Relationship between Monounsaturated Fatty Acid Composition and Stearoyl-CoA Desaturase mRNA Level in Hanwoo Liver and Loin Muscle (한우 간 및 등심 조직에서 불포화지방산의 조성비율과 Stearoyl-CoA Desaturase mRNA의 발현 양상)

  • The Stearoyl-CoA Desaturase(SCD) is a key enzyme, which converting palmitic acid(16:0) and stearic acid (18:0) to pahnitoleic acid(16:1) and oleic acid(l8:1), respectively. The concentration of oleic acid(18:1) in meat of beef cattle could influence both palatability and perception of meat. This experiment has conducted to determine relationship between the compositions of monounsaturated fatty acids and the SCD mRNA level in bovine liver and loin muscle tissue. The compositions of palmitoleic acid(16:1) and oleic acid(18:1) in loin muscle were 5% and 46% of total lipid and in liver were 2% and 20% of total lipid, respectively. On the other hand, the compositions of palmitic acid(16:0) and stearic acid(18:0) in loin muscle were 25% and 45% of total lipid and in liver were 14% and 43% of total lipid, respectively. The ratio of monounsaturated to saturated fatty acids(the desaturation index) was used as a measure of SCD activity in tissues. The average desaturation index in loin muscle was higher about 3.6-fold than that in liver. The desaturation index of oleate/stearate and palmitoleatelpalmitate in loin muscle were higher 8-fold and 1.8-fold than those in liver, respectively, showing that the substrate specificity of SCD enzyme was very different between liver and muscle tissues. To determine whether the composition of monounsaturated fatty acids in liver and muscle are dependent on SCD expression, SCD mRNA level was examined by RT-PCR analysis. The SCD mRNA level in loin muscle was higher about 3-fold than that in liver. Thus, the quantitative relationship between the desaturation index of fatty acid and SCD mRNA was observed in liver and muscle. The difference in the compositions of monounsaturated fatty acids between bovine liver and muscle tissues may be due to different level of Stearoyl-CoA Desaturase mRNA.

Physicochemical Changes of Food Waste Slurry Co-fermented with Pig Manure Slurry (음식물쓰레기와 돈분 액상물의 혼합부숙시 이화학적 특성 변화)

  • To find a feasibility of utilization of food waste slurry (FWS) generated during composting, FWS was combined with pig manure slurry (PMS) in various ratios and the change of nutrient contents and offensive odor of the combined slurries before and after fermentation were studied. The initial pH was 7.67 for PMS and 8.45 for FWS. However, during the fermentation, pH increased in the combined slurries with the higher FWS rate among the treatments while decreased in thosewith higher PMS rate. EC of each slurry sample showed that the difference among combined slurry samples has been reduced during fermentation and became stabilized in $21{\sim}23dS\;m^{-1}$ after 180 days. After 180 days fermentation, total nitrogen (T-N) decreased. T-N of mixture with a half and more FWS decreased up to 0.1%, less than the critical level (0.3%). The contents of O.M., T-N, phosphorus, calcium and magnesium decreased with fermentation while those of potash and salinity increased. From initial fermentation until 30 days, a lot of $NH_3$, as an offensive odor, was produced. However, it decreased steadily, except in higher PMS rate. In terms of producing $50{\mu}g\;ml^{-1}$ of $NH_3$, the top layer took 30 days after fertilization with FWS only, 45 days for utilized treatment with F75 (25 % of PMS), 75 days for utilized with F50 (50%) and F25 (75%) and 90 days for PMS only, respectively. $RNH_2$ also had similar trend with $NH_3$ but it was produced continuously as long fermentation proceeded. In terms of $RNH_2$, the decrease in concentration up to $50{\mu}g\;ml^{-1}$ were; 45 days for FWS only(F100), 105 days for F75 utilization, 120 daysfor F50, 165 days for F25, respectively. ethyl mercaptan was produced in PMS until 180 days after fertilization but it was not produced in FWS. Sensory tests as an integrated test of offensive odor were also done. FWS showed lower than 1 after 30 days from initial fermentation, while PMS had still offensive odor even up to 180 days from initial fermentation. It is probably affected by the continuous production of ethyl mercaptan and amines. However, considering in decrease T-N content caused by volatilization while offensive odor intensity according to official standard of fertilizer is lower than 2. Further study on controlling offensive odor needs to be done.

