Regulation properties of phospholipase C$\delta$ cloned from Misgurnus mizolepis

  • Phosphoinositide-specific phospholipase Cδ (PLCδ) plays an important role in many cellular responses and is involved in the production of second messenger. The present study was conducted to characterize the catalytic and regulatory properties of the PLCδ of Misgurnus mizolepis (ML-PLCδ). The ML-PLCδ gene was cloned and expressed under according to the method of the previous report (Kim et al., 2004), and its recombinant protein was purified by successive chromatography using Ni2+-NTA affinity column. The recombinant ML-PLCδ showed a concentration-dependent PLC activity to phosphatidylinositol 4,5-bisphosphate (PIP2) or phosphatidylinositol (PI). Its activity was absolutely Ca2+-dependence, which was similar to mammalian PLCδ isozymes. The Ca2+ concentration yielding maximal activation of ML-PLCδ was 100 μM. However, the activity was decreased interestingly by a polyamine, such as spermine and spermidine. In vitro assay using cholate-micelle cell, ML-PLCδ activity was inhibited in dose-dependent manner by sphinogosine but increased by phosphocholine . In the lipid-binding assay, ML-PLCδ was strongly bound to LPA, PI(3)P, PI(4)P, PI(5)P, PI(3,5)P2, PI(4,5)P2, PI(3,4,5)P3 and PA, but it showed the low affinity to S1P, PI(3,4)P2 and PS. Taken together our results, it is suggested that the general catalytic and regulatory properties of ML-PLCδ are similar with those of mammalian PLCδ1 isozymes, but the N-terminal extended piscine phospholipase Cδ1 (ML-PLCδ) might reflect some distinctions in regulatory properties and inositol-lipid binding specificity between piscine ML-PLCδ and mammalian PLCδ isozymes.

Pseudomonas sp.가 생산하는 Inulinase에 관한 연구 -효소의 정제와 성질 - (Purification and Properties of Extracellular Inulinase of Pseudomouas sp.)

  • Pseudomonas sp.로 동정된 토양분리균의 inulinase를 ammonium sulfate 분획, DEAE Sephadex A-50 Chromatography, Sephadex G 100 및 Sephadex G200 gel여과 등의 과정을 거쳐 정제 한 결과 inulinase PI과 PII의 두 isoenzyme으로 분리되었으며 각각 약 24배와 17배 정제되어 단일단백질로 분리되었다. PI, PII 두 isoenzyme은 다같이 sucrose, raffinose 및 levan은 분해하지 못하고 inulin만을 endo-type로 분해 절단하는 $\beta$-2,1-fructanfructano hydrolase(EC 3.2.1.7)임이 밝혀졌다. 효소활성 최적온도는 두 효소가 같은데 비해 (55$^{\circ}C$), 최적 pH는 PI이 pH5.5, PII가 pH6.0으로 약간의 차이를 보였다. Inulin에 대한 Km값은 PI ; 2$\times$$10^{-3}$M, PII : 5$\times$$10^{-3}$M로써 PI효소가 PII보다 약간 높은 친화력을 보였다.

Pseudomonas sp.의 균체외 Endo-Inulinase 특성 (Characteristics of Extracellular Endo-Inulinase Produced by Pseudomonas sp.)

  • 토양분리균 Pseudomonas sp.가 생산하는 inulinase를 분리.정제하여 얻은 단일 단백질 효소 PI과 PII는 탄수화물 함량이 각각 15%와 2.4%인 당 단백질 형태의 endo-inulinase로서 두 효소가 모두 촉매활성에 필수적인 tryptophan 잔기를 가지고 있었다. 분자량은 PI 210,000, PII 170,000으로 측정되었다. 1mM pCMB 존재에 의해 두 효소가 약80% 정도의 활성저해를 보였으나 5mM cysteine 또는 1mM dithiothreitol을 첨가하면 효소활성이 거의 완전 회복되는 특성을 나타내었다. 최종 가수분해산물인 fructose(1mM)에 의해 PI, PII 효소가 각각 15% 정도의 활성저해를 받는 반면 Co$^{+2}$ 이온은 50~60%의 높은 활성화 효과를 보였다. 두 효소는 pH 4.0~7.5사이에서 매우 안정하였으며, 열에 대하여서도 비교적 안정하여 6$0^{\circ}C$, 120분 가열에 의해 PI이 약 27%, PII가 약 40%의 실활을 나타낼 뿐이다. 또한 60units의 효소를 사용, 2% inulin을 5$0^{\circ}C$에서 72시간 가수분해 시켰을 때 PI약 70%, PII약56%의 기질 분해율을 보였다.

