Pharmacokinetics and urinary excretion of sulfamethoxazole were investigated in healthy sheep. From the plasma disappearance curves after intravenous bolus injection (50 mg/kg), the half-life and volume of distribution were found to be 76 $\PM$14 min and 0.41 $\PM$ 0.18 lit/kg respectively. Body clearance was 4.06 $\PM$ 1.03 ml/kg/min. Very low Concentration of ddrug was present in plasma after 3 hours of administration and plasma level at 6 hour was only 4.4 $\PM$ 2.0 $\mu$g/ml. The renal clearance of sulfamethoxazole (22 $\PM$ 2.17 ml/min/10 kg) exeeded the creatinine clearance (9.78 $\PM$ 1, 57 ml/min/ 10 kg) which may be due to involvement of active tubular secretion and pH dependent back diffusion. Half of the dose of sulfamethoxazole was excreted as unchanged free drug while acetylated amine comprised of 20 percent within the first 6 hours of drug administration.
The metabolic fate of ginsenosides including gastrointestinat absorption, organ distribution, excretion and metabolism in liver was investigated by tracer studies using the radio-labeled ginsenosides. 3H-ginsenosides were shown to be absorbed from the mouse digestive tract and then to be excreted rapidly into urine and/or bile. Bile juice was concluded to play a significant role in absorption of ginsenosides. The total concentration of radioactivity persisted in tissues 24 hrs after oral administration was less than 1.3% of the administered dose and Rbl showed the highest value. The concentrations of radioactivity were relatively high in the liver and kidney. After administration of Rbl radioactivity was detected in the brain. After oral administration of 8H-ginsenosides, major component excreted into urine was found to be the intact ginsenosides and decomposed and/or metabolized products were found in GIT in the case of Rbl. 3H-ginsenoside Rbl was shown to be metabolized in the liver and the metabolite was suggested to be an acylated compound of Rbl by a certain organic acid.

An experiment was conducted to investigate the effects of environmentally friendly agents (yucca extract, mineral feed additive, acidifier, nonspecific immunostimulating anionic alkali solution) on the growth performance, nutrient digestibility, nutrient excretion and carcass characteristics in growing-finishing pigs. A total of 125 pigs were allotted into 5 treatments with 5 replications, 5 pigs per pen in a completely randomized block design. Dietary treatments consisted of 1) control: basal diet, 2) YE: basal diet+120 mg/kg of yucca extract, 3) MFA: basal diet+50 mg/kg of mineral feed additive, 4) NIS: basal diet+3% of NIS (nonspecific immunostimulating anionic alkali solution), and 5) Acidifier: basal diet+0.5% of acidifier (organic acid complex). During the growing phase of the feeding trial, There were no significant differences in average daily feed intake (ADFI) and feed conversion ratio (F/G) among treatments, but the Acidifier group showed higher average daily gain (ADG) than the MFA group (p<0.05). During the finishing phase, there were no significant differences in growth performance among treatments, however YE supplementation increased ADG and F/G by 3.1 and 6.15%, respectively, when compared to control group. Proximate nutrients (dry matter, DM; crude protein, CP; crude ash, CA; crude fat, CF; gross energy, GE; calcium, Ca and phosphorus, P) digestibility did not show any significant differences among the treatments. And there were no significant differences in DM, N and P excretion as well. The supplementation of environmentally friendly agents tended to increase carcass weight compared to control group. Pigs fed MFA showed significantly heavier (p<0.05) carcass weight than those fed control or Acidifier diets. Addition of these agents except for NIS to diet for growing-finishing pigs caused a similar feed cost/kg weight gain than control group. Therefore, present study suggested that the optimum environmentally friendly agent would be yucca extracts, considering the effects on growth performance, nutrient digestibility, nutrient excretion, carcass characteristics and feed cost in growing-finishing pigs.

