[ $W_{1}/O/W_{2}$ ] Multi-emulsion Capsules Containing Ascrobic Acid-2-Glucoside Prepared Using PCL-based Amphiphilic Di- and Tri-block Copolymers

  • Biodegradable and amphiphilic di-block and tri-block copolymers, prepared with monomethoxy poly ethylene glycol (MPEG) and ${\varepsilon}-caprolactone\;({\varepsilon}-CL)$, were used for the application of W1/O/W2 multi- emulsion capsules. The effects of topology and the ratio of hydrophilic moiety of PCL-based polymers on the encapsulation efficiency of the $W_{1}/O/W_{2}$ multi-emulsion capsules containing Ascorbic Acid-2-Glucoside (AA-2-G) were investigated. The ratio of PEG and PCL was 1:0.5, 1:0.75, 1:1, and 1:1.25. PEG-PCL block copolymers were added to the first step of the preparation of $W_{1}/O$ emulsions. The dispersion stability, the particle size, the morphology of the $W_{1}/O/W_{2}$ multi-emulsion capsules were observed using an on-line turbidity meter, dynamic light scattering (DLS), a confocal microscopy (with FITC) and an optical microscopy. Biodegradable behavior of the PEG-PCL block copolymers and release behavior of AA-2-G were also observed by Gel Permeation Chromatography (GPC) and High Performance Liquid Chromatography (HPLC).

  • Poly-carprolactone (PCL)은 생분해성 고분자로 장기간의 임상실험 결과 생체에 독성이 없으며 생체친화성이 우수한 소재로 확인되어 PLGA, PLLA 등과 더불어 조직공학 분야에서 널리 사용되고 있는 생체재료이다. 그러나 PCL은 5개의 비극성 methylene group과 1개의 극성 ester group이 반복되는 지방족의 polyester로 구조상 탄소수가 많아 소수성을 띄는 단점을 가지고 있다. 이러한 표면이 소수성인 재료의 경우, 초기 단백질 흡착능이 떨어져 세포의 부착이 느린 속도로 일어나므로 세포 분화 및 조직 재생이 더디게 일어난다. 본 연구에서는 소수성의 PCL 표면의 단백질 흡착능을 증가시키기 위해 기능성 amine group을 부착하였으며, 또한 골재생을 촉진시킬 수 있는 세포외 기질인 collagen과 osteopontin을 부착함으로써 고기능성 PCL membrane을 제조하였다. 제조된 PCL membrane은 골재생용 조직공학에의 응용을 위해 지방유래 줄기세포를 이용하여 부착능 및 골세포로의 분화능을 확인하였다. 표면 성질의 변화에 의한 세포의 부착능의 변화를 confocal microscopy을 이용하여 부착에 관여하는 단백질의 발현을 확인하였으며, collagen과 osteopontin에 의한 골세포로의 분화능을 확인하기 위해 real time PCR을 통해 골세포의 분화 표지 유전자의 발현을 비교 분석하였다.

  • Recombinant human bone morphogenic protein-2 (rhBMP-2), which is known as one of the major local stimuli for osteogenic differentiation, was immobilized on the surface of hyaluronic acid (HA)-modified poly$(\varepsilon-caprolactone)$ (PCL) (HA-PCL) scaffolds to improve the attachment, proliferation, and differentiation of human bone marrow stem cells (hBMSCs) for bone tissue engineering. The rhBMP-2 proteins were directly immobilized onto the HA-modified PCL scaffolds by the chemical grafting the amine groups of proteins to carboxylic acid groups of HA. The amount of covalently bounded rhBMP-2 was measured to 1.6 pg/mg (rhBMP/HA-PCL scaffold) by using a sandwich enzyme-linked immunosorbant assay. The rhBMP-2 immobilized HA-modified-PCL scaffold exhibited the good colonization, by the newly differentiated osteoblasts, with a statistically significant increase of the rhBMP-2 release and alkaline phosphatase activity as compared with the control groups both PCL and HA-PCL scaffolds. We also found enhanced mineralization and elevated osteocalcin detection for the rhBMP-2 immobilized HA-PCL scaffolds, in vitro.

