Objectives : This study was performed to investigate the effects of fermented Ephedra sinica (FMH) nano-extract compared Ephedra sinica (MH) on 3T3 L1 cell viability, lipid and glycometabolism, Obes rats' behavior and state-trait anxiety. Methods and materials : Each of 6 rats was divided into a normal diet, HFD, MH, and FMH. We fed a HFD group of rats a high fat diet and administered normal saline for 8 weeks. And we fed an experimental group of rats a high fat diet and administered an extract of Ephedra sinica and fermentated Ephedra sinica for 8 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. Ephedra sinica and fermentated Ephedra sinica was examined in effects of 3T3 L1 cell viability, lipid and glycometabolism, rats' behavior and state-trait anxiety. Results : 1. Ephedra sinica and fermentated Ephedra sinica didn't induce cytotoxicity at 3L3 L1 cell. 2. Fermentated Ephedra sinica by Lactococcus confusus inhibit $PPAR-{\gamma}$ activation and promote $TNF-{\alpha}$ activation. 3. In MH and FMH group, the body weight and FER decrease occured significantly than in HFD group. 4. No side effect of Ephedra sinica appeared in FMH group, so fermentated Ephedra sinica have a capacity of anti-anxiety. 5. Serum total cholesterol, triglyceride, HDL-cholesterol and creatine of HFD+Ephedra sinica, HFD+LO group were decreased compared with that of HFD group. Decreasing rates of sample group was significant(p<0.05). 6. In both MH and FMH group serum HDL was increased(P>0.05). 7. In MH group only TG was decreased, but in FMH group total cholesterol, TG, and glucose were decreased(P>0.05). Conclusions : Fermentated Ephedra sinica by Lactococcus confusus was superior to Ephedra sinica in capacity of anti-obesity. Further fermentated Ephedra sinica have no Ephedra sinica's side effect.
Objectives: This study was designed to investigate anti-obesity effects the improvement effects of Gyejibongnyeong-hwan and Gyejibongnyeong-hwan-Gangji-hwan (CIPPDF) in a ob/ob mouse model. Methods: Seven-week old mice (wild-type C57BL/6J and ob/ob) were used for all experiments. Wild-type C57BL/6J mice were used as normal group and obese ob/ob mice were randomly divided into 4 groups. a normal group given a standard diet, an obese control group given a standard diet with CIPP (300 mg/kg), CIPPDF (1) (300+300 mg/kg), CIPPDF (2) (300+600 mg/kg) respectively. After 10 weeks of treatment, body weight gain, feeding efficiency ratio, blood lipid markers, mRNA levels of genes involved in fatty acid ${\beta}-oxidation$ and lipogenesis in in-vivo, were examined. Results: 1. Body weight gain and Feeding efficiency ratio were significantly decreased in CIPPDF (1) compared with control. Fat mass was significantly decreased in CIPPDF (2) in EAT compared with control. 2. Consistent their effects on body weight gain and fat mass, circulating concentrations of LDL-cholesterol were decreased in CIPPDF (1), CIPPDF (2) groups compared with control. 3. MCAD mRNA levels of genes was increased in CIPPDF (1), CIPPDF (2) groups in the liver, epididymal adipose tissue compared with control. VLCAD mRNA levels of genes was increased in CIPPDF (1), CIPPDF (2) groups in the skeletal muscle compared with control. 4. $PPAR{\gamma}$ mRNA was decreased in CIPPDF (1) in the liver compared with control. SCD1 mRNA was decreased in CIPPDF (1), CIPPDF (2) groups in the epididymal adipose tissue compared with control. Conclusions: In conclusion, These results suggest that CIPPDF not only decrease feeding efficiency ratio, and LDL-cholesterol, but also reduce EAT fat mass contributing to the improvement of ovesity. CIPPDF also were increased in mRNA levels of genes involved in fatty acid ${\beta}-oxidation$ and decreased in mRNA levels of genes involved in lipogenesis.