  • This study was to examine the physicochemical characteristics of coagulation reaction between ignited oyster shell powder (IOSP) and red tide organisms (RTO), and its feasibility, in developing a technology for the removal of RTO bloom in coastal sea,IOSP was made from oyster shell and its physicochemical characteristics were examined for particle size distribution, surface characteristic by scanning electron microscope, zeta potential, and alkalinity and pH variations in sea water. Two kinds of RTO that were used in this study, Cylindrotheca closterium and Skeletonema costatum, were sampled in Masan bay and were cultured in laboratory. Coagulation experiments were conducted using various c(Incentrations of IOSP, RTO, and a jar tester. The supernatant and RTO culture solution were analyzed for pH, alkalinity, RTO cell number, IOSP showed positive zeta potentials of $11.1{\~}50.1\;mV\;at\;pH\;6.2{\~}12.7$, A positive zeta potential of IOSP slowly decreased with decreasing pNa 4,0 to 2,0. When pNa reached zero, the zeta potential approached zero, When a pMg value was decreased, the positive zeta potential of IOSP increased until pMg 3.0 and decreased below pMg 3.0. IOSP showed 4.8 mV of positive zeta potential while RTO showed -9.2 mV of negative zeta potential in sea water. A positive-negative EDL (electrical double-layer) interaction occurred between $Mg(OH)_2$ adsorption layer of IOSP and RTO in sea water so that EDL attractive force always worked between them. Hence, their coagulation reaction occurred at primary minimum on which an extreme attractive force acted because of charge neutralization by $Mg(OH)_2$ adsorption layer of IOSP. As a result, the coagulation reaction was rapidly processed and was irreversible according to DLVO (Deriaguin-Landau-Verwey-Overbeek) theory. Removal rates of RTO were exponentially increased with increasing both IOSP concentration and G-value. The removal rates were steeply increased until 50 mg/l of IOSP and reached $100{\%}\;at\;400\;mg/l$ of IOSP. Removal rates of RTO were $70.5,\;70.5,\;81.7,\;85.3{\%}$ for G-values of $1,\;6,\;29,\;139\;sec^(-1)$at IOSP 100 mg/l, respectively. This indicated that mixing (i.e., collision among particles) was very important for a coagulation reaction.

Geochemistry of Geothermal Waters in Korea: Environmental Isotope and Hydrochemical Characteristics I. Bugok Area (한반도 지열수의 지화학적 연구: 환경동위원소 및 수문화학적 특성 I. 부곡 지역)

  • Hydrogeochemical and environmental isotope studies were undertaken for various kinds of water samples collected in 1995-1996 from the Bugok geothermal area. Physicochemical data indicate the occurrence of three distinct groups of natural water: Group I ($Na-S0_4$ type water with high temperatures up to $77^{\circ}C$, occurring from the central part of the geothermal area), Group II (warm $Na-HCO_{3}-SO_{4}$ type water, occurring from peripheral sites), Group III ($Ca-HCO_3$ type water, occurring as surface waters and/or shallow cold groundwaters). The Group I waters are further divided into two SUbtypes: Subgroup Ia and Subgroup lb. The general order of increasing degrees of hydrogeochemical evolution (due to the degrees of water-rock interaction) is: Group III$\rightarrow$Group II$\rightarrow$Group I. The Group II and III waters show smaller degrees of interaction with rocks (largely calcite and Na-plagioclase), whereas the Group I waters record the stronger interaction with plagioclase, K-feldspar, mica, chlorite and pyrite. The concentration and sulfur isotope composition of dissolved sulfate appear as a key parameter to understand the origin and evolution of geothermal waters. The sulfate was derived not only from oxidation of sedimentary pyrites in surrounding rocks (especially for the Subgroup Ib waters) but also from magmatic hydrothermal pyrites occurring in restricted fracture channels which extend down to a deep geothermal reservoir (typically for the Subgroup Ia waters). It is shown that the applicability of alkaliion geothermometer calculations for these waters is hampered by several processes (especially the mixing with Mg-rich near-surface waters) that modify the chemical composition. However, the multi-component mineral/water equilibria calculation and available fluid inclusion data indicate that geothermal waters of the Bugok area reach temperatures around $125^{\circ}C$ at deep geothermal reservoir (possibly a cooling pluton). Environmental isotope data (oxygen-18, deuterium and tritium) indicate the origin of all groups of waters from diverse meteoric waters. The Subgroup Ia waters are typically lower in O-H isotope values and tritium content, indicating their derivation from distinct meteoric waters. Combined with tritium isotope data, the Subgroup Ia waters likely represent the older (at least 45 years old) meteoric waters circuated down to the deep geothermal reservoir and record the lesser degrees of mixing with near-surface waters. We propose a model for the genesis and evolution of sulfate-rich geothermal waters.