Growth Factor Releasing Porous Poly (${\varepsilon}-caprolactone$)-Chitosan Matrices for Enhanced Bone Regenerative Therapy

  • Drug releasing porous poly($\varepsilon$-caprolactone) (PCL)-chitosan matrices were fabricated for bone regenerative therapy. Porous matrices made of biodegradable polymers have been playing a crucial role as bone substitutes and as tissue-engineered scaffolds in bone regenerative therapy. The matrices provided mechanical support for the developing tissue and enhanced tissue formation by releasing active agent in controlled manner. Chitosan was employed to enhance hydrophilicity and biocompatibility of the PCL matrices. PDGF-BB was incorporated into PCL-chitosan matrices to induce enhanced bone regeneration efficacy. PCL-chitosan matrices retained a porous structure with a 100-200 $\mu$m pore diameter that was suitable for cellular migration and osteoid ingrowth. $NaHCO_3$ as a porogen was incorporated 5% ratio to polymer weight to form highly porous scaffolds. PDGF-BB was released from PCL-chitosan matrices maintaining therapeutic concentration for 4 week. High osteoblasts attachment level and proliferation was observed from PCL-chitosan matrices. Scanning electron microscopic examination indicated that cultured osteoblasts showed round form and spread pseudopods after 1 day and showed broad cytoplasmic extension after 14 days. PCL-chitosan matrices promoted bone regeneration and PDGF-BB loaded matrices obtained enhanced bone formation in rat calvarial defect. These results suggested that the PDGF-BB releasing PCL-chitosan porous matrices may be potentially used as tissue engineering scaffolds or bone substitutes with high bone regenerative efficacy.

Posterior Tibial Nerve Somatosensory Evoked Potentials Recorded on Subdural Electrodes around Paracentral Lobule (부중심소엽 주변 경막하 전극들에서 기록된 후경골신경 체성감각유발전위)

  • Background : Posterior tibial nerve somatosensory evoked potentials (PTSEP) have cortical potentials on primary sensory area of foot around 40 msec. The direct cortical recordings of the cortical potentials shows high voltage positive wave on medial hemisphere, especially on paracentral lobule (PCL). However, it is so difficult to record the potential directly on PCL that the cortical potential of PTSEP is not well understood. We investigated the cortical potential of PTSEP on subdural electrodes. Methods : We recorded cortical potentials to posterior tibial nerve stimulation on subdural electrodes which were on medial hemisphere near PCL in 15 intractable neocortical epilepsy patients. The numbers of subdural electrodes were 8 in 10 subjects ($1{\times}8array$) and 16 in 5 subjects ($2{\times}8arrays$). Seven subjects had three-dimensional imaging fusion (3D-fusion) of MRI and the electrodes using Analyze program. We investigated the amplitude, latency, polarity, and phase of the waves regarding location. Results : The waves had maximal amplitude on PCL in 4 subjects, precuneus in 1, cingulate gyrus nearest to PCL in 2 among 7 subjects with 3D-fusion. Also the electrodes were located on posterior area of PCL (2 out of 2 subjects with more than two electrodes put on PCL in 3D-fusion) and superior area of it (5 out of 5 subjects with $2{\times}8arrays $). All the high (more than 20 uV) amplitude around 40msec had positive polarity in 7 subjects. The phase reversals were detected between the electrodes with the highest amplitude and the just posterior (2 subjects) or anterior (6 subjects) located electrodes. The just posterior located electrodes had sharper phase reversal than the anterior one. Conclusion : PTSEP might have maximal amplitude of cortical potentials on the more superior and posterior area of PCL. The highest amplitude potential has positivity. The wave with maximal amplitude could have phase reversal of cortical potentials with surrounding electrodes, especially shaper with posterior part than with anterior one.

Preparation and Characteristics of a Single-layer PVA Laminated CTA/PCL Membrane for Oxygen Biosensor Electrode (산소센서용 CTA/PCL 효소고정화막과 반투막을 단일화한 PVA적층막의 제조 및 특성)

  • The oxygen electrode of a biosensor needs enzyme immobilized membrane and a dialysis membrane to measure the oxygen concentration that remains after an enzyme reacts with its substrate. Accodingly, a single-layer PVA laminated CTA/PCL membrane was developed as an oxygen biosensor electrode. The enzymes were immobilized on a cellulose triacetate/polycarprolactone membrane using the 1,1'-carbonyl diimidazole(CDI) method, and then laminated with polyvinyl alcohol, aldehyde and acid. The alcohol oxidase and PVA laminated CTA/PCL membrane was tested with various concentration of enzyme substrates using a Yellow Springs Instrument(YSI) oxygen sensor. Under 5-10mmol substrates produced $0.37{\sim}0.83{\mu}A$(r=0.995) currents, and ater 8 weeks the glucose oxidase activity remained at about 56%, while the other activities remained very low. A SEM indicated a smooth surface and tightly attached PVA on the enzyme-immobilized CTA/PCL membranes.

