The Formation of Trans Fatty Acids with Heat Treatment and Storage of Fats and Oils (II) -The contents and the kinds of Trans fatty acids- (유지의 가열 및 저장에 따른 Trans지방산 생성에 관한 연구 -제2보 ; Trans지방산 함량 및 조성 변화를 중심으로-)

  • In this study, the cis to trans isomerization of unsaturated fatty acids were investigated about Soybean oil (SBO), Corn germ oil (CGO), Cottonseed oil (CSO), Margarine (MG), Shortening (ST) incubated at $40{\pm}2^{\circ}C$ for 35 days and heated at $185{\pm}2^{\circ}C$ for 24 hours. The contents and kinds of trans fatty acids in each sample were determined by GLC after seperating by HPLC. The results were obtained as follows; 1. When samples were incubated at $40{\pm}2^{\circ}C$ for 35 days, the contents of total trans fatty acids were increased from $0.5{\sim}12.3%$ to $0.8{\sim}20.5%$. The kinds of trans fatty acids found were $t-C_{18:1},\;t,c-C_{18:2},\;t,t-C_{18:2},\;t,c,c-C_{18:3}$ in $SBO,\;t-C_{18:1},\;t,c-C_{18:2}$ in $CGO,\;t-C_{16:1},\;t-C_{18:1},\;t,c-C_{18:2},\;t,t-C_{18:2}$ in CSO. Processed oil such as MG and ST showed more complicated composition of trans fatty acids than SBO, CGO and CSO. $t-C_{18:1},\;t,c-C_{18:2},\;t,t-C_{18:2},\;t,c,t-C_{18:3},\;t-C_{20:1}$ were detected in ST. 2. In the case of heating at $185{\pm}2^{\circ}C$ for 24 hours, the contents of total trans fatty acid were $1.6{\sim}37%$. 2% in all samples. Heating made more remarkable isomerization than incubation $(40{\pm}2^{\circ}C)$. Specially, $c,c,t-C_{18:3}$ in $SBO,\;t,t,t-C_{18:3}$ in $MG,\;t,c,t-C_{18:3},\;t,t,t-C_{18:3}$ in ST were detected. 3. The total contents of trans fatty acids of processed oil were higher than vegetable oils. During incubation, trans fatty acids increased in the order of MG>ST>CGO>SBO>CSO, and during heat treatment, MG>ST>SBO>CGO>CSO.

Effects of Caprylic Acid and Cyclodextrin Complex on In vitro Fermentation Characteristics and Methane Production (Caprylic Acid와 Cyclodextrin 복합물이 In vitro 반추위 발효성상 및 메탄 생성에 미치는 영향)

  • This study was conducted to evaluate the effects of dietary addition of caprylic acid(CA)-cyclodextrin (CD) complex on in vitro fermentation characteristics, total gas and methane production. Experiment was done with six treatment groups; 1) no CA-CD complex(control), 2) CA 20 mg(T1), 3) CD 830 mg(T2), 4) CA-CD complex 425 mg(T3), CA-CD complex 850mg(T4), CA-CD complex 1,700 mg(T5). Ruminal pH, ammonia and total VFA concentrations of T2, T3, T4 and T5 were lower(P<0.05) than those of control and T1 for the 12h incubation. The increase in molar percentage of propionate was observed in T4 and T5 compared with control and T2 for the 8h incubation(P<0.05), however, the ratio of acetate to propionate was unchanged in all treatments. Total gas of T1 was lower than that of control, but T2, T3, T4 and T5 were higher compared with control for 12h incubation(P<0.05). If the methane ratio (as %) to total gas for all treatments was compared, T3, T4 and T5(CA-CD supplemented groups) averaged 2.7% whereas control, T1 and T2 showed 3.4, 2.8 and 5.1%, respectively. Therefore, according to these results, it might be concluded that supplementation of CA-CD complex could reduce methane production without disrupting ruminal fermentation.

