Single Oral Dose Toxicity Test of Fermented Sipjeondaebo-tang Extracts in Mice (마우스를 이용한 십전대보탕(十全大補湯) 발효물의 단회투여 독성 연구)

  • Objectives : Sipjeondaebo-tang is a medicine traditionally prescribed as a restorative. The aim of this study was to investigate the single oral dose toxicity and safety of extract of fermented Sipjeondaebo-tang in ICR mice. Methods : In single oral dose toxicity study, non-fermented or fermented Sipjeondaebo-tang were administered by oral gavage to ICR mice (5 males, 5 females) at single doses of varying concentrations: 1250, 2500 and 5000 mg/kg. Changes of body weight, general behavior, adverse effects and mortality were determined throughout the experimental period. Hematological parameters, organ weights and necropsy findings were evaluated at the end of the experiment. Results : There were no mortality or signs of toxicity in single oral dose toxicity studies. There were also no significant differences in body weight, organ weight, or hematological parameters between the treatment and control groups. Conclusions : Fermented Sipjeondaebo-tang did not cause remarkable adverse effects in ICR mice. The oral lethal dose of fermented Sipjeondaebo-tang is more than 5000 mg/kg and no-observed-adverse-effect level (NOAEL) of the extract for both male and female mice is 5000 mg/kg.

Effects of Seed Coating, Slope Control and Soil Mulching on Seed Germination and Seedling Growth of Rehabilitation Plants (종자피복, 경사조절 및 토양멀칭 처리가 녹화식물의 발아와 생장에 미치는 영향)

  • This study was carried out to improve rehabilitation effect by hydro-seeding methods on denuded slopes. Lespedeza cyrtobotrya, Indigofera pseudotinctoria, Arudinella hirta, Poa pratensis, and Lolium perenne were used in this study. The results of the germination and growth characteristics by seed coating, slope control and soil mulching treatments are summarized here. Seed coating of those plants had an effect on germination ratio, germination force, $LD_{50}$, and seedling growth of stem, leaf and root. The seed coating effect was highest for I. pseudotinctoria while the seedling growth was best for L. cyrtobotrya. Vermiculite+Talcum was the best coating material for germination and seedling growth of the plants. As the slopes were steeper, germination ratio was lower and seedling growth of stem and root decreased. Lots of seeds and soils were swept away when the slope was steep. Soil mulching was effective for germination, seedling growth of stem and root, and soil stabilization. It was more effective when the slope was steeper. Coir net was the most effective soil mulching material n this study.

Acaricidal Components of Medicinal Plant Oils Against Dermatophagoides farinae and Dermatophagoides pteronyssinus

  • The oils of Acorus gramineus, Cinnamomum sieboldii, Eugenia aromatica, and Inula helenium were tested for their acaricidal activity against Dermatophagoides farinae and D. pteronyssinus. Responses varied according to dose and mite species. As compared to the oils, the oil most toxic to D. farinae and D. pteronyssinus was E. aromatica, followed by C. sieboldii, A. gramineus, and I. helenium. On the basis of $LD_{50}$ values of the oils in A. gramineus, C. sieboldii, and E. aromatica, the compound most toxic against D. farinae and D. pteronyssinus was eugenol congeners (isoeugenol>eugenol>acetyleugenol) followed by benzyl benzoate, salicylaldehyde, safro1, DEET, cinnamyl alcohol, and 3-carene. As a naturally occurring acaricide, these oils and eugenol congeners could be useful as new acaricidal agents against Dermatophagoides spp.

Identification of Virulence Factors in Vibrio vulnificus by Comparative Transcriptomic Analyses between Clinical and Environmental Isolates Using cDNA Microarray

  • We compared the gene expression among four clinical and five environmental V. vulnificus isolates, using a cDNA microarray containing 131 genes possibly associated with pathogenicity, transport, signal transduction, and gene regulations in the pathogen. cDNAs from total RNAs of these isolates were hybridized into the cDNA microarray using the cDNA of the wild-type strain MO6-24/O as a reference. We focused on selecting differentially expressed (DE) genes between clinical and environmental isolates using a modified t-statistic. We could detect two statistically significant DE genes between virulent isolates and less-virulent isolates with a marginal statistical significance (p-value of 0.008). These were genes putatively encoding pilin and adenlyate cylase. Real time-PCR confirmed that these two selected genes transcribed in significantly higher levels in virulent isolates than in less-virulent isolates. Mutants with lesions in the gene encoding pilin showed significantly higher $LD_{50}$ values than that of wild type.