제2형 당뇨병은 대사성 질환으로 간, 근육 그리고 지방 조직 세포에서 인슐린 작용의 장애로 나타나는 인슐린 저항성으로 혈당의 이용이 감소하여 혈당이 높아짐에도 불구하고 췌장의 베타세포에서 인슐린 분비가 충분하지 못할 때 유발된다. 서구에서는 비만 등으로 인해 인슐린 저항성이 증가하면 인슐린 분비가 높은 고인슐린혈증을 나타내어 당뇨병으로의 진전은 늦다. 하지만 우리나라를 비롯한 아시아의 사람들은 인슐린 저항성이 증가할 때 인슐린 분비가 충분치 못해 혈청 인슐린 농도가 정상인과 비슷하거나 더 낮은 상태에서 당뇨병으로 진전된다. 이러한 차이는 우리나라를 비롯한 아시아 사람들에게서 제2형 당뇨병의 발생이 급격하게 증가할 것이라는 보고되었다. 결국 당뇨병은 간, 근육 및 지방조직에서의 인슐린 작용의 장애와 췌장의 베타세포에서 인슐린 분비의 부족의 복합적인 장애에 의해서 나타나고 이것은 공통적으로 각 조직에서의 인슐린/insulin growth factor (IGF)-1 신호전달의 장애와 관련이 있다. 베타세포에서의 인슐린분비 자체는 인슐린/IGF-1 신호전달과 관계가 없지만 간접적으로 관련이 있다. 인슐린 분비능은 베타세포의 증식과 생존에 의한 베타세포의 양과 밀접한 관련이 있는데 인슐린/IGF-1 신호전달은 베타세포의 증식과 생존을 조절한다. 그러므로 혈당 조절에 관여하는 기능성 식품은 인슐린 작용을 향상시키는 인슐린 민감성 특성을 가지거나, 혈당이 높아질 때 인슐린 분비를 촉진시키는 insulinotropic 작용을 하는 성질을 가지고 있어야 하겠다. 전자의 대표적인 약은 1999년에 미국 FDA에서 승인 받은 peroxisome proliferator-activated receptor $(PPAR)-{\gamma}$ agonist 인 thiazolidinedione 계통의 약물인 troglitazone, pioglitazone, rosiglitazone 등이 있고, 후자는 2007년에 승인 받은 Exenatide는 glucagon like peptide (GLP)-1 agonist이다. 이 두 가지 약은 모두 자연계에 존재하는 동식물에서 유래된 것으로 식품에도 많이 다양한 종류의 인슐린 민감성 물질이나 insulinotropic 작용을 하는 물질이 함유되어 있을 것이다. 이러한 기능 이외에 혈당조절 약이나 식품으로 사용되는 것은 탄수화물의 소화를 방해하는 것으로 탄수화물 소화효소인 a-amylase 또는 maltase의 활성을 억제하여 식후 혈당의 급격한 상승을 방지하는 것이 있다. 우리나라 사람들은 탄수화물의 섭취가 너무 많아서 실제로 이러한 식품이나 약의 효능이 높지 않을 것이다. 혈당을 조절하는 기능성 식품은 이 세 가지 효능 중 일부를 가지고 있는 것이 될 수 있다. 이러한 기능을 스크리닝하기 위해서 3가지 단계를 거쳐야 한다. 먼저 시험관에서 또는 세포 실험을 통해서 앞서 언급한 3가지 기능을 가지고 있는 지 여부를 각각 조사한다. 이중에서 효과가 있는 것은 당뇨 동물 모델을 사용하여 in vivo에서 혈당 강하기능과 혈당 강하기전을 조사하는 실험을 한다. 효과가 있는 식품이 우리가 전통적으로 식품으로 섭취해 왔다면 독성 검사를 거쳐야 할 필요가 없지만 한약재이거나 특수 식품의 경우에는 in vivo 실험 전에 GLP 기관에서 반드시 독성 실험을 거쳐 독성 유무를 확인할 필요가 있다. 동물 실험에서 효과적인 것은 인체 실험을 거쳐 혈당 조절 기능성 식품으로 식약청에서 허가를 받을 수 있겠다. 결론적으로 식품에는 항당뇨 특성을 가진 물질들이 함유되어 있는 것들이 상당히 많다. 혈당 조절기능이 있는 기능성 식품으로 개발할 때 고려해야 할 것은 1) 그 양이 혈당 강하 기능성 식품으로 지정받을 수 있을 정도로 충분히 함유되어 있느냐, 2) 혈당을 강하시키는 기전이 단순히 당의 배설을 촉진시켜서 혈당을 저하시키는 것이 아니라, 인슐린 작용을 촉진시키거나, 포도당 자극에 의한 인슐린 분비를 촉진시키거나 탄수화물의 소화 흡수를 억제시킴으로 혈당을 강하시키는 지 등을 파악하는 것이다. 이러한 조건을 만족시키는 식품은 지속적으로 섭취할 때 당뇨병을 예방하거나 진전을 지연시킬 수 있는 혈당조절기능이 있는 기능성 식품으로 개발 가능성이 있겠다.