The objective of the present study was to examine whether yaks possess any adaptive mechanisms of nitrogen (N) metabolism to survive in the harsh foraging environment of the Qinghai-Tibetan Plateau. A grazing experiment on native alpine meadows was conducted to determine availability of herbage biomass and body weight (BW) change of yaks over the year, followed by two indoor feeding trials to investigate adaptation mechanisms of N metabolism in yaks fed at similar intake level to grazing conditions. Three castrated males of each of three genotypes; yak (Bos grunniens), indigenous cattle (Bos taurus) and their crossbred - cattleyak (Bos taurus male${\times}$Bos grunniens female), were used in the housed trials. Results showed that: i) Monthly herbage biomass production and daily grazing intakes by yaks over the year ranged from 220 to 4,664 kg DM per ha, and 1.90 to 8.50 kg DM, respectively. For about seven months each year, yaks suffer from malnutrition as a result of inadequate pasture conditions; ii) Urinary N excretion and N retention by yaks were significantly affected by feeding level, and yaks had a lower (p<0.05) average daily urinary N excretion (0.39 g/kg $BW^{0.75}$) and a greater (p<0.05) N retention (-0.09 g/kg $BW^{0.75}$) than indigenous cattle (0.47 and -0.16 g/kg $BW^{0.75}$, respectively). Fasting daily urinary N excretion was greater (p<0.05) for indigenous cattle than yaks (353 vs. 248 mg/kg $BW^{0.75}$). Purine derivative N excretion and purine derivative N index (PNI) increased with increasing feeding level, while the value of PNI was greater (p<0.05) for yaks and cattleyak (0.11 and 0.12, respectively) than for indigenous cattle (0.09) during the feeding trials. 목적: Tc-99m DISIDA 신티그라피에서 간 실질에 분절형으로 배설지연을 보이는 소견은 분절형 간내담관폐쇄의 소견으로 보고되고 있으나, 이러한 소견의 분절형 간내 담관폐쇄에서의 진단적 가치는 알려져 있지 않다. 본 연구에서는 간 실질 분절형 배설지연 소견의 분절형 간내 담관폐쇄 진단에서의 양성예측도와, 방사선학적 검사와 비교한 진단적 가치를 알고자하였다. 대상 및 방법: Tc-99m DISIDA 신티그라피에서 간 실질에 분절형 배설지연 소견을 보인 43명의 환자(48개 영상)를 대상으로하여 그 원인을 분석하고, 1개월 이내에 시행된 CT와 US와 비교하였다. 결과: 31예는 ERC나 PTC에 의해, 13예는 임상적 경과 관찰에 의해 분절형 담도 폐쇄가 확진되었고, 4예는 원인이 규명되지 않았다. 분절형 담관폐쇄에 대한 Tc-99m DISIDA 간담도 신티그라피의 양성예측도는 92% (44/48)이었다. US의 28%(5/ 18), CT의 13% (5/38)는 정상 소견이고, US의 17% (3/18), CT의 18% (7/38)에서는 담관 확장 소견만이 있었고, 원인이 되는 병변을 알 수 없었다. 결론: Tc-99m DISIDA 신티그라피에서 간 실질에 분절형 배설지연을 보이는 소견은 분절형 간내 담관폐쇄 진단에 높은 양성 예측도를 가진다. Tc-99m DISIDA 신티그라피는 특히 US나 CT가 정상인 경우에 분절형 담관폐쇄 진단에 선별검사로서 이용되어질 수 있을 것으로 생각된다. Tc-99m DISIDA 신티그라피의 분절형 담도관쇄 진단에서의 진단적 가치에 대한 전향적인 연구가 요구된다.
Four local cattle were ($145{\pm}9.7kg$) used in a $4{\times}4$ Latin square design to study the effect of different levels of rumen degradable protein (RDP) in straw based ration on purine derivatives excretion and microbial N supply in cattle. The four rations were formulated at the same amount of energy but varying RDP approximately 50 (U0), 75 (U1), 100 (U2) and 150 (U3) percent levels of RDP requirement for maintenance. They were fed ranged from 101 to 304 g RDP/d. Apparent digestibility of all nutrients increased significantly (p < 0.01) in cattle fed ration U2 than other rations. Rumen $NH_3-N$ concentration increased from 43 to 130 mg/l in response of RDP intake. Purine derivatives excretion increased significantly (p < 0.01) with incremental level of 203 g RDP/d (U2) intake and positively correlated (r=0.69, p < 0.01, n=16) with amount of RDP intake. The rates of rumen microbial N supply were 16.8, 27.2, 39.1 and 32.9 g/d for rations U0, U1, U2 and U3 respectively. Efficiency of microbial N supply (EMNS) per kg of DOMR were 19.0, 25.3, 33.0, and 28.6 g and per MJ of ME. Intake were 0.62, 1.00, 1.44 and 1.21 g for U0, U1, U2 and U3 respectively and highest results were obtained in cattle fed U2 ration. Results of this study suggest that PD excretion and EMNS were increased as incremental level of RDP intake (U2) in local cattle.

