Inhibition of Dermatitis Development by Sopungsan in Nc/Nga Mice

  • Sopungsan (SS) is a traditional Korean decoction used for the treatment of dermatitis. The aim of this study is to confirm whether or not SS has a preventive effect on the development of atopic dermatitis in dinitrochlorobenzene-applied Nc/Nga mice. SS was administered orally to Nc/Nga mice, which led to the remarkable suppression of the development of dermatitis, as determined by a histological examination and the serum IgE levels. Moreover, SS inhibited the production of thymus- and activation-regulated chemokine (TARC) and its mRNA expression in a keratinocyte cell line, HaCaT, which had been stimulated with tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interferon-${\gamma}$ (IFN-${\gamma}$). Activation of the nuclear factor-${\kappa}B$ (NF-${\kappa}B$) or activator protein-1 (AP-1) is one of key steps in the signaling pathways mediating induction of TARC. In this study, SS selectively suppressed NF-${\kappa}B$ activation which may be essential for TARC expression in $TNF-{\alpha}/IFN-{\gamma}$ treated keratinocytes. The inhibitory effect of SS on NF-${\kappa}B$ activation and TARC production might be associated with the anti-dermatitic effects of SS.

The Effects of Gardenia jasminoides J.Ellis on Allergic Inflammation in RBL-2H3 Mast Cells and OVA/alum-sensitized Mice (쥐 비만세포 (RBL-2H3)와 OVA/alum 감작 마우스에서 치자 추출물의 알레르기 염증 억제 효과)

  • Objectives The purpose of this study is to investigate the effects of Gardenia jasminoides for. grandiflora extracts' (GAJ) anti-inflammatory effect on RBL-2H3 mast cells and OVA/alum-sensitized mice. Methods In this study, IL-4 and IL-13 production was measured via ELISA analysis, and mRNA expressions of GM-CSF, IL-4, IL-5, $TNF-{\alpha}$, IL-6 were analyzed by real-time PCR. In addition, MAPKs and $NF-{\kappa}B$ p65 transcription factors were examined using western blotting, and ELISA was used to understand IgE, IL-4, and IL-13 production in ovalbumin-allergic mice in in vitro study. Results As a result of this study, 1. GAJ were observed to suppress the mRNA expression of GM-CSF, IL-4, IL-13, IL-5, $TNF-{\alpha}$, IL-6 in comparison to PMA 50 ng/ml, ionomycin $0.5{\mu}M$ (PI) control group. 2. GAJ also inhibited the IL-4, IL-13 production in comparison to PI control group. 3. Western blot analysis showed decrease on the expression of mast-cell-specific transcription factors, including MAKPs (ERK, JNK, p38) and $NF-{\kappa}B$ p65. 4. Orally-administered GAJ group in OVA/alum induced Balb/c mice showed decreased level of OVA-specific IgE in the serum. This group also has shown decreased the level of IL-4, IL-13 in the splenocyte culture supernatant. Conclusions Obtained results suggest that GAJ may regulate the allergic inflammation by transcription factors MAKPs (ERK, JNK, p38) and $NF-{\kappa}B$ p65 causing inhibition of Th2 cytokines in mast cells and OVA/alum-sensitized mice.

Suppressive Effect of Kwanjulbang-6 on Collagen-Induced Arthritis in Mice (관절6호방이 Type II collagen으로 유발된 관절염에 미치는 영향)

  • This study was carried out to know the effects of Kwanjulbang-6(GJB-6) on the inhibition of arthritis. GJB-6 was orally administered to mouse with arthritis induced by collagen II . Cytotoxicity, hepatotoxicity, arthritis index, value of immunocyte in draining lymph node and paw joint, rheumatoid factor in serum were measured in vivo. The incidence of arthritis was significantly decreased. Total cell number of draining lymph node was significantly increased compared with control. Total cell number of paw joint was significantly decreased compared with control. The absolute number of $CD19^+$, $CD8^+$, $CD3^+/CD69^+$, $CD3^+/CD49b^+$, $CD4^+/CD44^+$, $CD3^+$, $CD4^+$, $CD4^+/CD25^+$ and $CD3^+/CD8^+$ cells in draining lymph node were significantly increased compared with control. The absolute number of $CD3^+$, $CD4^+$, $CD4^+/CD25^+$ and $CD11b^+/Gr-1^+$ cells in paw joint were significantly decreased compared with control. The absolute number of $B220^+/CD23^+$ and $MHCII^+/CD11c^+$ cell in draining lymph node were significantly decreased compared with control. The levels of IgG was decreased and The levels of IgM was significantly decreased compared with control. Anti-collagen II in serum was significantly decreased compared with control. With the hematoxylin and eosin stain, the cartilage destruction and synovial cell proliferation were decreased compared with control. With the Masson's trichrome stain, the expression of collagen fibers was decreased compared with control. Results showed that GJB-6 had immunomodulatory effects. So we expect that GJB-6 should be used as a effective drugs for not only rheumatoid arthritis but also another auto-immune disease.