Phantom-Validated Reference Values of Myocardial Mapping and Extracellular Volume at 3T in Healthy Koreans

  • Purpose: Myocardial T1 and T2 relaxation times are affected by technical factors such as cardiovascular magnetic resonance platform/vendor. We aimed to validate T1 and T2 mapping sequences using a phantom; establish reference T1, T2, and extracellular volume (ECV) measurements using two sequences at 3T in normal Koreans; and compare the protocols and evaluate the differences from previously reported measurements. Materials and Methods: Eleven healthy subjects underwent cardiac magnetic resonance imaging (MRI) using 3T MRI equipment (Verio, Siemens, Erlangen, Germany). We did phantom validation before volunteer scanning: T1 mapping with modified look locker inversion recovery (MOLLI) with 5(3)3 and 4(1)3(1)2 sequences, and T2 mapping with gradient echo (GRE) and TrueFISP sequences. We did T1 and T2 mappings on the volunteers with the same sequences. ECV was also calculated with both sequences after gadolinium enhancement. Results: The phantom study showed no significant differences from the gold standard T1 and T2 values in either sequence. Pre-contrast T1 relaxation times of the 4(1)3(1)2 protocol was 1142.27 ± 36.64 ms and of the 5(3)3 was 1266.03 ± 32.86 ms on the volunteer study. T2 relaxation times of GRE were 40.09 ± 2.45 ms and T2 relaxation times of TrueFISP were 38.20 ± 1.64 ms in each. ECV calculation was 24.42% ± 2.41% and 26.11% ± 2.39% in the 4(1)3(1)2 and 5(3)3 protocols, respectively, and showed no differences at any segment or slice between the sequences. We also calculated ECV from the pre-enhancement T1 relaxation time of MOLLI 5(3)3 and the post-enhancement T1 relaxation time of MOLLI 4(1)3(1)2, with no significant differences between the combinations. Conclusion: Using phantom-validated sequences, we reported the normal myocardial T1, T2, and ECV reference values of healthy Koreans at 3T. There were no statistically significant differences between the sequences, although it has limited statistical value due to the small number of subjects studied. ECV showed no significant differences between calculations based on various pre- and post-mapping combinations.

Study on brewing of sweet potato starch (고구마 전분질원료를 이용한 주류제조에 관한 연구)

  • We have been studied on brewing sweet starch. We obtained the results as follows ; 1) 5 strains, T-T-2, T-T-4, T-K-2, T-T-18, T-T-1, were the most available in view of fermentative power by capacity of $CO_2$. 2) 5 strains, T-T-4, T-T-2, T-T-1, T-T-3, T-K-2, produced capacity of alcohol more than 5.78%. 3) 6 strains, T-T-2, T-K-2, T-T-4, T-S-2, T-I-3, T-I-1, are available not only taste and flavour, but productive power of alcohol in sweet potato starch. 4) The form of 6 strains are long oval and round and most of them are similar to the other yeast in size. 5) In giant colony the color was cream color and cream buff, and T-K-2 was formed by $15{\times}12mm$ on diameter and by 3.5mm on high. 6) Optimum temperature of most of all strains is 25~ $300^{\circ}C$but T-K-4 is 28-30.deg.C. 7) Optimum pH is 3.4-4.6. 8) T-S-2 was died off at 65.deg.C, the other strains died $60^{\circ}C$. 9( Making Bun-kok with non-heated wheat bran .alpha.-amylase was more increased by 4.5-13.5 mg of glucose in reaction solution and .betha.-amylase more 1.6-3.4ml of N/10-$KMnO_4$ Solution than Bun-kok with heated wheat bran. 10) It seems that mycellium grows better than original in substance containing 0.4 ~ 1.2% of HCl. 11) Making Bun-kok to add 0.8% HCl, .alpha.-amylase was increased 9.93-20.7mg of glucose and .betha.-amylase ws increased 2.6~4.3ml of N/10-$KMnO_4$ solution to reaction solution. 12) 1.2%-HCl, or higher concentration, acts as inhibitor, in the meanwhile the concentration between 0.4~0.8% of HCl acts as activator. 13) We must make Bun-kok for 42 hours, at 28~$30^{\circ}C$) After we made Bun-kok using S-O-II and R-J-I one by one, Bun-kok which mix each other in equal quantity is increased more than original on enzyme acrivity. 15) Oxidation is the best way of refining sweet potato starch in N/10-phosphate buffer solution (pH 7.5). 16) When we prepared sweet potato starch, first pH was 3.0.

