Biotransformation of Ginsenoside Rd from Red Ginseng Saponin using Commercial β-glucanase (상업용 β-glucanase를 이용한 홍삼유래 사포닌으로부터 Ginsnoside Rd 의 생물 전환)

  • Bio-conversion manufacturing technology has been developed to produce ginsenoside Rd which is increasingly in demand as a cosmetic material due to various possibilities related to improving skin function. In order to convert ginsenoside Rb1 which is contained in red ginseng saponin (RGS) into Rd, several commercial enzymes were tested. Viscoflow MG was found to be the most efficient. In order to optimize the conversion of RGS to ginsenoside Rd by enzymatic transition was carried out using response surface methodology (RSM) based on Box-Behnken design (BBD). The main independent variables were RGS concentration, enzyme concentration, and reaction time. Conversion of ginsenoside Rd was performed under 17 conditions selected according to BBD model and optimization conditions were analyzed. The concentration of the converted ginsenoside Rd ranged from 0.3113 g/L to 0.5277 g/L, and the highest production volume was obtained under condition of reacting 2% RGS and 1.25% enzyme for 13.5 hours. Consequently, RGS concentration, enzyme concentration which is 0.05 less than p-value and among the interactions between the independent variables, the interaction between enzyme concentration and reaction time was confirmed to be the most influential.

Anti-diabetic effect and mechanism of Korean red ginseng extract in C57BL/KsJ db/db mice

  • Purpose: Ginseng is a well-known medical plant used in traditional Oriental medicine. Korean red ginseng (KRG) has been known to have potent biological activities such as radical scavenging, vasodilating, anti-tumor and anti-diabetic activities. However, the mechanism of the beneficial effects of KRG on diabetes is yet to be elucidated. The present study was designed to investigate the anti-diabetic effect and mechanism of KRG extract in C57BL/KsJ db/db mice. Methods: The db/db mice were randomly divided into six groups: diabetic control group (DC), red ginseng extract low dose group (RGL, 100 mg/kg), red ginseng extract high dose group (RGH, 200 mg/kg), metformin group (MET, 300 mg/kg), glipizide group (GPZ, 15 mg/kg) and pioglitazone group (PIO, 30 mg/kg), and treated with drugs once per day for 10 weeks. During the experiment, body weight and blood glucose levels were measured once every week. At the end of treatment, we measured Hemoglobin A1c (HbA1c), blood glucose, insulin, triglyceride (TG), adiponectin, leptin, non-esterified fatty acid (NEFA). Morphological analyses of liver, pancreas and white adipose tissue were done by histological observation through hematoxylin-eosin staining. Pancreatic islet insulin and glucagon levels were detected by double-immunofluorescence staining. To elucidate an action of mechanism of KRG, DNA microarray analyses were performed, and western blot and RT-PCR were conducted for validation. Results: Compared to the DC group mice, body weight gain of PIO treated group mice showed 15.2% increase, but the other group mice did not showed significant differences. Compared to the DC group, fasting blood glucose levels were decreased by 19.8% in RGL, 18.3% in RGH, 67.7% in MET, 52.3% in GPZ, 56.9% in PIO-treated group. With decreased plasma glucose levels, the insulin resistance index of the RGL-treated group was reduced by 27.7% compared to the DC group. Insulin resistance values for positive drugs were all markedly decreased by 80.8%, 41.1% and 68.9%, compared to that of DC group. HbA1c levels in RGL, RGH, MET, GPZ and PIO-treated groups were also decreased by 11.0%, 6.4%, 18.9%, 16.1% and 27.9% compared to that of DC group, and these figure revealed a similar trend shown in plasma glucose levels. Plasma TG and NEFA levels were decreased by 18.8% and 16.8%, respectively, and plasma adiponectin and leptin levels were increased by 20.6% and 12.1%, respectively, in the RGL-treated group compared to those in DC group. Histological analysis of the liver of mice treated with KRG revealed a significantly decreased number of lipid droplets compared to the DC group. The control mice exhibited definitive loss and degeneration of islet, whereas mice treated with KRG preserved islet architecture. Compared to the DC group mice, KRG resulted in significant reduction of adipocytes. From the pancreatic islet double-immunofluorescence staining, we observed KRG has increased insulin production, but decreased glucagon production. KRG treatment resulted in stimulation of AMP-activated protein kinase (AMPK) phosphorylation in the db/db mice liver. To elucidate mechanism of action of KRG extract, microarray analysis was conducted in the liver tissue of mice treated with KRG extract, and results suggest that red ginseng affects on hepatic expression of genes responsible for glycolysis, gluconeogenesis and fatty acid oxidation. In summary, multiple administration of KRG showed the hypoglycemic activity and improved glucose tolerance. In addition, KRG increased glucose utilization and improved insulin sensitivity through inhibition of lipogenesis and activation of fatty acid $\beta$-oxidation in the liver tissue. In view of our present data, we may suggest that KRG could provide a solid basis for the development of new anti-diabetic drug.

