5년(年)동안 답전수환시(畓田輸換時) 작부체계(作付體系)에 따른 토양(土壤) 층위별(層位別) 양분분포(養分分布)와 양분수지(養分收支)를 검토(檢討)한 결과(結果)는 아래와 같았다. 1. 질산태질소(窒酸態室素), 치환성가리(置換性加里) 및 유효인산(有效燐酸) 함량(含量)과 EC는 표층(表層) 0~20cm에서 약간 높았을뿐 토층하부(土層下部)로 이동집적(移動集積)이 적었고, 작부체계(作付體系)에 따른 표토중(表土中) 함량은 전이환(田轉換) 감자-배추>2년 수환(輸換) 감자-배추>전전환(田轉換) 대두(大豆)>수도연작구(水稻連作區) 순(順)으로 높아 처리별(處理別) 시비량을 반영(反影)하였다. 2. 치환성(置換性) 석회(石灰)와 고토(苦土) 함량 및 pH는 심토(深土)로 갈수록 높아졌고, 이러한 경향은 특히, 수도연작구(水稻連作區)에서 현저(顯著)하였다. 작부체계(作付體系)에 따라서는 전(全) 토층(土層)에서 전전환(田轉換) 대두(大豆)>전전환(田轉換) 감자배추>2년(年) 윤환(輪換) 감자-배추>수도연작구(水稻連作區) 순(順)이었고 처리별 식물체 흡수량(吸收量) 차이가 크게 영향하였다. 3. 전전환(田轉換) 감자-배추구(區)에서 질소(窒素), 인산(燐酸), 가리(加里)의 양분수지(養分收支)는 투입량(投入量)보다 탈취량(奪取量)(흡수량)이 각각(各各) 21.5, 26.8, 9.2kg/10a 많았으나 투입량(投入量)중 화학비료량(化學肥料量)은 질소(室素)와 가리(加里)의 경우 탈취량(奪取量)에 비해 적었던 반면 인산(燐酸)은 많았다. 4. 전전환(田轉換) 대두구(大豆區)의 양분수지(養分收支)는 질소(窒素), 인산(燐酸), 가리(加里) 각각(各各) -12.8, 4.1, -1.0kg/10a로 질소(窒素)와 가리(加里)의 투입량(投入量)이 탈취량(奪取量)보다 적었으나 질소(窒素)는 근류균(根瘤菌)의 질소고정(窒素固定)으로 결핍되지 않았던 반면 가리(加里)는 생육중기(生育中期)에 결핍증상(缺乏症狀)을 나타내었다.

Effect of continuous maize cultivation on soil condition and yield in Northern Laos

  • In Northern Laos, maize is cultivated in continuous cropping without fertilizer, fallowing nor any other soil conservation practice. It is expected that this inadequate management in maize cultivation will degrade soil and decrease yield. However, there is limited information about the change of soil condition and yield under continuous maize cultivation. We tried to evaluate the change of soil condition and yield under continuous maize cultivation in Northern Laos. Our study was conducted in farmer's flat and slope fields in Sainyabuli province where maize cultivation had been introduced earlier than the other provinces of Northern Laos. Our study was conducted in 21 fields in 2014, and in 19 fields in 2015. We analyzed grain yield and soil characteristic (total carbon (TC), total nitrogen (TN), available phosphorus (Av-P), exchangeable cation, pH, soil texture) at 3 places in each field. The 3 places were set at different elevation level (lower position, middle position, upper position) in slope fields. Further, the period of continuous maize cultivation and crop management practice were investigated. Then, by evaluating the relation between the period of continuous maize cultivation and yield and the soil characteristics, the effect of maize cultivation was estimated. Crop management practices were similar among the investigated fields. Maize was cultivated in rain season. Grain seed and cob were harvested on September or October, but shoot was left on the fields. No crop was cultivated during dry season. Fertilization and fallowing has never been conducted under continuous maize cultivation. On the other hand, the period of maize cultivation was different among the fields, and ranged from 2 years to 30 years. In the slope fields, as the period of continuous maize cultivation was longer, the contents of TC and TN were lower at all 3 positions, Av-P content was lower at the upper position, exchangeable potassium (Ex-K) content was lower at the middle and the upper positions. The other soil characteristics weren't related with the period of maize cultivation in the slope fields. In contrast, soil characteristics weren't related with the period of maize cultivation in the flat fields. Yield was lower as the period of maize cultivation was longer at the upper position of the slope fields. At middle position of slope fields, yield tended to be low with increase of the period of maize cultivation. In contrast, yield wasn't related with the period of continuous maize cultivation in flat fields. From the results about crop management, it was presumed that the period of maize cultivation was one of the main factors causing the difference of yield and soil characteristics among the fields. Therefore, from the result of yield and soil condition, it was considered that the continuous maize cultivation decreased soil productivity in the slope fields with decline of TC, TN, Av-P, Ex-K and yield at upper position of slope fields, and decline of TC and TN in the other positions in Sainyabuli province.

