Effect of Plant Growth Regulators on Regeneration from the Cotyledon Explants in Watermelon (Citrullus lanatus (thunb.) Matsum. & Nakai) (수박(Citrullus lanatus (thunb.) Matsum. & Nakai) 자엽 절편의 재분화에 미치는 생장조절물질의 영향)

  • In this study, we developed a high frequency watermelon regeneration system using three breeding lines ('B02', 'B05' and 'D04') of watermelon (Citrullus lanatus (thunb.) Matsum. & Nakai) which are differed in their fruits in shape, color of pericarp and flesh. The highest frequency of explants with callus was observed by using explants that consist of cotyledon proximal part end in all breeding lines, and the highest rate of callus induction was obtained on MS medium containing 1.0 mg/L BAP + 0.5 mg/L IAA for 'B02' (94%), 3.0 mg/L BAP for 'B05' (95%), 3.0 mg/L BAP + 0.1 mg/L IAA for 'D04' (90%). The highest shoot regeneration rates from derived callus were obtained on MS medium containing 1.0 mg/L BAP + 0.5 mg/L IAA for 'B05' (94%), and then a 'B02' (81%) with a same culture conditions, and the lowest regeneration was obtained on MS medium containing 1.0 mg/L BAP for 'D04' (56%). Regenerated plants showed the best rates of root formation on MS containing 0.1 mg/L IBA for 'B02' (67%), 0.1 mg/L NAA for 'B05' (87%), 0.5 mg/L IAA for 'D04' (88%). The regenerated plants showed a 100% survival rate in soil condition. The tissue culture and regeneration conditions obtained from this study will be useful for regenerating plants in breeding applications, and will be a useful tool for further genetic transformation studies on watermelons.

  • To set up the outstanding and scientific dyeing method in making the condensed liquid of pigment obtained from onion skins and the improved reliability, the following basic experiments were performed. The pigment was extracted in the distilled water at $70^{\circ}C$ and methanol at room temperature and then it was analyzed with LC/MS/MS system (Liquid Chomatography/Mass Spectroscopy/Mass Spectroscophy, LIQ Advantage Max, Thermo Finnigan, USA) for its pigmental characteristics. The unrefined silk and refined silk were dyed by making use of the derived pigment in such a way. The chromameter (CR-200, Minolta, Japan) was used to measure the change in surface color in textiles to be dyed by the extracting condition and the color difference ${\Delta}E$ was determined according to the color difference formula CIE LAB through measuring the psychometric lightness L* and chromaticity coordinates a* and b*.

Use of Resin Infiltrant to Prevent Discoloration after Teeth Whitening

  • Background: This study attempted to apply resin infiltrant (RI) as a method to maintain the effect of tooth bleaching treatment and compared it with fluoride varnish (FV) or artificial saliva to evaluate the effect. Methods: Sixty healthy lozenge specimens were classified into five groups. Group 1 was the negative control group, and discoloration was induced after artificial saliva treatment of the tooth specimen (G1S+C). Group 2 was a positive control group, in which pigmentation was induced after bleaching treatment and artificial saliva treatment (G2 B+S+C). Coloration was induced in group 3 (experimental group 1) after bleaching treatment and artificial saliva treatment, followed by application of fluorine varnish (G3B+FV+S+C). Coloration was induced in Group 4 (experimental group 2) after applying RI after bleaching treatment and artificial saliva treatment (G4B+RI+S+C). Pigmentation was induced in group 5 (experimental group 3) after bleaching treatment and artificial saliva treatment, followed by acid treatment (etching) and treatment with RI (G5B+E+RI+S+C). Coffee and wine were used to induce discoloration. The lightness value (L*) of the CIE L*a*b* color system was obtained by image analysis. Kruskal-Wallis H analysis was performed for the mean difference in L* values by group. Results: When coloration was induced with coffee, there was no significant difference in L* value between artificial saliva (G2 B+S+C), FV (G3B+FV+S+C), and RI (G4B+RI+S+C, G5B+E+RI+S+C) groups. There was no significant difference in L* values between the artificial saliva (G2 B+S+C), FV (G3B+FV+S+C), and RI (G4B+RI+S+C, G5B+E+RI+S+C) groups, even in the case of wine induced coloration. Conclusion: It was confirmed that artificial saliva or RI treatment had similar effects to the FV previously used to maintain the effect of tooth bleaching treatment.

