The application of LFH-PCR(long flanking homology region-PCR) for Bacillus subtilis gene disruption is presented. Without plasmid- or phage-vector construction, only by PCR, based on a DNA sequence retrieved from B. subtilis genome data base, kanamycin resistance gene was inserted into two genes of B. subtilis involved in sporulation, spoIIIE and spoIIIG. The effect of gene disruption on subtilisin expression was examined and the sporulation frequency of two mutants was compared to that of the host strain. For this purpose, only 2 or 3 rounds of PCR were required with 4 primers. We first demonstrated the possibility of LFH-PCR for rapid gene disruption to characterize an unknown functional gene of B. subtilis or other prokaryote in the genomic era.
목적 : 멀티 생체신호를 이용한 지능형 실시간 심장과 호흡에 동기화 하는 시스템을 사용하여 심장자기공명영상을 수행하였다. 또한 멀티 생체신호를 측정하는 과정에서 유기될 수 있는 eddy current의 특성을 분석하였다. 대상 및 방법 : 멀티 생체신호 동기화 시스템에서는 심전도 신호와 호흡신호 외에 추가로 $SPO_2$ 정보를 수집하여 심장의 움직임에 동기화 하였다. 심장운동의 동기화는 심전도와 함께 $SPO_2$의 이차미분 신호를 이용할 수 있음을 보였다. 심장 동기화 과정에서 피검자의 움직임과 부정맥에 의해 발생할 수 있는 잘못된 동기신호를 평균 R-R 시간을 이용하여 제거하였다. 심장 영상화를 위한 시퀀스에서 경사자계의 스위칭에 의해 유기될 수 있는 eddy current의 특성을 분석하여 하드웨어 및 소프트웨어 필터로 차단하였다. 결과 : 제안된 동기화 시스템을 이용하여 심장과 호흡 운동에 동기화된 심장자기공명영상을 얻었다. 심전도 신호에서 피검자의 움직임과 부정맥에 의해 발생할 수 있는 동기신호를 차단하였고, 심장 영상화 과정에서 유기될 수 있는 eddy current를 제거하였다. 또한 심전도 신호를 보완하여 $SPO_2$의 이차미분신호를 이용하여 심장 영상이 가능함을 보였다. 결론 : 본 논문에서 제안한 멀티 생체신호 동기화 시스템은 심장자기공명영상을 위해 여러 생체신호 (심전도, $SPO_2$, 호흡)를 이용하여 실시간으로 심장과 호흡 동기화를 수행한다. 심전도에서 피검사자의 움직임과 부정맥에 의해 발생할 수 있는 동기 신호를 차단하였다. 경사자계의 스위칭에 의해 생체신호에 유기될 수 있는 eddy current를 분석하였고, 심장과 호흡 동기를 병행하여 피검사자가 자유롭게 호흡하면서 심장 영상을 얻을 수 있었다.
산소포화도 계측기는 파장대가 다른 두 개의 광원을 사용하여 신체 일부분에 빛을 투과시켜 나타나는 두 신호를 측정하여 측정된 신호로부터 산소포화도를 구하게 되며, 이러한 광학적 방법은 혈액내의 산소가 빛을 흡수하는 성질을 가지고 있기 때문이다. 혈중 산소포화도는 제 5의 생체신호라 불릴 정도로 현대 의학에서 중요한 위치를 차지하고 있다. 하지만 기존의 산소포화도 계측기는 사용자가 산소포화도를 보는 용도로서 사용하였고, 통신을 사용한 모니터링 기능이 부족하였다. 본 논문에서는 기존의 산소포화도 계측기에 블루투스 기능을 활용하여 외부의 Gateway와의 통신을 위해 의료기기 표준화 프로토콜인 IEEE11073-10404를 내장한 산소포화도 계측기를 설계하였다.
본 연구에서는 안정 상태에서 일반 공기 중에 산소 농도(21%) 환경에 비해 외부에서 고 농도(35%)의 산소 공급이 혈중 산소 포화도(SPO2), 심박동율(Heart rate), 피부전도도(Galvanic skin response)에 어떠한 영향을 미치는지를 검증하고자 한다. 35%의 고농도 산소를 2L/min의 양으로 일정하게 공급할 수 있는 산소 공급 장치를 이용하였고, 뇌 질병이 없는 5명의 대학생들이 피험자로 참여하였다. 21%의 비해 35% 산소 농도에서 모든 실험 참여자의 3분 동안의 평균 심박동율은 감소하였고 평균 혈중 산소 포화도는 증가하였다. 그러나 피부전도도는 차이가 없었다.