소세포폐암 환자의 암성 통증에 대한 전침치료의 즉각적 진통 효과 1례 (Acute Analgesic Effect of Electroacupuncture on a Cancer Pain in a Small Cell Lung Cancer Patient : a Case Report)

  • We report a female small cell lung cancer patient in the extensive stage(T3N3Mx). After 6 cycles of chemotherapy combined radiation therapy, she received inpatient Korean medical care including herbal medicine, acupuncture therapy and concurrent western oral medications of opioid analgesics and anti-anxiety agent. The chief complaint was right side thoracic wall pain which had started after chemotherapy and was not effectively controlled by analgesics. For this condition, we treated her with 2Hz of constant electrical stimulation on Jiaji (Ex-B2) points T5-T7 laterally (right) using three needles for 20 minutes once a day for 9 days. With every session of electrical acupuncture treatment, thoracic pain decreased acutely. Korean medicine treatments including Jiaji (Ex-B2) point stimulation might be tried for lung cancer patients with uncontrolled thoracic pain at least for the acute analgesic effect.

향장기성 두경부 편평세포암종의 미세잔존암 모델에서 GM-CSF 유전자를 이입시킨 제한복제성 헤르페스바이러스 벡터를 이용한 종양백신의 유전자 치료 (Gene Therapy Using GM-CSF Gene Transferred by a Defective Infectious Single-cycle Herpes Virus in Micro-residual Organotropic Head and Neck Squamous Cell Cancer Model)

  • Background and Objectives: The Herpes Simplex type 2 Defective Infectious Single Cycle virus (DISC virus) is attenuated virus originally produced as viral vaccines but are also efficient gene transfer vehicle. The main goals of this study were to examine the efficiencies of the gene transfer using DISC vectors for various head and neck squamous cell carcinoma cell lines and to evaluate the efficacy of vaccination with DISC virus carrying a immunomodulatory genes (GM-CSF) as cancer therapy in a organotopic oral cavity squamous cell cancer model. Materials and Methods : We determinated the gene transfer efficiency of DISC virus by x-gal stain method and proved gene and protein expression of DISC-GMCSF transfected SCCVII cells by RT-PCR and ELISA method. Also we evaluated the ex vivo vaccination effects of SCCVII/GMCSF (DISC-GMCSF transfected SCCVII vaccine) vaccine on preventing the recurrence of micro-residual tumor. After the vaccination of SCCVII/GMCSF, specific cytotoxic T-cell responses was evaluated by CTL assay. Results: At an MOI of 10 DISC virus showed 64-88% of transfection rates in various head and neck squamous cancer cell lines. SCCVII cells transduced by DISC virus vector (MOI=10) carrying the GM-CSF gene, produced 4.5 nanogram quantities of GM-CSF per $10^6$ cells. In vivo vaccination using tumor cells transduced ex vivo with DISC-GMCSF resulted in better protection rate against subsequent tumor recurrence in organotopic oral cavity cancer model. Although tumor free survival rate was not statistically significantly increased in vaccination group (p=0.078), tumor specific cytotocic T-cell responses were significantly increased in SCCVII/GMCSF vaccination group. Conclusion: These data demonstrate that; 1) The DISC virus vector is capable of efficient gene transfer to various head and neck squamous cancer cell lines, 2) GM-CSF secreting genetically modified tumor vaccine (SCCVII/GMCSF) efficiently protected against tumor recurrence in organotopic micro-residual oral cavity cancer model and produced tumor specific cytotoxic T-cell response. DISC virus-mediated, cytokine gene transfer may prove to be useful as a clinical therapy for head and neck cancers.