Gene Frequencies and Phenotypes of Transferrin C Subtypes and Haptoglobin in Korean Population (한국인집단의 Transferrin C Subtypes와 Haptoglogin Phenotypes의 분포와 유전자 빈도)

  • Genetic polymorphism of transferrin $(T_f)$ subtypes in Jeju population was studied by isoelectric focusing of human sera on polyacrylamide gels under high voltage, and haptolobin (Hp) polymorphism in Seoul and Jeju population was studied by polyacrylamide gel electrophoresis. Among 946 normal samples, three common types of transferrin, $T_{f}C_{1}, T_{f}C_{1}-C_{2} and T_{f}C_{2}$ were observed with some variants migrating slower than $T_{f}C$ subtypes, while among 139 patient (hepatitis) samples, only three common types were found. The gene frequencies were calculated as follows; in normal population, $T_{f}C^{1}$ was 0.7220; $T_{f}C^{2}, 0.2743; T_{f}D^{Jeju}, 0.0037$, and in patient population, $T_{f}C^{1} was 0.7194; T_{f}C^{2}, 0.2806$ respectively. Among 460 samples in Seoul and 502 in Jeju population, three types of haptoglobin, Hp 1-1, Hp 2-1 and Hp 2-2 were observed. The gene frequency of $Hp^1$ was 0.304, $Hp^2$, 0.696 in Seoul and in Jeju, $Hp^1$ was 0.269 and $Hp^2$, 0.731, respectively. The frequencies of the genes and the polymorphic phenotypes were discussed comparatively with the other populations.

Comparison of mDixon, T2 TSE, and T2 SPIR Images in Magnetic Resonance Imaging of Lumbar Sagittal Plane (요추 시상면 자기공명 영상검사에서 mDixon과 T2 TSE, T2 SPIR 영상의 비교 연구)

  • The purpose of this study was to compare and analyze the differences in scan time, signal-to-noise ratio (SNR), and contrast-to-noise ratio (CNR) in the third lumbar vertebral region including the back fat, spinal cord, and cerebrospinal fluid using the mDixon, T2 TSE, and T2 spectral pre-saturation with inversion-recovery (SPIR) techniques. With the factors affecting the SNR fixed, the lumbar sagittal plane images of 30 adults were compared on mDixon, T2 TSE, and T2 SPIR imaging tests. The test times for mDixon, T2 TSE, and T2 SPIR were 115 seconds, 60 seconds, and 60 seconds, respectively. The mDixon T2 images showed higher SNR than the T2 TSE images at the third lumbar vertebral region (p<0.05), lower SNR in the back fat and cerebrospinal fluid (p<0.05) areas, and comparable SNR in the spinal cord (p>0.05). The CNR between the third lumbar vertebral area and back fat was higher in the mDixon T2 images, and the CNR of the cerebrospinal fluid and spinal cord images was higher in the T2 TSE images (p<0.05). The mDixon T2 FS images CNR was lower for the 3rd lumbar vertebral body region and back fat than the T2 SPIR images, and higher for the spinal cord and cerebrospinal fluid images (p<0.05). The CNR between the third lumbar body and back fat areas was higher in the mDixon T2 FS images (p<0.05), and there was no difference in the CNR in the images of the cerebrospinal fluid and the spinal cord (p>0.05). It is difficult to determine whether the mDixon technique is superior to the conventional T2 TSE and T2 SPIR techniques in terms of test time, SNR, and CNR. This study was confined to patients with simple lower back pain and was limited by controlled experimental conditions. Studies using clinically applied protocols are warranted in the future.

