만성폐쇄성폐질환의 급성악화와 회복기에서 유도객담 내 Nuclear Factor-κB(NF-κB)의 활성도와 IL-6, IL-8 및 TNF-α의 농도 변화 (Nuclear Factor-κB(NF-κB) Activity and Levels of IL-6, IL-8 and TNF-α in Induced Sputum in the Exacerbation and Recovery of COPD Patients)

  • 연구배경 : COPD의 급성악화는 세균에 의한 감염, 바이러스성 상기도 감염, 대기오염, 기후변화 등에 의하며, COPD의 급성악화 시에 객담 내 호중구의 증가, IL-6와 IL-8 농도의 증가, 그리고 산화질소의 증가는 $NF-{\kappa}B$의 활성화와 관련된 것으로 알려져 있다. 그러므로 COPD의 병인 및 급성악화의 기전에 $NF-{\kappa}B$ 활성도와 IL-6, IL-8 및 $TNF-{\alpha}$가 관련이 있는지 연구하고자 하였다. 방 법 : 정상대조군 및 COPD로 입원하였던 환자들의 급성악화기 및 치료 후 회복기에 유도객담에서 IL-6, IL-8, $TNF-{\alpha}$, $NF-{\kappa}B$ 활성화 정도를 측정하여 비교하였다. 결 과 : 1) 유도객담내의 IL-6, IL-8 및 $TNF-{\alpha}$는 COPD 환자에서 대조군에 비해 유의하게 증가되었다(p<0.01). IL-8은 급성악화 시에 비해 회복기에 유의하게 감소되었고(p<0.05), IL-6와 $TNF-{\alpha}$는 회복기에도 차이가 없었다. 2) 유도객담 내 대식세포에서의 $NF-{\kappa}B$의 활성도는 COPD 환자에서 대조군에 비해 유의하게 증가 되었고(p<0.05), 회복기에는 악화 시에 비해 감소하는 경향을 보였다. 결 론 : COPD 환자에서 유도객담 내 IL-6, IL-8, $TNF-{\alpha}$$NF-{\kappa}B$의 활성도 등이 정상대조군에 비해 증가되었고, 회복기에 IL-8은 감소하였고, $NF-{\kappa}B$의 활성도, IL-6 및 $TNF-{\alpha}$는 감소하는 경향을 보여서, COPD의 급성악화 및 COPD의 병인에 $NF-{\kappa}B$가 일부 관여할 것으로 생각된다.

백화사설초 메탄올 추출물에 의한 HL-60 세포고사과정에 있어서의 transcriptional factors 활성변화 연구 (Study on Transcriptional Factors Activation Change of HL-60 cell Apoptosis by Hedyotis Diffusa's Methanol Extract)

  • Objective : Hedyotis diffusa has been used as an anticancer agent for several decades in oriental medicine. We test whether the methanol extract of the herb affects transcriptional activation factors including $NF-{\kappa}B$ and AP-1. Methods : 1. HL-60 cells were treated with various concentrations(from 200 to $50{\mu}g/ml$) of methanol extract and $H_2O$ extract($200{\mu}g/ml$)of hedyotis diffusa, After 48h later, the cells were tested for viability by MTT assay. 2. The HL-60 cells were treated with $200{\mu}g/ml$ of methanol extract for the indicated periods. First. Nuclear extracts were isolated and incubated with oligonucleotide probe of $NF-{\kappa}B$ and AP-1. Second. Nuclear extracts were isolated and reacted with p50, p65. c-rel pan-Jun, c-Jun, JunB. JunD antibody on ice for 30min. Finally The cell lysates were prepared and analyzed by western blotting using anti-Fas, anti-FasL and anti-p53 antibody. Results : 1. The methanol extract decreases the viability of human lymphoid origin leukemia HL-60 cells in a dose-dependent manner. 2. $NF-{\kappa}B$ is rapidly activated by the addition of the methanol extract, reaches a peak at 30min and gradually returns to resting level. We confirm that $NF-{\kappa}B$ is a heterodimer mainly composed of p65 subunit with c-Rel. 3. Transcriptional activation of AP-1 is detected at 30min and reaches a maximum at 1hr after stimulation of the cells with the methanol extract. AP-1 is mainly composed with Jur-D and partially Jug-B proteins. 4. the methanol extract of Hedyotis diffusa induces the expression of Fas, Fas ligand and p53 proteins of HL-60 cells in a time dependent fashion. Conclusions : These results suggest that the methanol extract of Hedyotis diffusa exerts anticancer effects to induce the death of human leukomic HL-60 cells via activation of trascriptional factors such as $NF-{\kappa}B$ and AP-1, increase in expression of Fas mediated signalling proteins, and induction of tumor suppressor gene. p53.

