Elastic analysis of arbitrary shape plates using Meshless local Petrov-Galerkin method

  • In this study the stress analysis of orthotropic thin plate with arbitrary shapes for different boundary conditionsis investigated. Meshfreemethod is applied to static analysis of thin plates with various geometries based on the Kirchhoff classical plate theory. According to the meshfree method the domain of the plates are expressed through a set of nodes without using mesh. In this method, a set of nodes are defined in a standard rectangular domain, then via a third order map, these nodes are transferred to the main domain of the original geometry; therefore the analysis of the plates can be done. Herein, Meshless local Petrov-Galerkin (MLPG) as a meshfree numerical method is utilized. The MLS function in MLPG does not satisfy essential boundary conditions using Delta Kronecker. In the MLPG method, direct interpolation of the boundary conditions can be applied due to constructing node by node of the system equations. The detailed parametric study is conducted, focusing on the arbitrary geometries of the thin plates. Results show that the meshfree method provides better accuracy rather than finite element method. Also, it is found that trend of the figures have good agreement with relevant published papers.

An Easy Method of Disk Diffusion Antibiotic Susceptibility Test for Detection of Erythromycin-induced Resistance to Clindamycin in Staphylococci (포도구균의 Erythromycin 유도성 Clindamycin 내성검출을 위한 간편한 디스크 확산법의 유용성)

  • A simple and easy modification of AST by disk diffusion was tested for the detection of induced clindamycin resistant Staphylococci and their antimicrobial susceptibility at the same time. The incidence of inducible clindamycin resistant staphylococci in blood culture and their MIC characterization at Seoul National University Hospital was analyzed by an AST contained disk approximation test (D-zone test) and Etest, respectively. Of the total 309 staphylococcal isolates, 139 (45%) isolates presented constitutive resistance to ERY and CLI (ERY-R, CLI-R phenotype), and 59 were ERY-I/R and CLI-S phenotypes. Of the 59 isolates, 19 (32%) isolates were inducible resistant to CLI. The incidence was higher in S. aureus (66.7%) than coagulase-negative staphylococci (CNS, 26.0%). Especially, methicillin-resistant staphylococci (MRSA, 100%; MRCNS, 45.5%) presented higher inducibility than methicillin susceptible (MSSA, 50%; MSCNS, 20%). For most of the inducible clindamycin resistant staphylococci (15 of 19 isolates), their ERY MIC were high (>$128_{\mu}g/mL$) and were methicillin resistant. The remaining 4 isolates were methicillin susceptible and their ERY MIC were of intermediate concentrations ($1-4_{\mu}g/mL$). We concluded that suscetibility testing of staphylococci, especially methicillin resistant, should include the D-zone test.

Mutational Analysis Elucidates the Role of Conserved 237 Arginine in 23S rRNA Methylation, Which is in the Concave Cleft Region of ErmSF (ErmSF에서 두 도메인 사이에 존재하는 잘 보존된 237번 아르지닌 잔기의 위치 지정 치환 변이의 효소 활성 검색을 통한 역할 규명)

  • The Erm family of adenine-$N^6$ methyltransferases (MTases) is responsible for the development of resistance to macrolide-lincosamide-streptogramin B antibiotics through the methylation of 23S ribosomal RNA. Recently, it has been proposed that well conserved amino acids in ErnC' located in concave cleft between N-terminal 'catalytic' domain and C-terminal 'RNA-binding' domain interacts with substrate RNA. We carried out the site-directed mutagenesis and studied the function of the ErmSF R237 mutant in vitro and in vivo. R237 amino acid residue is located in the concave cleft between two domains. Furthermore this residue is very highly conserved in almost all the Erm family. Purified mutant protein exhibited only 51% enzyme activity compared to wild-type. Escherichia coli with R237A mutant protein compared to the wild-type protein expressing E. coli did not show any difference in its MIC (minimal inhibitory concentration) suggesting that even with lowered enzyme activity, mutant protein was able to efficiently methylate 23S rRNA to confer the resistance on E. coli expressing this protein. But this observation strongly suggests that R237 of ErmSF probably interacts with substrate RNA affecting enzyme activity significantly.

Change of Water Quality and Growth of Leiocassis ussuriensis Cultivated in a Biofloc System using Bacillus subtilis (Bacillus subtilis을 활용한 바이오플락 시스템에서 사육한 대농갱이(Leiocassis ussuriensis)의 성장 및 사육수 수질 변화)

  • This study observed changes in survival, growth performance and water quality for 90 days to confirm the possibility of Leiocassis ussuriensis farming using Biofloc technology (BFT) using Bacillus subtilis. Feed and molasses were added to the experimental tank to produce BFT water before planting the experiment, and B. subtilis was inoculated to stabilize the water quality for 40 days. The survival rate of the experimental fish was 92.7±3.2% in the control group and 95.8±3.3% in the BFT group. The Weight gain (WG) was 118.1±9.0% of the control and 197.7±15.6% of the BFT, and the Specific growth rate (SGR) was 0.87±0.5% of the control and 1.21±0.06% of the BFT. As for the feed efficiency, the control was 43.7±2.6% and the BFT was measured at 70.1±4.1%, indicating that the feed efficiency of the BFT was higher. As a result of measuring the water quality change during the experimental period, pH was reduced in both the control and the BFT, and Mixed Liquor Suspended Solids (MLSS) did not show any change in the control, but the BFT showed a significant increase at 90 days. NH4+-N and NO2--N showed a significant increase from the 30 days of the experiment in the control, but showed no change in the BFT. In conclusion, as a result of applying the BFT system using B. subtilis to the process of cultivating Leiocassis ussuriensis, the water quality tended to stabilize, and the growth rate and feed efficiency were found to be higher than those of the control, confirmiWng that it had a positive effect.