Changes in Physicochemical Properties of Pork Patty with Dandelion Extract during Refrigerated Storage (민들레 추출액을 첨가한 돈육패티의 냉장저장 중 품질변화)

  • This study was carried out to investigate the effects of dandelion leaf, root and whole part extracts on the physicochemical characteristics of pork patties stored at $4^{\circ}C$ for 10 days. Four types of pork patties were evaluated: ice-water-added (T0), dandelion leaf extract (T1), dandelion root extract (T2) and dandelion whole part extract (T3). The pH decreased significantly during storage, but increased after 10 days of storage. The pH was highest in T0 during storage. The L value of T0 increased whereas T1, T2 and T3 did not significantly changed during storage. The a value decreased with a longer storage period, and the a values for T1, T2 and T3 were higher than that of T0 during storage. The b value did not change significantly during storage. The TBARS increased with a longer storage period, and the values for T0, T1, T2 and T3 were 0.87, 0.29, 0.47 and 0.31 mg MA/kg, respectively, after 10 days of storage. The VBN content value of T0 increased, but those of T1, T2 and T3 did not significantly change during storage. The water holding capacity of T0 decreased, but those of T1, T2 and T3 did not significantly change during storage. Cooking loss increased during storage and that of T0 was higher than those of T1, T2 and T3. Hardness and chewiness decreased while springiness and cohesiveness increased during storage.

The Detection of T-2 toxin in Serum and Organ of Mouse by ELISA (ELISA법에 의한 mouse의 혈청 및 조직중의 T-2 toxin의 검색)

  • In order to detect the T-2 toxin accumulation in the animal tissues, T-2 toxin, produced by Fusarium sporotrichioides M-1-1, was injected to mouse by 0, 1 and 2 mg per kilogram of body weight, respectively, and T-2 toxin extracted from serum and organs were analyzed by the indirected competitive ELISA. The indirect competitive ELISA established in the laboratory can be check less than 0.1 ppb level of T-2 toxin and average recovery of T-2 toxin spiked was 80~113% in animal samples such as serum, liver and kidney. After 6 weeks of treatment with 2 mg of T-2 toxin per kg body weight, T-2 toxin was accumulated in serum (133.0 ng/ml), liver(1.4 ng/g) and kidney(14.3 ng/g) of mouse injected with 2 mg of toxin per kg body weight.

The Effects of Live Yeast(Saaccharomyces cerevisiae) Supplementation on the Performance of Laying Hens (활성효모 첨가가 산란계의 생산성에 미치는 영향)