Salicortin suppresses lipopolysaccharide-stimulated inflammatory responses via blockade of NF-κB and JNK activation in RAW 264.7 macrophages

  • We isolated the phenolic glucoside salicortin from a Populus euramericana bark extract, and examined its ability to suppress inflammatory responses as well as the molecular mechanisms underlying these abilities, using lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Salicortin inhibited iNOS expression and the subsequent production of NO in a dose-dependent manner in the LPS-stimulated RAW 264.7 cells. Salicortin significantly suppressed LPS-induced signal cascades of NF-${\kappa}B$ activation, such as IKK activation, $I{\kappa}B{\alpha}$ phosphorylation and p65 phosphorylation in RAW 264.7 cells. In addition, salicortin inhibited the LPS-induced activation of JNK, but not ERK or p38 MAPK. Furthermore, salicortin significantly inhibited production of pro-inflammatory cytokines, such as TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 in the LPS-stimulated RAW 264.7 cells. These findings suggest that salicortin may show its anti-inflammatory activity by suppressing the LPS-induced expression of pro-inflammatory mediators through inhibition of NF-${\kappa}B$ and JNK MAPK signaling cascades in macrophages.

자하차약침(紫何車藥針)의 항염증능(亢炎症能)이 LPS 유발 관절염(關節炎) 치료(治療)에 미치는 영향(影響) (The Effect of Homnis Placenta Herbal Acupuncture on Reducing Expression of LPS-induced Arthritis Model as an Anti-inflammatory Agent)

  • Objectives : Rheumatoid arthritis(RA) is a systemic & a chronic inflammatory autoimmune disease . A chronic , locally destructive inflammmatory reaction in human is examplified by the synovitis present in some connective tissue disorder. The presence of a number of cytokines, $TNF-{\alpha}$, iNOS & expression of nitric oxide, NF-kB p65 activation implies an important role of cellular immune response in RA inflammatory reaction. This study was designed to evaluate on the effects of the Homnis Placenta herbal acupuncture on EX-LE201 & ST 35 reducing expression of LPS-induced arthritis model in mice. Materials and Methods : Homnis Placenta herbal acupuncture was inserted into 10 rats induced rheumatoid arthritis. The acupunctures were injected into the EX-LE201 and ST35 points. Such indexes were measured the inhibition of inducible nitric oxide synthase(iNOS) expression, nitric oxide(NO) production in vitro experiment and Tumor Necrosis $Factor-{\alpha}(TNF-{\alpha})$ & Nuclear Factor kappa $B(NF-{\kappa}B)$ p65 activation, synovial hyperplasia, angiogenesis and fibrosis in synovial membrane of knee joint of mice in vivo experiment. Results : 1.Homnis Placenta Herbal acupuncture inhibited iNOS mRNA and NO in RAW 264.7 cell of LPS-induced rheumatoid arthritis in a dose dependent manner. 2.Homnis Placenta Herbal acupuncture also showed significant inhibition of $TNF-{\alpha}$ & $NF-{\kappa}B$ p65, activation, synovial hyperplasia, angiogenesis and fibrosis in synovial membrane of knee joint of mice. Conclusion : These results suggest that Homnis Placenta Herbal acupuncture has an therapeutic effects on LPS induced-rheumatoid arthritis by inhibiting $TNF-{\alpha}$ activation.