Edwardsiella ictaluri Infection in Cultured Yellow Catfish Pelteobagrus fulvidraco Fingerlings in Korea (양식 동자개(Pelteobagrus fulvidraco)의 Edwardsiella ictaluri 감염)

  • We observed yellow catfish Pelteobagrus fulvidraco fingerlings cultured in land ponds in Korea swimming in a corkscrew spiral pattern while hanging head-up and tail-down at the water surface, before eventually dying. Externally, these fish displayed “hole in the head” disease, pale gills, and hemorrhages in the base of the pectoral and caudal fins; internally they had liver hemorrhages and kidney discoloration. The bacterium Edwardsiella ictaluri (YCK-01 and YCL-01) was identified in the kidneys and livers of diseased fish via phenotypic characteristics and PCR analysis using the ictaluri-specific primers IVS (an intervening sequence) and IRS (the inter-ribosomal spacer). Infectivity challenges by intraperitoneal and immersion routes showed that a representative bacterial strain (YCK) exhibited strong virulence to yellow catfish, with an LD50 of 3.2×104 CFU/fish and 2.5×106 CFU/mL, respectively. This is the first report of E. ictaluri infection in yellow catfish from Korea.

Outer Membrane Vesicles Derived from Salmonella Enteritidis Protect against the Virulent Wild-Type Strain Infection in a Mouse Model

  • Foodborne contamination and salmonellosis caused by Salmonella Enteritidis (S. Enteritidis) are a significant threat to human health and poultry enterprises. Outer membrane vesicles (OMVs), which are naturally secreted by gram-negative bacteria, could be a good vaccine option because they have many biologically active substances, including lipopolysaccharides (LPS), outer membrane proteins (OMPs), and phospholipids, as well as periplasmic components. In the present study, we purified OMVs derived from S. Enteritidis and analyzed their characteristics through silver staining and sodium dodecyl sulfate polyacrylamide gel electrophoresis. In total, 108 proteins were identified in S. Enteritidis OMVs through liquid chromatography tandem mass spectrometry analysis, and OMPs, periplasmic proteins, and extracellular proteins (49.9% of total proteins) were found to be enriched in the OMVs compared with bacterial cells. Furthermore, native OMVs used in immunizations by either the intranasal route or the intraperitoneal route could elicit significant humoral and mucosal immune responses and provide strong protective efficiency against a lethal dose (~100-fold $LD_{50}$) of the wild-type S. Enteritidis infection. These results indicated that S. Enteritidis OMVs might be an ideal vaccine strategy for preventing S. Enteritidis diseases.

Gamma-Radiation Induced Apoptotic and Inflammatory Degeneration of Mouse Ovarian Follicles : Informative Biological-End Point for Disaster-Prevention

  • In mammals, most of the follicles can not be ovulated, and instead, are degenerated throughout the entire reproductive period. However, the precise mechanism of follicle atresia is unknown. Three weeks old female mice (ICR strain) were ${\gamma}$-irradiated with a dose of LD$^{50}$ . Before irradiation (day 0) and at day 1, 2, and 3 after irradiation, the normal and atretic preantral and antral follicles of the left ovaries were morphologically observed. Atretic follicles at 2 days after irradiation had numerous cell debris, apoptotic cells and bodies, and polymorphonuclear leukocytes in the antral cavity. In severely atretic follicles, numerous polymorphonuclear leukocytes infiltrated into the follicle. The frequencies of atretic antral (58.0 $\pm$8.6) and preantral follicles (27.3$\pm$11.2) induced by ${\gamma}$-radiation increased to 94.0$\pm$3.4 and 86.9$\pm$7.6, respectively at 2 days after irradiation (p<0.05). The number of follicles with one or more neutrophils in the largest cross sections at 2 and 3 days after irradiation significantly increased (p<0.05). It can be concluded that ${\gamma}$-radiation triggers the recruitment of neutrophils into the follicles during degeneration. The ovarian follicles can make a role of informative biological end-point useful for disaster-prevention.