Antitumor and Free Radical-Scavenging Activities of Various Extract Fractions of Fruits and Leaves from Prunus mume (금매와 매화 잎 추출물의 프리라디칼 억제 활성 및 항암 효과)

  • This study investigated the free radical-scavenging and antitumor activities of hot water, water, acetone, ethanol, ethyl acetate, chloroform, and hexane extracts of fruits and leaves from Prunus mume. The various extracts were evaluated for their total polyphenol, flavonoid, and tannin contents, scavenging activities by DPPH and ABTS analyses, reducing power, protective effects against oxidative stress in L-132 cells, and antitumor activities against A549, HeLa, and U87 cancer cells. Ethanol extracts of fruits and leaves showed the highest total polyphenol content (336.41 and 523 mg GAE/100 g, respectively). DPPH and ABTS radical-scavenging activities increased according to concentration of fruit. DPPH radical-scavenging activity of ethanol extracts from leaves was 65.48% at $200{\mu}g/mL$. All extract fractions of leaves showed high ABTS radical-scavenging activities. The reducing power activities increased according to increasing concentration of fruits and leaves. All extracts of leaves performed better than extracts of fruits in terms of protective effects against oxidative stress in L-132 cells. Ethyl acetate, chloroform, hexane, ethanol extracts of fruits and leaves showed anticancer activities against A549, HeLa, and U87 cancer cells. However, ethanol extracts of fruits and leaves showed no toxicity in normal cells (BNLCL2). This study suggests that antioxidant activities of fruits and leaves from P. mume depend on polyphenol contents. Thus, fruits and leaves from P. mume can be useful as natural antioxidant compounds.

The Relationship between Intracellular Protein Kinase C Concentration and Invasiveness in U-87 Malignant Glioma Cells (교모세포종 세포주 U-87에서 세포내 PKC 농도와 종양침습성과의 상관 관계)

  • Objective : Glioblastomas, the most common type of primary brain tumors, are highly invasive and cause massive tissue destruction at both the tumor invading edges and in areas that are not in direct contact with glioma cells. As a result, patients with high-grade gliomas are faced with a poor prognosis. Such grim statistics emphasize the need to better understand the mechanisms that underlie glioma invasion, as these may lead to the identification of novel targets in the therapy of high grade gliomas. Protein kinase C(PKC) is a family of serine/threonine kinases and an important signal transduction enzyme that conveys signals generated by ligand-receptor interaction at the cell surface to the nucleus. PKC appears to be critical in regulating many aspects of glioma biology. The purpose of this study was to assess accurately the role of PKC in the invasion regulation of human gliomas based on hypothesis that protein kinase C(PKC) is functional in the process of glial tumor cell invasion. Method : To test this hypothesis, U-87 malignant glioma cell line intracellular PKC levels were up and down regulated and their invasiveness was tested. Intracellular PKC level was characterized using PKC activity assays. Invasion assays including barrier migration and spheroid confrontation were used to study the relationship between PKC concentration and invasiveness. Result : The cell line which were treated by PKC inhibitor tamoxifen and hypericin exhibited decreased PKC activity and decreased invasive abilities dose dependently both in matrigel invasion assay and tumor spheroid fetal rat brain aggregates(FRBA) confrontation assay. However, the cell line that was treated by PKC activator 12-O-tetradecanylphorbol-13acetate(TPA) did not exhibit increases in either PKC activity or invasive ability. Conclusion : These studies suggest that PKC may be a useful molecular target for the chemotherapy of glioblastoma and other malignancies and that a therapeutic approach based on the ability of PKC inhibitors may be helpful in preventing invasion.