Point Mutations in the Split PLC-γ1 PH Domain Modulate Phosphoinositide Binding

  • A number of signaling molecules contain small pleckstrin homology (PH) domains capable of binding phosphoinositides or proteins. Phospholipase C (PLC)-${\gamma}1$ has two putative PH domains, an $NH_2$-terminal (PH1) and a split PH domain ($nPH_2$ and $cPH_2$). We previously reported that the split PH domain of PLC-${\gamma}1$ binds to phosphatidylinositol 4-phosphate (PI(4)P) and phosphatidylinositol 4,5-bisphosphate (PI(4,5)$P_2$) (Chang et al., 2002). To identify the amino acid residues responsible for binding with PI(4)P and PI(4,5)$P_2$, we used site-directed mutagenesis to replace each amino acid in the variable loop-1 (VL-1) region of the PLC-${\gamma}1$ $nPH_2$ domain with alanine (a neutral amino acid). The phosphoinositide-binding affinity of these mutant molecules was analyzed by Dot-blot assay followed by ECL detection. We found that two PLC-${\gamma}1$ nPH2 domain mutants, P500A and H503A, showed reduced affinities for phosphoinositide binding. Furthermore, these mutant PLC-${\gamma}1$ molecules showed reduced PI(4,5)$P_2$ hydrolysis. Using green fluorescent protein (GFP) fusion protein system, we showed that both $PH_1$ and $nPH_2$ domains are responsible for membrane-targeted translocation of PLC-${\gamma}1$ upon serum stimulation. Together, our data reveal that the amino acid residues $Pro^{500}$ and $His^{503}$ are critical for binding of PLC-${\gamma}1$ to one of its substrates, PI(4,5)$P_2$ in the membrane.

  • 수목(樹木)에 있어서 내건성(耐乾性)의 크기를 진단(診斷)하기 위하여 P-V 곡선법(曲線法)에 의해 얻은 잣나무와 젓나무지엽(枝葉)(shoot)의 내적(內的) 수분특성인자(水分特性因子) ${\pi}_o$, ${\pi}_p$, $E_{max}$, $N_s/DW$, $V_o/W_s$, $RWC_{(tlp)}$ 등의 계절변화(季節變化)를 측정고찰(測定考察)하였다. 그 결과(結果)는 다음과 같다. 1) 잣나무지엽(枝葉)의 최대포수시(最大飽水時)의 침투압(浸透壓) ${\pi}_o$는 -1.2~-1.6 MPa, 젓나무지엽(枝葉)의 ${\pi}_p$는 -1.4~-1.7 MPa의 계절변화(季節變化)를 나타냈다. 2) 초기원형질분리점(初期原形質分離點)에 있어서 잣나무지엽(枝葉)의 침투압(浸透壓) ${\pi}_p$는 -1.8~-2.1MPa, 젓나무지엽(枝葉)의 ${\pi}_p$는 -1.6~-2.1MPa의 계절변화(季節變化)를 나타냈다. 3) 초기원형질분리점(初期原形質分離點)에 있어서 잣나무지엽(枝葉)의 상대함수율(相對含水率) $RWC_{(tlp)}$는 70~77%, 젓나무지엽(枝葉)의 $RWC_{(tlp)}$는 69~85%의 계절변화(季節變化)를 나타냈다. 4) 최대팽창시(最大膨壓時)의 잣나무지엽세포군(枝葉細胞群)의 탄성계수(彈性係數) $E_{max}$는 2.2~6.3 MPa, 젓나무의 $E_{max}$는 3.1~7.9 MPa의 계절변화(季節變化)를 나타냈다. 5) 잣나무지엽(枝葉)의 건물중(乾物重)에 대한 osmol 수(數) $N_5/DW$는 0.5~1.3, 젓나무지엽(枝葉)의 경우는 0.3~1.0의 계절변화(季節變化)를 나타냈다. 6) 잣나무지엽(枝葉)의 총함수량(總含水量)(symplasmic과 apoplastic water의 합계(合計))에 대한 총침투수량(總浸透水量) $V_o/W_s$는 55~65%, 젓나무지엽(枝葉)의 $V_o/W_s$는 40~65%의 계절변화(季節變化)를 나타냈다. 이상(以上)의 ${\pi}_o$, ${\pi}_p$, $E_{max}$, $N_s/DW$, $V_o/W_s$, $RWC_{(tlp)}$값의 특성(特性)을 고찰(考察)할 때 젓나무지엽(枝葉)이 잣나무지엽(枝葉)보다 내건성(耐乾性)이 약간 강한 수종(樹種)임을 알았다.