Indacaterol Inhibits Tumor Cell Invasiveness and MMP-9 Expression by Suppressing IKK/NF-κB Activation

  • The ${\beta}_2$ adrenergic receptor (ADRB2) is a G protein-coupled transmembrane receptor expressed in the human respiratory tract and widely recognized as a pharmacological target for treatments of asthma and chronic obstructive pulmonary disorder (COPD). Although a number of ADRB2 agonists have been developed for use in asthma therapy, indacaterol is the only ultra-long-acting inhaled ${\beta}_2$-agonist (LABA) approved by the FDA for relieving the symptoms in COPD patients. The precise molecular mechanism underlying the pharmacological effect of indacaterol, however, remains unclear. Here, we show that ${\beta}$-arrestin-2 mediates the internalization of ADRB2 following indacaterol treatment. Moreover, we demonstrate that indacaterol significantly inhibits tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$)-induced NF-${\kappa}B$ activity by reducing levels of both phosphorylated-IKK and -$I{\kappa}B{\alpha}$, thereby decreasing NF-${\kappa}B$ nuclear translocation and the expression of MMP-9, an NF-${\kappa}B$ target gene. Subsequently, we show that indacaterol significantly inhibits TNF-${\alpha}$/NF-${\kappa}B$-induced cell invasiveness and migration in a human cancer cell line. In conclusion, we propose that indacaterol may inhibit NF-${\kappa}B$ activity in a ${\beta}$-arrestin2-dependent manner, preventing further lung damage and improving lung function in COPD patients.

Cardamonin Inhibits the Expression of Inducible Nitric Oxide Synthase Induced by TLR2, 4, and 6 Agonists

  • Toll-like receptors (TLRs) play an important role for host defense against invading pathogens. The activation of TLRs signaling leads to the activation of $NF-{\kappa}B$ and the expression of pro-inflammatory gene products such as cytokines and inducible nitric oxide synthase (iNOS). To evaluate the therapeutic potential of cardamonin, which is a naturally occurring chalcone from Alpinia species (zingiberaceous plant species), $NF-{\kappa}B$ activation and iNOS expression induced by MALP-2 (TLR2 and TLR6 agonist) or LPS (TLR4 agonist) were examined. Cardamonin inhibited the activation of $NF-{\kappa}B$ induced by MALP-2 or LPS. Cardamonin also suppressed the iNOS expression induced by MALP-2 or LPS. These results suggest that cardamonin has the specific mechanism for anti-inflammatory responses by regulating of TLRs signaling pathway.

혈관평활근세포에서 HSP90에 의한 IL-6 발현에 TLR-4와 NF-κB의 작용 (Roles of TLR-4 and NF-κB in Interleukin-6 Expression Induced by Heat Shock Protein 90 in Vascular Smooth Muscle Cells)

  • HSP90에 노출된 혈관평활근세포에서 IL-6 transcript가 증가하고, IL-6 단백질의 분비가 증가하며, 또한 IL-6 유전자의 promote가 활성화되었다. HSP90에 의한 IL-6 유전자의 promoter 활성화는 dominant negative 형태의 TLR-4와 MyD88에 의하여 크게 감소되었지만, dominant negative 형태의 TLR-3와 TRIF의 영향을 받지 않았다. 그리고 TLR-4의 이합체화(dimerization)를 저해하는 curcumin은 HSP90에 의한 IL-6의 분비 및 IL-6 유전자 promoter 활성화를 억제하였다. 그리고 IL-6 유전자의 promoter의 NF-${\kappa}B$- 또는 C/EBP-binding sequence에 변이는 HSP90에 의한 IL-6 유전자의 promoter 활성화 억제하였다. 이러한 결과는 혈관평활근세포에서 HSP90에 의한 IL-6 유전자 활성화에 TLR-4와 NF-${\kappa}B$B가 관여함을 의미한다.

꽃송이버섯 추출물의 항염활성 효과 (Anti-inflammatory effects of Sparassis crispa extracts)