  • For thestudying and utilizing the thorn sea cucumber synthetically, the method preparing polysaccharide and polypeptide from sea cucumber was studied. The comparatively rational craft for preparing crude polysaccharide is fresh raw materials deal with oar form, $60\%$ of the ethanol precipitate after hydrolyzed by pretense and vibrated with ultrasonic wave auxiliary. The purification of polysaccharide and removal of protein fromcrude polysaccharide were made through precipitation method using the acetate. The polypeptide is obtained by concentrating in vacuum, freeze-drying after mixing precipitations of two times. Polysaccharide acute poisoning experiment indicate there is no bad reaction when LD50>5000g/kg, there strain rate of liver tumor H11 approach present generally acknowledged tumor treat medicine cyclophosphamide, and there is no side effect. The fatigue resistance function of polypeptide experiment also indicates that the fatigue resistance ability of mouse which fed on added sea cucumber polypeptide has a great improvement.

Tissue culture of the plant Pluchea indica (L.) Less. and evaluation of diuretic potential of its leaves

  • The present study focused on the establishment of micropropagation protocol for the high value Pluchea (P.) indica (L.) Less., genotype, an important medicinal plant and evaluation of the diuretic activity of the leaf extract of the tissue cultured plant. Leaf explants, nodal segments and shoot tips were cultured in MS medium supplemented with auxin and cytokinin and their combinations. With the objective of inducing callus giving rise to new adult plants, naphthalene acetic acid was found to be most effective for (80%) for callus induction. The methanolic extract of leaves of the micropropagated P. indica was investigated for its diuretic activity in Wistar albino rats. Urinary excretion parameters were studied for evaluation of diuretic activity using Frusemide (20 mg/kg, p.o.) as standard. The extract showed significant diuretic activity at the doses of 100, 200 and 300 mg/kg. p.o. An oral acute toxicity study for the extract was carried out and the $LD_{50}$ value was found to be 2,825 mg/kg body weight.

Characteristics of amorphous-to-crystalline phase transformation in the Al-added $Ge_2Sb_2Te_5$ films (Al을 첨가한 $Ge_2Sb_2Te_5$ 박막의 비정질-결정질 상변화 특성)

  • 본 논문에서는 PRAM 에서 기록매질로 이용될 수 있는 최적의 물질을 찾고자 $Ge_2Sb_2Te_5$ 박막에 Al을 첨가하여 비정질-결정질 천이시의 원자구조와 상변화 특성간의 관계를 연구하였다. 이 실험에 사용된 $Al_x(Ge_2Sb_2Te_5)_{1-x}$ 조성은 5N의 금속 파우더를 용융-냉각법으로 벌크를 제작하였고 열증착 방법으로 Si (100) 및 유리 (corning glass, 7059) 기판위에 200nm 두께로 박막을 증착하였다. 비정질 박막의 상변화에 따른 반사도 차이를 평가하기 위해서 658 nm의 LD가 장착된 나노펄스 스캐너를 이용하여 power; 1~17mW, pulse duration; 10~460 ns의 범위에서 각 조성의 비정질-결정질 상변화속도를 측정, 비교 분석하였다. 또한 각각의 박막을 $100^{\circ}C$ 에서 $400^{\circ}C$ 까지 $50^{\circ}C$ 간격으로 $N_2$ 분위기에서 1시간동안 열처리 한 후 XRD와 UV-Vis-NIR spectrophotometer를 사용하여 각 상의 구조분석 및 광학적 특성을 분석하였다. 또한 4-point probe로 면저항을 측정하였다.