Comparison of Water Relations of Three Cultivated Pleurotus Species and Trichoderma Green Moulds

  • The effect of ionic osmotic potential (Ψ$\pi$), and matric potential (Ψm) in the range of -0.2 to -4.0 Mpa on mycelial growth of three species of Pleurotus (P.florida, P.ostrenatus and P.safor-caju) were determined over a range of temperature (15-3$0^{\circ}C$) on a 2% malt extract agar medium and compared with the Ψ$\pi$ effect on growth of two strains of Trichoderma green mould. With the ionic solute KCl, optimun Ψ$\pi$for growth was -0.2 MPa for P.floreda and in the range of -0.2 to -0.5 MPa, with slight growth at -3.0 MPa and with nogrowth at -4.0 MPa. Of the species of Pleurotus, P.florida grew signigicantly slower than the other two species. Growt of the species of Pleurocus was significantly slower when water potential (Ψ$\omega$) was modified matrically with polyethylene glycol (PEG) 8000 then osmotically with KCl. They were also more sensitive to changes in Ψm than Ψ$\pi$The optimum Ψm of the Pleurotus was -0.5 Ψm, with no growth below -3.0 MPa. Of the species of Pleurotus, P.florida was most sensitive and P.sajor-caju was more tolerent to lowered Ψ$\pi$,but P.sajor-caju was most sensitive to lowered Ψm. The growth rate of the Trichoderma green mould strains was much faster than that observed for the Pleurotus spp. Optimum growth for bot strains of Trichoderma was in the range of -0.2 to -0.5 MPa. Strain CNU 503 was more tolerant to water stress than strain CNU 501. Both strains were able to grow up to 30% of optimum growth at -4.0 MPa at 25-3$0^{\circ}C$.

$\beta$-Mercaptoethanol과 Cysteamine 첨가와 소 난관상피세포 공동배양이 소 체외수정란의 체외발육과 세포내 Glutathione 농도 변화에 미치는 영향 (Effect of $\beta$-Mercaptoethanol and Cysteamine With Bovine Oviduct Epithelial Cells on Development and Intracellular Glutathione Concentrations of Bovine IVM/IVF Embryos)

  • The objective of this study was to investigate the effects of thiol compounds with bovine oviduct epithlial crlls(BOEC) co culture on development and intracellular glutathione(GSH) concentrations of bovine embryos derived from IVM /IVF oocytes. In experiment 1 and 2, embryos developed to 2~8 cell stage after in vitro fertilization were co-cultured with BOEC in CR$_1$aa with or without $\beta$-mercaptoethanol($\beta$-ME) and cysteamine. The percentage of embryos that developed to morulae and blastocysts in 0,10, 25 and 5O$\pi$M $\beta$-ME with BOEC was 48.1, 64.0, 72.9 and 75.9%, respectively. Twenty-five and 5O$\pi$M $\beta$-ME groups were significantly higher than in 0 and 1O$\pi$M $\beta$- -ME groups(P$\pi$M cysteamine with BOEC was 50.0, 53.2, 72.0 and 66.7%, respectively. Fifty $\pi$M cysteamine group was significantly higher than any other groups (P$_4$aa with 0 and 5O$\pi$M $\beta$-ME or cysteamine were 68.5, 77.8, 78.7 and 80.0pM, respectively. Fifty $\pi$M $\beta$-ME group was significantly higher than that of control(P<0.05), but cysteamine group was not. Cell numbers of blastocysts were not difference in all experimental groups. These experiments indicate that $\beta$-ME and cysteamine with BOEC co-culture can affect the development and intracellular GSH concentrations of bovine embryos produced by IVM /IVF docytes.