  • 꽃송이버섯 추출물은 염증반응 시 유도되는 NO 생성을 저해하는 활성이 있는 것으로 나타났으며, 200 ${\mu}g/ml$의 꽃송이버섯 추출물을 처리하였을 때 NO 저해능이 최대 효과를 보였고 RAW264.7 cell에서는 대조군과 유사한 수준으로 NO 생성을 억제하였다. 이러한 NO 생성의 저해는 iNOS의 발현이 감소한 것에 의한 결과임을 단백질과 mRNA의 발현량 변화를 통하여 확인하였으며, mRNA 발현 변화는 iNOS 유전자의 전사를 담당하는 전사인자인 $NF-{\kappa}B$와 STAT-1의 활성감소에 의한 결과임을 western blot을 통하여 확인하였다. 특히, $NF-{\kappa}B$의 활성감소는 $I{\kappa}B{\alpha}$의 활성증가에 의한 $NF-{\kappa}B$의 억제능 향상에 의한 것임을 확인하였고, 활성억제된 $NF-{\kappa}B$가 핵 내부로의 이동이 저해되면서 iNOS 유전자 발현에 영향을 미친 것임을 확인하였다. 그러므로, 꽃송이버섯 추출물은 NO 저해능을 이용한 항염증소재로서 염증성질환의 완하에 도움이 될 것으로 사료된다.

Quercetin Ameliorates NO Production via Down-regulation of iNOS Expression, $NF{\kappa}B$ Activation and Oxidative Stress in LPS-Stimulated Macrophages

  • Effect of quercetin on NO production and regulation mode of quercetin on oxidative stress, $NF{\kappa}B$ activation, and iNOS expression, possible mechanisms of NO suppression in LPS-stimulated macrophages were investigated. Treatment of RAW 264.7 cells with quercetin significantly reduced lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production dose-dependently ($IC_{50}$, $9.2\;{\mu}M$). Expression of iNOS and specific DNA binding activities of nuclear factor kB ($NF{\kappa}B$) were significantly suppressed by quercetin pretreatment. Quercetin reduced thiobarbituric acid-reactive substances (TBARS) accumulation, enhancing GSH level and antioxidant activities of enzymes, such as superoxide dismutase (SOD) and catalase. These results demonstrate quercetin may ameliorate inflammatory diseases by suppressing NO production through inhibition of iNOS expression, $NF{\kappa}B$ transactivation, and oxidative stress, which may be mediated partially by antioxidative effect of quercetin. Thus, quercetin appears to be used as a potential therapeutic agent for treating LPS-induced inflammatory processes.

Anti-inflammatory effects of proanthocyanidin-rich red rice extract via suppression of MAPK, AP-1 and NF-κB pathways in Raw 264.7 macrophages

  • BACKGROUND/OBJECTIVES: Several pharmacological properties of red rice extract have been reported including anti-oxidant, anti-tumor, and reduced cancer cell invasion. This study was conducted to evaluate the anti-inflammatory effects of red rice extract on the production of inflammatory mediators in lipopolysaccharide (LPS)-induced Raw 264.7 macrophages. MATERIALS/METHODS: Pro-inflammatory cytokines including tumor necrosis factor-${\alpha}$ and interleukin-6 were determined by ELISA and cyclooxygenase-2 and inducible nitric oxide synthase expression was evaluated using western blot analysis. In addition, the signaling pathway controlling the inflammatory cascade such as nuclear factor kappa B ($NF-{\kappa}B$), activator proteins-1 (AP-1), and mitogen-activated protein kinase (MAPK) was determined. RESULTS: Our results showed that red rice polar extract fraction (RR-P), but not non-polar extract fraction, inhibited interleukin-6, tumor necrosis factor-${\alpha}$, and nitric oxide production in LPS-induced Raw 264.7 cells. RR-P also reduced the expression of inflammatory enzymes, inducible nitric oxide synthase, and cyclooxygenase-2. In addition, activation of AP-1 and $NF-{\kappa}B$ transcription factor in the nucleus was abrogated by RR-P. RR-P inhibited the phosphorylation of extracellular signaling-regulated kinase 1/2, c-Jun NH2-terminal kinase, and p38 MAPK signaling responsible for the expression of inflammatory mediators in LPS-stimulated Raw 264.7 cells. Based on chemical analysis, high amounts of proanthocyanidin and catechins were detected in the RR-P fraction. However, only proanthocyanidin reduced $NF-{\kappa}B$ and AP-1 activation in LPS-activated Raw 264.7 cells. CONCLUSION: These observations suggest that the anti-inflammatory properties of RR-P may stem from the inhibition of pro-inflammatory mediators via suppression of the AP-1, $NF-{\kappa}B$, and MAPKs pathways.