  • In order to study the effects of supplementation of live yeast(Saccharomyces cerevisiae) on the performance of laying hens, five experiments were conducted. Two experiment were conducted during summer period, one with 37 wk old Dekalb-Delta strain laying birds(Exp. 1) and the other one with 100 wk old molted Nick Chick Brown laying birds(Esp.2) . In each experiment, 240 birds were divided into 12 groups of 20 birds each and randomly distributed. Each of the two experimental diets(Control 71 and 0.05% live yeast supplemented 72) was fed to 6 groups for 4 wks in Exp.1 and 3 wks in Exp.2. Three experiments were conducted during winter period, Exp.2 with 54 wk old Hy-Line strain laying birds, Exp.4 with 52 wk old Hy-Line strain laying birds, and Exp.5 with 36 wk old broiler breeder(Indian River strain). In each experiment, 540 birds were divided into 18 groups of 30 birds each and randomly distributed. Each of the 3 experimental diets(Control:$T_1$0.05% live yeast supplemented:$T_2$ and 0.1% live yeast supplemented : T$_3$) was fed to 9 groups for 6 wks in Exp.3, 9 wks in Exp.4 and 4 wks in Exp.5. In Exp.4, Latin Square design was employed to determine the effects of switching feeds at 3 wk intervals. All hens were housed in cages of commercial farm and experimental diets were made with commercial layer feeds. In Experiment 1, egg production was significantly(P<0.05) higher in $T_2$. Feed intake was significantly (P<higher in 72 at 1st wk but 4 wk average was not significantly different. Feed efficiency was significantly(P<0.01) better in 72 at End wk but 4 wk average was not significantly different. Other parameters, such as weight, soft egg production, cracked egg production and mortality were not significantly different. In Experiment 2, egg production was significantly(P<0.05) higher in $T_2$. Feed efficiency was significantly (P<0.05 and P<0.01) better at End wk and 3rd wk but 3 wk average was not significantly different. Soft egg production was significantly(P<0.05) higher in 72. Other parameters were not significantly different. In Experiment 3, egg productions were significantly(P<0.05) different among treatments : $T_3$ was higher than $T_1$ and $T_2$ was higher than $T_1$. Egg weight of $T_1$ and $T_2$was significantly(P<0.05) heavier than $T_3$. Feed intake of $T_2$ and $T_3$ was significantly(P<0.05) higher than $T_1$ at 6th wk but overall average was not significantly different. Soft egg production were significantly(P<0.01) different among treatments:$T_1$ was higher than $T_3$ was higher than $T_2$. Feed efficiency cracked e99 Production and mortality were not signifcantly different. In Experiment 4, egg production tended to increase as the level of live yeast supplementation increased but they were not statistically different. In Experiment 5, egg production of broiler breeders of $T_3$ was significantly(P<0.01) higher than $T_1$. Feed intake of $T_3$ was significantly(P<0.05) greater than $T_1$ and $T_2$ at 3rd wk but overall average was not significantly different. Fertility and hatchability tended to be higher in the supplemented groups than in the control.

Investigation on the Characteristics Variation According to Air Supply Capacity in Layer Manure by Composting (공기공급량에 따른 산란계분의 퇴비화 특성변화에 관한 연구)

  • The composting of layer manure is economical and efficiently process. In this study, the variation of composting characteristics in layer manure was investigated according to air supply capacity. The fermented compost was added in layer manure and mixed with sawdust inside composting reactors. The level of air supply capacity was varied in the range of $50{\sim}200\;{\ell}/m^3/min$. During composting the temperature variations of composting piles was different the temperatures of composting piles for T-1 ($50\;{\ell}/m^3/min$) and T-2 ($100\;{\ell}/m^3/min$) were reached at $40^{\circ}C$ and $50^{\circ}C$ within 2 days, respectively. For T-3 ($150\;{\ell}/m^3/min$) and T-4 ($200\;{\ell}/m^3/min$), their temperatures was $60^{\circ}C$ within same days and maintained during 8 days. Water contents decreased according to the air supply capacity; 8.9%, 15.4%, 18.0% and 18.6% for T-1, T-2, T-3 and T-4. The weight ratios of T-1, T-2, T-3 and T-4 were reduced to 12.8%, 15.6%, 18.1% and 17.9%, respectively. The decreasing volumetric ratios of T-1, T-2, T-3 and T-4 were 18.0%, 21.0%, 22.3% and 22.0%. The oxygen discharge concentrations during composting were 12 ppm for T-1, T-2 and 9 ppm for T-3 and T-4. After composting, fertilizer components such as total nitrogen (TN) and phosphorous pentoxide ($P_2O_5$) were examined at each air supply capacity. Nitrogen contents of the T-1, T-2, T-3 and T-4 were 0.75%, 0.74%, 0.72% and 0.64%. Also, The contents of $P_2O_5$ were 0.35%, 0.40%, 0.38% and 0.42% for T-1, T-2, T-3 and T-4.