  • 비선형 Hammett 관계를 고찰하기 위해서 $XC_6H_4CH_2Cl\;(X=H,\;p=CH_3,\;p-NH_2,\;p-NO_2)$와 benzyl radical, cation, anion에 대해서 CNDO/2 계산을 행하였다. 이로부터 얻은 주요 결론은 다음과 같다. 1. 염화벤질은 C-Cl 결합과 고리의 $\pi$-system 간의 ${\sigma}-{\pi}$ conjugation으로 기인되는 borderline 성질을 나타내고 있다. 2. Mutual conjugation의 정도는 $\pi$-charge와 bond alternation, 그리고 interfrontier level separation narrowing 효과로 확인된다. 3. 전자공여 파라치환체는 벤질탄소의 HOMO AO 계수를 감소시키는 반면, 전자흡인 파라치환체는 벤질탄소의 LUMO AO 계수를 감소시킨다.

Study of the Valence and Rydberg States of a Lithium Dimer by the Multi-reference Configuration-interaction Method

  • Convergent all-electron multi-reference configuration-interaction (MRCI) calculations are performed for a lithium dimer with Kaufmann's Rydberg basis functions. A comparison of the results of these calculations with those of the effective core potential/core polarization potential (ECP/CPP) method and experimental data reveals the deficiency of the all-electron ab initio method. The deficiency is related to the mere 51.9% attainment of electron correlation for the ground state. The percent attainment of electron correlation for the first excited state is slightly better than that for the ground state, preventing us from obtaining better agreements with experimental data by means of increasing the size of basis sets. The Kaufmann basis functions are then used with the ECP/CPP method to obtain the accurate convergent potential energy curves for the $^1\prod_u$ states correlated to Li(2p) + Li(2p) and Li(2s) + Li(n = 2, 3, 4). Quantum defect curves (QDCs) calculated for both the $X^2\sum_g$ and 1 $^2\prod_u$ states of the $Li{_2}^+$ ion and the Lu-Fano plot reveal a strong series-series interaction between the two $2snp{\pi}$ and $2pnp{\pi}$ Rydberg series. The QDCs are then used to resolve assignment problems in the literature. The reassignments, performed by Jedrzejewski-Szemek et al., of the dissociation product of the D $^1\prod$ state from (2s+3d) to (2s+3p) and that of the 6 $^1\prod_u$ from (2s+4d) to (2s+4p) are found to be incorrect. It may be more natural to assign their $2snp{\pi}$ Rydberg series as a $2snd{\pi}$ series. The state, assigned as 5p $^1\prod_u$ by Ross et al. and 4d $^1\prod$ by Jedrzejewski-Szemek et al., is assigned as the 7 $^1\prod_u$ state, correlated to the Li(2s) + Li(4f) limit.

Identification of phospholipase Cβ downstream effect on transient receptor potential canonical 1/4, transient receptor potential canonical 1/5 channels

  • $G{\alpha}_q$-coupled receptor stimulation was implied in the activation process of transient receptor potential canonical (TRPC)1/4 and TRPC1/5 heterotetrameric channels. The inactivation occurs due to phosphatidylinositol 4,5-biphosphate ($PI(4,5)P_2$) depletion. When $PI(4,5)P_2$ depletion was induced by muscarinic stimulation or inositol polyphosphate 5-phosphatase (Inp54p), however, the inactivation by muscarinic stimulation was greater compared to that by Inp54p. The aim of this study was to investigate the complete inactivation mechanism of the heteromeric channels upon $G{\alpha}_q$-phospholipase $C{\beta}$ ($G{\alpha}_q-PLC{\beta}$) activation. We evaluated the activity of heteromeric channels with electrophysiological recording in HEK293 cells expressing TRPC channels. TRPC1/4 and TRPC1/5 heteromers undergo further inhibition in $PLC{\beta}$ activation and calcium/protein kinase C (PKC) signaling. Nevertheless, the key factors differ. For TRPC1/4, the inactivation process was facilitated by $Ca^{2+}$ release from the endoplasmic reticulum, and for TRPC1/5, activation of PKC was concerned mostly. We conclude that the subsequent increase in cytoplasmic $Ca^{2+}$ due to $Ca^{2+}$ release from the endoplasmic reticulum and activation of PKC resulted in a second phase of channel inhibition following $PI(4,5)P_2$ depletion.