A New Method For Measuring Acupoint Pigmentation After Cupping Using Cross Polarization (교차편광 촬영술(Cross Polarization Photographic Technique)를 이용한 부항요법의 배수혈 피부 색소 침착 변화 측정 평가)

  • Objectives : Skin color deformation by cupping has been widely used as a diagnostic parameter in Traditional Korean Medicine(TKM). Skin color deformation such as ecchymoses and purpura is induced by local vacuum in a suction cup. Since existing studies have relied on a visual diagnostic method, there is a need to use the quantitative measurement method. Methods : We conducted an analysis of cross-polarization photographic images to assess the changes in skin color deformation. The skin color variation was analyzed using $L^*a^*b^*$ space and the skin erythema index(E.I.). The meridian theory in TKM indicates that the condition of primary internal organs is closely related to the skin color deformation at special acupoints. Before conducting these studies, it is necessary to evaluate whether or not skin color deformation is influenced by muscle condition. Hence, we applied cupping at BL13, BL15, BL18, BL20 and BL23 at Bladder Meridian(BL) and measured blood lactate at every acupoint. Results : We confirmed the high system measurement accuracy, and observed the diverse skin color deformations. Moreover, we confirmed that the $L^*$, $a^*$ and E.I. had not changed after 40 minutes(p>0.05). The distribution of blood lactate levels at each part was observed differently. Blood lactate level and skin color deformation at each part was independent of each other. Conclusions : The negative pressure produced by the suction cup induces a reduction in the volumetric fraction of melanosomes and subsequent reduction in epidermal thickness. The relationship between variations of tissue and skin properties and skin color deformation degree must be investigated prior to considering the relationship between internal organ dysfunction and skin color deformation.

The metameric effect of monolithic zirconias with varying yttrium ratios

  • PURPOSE. To evaluate the metameric disparities among monolithic zirconia materials with differing yttrium compositions across various lighting conditions. MATERIALS AND METHODS. Thirty-six square-shaped zirconia samples measuring 10 × 10 × 0.5 mm were prepared from monolithic zirconia materials with three different yttrium contents. A 0.2 mm thick layer of polymerized dual-polymerizable self-adhesive resin cement was created using a silicone mold with the same dimensions as the prepared zirconia specimens. To evaluate metamerism, color measurements were conducted using a spectrophotometer device on a neutral gray background in a color measurement cabinet that offers four different illumination environments. All samples underwent aging by subjecting them to 10000 thermal cycles using a thermal cycle tester. Following thermal aging, color measurements were taken once more, and the data were recorded using the CIE L*, a*, b* color system. Two-way ANOVA and Post-hoc Bonferroni tests were employed to analyze the data. RESULTS. It was observed that there was no statistical difference among the color measurements made in different illumination environments of the monolithic zirconia ceramics used to evaluate metamerism (P > .05). This observation remained consistent both before and after thermal aging. After thermal aging, the color of monolithic zirconia materials exhibited a tendency towards red and yellow hues, accompanied by a decrease in brightness levels. CONCLUSION. It can be stated that different illumination conditions did not affect the metamerism of monolithic zirconia materials, but there was a color change in monolithic zirconia materials after a thermal aging period equivalent to one year.

Variation and Sex-limited Expression of Fluorescent Color by Ultraviolet Spectrum on the Silkworm Cocoon (누에고치의 분광성에 관한 계통별 변이 및 한성적 발현)

  • Ultraviolet weavelength (UV) of 366 nm produced clearer fluorescent dolor than that of 254 nm for the inspection of silkworm cocoons. Fluorescent color of silkworm cocoons varied in color, appears no relationship with the natural color under the normal light. Uniformity of fluorescent color was improved by selection of blue or yellow line from wild types. Blue and yellow, located at the opposite poles on the color solid and L*a*b* color system, confirmed as pure standard of fluorescent color in the silkworm races for commercial white cocoons. the cocoons with blue fluorescence occupied as high as 1.7 to 8.6 times than those with yellow in the Japanese silkworm races. Fluorescence of silkworm cocoon was not affected by forced flow dry at 70$^{\circ}C$ for 6 hrs. While the Japanese races revealed no sexual difference in fluorescent color, sex-dependence of the color was common in the Chinese races for commercial white cocoon. The fluorescence of cocoon shell of Chinese races showed clear separation of blue of median color. Silkworm strain of Dc20 and Fc24 were sexualy segregated 98.8${\pm}$1.20%, 99.0${\pm}$1.00% by cocoon fluorescence, as that of 99.3${\pm}$0.44% by typical larval marking of sex-limited inheritance. Specific expression of cocoon fluorescence, applicable to breeding of simple discrimination of sex for Chinese races, inspected thoroughly on the surface and inner layer of cocoon shell.