Evaluation on Cooling Effects of Geothermal Heat Pump System in Farrowing House (지열 냉방시스템을 이용한 분만돈사의 냉방효과 분석)

  • The principal objective of this study was to investigate the cooling effects of geothermal heat pump system (GHPS) in farrowing house. A total of 96 sows were allocated to 2 pig housings (GHPS and conventional housing) with 48 for four weeks in summer season. During the experimental period of four weeks, the highest outside temperature observed was approximately $34.1^{\circ}C$, GHPS decrease indoor temperature of pig housing up to $30.9^{\circ}C$, but conventional pig housing was similar to outside temperature. Dust concentrations (maximum 61.4%) of particulate matter less than $10{\mu}m$ (PM 10) in GHPS-housing were lower than the conventional housing. GHPS showed no signigicant difference in carbon dioxide emission, whereas the ammonia gas concentration was significantly decreased in GHPS-housing compared to that of conventional housing. Sows in GHPS-housing showed significantly lower respiratory rate than those of the control group. GHPS did not affect hormone level, litter size and birth weight, but weaning weight of piglets was influenced by GHPS. Feed consumption of sows was significantly increased in GHPS-housing compared to the conventional hosing. These results suggest that GHPS decrease dust concentration, ammonia gas emission and indoor temperature of pig housing and may affect performance in sows and weaned piglets.

The Risk Assessment of Butachlor for the Freshwater Aquatic Organisms (Butachlor의 수서생물에 대한 위해성 평가)

  • To assess the effect of butachlor on freshwater aquatic organisms, acute toxicity studies for algae, invertebrate and fishes were conducted. The algae grow inhibition studies were carried out to determine the growth inhibition effects of butachlor (Tech. 93.4%) in Pseudokirchneriella subcapitata (formerly knows as Selenastrum capriconutum), Desmodesmus subspicatus (formerly known as Scendusmus subspicatus), and Chlorella vulgaris during the exposure period of 72 hours. The toxicological responses of P. subcapitata, D. subspicatus, and C. vulgaris to butachlor, expressed in individual $ErC_{50}$ values were 0.002, 0.019, and $10.4mgL^{-1}$, respectively and NOEC values were 0.0008, 0.0016, and $5.34mg\;L^{-1}$, respectively. P. subcapitata was more sensitive than any other algae species. Butachlor has very high toxicity to the algae, such as P. subcapitata and D. subspicatu. In the acute immobilisation test for Daphnia magna, the 24 and $48h-EC_{50}$ values were 2.55 and $1.50mg\;L^{-1}$, respectively. As the results of the acute toxicity test on Cyprinus carpio, Oryzias latipes and Misgurnus anguillicaudatus, the $96h-LC_{50}s$ were 0.62, 0.41 and $0.24mg\;L^{-1}$, respectively. The following ecological risk assessment of butachlor was performed on the basis of the toxicological data of algae, invertebrate and fish and exposure concentrations in rice paddy, drain and river. When a butachlor formulation is applied in rice paddy field according to label recommendation, the measured concentration of butachlor in paddy water was $0.41mg\;L^{-1}$ and the predicted environmental concentration (PEC) of butachlor in drain water was $0.03 mg\;L^{-1}$. Residues of butachlor detected in major rivers between 1997 and 1998 were ranged from $0.0004mg\;L^{-1}$ to $0.0029mg\;L^{-1}$. Toxicity exposure ratios (TERs) of algae in rice paddy, drain and river were 0.004, 0.05 and 0.36, respectively and indicated that butachlor has a risk to algae in rice paddy, drain and river. On the other hand, TERs of invertebrate in rice paddy, drain and river were 3.6, 50 and 357, respectively, well above 2, indicating no risk to invertebrate. TERs of fish in rice paddy, drain and river were 0.58, 8 and 57, respectively. The TERs for fish indicated that butachlor poses a risk to fish in rice paddy but has no risk to fish in agricultural drain and river. In conclusion, butachlor has a minimal risk to algae in agricultural drain and river exposed from rice drainage but has no risk to invertebrate and fish.