자생식물 Essential Oil 5 종의 항 아토피피부염 활성 연구 (A Study on the Activities of Five Natural Plant Essential Oils on Atopic Dermatitis)

  • 본 연구는 개똥쑥(Artemisia annua L., AA), 유자(Citrus junos Sieb. ex TANAKA, CJ), 산국화(Chrysanthemum boreale Makino, CB), 잣나무(Pinus koraiensis, PK), 금강송(Pinus densiflora for. erecta, PD) 총 5 종의 자생식물 essential oil의 항 아토피 효능을 확인하기 위한 실험이다. 항균 효과를 확인하기 위해 자생식물 essential oil 5 종을 Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans 총 4 종류의 균에 처리했다. 항염증 효과를 확인하기 위해 TNF-α와 IFN-γ (TI)를 처리한 HaCaT 세포에 5 종의 자생식물 essential oil을 처리했다. AA, CJ, CB, PK, PD은 10 mg/mL 농도에서 Candida albicans에 대한 항균 효과를 나타냈다. AA (1 ㎍/mL), CB (1 ㎍/mL), PK (0.1, 1 ㎍/mL)에서 thymus and activation-regulated chemokine (TARC) 생성량이 억제되었고, AA 및 PK (1 ㎍/mL)에서 macrophage-derived chemokine (MDC) 생성량이 억제되었으며, AA (0.1, 1 ㎍/mL), PK (0.1, 1 ㎍/mL)에서 IL-6 생성량이 억제되었다. AA, CB, PK essential oil의 항균 및 항염증 효과가 확인되었으며, 이에 아토피 피부염 완화에 기여할 수 있음을 기대한다.

12주간 수중걷기운동이 비만 노인 여성의 신체조성, hs-CRP, 인슐린 저항성 및 혈압에 미치는 영향 (Effect of aquatic walking exercise on body composition, hs-CRP, insulin resistance and blood pressure in obese elderly women)

  • 본 연구의 목적은 12주간 수중걷기운동이 비만 노인 여성의 신체조성, hs-CRP, HOMA-IR 및 혈압에 미치는 영향을 알아보기 위하여 실시하였다. 체지방률 30% 이상인 비만 노인 여성 중 운동군(10명), 대조군(10명)으로 총 20명으로 실시하였으며, 모든 검사항목은 동일한 방법과 조건으로 신체조성, hs-CRP, HOMA-IR 및 혈압을 사전, 사후 총 2회 측정하였다. 수중걷기운동은 12주간, 주 3회, 1회 50분, RPE 11-14 강도로 실시하였다. 측정변인에 대해 평균값과 표준편차(M±SD)를 산출한 후 사전 운동군과 대조군의 신체적 특성과 각 측정변인에 대한 동질성 검정을 위하여 독립 t-test를, 집단 내의 사전·사후 평균치 변화에 대한 차이 검증은 종속 t-test를, 집단 간 차이에 대한 주효과 검정 및 집단 간 시기 간 상호작용 효과는 반복측정 분산분석을 이용하였다. 본 연구결과 운동군에서 신체조성, 체지방률(p<.05), 인슐린(p<.01), HOMA-IR(p<.01) 및 수축기 혈압(p<.05)이 유의하게 감소하였다. 따라서 수중걷기운동이 비만 노인 여성의 비만 및 심혈관질환의 개선에 도움을 줄 수 있을 것으로 사료된다.

High-Level Expression of T4 Endonuclease V in Insect Cells as Biologically Active Form

  • T4 endonuclease V (T4 endo V) [EC 3. 1. 25. 1], found in bacteriophage T4, is responsible for excision repair of damaged DNA. The enzyme possesses two activities: a cyclobutane pyrimidine dimer DNA glycosylase (CPD glycosylase) and an apyrimidic/apurinic endonuclease (AP lyase). T4 denV (414 bp cDNA) encoding T4 en do V (138 amino acid) was synthesized and expressed using either an expression vector, pTriEx-4, in E. coli or a baculovirus AcNPV vector, pBacPAK8, in insect cells. The recombinant His-Tag/T4 endo V (rHis-Tag/T4 endo V) protein expressed from bacteria was purified using one-step affinity chromatography with a HiTrap Chelating HP column and used to make rabbit anti-His-Tag/T4 endo V polyclonal antibody for detection of recombinant T4 endo V (rT4 endo V) expressed in insect cells. In the meantime, the recombinant baculovirus was obtained by cotransfection of BacPAK6 viral DNA and pBP/T4 endo V in Spodoptera frugiperda (Sf21) insect cells, and used to infect Sf21 cells to overexpress T4 endo V protein. The level of rT4 endo V protein expressed in Sf21 cells was optimized by varying the virus titers and time course of infection. The optimal expression condition was set as follows; infection of the cells at a MOI of 10 and harvest at 96 h post-infection. Under these conditions, we estimated the amount of rT4 endo V produced in the baculovirus expression vector system to be 125 mg/l. The rT4 endo V was purified to homogeneity by a rapid procedure, consisting of ion-exchange, affinity, and reversed phase chromatographies, based on FPLC. The rT4 endo V positively reacted to an antiserum made against rHis-Tag/T4 endo V and showed a residual nicking activity against CPD-containing DNA caused by UV. This is the first report to have T4 endo V expressed in an insect system to exclude the toxic effect of a bacterial expression system, retaining enzymatic activity.