Study of Tongue Color Histogram in Elderly People with Evacuation Disorder (대변장애를 주증으로 하는 고령자의 혀 색상 히스토그램 특성 연구)

  • Evacuation disorder(ED) is frequently observed in the elderly people. In this study, we investigated the tongue color properties in the elderly. 327 subjects were participated in this study and classified into normal group (n=95) and ED group (n=23) by two Korean Oriental Medicine doctors. The tongue images were acquired by using computerized tongue diagnosis system, and its color were linearly corrected base on CIE $L^*a^*b^*$ values of 12 color samples. The tongue region was segmented from acquired image and divided into two regions along the vertical direction. In order to estimate color properties of the tongue, a color histogram was calculated for the root region based on the CIE $L^*$ and $a^*$ values, and differences of color histogram values between normal and ED groups were computed based on the Mann-Whitney U test. As results, pixels corresponding to typical colors of the pale tongue and thin tongue coating were significantly more distributed in ED than those in normal group(p<0.05). The tongue color of the root region in ED was revealed to be different from those in healthy subjects.

The Development of a Tongue Diagnosis System and the Evaluation of Reproducibility (설진 시스템 개발 및 재현성 평가)

  • The tongue diagnosis is a diagnostic method in the oriental medicine that uses shape, substance, coating, and movement of the tongue to determine the condition of health and disease characteristics in human. Since this information, however, could be affected by subjective sense and visual information, it is difficult to obtain the objective and reproducible results. This research aims at building a reproducible tongue diagnosis system using color chart that is attached close to the face contact region. The picture of color chart is taken simultaneously with a tongue and applied to color revision. The system, in addition, is focused on providing a clear tongue image through securing a sufficient photographing distance with a surface coating mirror. The lightning part which can suppress the reflection by sputum in maximum is implemented for the objectification and quantification of the tongue diagnosis system. The face contact region is designed for consideration of a testee's convenience. To evaluate the reproducibility of the system, the CVs (coefficient of variance, %) of $L{\ast}$, $a{\ast}$ and $b{\ast}$ of red, green and blue regions in color chart are calculated, respectively. The results of all CVs shows that the tongue diagnosis system is re liable and those consequences contribute to the objectification and quantification of the tongue diagnosis system.

Evaluation of the color reproducibility of all-ceramic restorations fabricated by the digital veneering method

  • PURPOSE. The objective of this study was to evaluate the clinical acceptability of all-ceramic crowns fabricated by the digital veneering method vis-$\grave{a}$-vis the traditional method. MATERIALS AND METHODS. Zirconia specimens manufactures by two different manufacturing method, conventional vs digital veneering, with three different thickness (0.3 mm, 0.5 mm, 0.7 mm) were prepared for analysis. Color measurement was performed using a spectrophotometer for the prepared specimens. The differences in shade in relation to the build-up method were calculated by quantifying ${\Delta}E^*$ (mean color difference), with the use of color difference equations representing the distance from the measured values $L^*$, $a^*$, and $b^*$, to the three-dimensional space of two colors. Two-way analysis of variance (ANOVA) combined with a Tukey multiple-range test was used to analyze the data (${\alpha}$=0.05). RESULTS. In comparing means and standard deviations of $L^*$, $a^*$*, and $b^*$ color values there was no significant difference by the manufacturing method and zirconia core thickness according to a two-way ANOVA. The color differences between two manufacturing methods were in a clinically acceptable range less than or equal to 3.7 in all the specimens. CONCLUSION. Based on the results of this study, a carefully consideration is necessary while selecting upper porcelain materials, even if it is performed on a small scale. However, because the color reproducibility of the digital veneering system was within the clinically acceptable range when comparing with conventional layering system, it was possible to estimate the possibility of successful aesthetic prostheses in the latest technology.

Study of Reaction Products and Color Changes in Glutamine-Glucose Model System during Heating (Glutamine-Glucose Model System에서 가열조건별 반응생성물과 색의 변화)

  • The product formation and changes in color of glucose/glutamine model system were investigated in relation to heating temperature and time. The mixtures of glucose and glutamine in equal molar ratio were heated at 125, 150 and $175^{\circ}C$ for 10, 20 and 30 minute, respectively. Acetic acid, butanoic acid, 2-butenoic acid, di-(2-cthylhexyl)phthalate, 2,3-dihydro-3,5-dihydroxy-6-methly-4H-pyran-4-one and 5-hydroxymethylfurfural were identified as a major compounds, and 1,3-dimethylbenzene, 2-ethylhexanol, furfural, 5-methylfurfural, 2-pyrrolidinone, and 2,6-di(t-butyl)-4-hydroxy-4-methyl-2.5-cyclohexadien-1-one as 6 minor compounds by using GC/MS. The contents of acetic acid, 2-ehylhexanol and 2-pyrrolidinone increased with increased heating temperature and time, whereas the formation of the other 9 compounds increased up th heating conditions of $150^{\circ}C$ for 10 or 20 min or $175^{\circ}C$ for 10 min, and decreased dramatically with heating above those conditions. Color parameter $L^*$ decreased with increasing heating conition, resulting in dark brown color in final products. Changes of redness parameter $a^*$ and yellowness $b^*$ showed similar to those of the contents of 9 compounds mentioned as above.