Preparation of Core-shell Type Nanoparticles of Poly($\varepsilon$-caprolactone) /Poly(ethylene glycol)/Poly( $\varepsilon$-caprolactone) Triblock Copolymers

  • A triblock copolymer based on $poly(\varepsilon-caprolactone)$ (PCL) as the hydrophobic part and poly(ethylene glycol) (PEG) as the hydrophilic portion was synthesized by a ring-opening mechanism of ${\varepsilon}-caprolactone$ with PEG containing a hydroxyl group at bot h ends as an initiator. The synthesized block copolymers of PCL/PEG/PCL (CEC) were confirmed and characterized using various analysis equipment such as 1H NMR, DSC, FT-IR, and WAXD. Core-shell type nanoparticles of CEC triblock copolymers were prepared using a dialysis technique to estimate their potential as a colloidal drug carrier using a hydrophobic drug. From the results of particle size analysis and transmission electron microscopy, the particle size of CEC core-shell type nanoparticles was determined to be about 20-60 nm with a spherical shape. Since CEC block copolymer nanoparticles have a core-shell type micellar structure and small particle size similar to polymeric micelles, CEC block copolymer can self-associate at certain concentrations and the critical association concentration (CAC) was able to be determined by fluorescence probe techniques. The CAC values of the CEC block copolymers were dependent on the PCL block length. In addition, drug loading contents were dependent on the PCL block length: the larger the PCL block length, the higher the drug loading content. Drug release from CEC core-shell type nanoparticles showed an initial burst release for the first 12 hrs followed by pseudo-zero order release kinetics for 2 or 3 days. CEC-2 block copolymer core-shell type nanoparticles were degraded very slowly, suggesting that the drug release kinetics were governed by a diffusion mechanism rather than a degradation mechanism irrelevant to the CEC block copolymer composition.

Apoptosis-inducing effect and structural basis of Polygonatum cyrtonema lectin and chemical modification properties on its mannose-binding sites

  • Polygonatum cyrtonema Lectin (PCL), which is classified as a monocot mannose-binding lectin, has received great regards for its uniquely biological activities and potentially medical applications in cancer cells. This paper was initially aimed to study apoptosis of PCL on Hela cells. Thus, 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) method was carried out. Through observation of cell morphologic changes and Lactate dehydrogenase (LDH) activity-based cytotoxicity assays, PCL induced HeLa cell apoptosis in a dose-dependent manner. To further gain structural basis, multiple alignments, homology modeling and docking experiments were performed to analyze the correlation between its biological activities and mannose-binding sites. Eventually, considering docking data, chemical modification properties on the three mannose-binding sites were analyzed by a series of biological experiments (e.g., hemagglutinating and mitogenic activity assays, fluorescence and Circular Dichrosim (CD) spectroscopy) to profoundly identify the role of some key amino acids in the structure-function relationship of PCL.

Development of Polymer Coating Method for Stable Stent Coating Using Chemical Bond Between Metal Surface and Polymer (안정된 스텐트 코팅막을 형성하기 위해 금속표면과 고분자 사이의 화학적 결합을 이용한 고분자 코팅법 개발)

  • To produce stable polymer coating layer using the interaction between metal stent and polymer layer, Ahx-HSAB was synthesized by coupling 6-aminoheanoic acid (Ahx) with N-Hydroxy succinimidyl 4-azidobenzonate (HSAB) containing photo reactive group. Then, Ahx-HSAB was applied to self·assembled monolayer (SAM) on $TiO_2$-coated surface, since one end of Ahx-HSAB was carboxyl acid which was known to be able to interact with $TiO_2$ surface. That SAM layer was incubated in 1% polycaprolacton (PCL) solution and photoreacted by ultraviolet light (254 nm) to produce the chemical bond between SAM and polymer layer, followed by PCL polymer coating ({\sim}5\;{\mu}m$) by the method of spray coating. The surface change was investigated by measuring of contact angle of the surface. The contact angle values of stainless steel (SS) surface, $TiO_2$-coated surface, SAM layer by Ahx-HSAB, photoreacted surface with PCL and PCL layer by spray coating were 70.48${\pm}$1.89, 38.57${\pm}$3.31, 60.14${\pm}$2.21, 54.91${\pm}$2.70 and 56.47${\pm}$2.12, respectively. The stability of polymer layers was tested by incubation of PCL-coated plates in 0.1M PBS buffer (pH 7.4, 0.05%, Tween 80) with vigorous shaking (200 rpm). While the poiymer layer prepared by these processes showed the intact surface morphology over 3 days, the polymer layers prepared by spray coating of PCL onto SS plate (control 1) and $TiO_2$-coated SS plate (control 2) were Peeled off in 3 days. Thus, the polymer coating method using SAM and photoreaction seems to be a effective method to obtain the stable polymer layer onto SS surface.