Improvement and Validation of an Analytical Method for Quercetin-3-𝑜-gentiobioside and Isoquercitrin in Abelmoschus esculentus L. Moench (오크라 분말의 Quercetin-3-𝑜-Gentiobioside 및 Isoquercitrin의 분석법 개선 및 검증)

  • This study aimed to investigate the validation and modify the analytical method to determine quercetin-3-𝑜-gentiobioside and isoquercitrin in Abelmoschus esculentus L. Moench for the standardization of ingredients in development of functional health products. The analytical method was validated based on the ICH (International Conference for Harmonization) guidelines to verify the reliability and validity there of on the specificity, linearity, accuracy, precision, detection limit and quantification limit. For the HPLC analysis method, the peak retention time of the index component of the standard solution and the peak retention time of the index component of A. esculentus L. Moench powder sample were consistent with the spectra thereof, confirming the specificity. The calibration curves of quercetin-3-𝑜-gentiobioside and isoquercitrin showed a linearity with a near-one correlation coefficient (0.9999 and 0.9999), indicating the high suitability thereof for the analysis. A. esculentus L. Moench powder sample of a known concentration were prepared with low, medium, and high concentrations of standard substances and were calculated for the precision and accuracy. The precision of quercetin-3-𝑜-gentiobioside and isoquercitrin was confirmed for intra-day and daily. As a result, the intra-day precision was found to be 0.50-1.48% and 0.77-2.87%, and the daily precision to be 0.07-3.37% and 0.58-1.37%, implying an excellent precision at level below 5%. As a result of accuracy measurement, the intra-day accuracy of quercetin-3-𝑜-gentiobioside and isoquercitrin was found to be 104.87-109.64% and the daily accuracy thereof was found to be 106.85-109.06%, reflecting high level of accuracy. The detection limits of quercetin-3-𝑜-gentiobioside and isoquercitrin were 0.24 ㎍/mL and 0.16 ㎍/mL, respectively, whereas the quantitation limits were 0.71 ㎍/mL and 0.49 ㎍/mL, confirming that detection was valid at the low concentrations as well. From the analysis, the established analytical method was proven to be excellent with high level of results from the verification on the specificity, linearity, precision, accuracy, detection limit and quantitation limit thereof. In addition, as a result of analyzing the content of A. esculentus L. Moench powder samples using a validated analytical method, quercetin-3-𝑜-gentiobioside was analyzed to contain 1.49±0.01 mg/dry weight g, while isoquercitrin contained 1.39±0.01 mg/dry weight g. The study was conducted to verify that the simultaneous analysis on quercetin-3-𝑜-gentiobioside and isoquercitrin, the indicators of A. esculentus L. Moench, is a scientifically reliable and suitable analytical method.

Removal of 1,4-dioxane in Ozone and Activated Carbon Process (오존과 활성탄 공정해서의 1,4-Dioxane 제거 특성)

  • Three different virgin activated carbons made of each coal(Calgon), coconut(Samchully) and wood(Picabiol) based activated carbon(AC) were tested for an adsorption performance of 1,4-dioxane in a continuous adsorption column. Breakthrough behavior was Investigated that the breakthrough points of coal, coconut and wood based AC were observed as 3600 bed volumn(BV), 1440 BV and 144 BV respectively. Adsorption capacity(X/M) of coal, coconut and wood based AC was observed. The reported results of adsorption capacity showed that coal based AC was highest(578.9 ${\mu}g/g$), coconut based AC was intermediate(142.3 ${\mu}g/g$) and wood based AC was lowest(7.4 ${\mu}g/g$) due to increasing specific surface area. Moreover, carbon usage rates(CURs) for coal, coconut and wood based AC had been shown as 0.48 g/day, 1.41 g/day and 6.9 g/day respectively. The constant characteristic of the system, k of coal based AC was found to be 91.5 and k of coconut based AC was found to be 17.9. Removal efficiencies of 1,4-dioxane with different ozonation dosages(2 and 5 mg/L) for 20 min ozonation had been shown 38% and 87% respectively. There was no observation for biological removal of 1.4-dioxane by attached micro-organisms when used(3.1 years and over 5 years) biological activated carbon(BAC) without pretreatment of oxidation were employed. When a combination of ozonation(2 mg/L and 5 mg/L) and BAC process for $10{\sim}30$ min was applied, removal efficiency for 1,4-dioxine increased only $2{\sim}6%$ compared to only applying ozonation. Therefore removal efficiency of BAC process prior to using oxidation was proven to negligible. Consequently, the results presented in this paper provide a better insight into the adsorption performance of 1,4-dioxane. This observation suggests that using virgin activated carbon made of coal is the best selection for removal of 1,4-dioxane in the water treatment for an advanced treatment. It is clear from this research that longer EBCT for ozonation or higher ozone concentration are more effective operation methods for removal of 1,4-dioxane than longer EBCT in the BAC process.