Patients with diabetes mellitus (DM) often suffer from diverse skin disorders, which might be attributable to skin barrier dysfunction. To explore the role of lipid alterations in the epidermis in DM skin disorders, we quantitated 49 lipids (34 ceramides, 14 free fatty acids (FFAs), and cholesterol) in the skin epidermis, liver, and kidneys of db/db mice, a Type 2 DM model, using UPLC-MS/MS. The expression of genes involved in lipid synthesis was also evaluated. With the full establishment of hyperglycemia at the age of 20 weeks, remarkable lipid enrichment was noted in the skin of the db/db mice, especially at the epidermis and subcutaneous fat bed. Prominent increases in the ceramides and FFAs (>3 fold) with short or medium chains ($LXR{\alpha}/{\beta}$ and $PPAR{\gamma}$, nuclear receptors promoting lipid synthesis, lipid synthesis enzymes such as elongases 1, 4, and 6, and fatty acid synthase and stearoyl-CoA desaturase were highly expressed in the skin and livers of the db/db mice. Collectively, our study demonstrates an extensive alteration in the skin and systemic lipid profiles of db/db mice, which could contribute to the development of skin disorders in DM.

The objective of this study was to determine the effect of different stocker management programs on skeletal muscle development and growth characteristics, satellite cell (SC) activity in growing-finishing beef cattle as well as the effects of SC-conditioned media on preadipocyte gene expression and differentiation. Fall-weaned Angus steers (n = 76; $258{\pm}28kg$) were randomly assigned to 1 of 4 stocker production systems: 1) grazing dormant native range (NR) supplemented with a 40% CP cottonseed meal-based supplement ($1.02kg{\cdot}steer^{-1}{\cdot}d^{-1}$) followed by long-season summer grazing (CON, 0.46 kg/d); 2) grazing dormant NR supplemented with a ground corn and soybean meal-based supplement fed at 1% of BW followed by short-season summer grazing (CORN, 0.61 kg/d); 3) grazing winter wheat pasture (WP) at high stocking density (3.21 steers/ha) to achieve a moderate rate of gain (LGWP, 0.83 kg/d); and 4) grazing winter WP at low stocking density (0.99 steers/ha) to achieve a high rate of gain (HGWP, 1.29 kg/d). At the end of the stocker (intermediate harvest, IH) and finishing (final harvest, FH) phases, 4 steers / treatment were harvested and longissimus muscles (LM) sampled for cryohistological immunofluorescence analysis and SC culture assays. At IH, WP steers had greater LM fiber cross-sectional area than NR steers; however, at FH, the opposite was observed (p < 0.0001). At IH, CORN steers had the lowest Myf-5+:Pax7+ SC density (p = 0.020), while LGWP steers had the most Pax7+ SC (p = 0.043). At FH, CON steers had the highest LM capillary density (p = 0.003) and their cultured SC differentiated more readily than all other treatments (p = 0.017). At FH, Pax7 mRNA was more abundant in 14 d-old SC cultures from HGWP cattle (p = 0.03). Preadipocytes exposed to culture media from proliferating SC cultures from WP cattle isolated at FH had more $PPAR{\gamma}$ (p = 0.037) and less FABP4 (p = 0.030) mRNA expression compared with NR cattle. These data suggest that different stocker management strategies can impact skeletal muscle growth, SC function, and potentially impact marbling development in growing-finishing beef cattle.