Dyeing of Cotton and Polyester/Cotton Blend with Disperse Dyes Using Sodium 2-(2,3-dibromopropionylamino)-5-(4,6-dichloro-1,3,5-triazinylamino)-benzenesulfonate

  • The cotton fabrics were pretreated by sodium 2-(2,3-dibromopropionylamino)-5-(4,6-dichloro-1,3,5-triazinylamino) benzenesulfonate (DBDCBS) at alkaline condition ofroom temperature and then dyed with four disperse dyes having amino groups (C.I. Disperse Yellow 9, C.I. Disperse Red 11, C.I. Disperse Blue 56 and C.I. Disperse Violet 1) at acidic condition of high temperature. A novel hetero-bifunctional bridge compound, DBDCBS, has two reactive groups such as dichloro-striazinyl group and ${\alpha},\;{\beta}$-dibromopropionylamido group. The first has reactivity towards hydroxy group of cellulosic fiber and the second shows reactivity towards amino groups of disperse dye containing amino groups. The results indicate that it is possible to dye polyester/cotton blend at one-bath dyeing using one kind of disperse dye containing amino groups. Therefore, two kinds of dyeing methods such as two-bath process one-bath dyeing and one-bath process one-bath dyeing were investigated and their dyeabilities were compared. The differences between these two methods were negligibly small so that perfect one-bath one-step dyeing of polyester/cotton blend by one kind of disperse dye was achieved.

Dyeing of Fibers Using Extract of Catalpa ovata Bark (오동나무 수피 추출액에 의한 섬유의 염색)

  • Recently there has been a growing interest in the use of natural dyes in textile applications. Natural dyes can exhibit better biodegradability and generally have a higher compatibility with the environment. In this study, the colorants of Catalpa ovata bark were extracted with water and analyzed by spectrophotometry for their main colorant species. Cotton, silk and wool fabrics have been dyed with aqueous extract of Catalpa ovata bark and their dyeabilities on the fibers were studied. Additionally the fastness to washing, drycleaning and light, and the effects on bacteriostatic rate and UV-B protection rate were also investigated. The major colorant of the extract of Catalpa ovata bark was shown to be 6-O-trans-caffeoyl-$\beta$-D-glucopyranoside. Cotton, silk and wool fabrics dyed with the extract of Catnip ovata bark were colored in yellowish red tint. The optimum dyeing condition of the colorants extracted from Catalp ovata bark was three repeated dyeing at $95^\circ{C}$ for 1 hr using post mordanting. For dyed silk and wool fabric, the fastness to washing were improved by mordanting, and the fastness to drycleaning were very outstanding. In case of wool fabric dyed with the extract of Catalpa ovata bark, the bacteriostatic rate was increased drastically by 98.0%, and UV-B protection rate was increased by 97.3%.

Molecular and Cultural Characterization of Colletotrichum spp. Causing Bitter Rot of Apples in Korea

  • Colletotrichum contains many important pathogens which cause economically significant diseases of crops like pepper, strawberry, tomato and apple. Forty four isolates were collected to characterize the diversity of Colletotrichum causing apple anthracnose in various regions of Korea. They were analyzed by random amplified polymorphic DNA (RAPD), internal transcribed spacer (ITS) of rDNA and partial $\beta$-tubulin gene DNA sequence, and culture characteristics on PDA and PDA-Benomyl. From the results of molecular analyses, 31 strains belonged to Colletotrichum gloeosporioides, ribosomal DNA group (RG) 4 of Moriwaki et al. (2002), 8 strains belonged to C. acutatum, A2 group of Talhinhas et al. (2005) and 5 strains to C. acutatum, A3 group of Talhinhas et al. (2005). Most isolates of C. gloeosporioides RG4 grew faster on PDA than strains of C. acutatum, A2 and A3 groups and most RG4 strains were sensitive to Benomyl. However, a few strains of RG4 grew slower and were resistant to Benomyl. On the basis of molecular characteristics, apple isolates of C. acutatum were clearly differentiated from red pepper isolates of the species, but apple isolates of C. gloeosporioides were not.

First Report of Botryosphaeria parva Causing Stem Blight on Rubus crataegifolius in Korea

  • In 2015, stem blight of Rubus crataegifolius was observed in Pohang, Korea. The symptoms began as dark red spots in the stem, which led to stem blight, then leaf blight, and eventually resulted in death. A fungal isolate was obtained from a symptomatic stem and incubated on a potato dextrose agar plate. The isolated fungus produced white, cloudy mycelia turned black in 3 days. Based on the morphological characteristics, the causal fungus was assumed to be Botryosphaeria sp. A pathogenicity test was conducted according to Koch's postulates. To identify the causal agent, the combined sequence of the internal transcribed spacer, ${\beta}$-tubulin, and translation elongation factor $1{\alpha}$ genes were used for phylogenetic analysis. Approximately 1,200 bp of the combined sequence clearly suggested that the isolated pathogen was Botryosphaeria parva. This is the first report on stem blight in R. crataegifolius caused by B. parva in Korea.