CORIUM COOLABILITY UNDER EX-VESSEL ACCIDENT CONDITIONS FOR LWRs

  • In the wake of the Three Mile Island accident, vigorous research efforts were initiated to acquire a basic knowledge of the progression and consequences of accidents that involve a substantial degree of core degradation and melting. The primary emphasis of this research was placed on containment integrity, with: i) hydrogen combustion-detonation, ii) steam explosion, iii) direct containment heating (DCH), and iv) melt attack on the BWR Mark-I containment shell identified as energetic processes that could lead to early containment failure (i.e., within the first 24 hours of the accident). Should the core melt fail the reactor vessel, then non-condensable gas production from Molten Core-Concrete Interaction (MCCI) was identified as a mechanism that could fail the containment by pressurization over the long term. One signification question that arose as part of this investigation was the effectiveness of water in terminating an MCCI by flooding the interacting masses from above, thereby quenching the molten core debris and rendering it permanently coolable. Successful quenching of the core melt would prevent basemat melt through, as well as continued containment pressurization by non-condensable gas production, and so the accident progression would be successfully terminated without release of radioactivity to the environment. Based on these potential merits, ex-vessel corium coolability has been the focus of extensive research over the last 20 years as a potential accident management strategy for current plants. In addition, outcomes from this research have impacted the accident management strategies for the Gen III+LWR plant designs that are currently being deployed around the world. This paper provides: i) an historical overview of corium coolability research, ii) summarizes the current status of research in this area, and iii) highlights trends in severe accident management strategies that have evolved based on the findings from this work.

식물의 희소성 평가를 위한 환경영향평가기법 개발 (Development of the Environmental Impact Assessment Techniques for the Grading of Rareness in Plant)

  • This study was conducted to develop the new Environmental Impact Assessment techniques for the grading of rareness in plant. 17 EISs which were submitted in 1996 were analyzed to know the problems in existent EIA techniques for the conservation of rare plants. Category of rare plant was reevaluated and evaluation technique for the grading of rareness in plant was developed. The results obtained from this study were as follows: 1. The evaluation technique of rareness in plant was not reflected in the current preparation provision for EIS. And t his fact also appeared in most of the EIS which were reexamined in this study. 2. The category of rare species which have been considered as the subject of protection in EIA have to be enlarged to more than legally protected species designated by MOE. 3. The taxonomic characteristics and status of species, characteristics of population, geographical characters, extent of threat and its possibility by man resulted from the habitat loss were investigated. Each item was endowed scores from 1 to 5 and all the scores were summed, and then this value was used to evaluate the order of conservation. 4. Conservation measures for the rare plants have to be applied according to the levels of rareness grade, and they are absolute conservation, in situ conservation, transplanting and ex situ conservation, post observation and none by the levels from A to E. 5. Considering the phenology of rare plants, investigation for the rare plants have to be made a1 least three times. 6. Size of the MVP(Minimum Viable Population), pollination ecology also have to be considered for the conservation of rare plants. And in the case of adopting ex situ conservation measure, the site and methods for the conservation of rare plants have to be proposed in detail.

적외선 체열 촬영을 이용한 안면홍조 진단의 절단값 산정 (The Cut Off Values for Diagnosing Hot flashes by Using Digital Infrared Thermographic Imaging)

  • Objectives: The purpose of this study is to find diagnostic points and define the cut off values of hot flashes by using digital infrared thermographic imaging. Methods: Thermographic images of 75 patients with hot flashes (HF, n=35) and non-hot flashes (NHF, n=40) were retrospectively reviewed. We used the temperature difference between Ex-HN3 and CV17, LU4, CV12, CV4 for diagnosing hot flashes. The temperature differences of between two groups were analysed using independent samples t-tests. The cut off values were calculated by received operating characteristic curve analysis. Analyses were undertaken using SPSS version 17.0. and p-value of <0.05 was considered significant. Results: The temperature difference Ex-HN3 and LU4 were the most significantly different between groups (p<0.001). Using receiver operating characteristic curve analysis, the sensitivity, specificity, and area under the curve were 65.7%, 72.5%, 0.729, respectively. The optimum cut off value was defined as $1.00^{\circ}C$. Conclusions: These results suggest that the digital infrared thermographic imaging is a reliable instrument for estimating hot flashes.