In this study, we investigated in vitro immune-enhancing and anti-obesity activity of Abelmoschus manihot roots (AMR) in mouse macrophage RAW264.7 cells and mouse adipocytes 3T3-L1 cells. AMR increased the production of immunostimulatory factors such as nitric oxide (NO), inducible nitric oxide synthase (iNOS), interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in RAW264.7 cells. The inhibition of toll like receptor (TLR) 2 and 4 blocked AMR-mediated production of immunostimulatory factors in RAW264.7 cells. In addition, the inhibition of mitogen-activated protein kinases (MAPKs) signaling pathway reduced AMR-mediated production of immunostimulatory factors. From these results, AMR is considered to have immune-enhancing activity through TLR2/4-mediated activation of MAPKs signaling pathway. In addition, AMR inhibited lipid accumulation and reduced the protein level such as CCAAT enhancer-binding protein alpha (CEBPα), peroxisome proliferator-activated receptor gamma (PPARγ), perilipin-1, adiponectin and fatty acid binding protein 4 (FABP4) associated with lipid accumulation in 3T3-L1 cells, indicating that AMR may have anti-obesity activity. Based on these results, AMR is expected to be used as a potential functional agent for immune enhancement and anti-obesity.

Objectives : Non-alcoholic fatty liver disease (NAFLD) is characterized by the accumulation of hepatic triglycerides (TG) that leads to inflammation and fibrosis. Crataegi Fructus ethanol extract (CE) is a korean traditional herb that used for digestive diseases. It has been investigated that CE has the effect that prevent hepatotoxicity caused by CCl4 or GaIN and regulate the inflammatory in several organs. However, a hypolipidemic effect of CF has not been reported. Methods : The purpose of this study is that examine the lipid accumulation inhibitory effect of CE on NAFLD. We checked the body and liver weight change of MCD-diet induced mice with/without administration of CE. The blood lipid levels of C57BL/6J mice were checked by biochemistry. Also we observed the liver histology of MCD-diet induced mice and investigate the molecular mechanisms in MCD-diet-induced NAFLD in C57BL/6J mice. Results : CE improved MCD-diet-induced lipid accumulation and TG and TC levels. Also, CE decreased hepatic lipogenesis such as SREBP-1, $C/EBP{\alpha}$, $PPAR{\gamma}$, ACC and FAS. Besides, we also found out that CE increased AMPK phosphorylation. These results indicated that CE has the same ability to activate AMPK and then reduce SREBP-1, and FAS expression, finally leading to inhibit hepatic lipogenesis and hepatic antioxidative ability. Conclusions : In this report, we found CE exerted a regulatory effect on lipid accumulation by decreasing lipogenesis in MCD-diet induced NAFLD model. Therefore, CE extract may be active in the prevention of fatty liver.

Purpose: Although activating thermogenic adipocytes is a promising strategy to reduce the risk of obesity and related metabolic disorders, emerging evidence suggests that it is difficult to induce adipocyte thermogenesis in obesity. Therefore, this study aimed to investigate the regulation of adipocyte thermogenesis in diet-induced obesity. Methods: Adipose progenitor cells were isolated from the white and brown adipose tissues of control diet (CD) or high-fat diet (HFD) fed mice, and fully differentiated white and brown adipocytes were treated with β-agonists or 18-carbon fatty acids for β-adrenergic activation or peroxisome proliferator-activated receptor (PPAR) activation. Results: Compared to the CD-fed mice, the expression of uncoupling protein 1 (Ucp1) was lower in the white adipose tissue of the HFD-fed mice; however, this was not observed in the brown adipose tissue. The expression of peroxisome proliferator-activated receptor gamma (Pparg) was lower in the brown adipose progenitor cells isolated from HFD-fed mice than in those isolated from the CD-fed mice. Norepinephrine (NE) treatment exerted lesser effect on peroxisome proliferator-activated receptor-γ coactivator (Pgc1a) upregulation in white adipocytes derived from HFD-fed mice than those derived from CD-fed mice. Regardless which 18-carbon fatty acids were treated, the expression levels of thermogenic genes including Ucp1, Pgc1a, and positive regulatory domain zinc finger region protein 16 (Prdm16) were higher in the white adipocytes derived from HFD-fed mice. Oleic acid (OLA) and γ-linolenic acid (GLA) upregulated Pgc1a expression in white adipocytes derived from HFD-fed mice. Brown adipocytes derived from HFD-fed mice had higher expression levels of Pgc1a and Prdm16 compared to their counterparts. Conclusion: These results indicate that diet-induced obesity may downregulate brown adipogenesis and NE-induced thermogenesis in white adipocytes. Also, HFD feeding may induce thermogenic gene expression in white and brown primary adipocytes, and OLA and GLA could augment the expression levels.