Cerebrospinal fluid analysis in 13 clinically healthy Beagle dogs; hematological, biochemical and electrophoretic findings

  • The purpose of this study is to define the normal findings of cerebrospinal fluid (CSF) of the clinically healthy Beagle dogs and to provide basic information in diagnosis of neurologic disorders. CSF obtained from 13 clinically healthy dogs was examined for total and differential cell counts, total protein concentration, glucose and lactate dehydrogenase (LDH) concentration, specific gravity, turbidity, and protein electrophoresis. On gross examination, CSF samples evaluated were clear and colorless. Few red blood cells and nucleated cells were present. The mean concentration of glucose and LDH examined were 65.8 mg/dl and 2.7 mg/dl, respectively. The cellular components of CSF samples based on differential counts were monocytes (41.9%), activated macrophages (35.8%), lymphocytes (20.0%), neutrophils (1.6%), and eosinophils (0.7%). The fractions of electrophoretic protein in CSF were albumin (52.7%), alpha-globulin (16.5%), beta-globulin (24.8%), and gamma-globulin (3.0%). Results of albumin quota were ranged from 0.15 to 0.38. In conclusion, this study provided normal composition of CSF in Beagle dogs.

Development and Immunochemical Properties of Two Monoclonal Antibodies Specific to Human Chorionic Gonadotropin

  • Using a hybridoma technique, spleen cells of Balb/c mice immunized with human chorionic gonadotropin (hCG) were fused with NS-1 mouse myeloma cells. Two hybrid cell lines, clones KS-8 and KS-19, secreting monoclonal antibodies to hCG, were isolated. KS-8 and KS-19 belong to the immunoglobulin $G_1$ subclass. With the aid of a double-antibody radioimmunoassay, it was established that the KS-8 monoclonal antibody recognizes an immunodeterminant of the $\beta$-subunit of hCG, whereas the KS-19 monoclonal antibody recognizes an epitope present on the $\alpha$-subunit of hCG. The KS-8 monoclonal antibody specifically reacts with human chorionic gonadotropin and shows cross-reactivity of less than 0.3% to other related human glycoprotein hormones. On the other hand, using a hemagglutination test based on antibody-induced agglutination of sheep red blood cells coated with hCG, It was shown that only the KS-19 monoclonal antibody was capable of inducing a positive reaction, although both monoclonal antibodies had similar binding capacity to the coated cells. The results from the dual screening procedures demonstrate that KS-8 and KS-19 monoclonal antibodies show high sensitivity in two different assays, and are hence useful for the qualitative and quantitative determination of hCG by both radioimmunoassay and hemagglutination inhibition tests.

Fabrication of Si quantum dots superlattice embedded in SiC matrix (SiC 매트릭스를 이용한 실리콘 양자점 초격자 박막 제조)

  • 다중접합 초 고효율 태양전지 제조를 위해 SiC 매트릭스를 이용한 실리콘 양자점 초격자 박막을 제조하고 특성을 분석하였다. $SiC/Si_{1-x}C_x$(x ~ 0.31)로 실리콘 양자점 초격자 박막을 Si과 C target을 이용한 co-sputtering법으로 초격자 박막을 제조하고, $1000^{\circ}C$에서 20분간 열처리를 하였다. high resolution transmission electron microscopy 사진으로 약1~7nm 크기인 양자점 생성과 분포 밀도를 확인할 수 있었으며, grazing incident X-ray diffraction (GIXRD)를 통해서 Si(111)과 $\beta$-SiC(111)이 생성되었음을 알 수 있었다. Auger electron spectroscopy (AES)측정에서 stoichiometric SiC층과 Si-rich SiC층의 Si 원자농도 (56%, 69%)와 C 원자 농도 (44%, 31%)를 알 수 있었으며, Fourier transform infra-red spectroscopy (FTIR)측정에서 SiC 픽의 위치가 767에서 $800cm^{-1}$으로 이동하는 것을 알 수 있었다.

  • This was aim to study the changes of components by different parts and maturity of tomato. Tomato include the green pigment chlorophyll, the yellow pigment-carotene. the red pigment lycopene and the other component. These colors of tomatoes are reported to have important functional roles to promote health in human. The main objective of this study was to define changes in the content of the four tomato maturation that is ascorbic acid, chlorophyll, carotene, lycopene. A ascorbic acid contents of tomato fruits was the highest at 50th day. Chlorophyll contents of tomatoes fruits was the highest at 10th days after flowering. ${\alpha}$-carotene contained low-quantity while all growth period. ${\beta}$-carotene contained the highest at 50th days after flowering, $1036.97{\mu}g$. Lycopene contained the highest at 50th days after flowering, $5800{\mu}g$.