본 연구의 목적은 21%농도의 산소와 비교하여 30%의 고농도 산소 흡입이 사이클 등속 운동 중에 심박동률(heart rate)과 혈중 산소 포화도($SPO_2$)에 미치는 영향을 분석하는 것이다. 21%와 30% 농도의 산소를 각각 8liter/min의 양으로 일정하게 공급할 수 있는 산소 공급 장치를 이용하였다. 신체 건강한 10명의 남자 대학생($25.2{\pm}2.2$세)이 21%와 30%의 두 종류의 산소 농도를 대상으로 각각 두 번의 실험에 참여하였다. 실험은 안정(2분), $20{\pm}1km/h$의 사이클 등속 운동(10분), 회복(6분)의 세 구간(총 18분)으로 구성되어 있고, 21%또는 30%의 산소는 등속 운동과 회복 구간에만 공급되었다. 21%에 비해 30%산소를 흡입했을 때 사이클 등속 운동과 회복 기간 중에 심박동률은 감소하였고, 혈중 산소 포화도는 .차이가 없었다. 21%와 30% 두 그룹 사이에 동일한 운동부하가 주어졌기 때문에 혈중 산소 포화도는 차이가 없는 것으로 판단되고, 30%의 고농도 산소 흡입으로 동일운동 부하에 필요한 산소 요구량이 원활히 공급되고 있기 때문에 심박동률은 감소한 것으로 판단된다.
Onions contain antioxidant flavonoids and bioactive sulfur compounds. These substances are more abundant in the peel than in onion flesh. For this reason, whole onions including peels were added to soy milk to produce soy milk with whole onions (SWO), whereas peeled onions were added to soy milk to produce soy milk with peeled onions (SPO). The functional and antioxidant properties of these two kinds of soy milk were then analyzed and compared. Compared to control soy milk (CS) without onion powder, treated samples (to which freeze dried onion powder was added at 1.4, 1.6, 1.8, and 2.0%, respectively) showed significantly increased amounts of quercetin, isoflavone, and total phenol (p=0.05). The magnitude of the increase rose as the amount of added onion powder increased and when onion powder contained onion peels. With regards to antioxidant activity (DPPH and ABTS), SWO showed a greater value than SPO. The sensory evaluation scores of SWO and SPO were lower than CS for roughness and swallowing, as adding onion powder increased solid contents and viscosities. However, higher overall acceptability were obtained 1.8SPO and 1.8SWO.
High spatial and spectral resolution observations have been made over a sunspot (SPO 6403) with the Echelle Spectrograph at the Vacuum Tower Telescope, Sacramento Peak Observatory. Our observed spectra, scanned with SPO's fast microdensitometer, clearly show strengthening of $C_2$ lines in the penumbra relative to the photosphere and much weakening in the umbra in agreement with the predictions made by our molecular equilibrium calculations (Lee et al. 1981).
무포자형성 자연돌연변이 균주의 선발 및 특성검정 결과 ASI 2069 균주가 무포자이면서 수량성이 높으나 상품적 가치가 없어 원형질체 재생에 의한 단핵화로 Nh36 (neohaplont 36) 등 4균주를 분리하였다. 원형1호(ASI 2180) 및 무포자느타리간의 단핵교배 (Mon-Mon)를 시도한 결과 128교배조합수를 얻어 이 중 30균주를 특성 검정한 다음 자실체형성 및 포자비산량 조사에 의해 소포자형성 유망 13균주를 선발하였다. 무포자균주의 스크리닝법을 개발하기위해 포자형성 및 감수분열에 관여하는 helicase, DMC1(recombinase) 및 Spo11(topoisomerase II)유전자를 이용하여 PCR 증폭한 결과 단핵화균주 Nh36 및 교배체 2균주 중에서 Spo11유전자가 증폭되지않아 무포자 균주 스크리닝 방법으로 가능하리라 사료된다.
Purpose: This study was performed to examine the effect of action observation training on sit to-walk times in chronic stroke patients. Methods: Twelve stroke patients were randomly allocated to two groups, as follows: an action observation training (AOT) group and a scenery picture observation (SPO) group. The AOT group was engaged in video observation for 2 minutes 30 seconds with 12 ADL functional activities related to the sit-to-walk task: physical training was carried out in two repetitions lasting 2 minutes 30 seconds. The landscape imagery control group observed landscape picture for 2 minutes 30 seconds and then carried out the same physical training. The intervention involved a total of 12 sessions over 4 weeks, and each session lasted 30 minutes. The pre and post-tests measured the timed up and go test (TUG), Sit to stand test (STS), and Dynamic gait index (DGI). The Wilcoxon signed ranks test was used to compare pre-test and post-test result, and the Mann-Whitney U test was employed for comparison between groups. Results: The TUG time was significantly different between the AOT group and the SPO group. According to the findings, the experimental group's TUG was significantly decreased (p<0.05). In comparison of the two groups, there was no significant in STS or DGI between the AOT group and SPO group. Conclusion: This study showed that the intervention can be used to decrease TUG time. Our research suggests that action observation training has a positive effect on sit-to-walk times in patients with chronic stroke.