The uptake of glucose for metabolism and growth is essential to most animal cells and is mediated by glucose transport protein. In the glucose transport protein family, GLUT4 plays a key role in cellular glucose uptake stimulated by insulin in skeletal muscles and adipose tissue in rodents and human. In this studies, we reported the identification, characterization, and expression of Hanwoo GLUT4 gene. The Hanwoo GLUT4 cDNA includes a 1527 bp open reading frame encoding a protein of 509 amino acids. The GLUT4 amino acid sequences of the Hanwoo show strong conservation with the corresponding sequences reported in other species. The highest mRNA expression of GLUT4 was detected in heart and lower expression was detected in rib meat, sirloin, and colon. We confirmed the expression of GLUT4 in the subcutaneous and small intestinal adipose tissue using RT-PCR. To investigate the expression of GLUT4 in the bovine intramuscular adipose differentiation, fibroblast-like cells were isolated from the sirloin of Hanwoo bull aged 12 months by collagenase digestion of minced tissue and cultured with activators of PPAR gamma. We identified that GLUT4 mRNA expression decreased during differentiation of preadipocytes into adipocyte in Korean cattle. These results indicated that function of GLUT4 in bovine adipose tissue was different from that of mouse and human.

Diabetes has been one of major health risks in industrialized countries. Allium hookeri is a wild herb distributed in India and Myanmar. The root of the plant has been used as food and medicine in Southeast Asia. We investigated Allium hookeri extract improves type 2 diabetes mellitus in C57BL/KSJ db/db obese mouse. C57BL/KSJ db/db obese mouse arise out of Type 2 diabetes and we treated Allium hookeri methanol extract 400 mg/kg (AH 400), 800 mg/kg (AH 800), positive control group (thiazolidinedine;TZDs) were administered orally for 8weeks. AH treated group normalized lipid enzyme system (triglyceride, total cholesterol, HDL-cholesterol and LDL-cholesterol) and serum glucose, HbA1c and plasma insulin level. AH treated group recovered β-cell damage by hyperglycemia and fatty liver disease. AH treated group significantly up regulated expression of Peroxisome proliferator-activated receptor gamma (PPAR-γ), pyruvate dehydrogenase kinase4 (PDK4), Sterol regulatory element-binding protein 1c (SREBP 1) and fork head box O1 (FOX 01) proteins in C57BL/KSJ db/db obese mouse liver. And we found that AH treated group decreased hepatic malondialdehyde formation in C57BL/KSJ db/db obese mouse liver. These results indicate that Allium hookeri methanol extract might be a potential anti-diabetic agent and could be useful in the treatment of type 2 diabetes mellitus.

제 3 장 계약 당사자의 의무
제 10 조 (KISTI의 의무)
① 당 사이트는 이용고객이 희망한 서비스 제공 개시일에 특별한 사정이 없는 한 서비스를 이용할 수 있도록
하여야 합니다.
② 당 사이트는 개인정보 보호를 위해 보안시스템을 구축하며 개인정보 보호정책을 공시하고 준수합니다.
③ 당 사이트는 회원으로부터 제기되는 의견이나 불만이 정당하다고 객관적으로 인정될 경우에는 적절한 절차를
거쳐 즉시 처리하여야 합니다. 다만, 즉시 처리가 곤란한 경우는 회원에게 그 사유와 처리일정을 통보하여야
합니다.
제 11 조 (회원의 의무)
① 이용자는 회원가입 신청 또는 회원정보 변경 시 실명으로 모든 사항을 사실에 근거하여 작성하여야 하며,
허위 또는 타인의 정보를 등록할 경우 일체의 권리를 주장할 수 없습니다.
② 당 사이트가 관계법령 및 개인정보 보호정책에 의거하여 그 책임을 지는 경우를 제외하고 회원에게 부여된
ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.
③ 회원은 당 사이트 및 제 3자의 지적 재산권을 침해해서는 안 됩니다.
제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