In an attempt to dofine the early biochemical determinants that participate in the pathogenesis of glycerol-induced nephrotoxicity, especially focusing on oxygen free radicals and $N-acetyl-{\beta}-D-glucosaminidase$ (NAG) activity, we studied 24-hours urine outflow, 24-hours urinary protein excretion and urinary NAG activity after the injection of glycerol and also we studied malondialdehyde(MDA) level and superoxide dismutase(SOD) activity in the kidney of rats at 24hr after the injection of glycerol. Sprague-Dawley albino rats weighing 240 to 260 gm were injected intramuscularly with a 50% solution of glycerol(2ml/kg, 4ml/kg and 8ml/kg). The group treated with glycerol showed significantly lower urine outflow level and urinary protein excretion level and higher urinary NAG activity after the injection as compared to those of control group. Also the group treated with glycerol showed significantly higher MDA level and lower SOD activity at 24hr after the injection as compared to those of control group. These results suggest that the excessive oxygen free radicals resulting from the depression of SOD activity is an important determinant in the pathogenesis of glycerol-induced nephrotoxicity and higher urinary NAG activity is an index of renal tubular cell damage in the glycerol-induced nephrotoxicity.
Plasma phamacokinetics and renal excretion of theophylline (TP) and its metabolities were ivnestigated in rats. Plasma concentrations of TP declined in a monoexponential manner, while those of 1-methyluric (MU) and 1,3-dimethyluric(DMU) declined in a biexponential manner upon respective iv bolus injection of each compound at 6mg/kg dose. The total body clearances $(CL_r)$ of the metabolites were 4-6 fold larger than that of TP, while the distribution volumes of them at steady-state $(Vd_{ss})$ were 40-50% smaller than that of TP. The metabolites showed their plasma peaks in 30 min after iv injection of TP indicating than that to MU. Renal excretion of TP and its metabolites was studied in urine flow rate (UFR)-controlled rats. The renal clearance $(CL_r)$ of TP was inversely related to pasma TP concentrations, and much smaller than the glomerular filtration rate (GFR) suggesting tubular secretion and profound reabsorption in the renal tubule. The $(CL_r)$ of each metabolite also showed that inverse relationship, but far exceeded GFR suggesting that tubular secretion than GFR by ip injection of probenecid (142.7 mg/kg). It supports that the metabolies are secreted in the renal tubule, and suggests that they share a common transport system in their sectrtion processes with probenecid. On the other hand, the $(CL_r)$ of TP was not affected significantly by the probenecid treatment. Considering the inverse relationship of TP between the $(CL_r)$ and its ploasma concentrations,no effect of probenecid on $(CL_r)$ of TP is most likely due to negligible contribution of the secretion to the overall $(CL_r)$ of TP.
In this study, Schizandriae fructus which has been used in oriental medicine and folks remedy, was studied to apply to functional foods and oriental medicinal cuisine. The aim of this experiments was to investigate the effects of Schizandriae fructus water extract(SFE) on the renal function, plasma renin activity, plasma levels of aldosterone and arterial natriuretic peptide(ANP) in rats. Spargue-Dawley rats weigh 200g, were randomly assigned to 3 groups such as basal diet only(BDG), basal diet with $0.5{\mu}L/g$ SFE(LAG) and basal diet with $1.0{\mu}L/g$ SFE(HAG). The results were as follows. Water balance decreased significantly after administration for 2 weeks compared with the control period in HAG. Urine volume increased significantly after administration for 1 week compared with the control period in LAG and HAG. Urinary excretion of sodium increased significantly after administration for 1 week and for 2 weeks compared with the control period in LAG and HAG. Urinary excretion of creatinine increased significantly after administration for 2 weeks compared with the control period in HAG. Plama levels of ANL decreased significantly after administration of $SFE(0.5{\mu}L/g)$. Plasma levels of aldosterone decreased significantly after administration of $SFE(1.0{\mu}L/g)$. There results indicated that Schizandriae fructus can improve the renal function through increased urine volume and sodium excretion. These results imply that SFE could be used as a potent food resource for diet therapy or clinical nutrition.