The Effects of Forsythiae Fructus n-BuOH Fraction on Atopic Dermatitis (연교(連翹) n-BuOH 분획물의 아토피 피부염 억제 효과)

  • Objectives Previous studies have found out that Forsythiae Fructus (FF) extracts have anti-atopic activities by in vitro experiment. In order to understand more about FF extracts' benefit, we subdivided FF extracts depending on systematic fractionation method by using Methylene chloride (MC), Ethyl acetate (EtOAc), n-BuOH and n-hexane (n-Hx). This study is designed to examine the effect of FF fractions on the PMA- ionomycin-induced activation of RBL-2H3 mast cell lines in vitro and on the DNCB-induced activation of NC/Nga mice in vivo. Methods For this study, we examined IL-4, IL-13 production by ELISA analysis, IL-4, IL-13, IL-31, IL-31RA and TNF-${\alpha}$ mRNA expression by real-time PCR and manifestations of AP-1 and MAPKs transcription factors by western blotting in vitro. Through in vitro experiment, we selected FF n-BuOH fraction that seems the best effective in atopic dermatitis then induced it on NC/Nga mice by DNCB. We measured mice's WBC, eosinophil and neutrophil in heart blood, IL-4, IL-5, IFN-${\gamma}$ in the spleenocyte culture supernatant, the absolute cell numbers of CD4+, CD8+, B220+CD23+, CD3+CD69+ and Gr-1+CD11b+ in the PBMCs, ALN and dorsal skin, IL-5, IL-13, IL-31, IL-31RA in the dorsal skin by real-time PCR and the distribution of immune cells by H&E on dorsal skin and ANL and toluidine blue staining on dorsal skin. Results FF n-BuOH fraction suppressed IL-4, IL-13 production and mRNA expression of IL-4, IL-13, IL-31, IL-31RA and TNF-${\alpha}$. Results from the western blot analysis showed that FF n-BuOH fraction reduced the activation of the mast cell specific transduction factors involved in AP-1 by suppressing JNK and ERK phosphorylation. In the gross, atopic dermatitis induced by DNCB in NC/Nga mice were improved by oral administration of FF n-BuOH fraction. Oral FF n-BuOH fraction also decreased the level of IgE in mice's serum and the level of IL-4 and IL-5 in the spleenocyte culture supernatant, cell numbers of CD8+, B220+CD23+ in the PBMCs, CD4+ in the ALN and CD4+, Gr-1+CD11b+ in the dorsal skin and suppressed mRNA expression of IL-5, IL-13, IL-31, IL-31RA in the dorsal skin. Histological examination showed that infiltration levels of immune cells in atopic dermatitis induced NC/Nga mice were improved by FF n-BuOH fraction. Conclusions FF n-BuOH fraction can reduce pruritus by suppressing IL-31, IL-31RA secretion and modulate molecular mediators and immune cells associated with atopic dermatitis induced in NC/Nga mice which may have played a significant role in recovering atopic dermatitis symptoms.

Suppressive Effects of GHS in Knee Joint, Regional Lymph Nodes, and Spleen on Collagen-induced Arthritis in Mice (Collagen II로 유도된 CIA 관절염 생쥐의 대한 가미해동피산(加味海桐皮散)의 억제 효과)

  • Rheumatoid arthritis (RA) is a systemic autoimmune disease with chronic inflammation characterized by hyperplasia of synovial cells in affected joints, which might be mediated by the altered activation of Immune system, ultimately leading to the destruction of cartilage and bone. To examine effects of GHS on rheumatoid arthritis DBA/1J mice were immunized with bovine type II collagen to induced arthritis and then treated with GHS once a day for 7 weeks. Oral administration of GHS (200 mg/Kg) significantly suppressed the progression of CIA, which extend is comparable to that of methotrexate (MTX, 0.3 mg/Kg), a positive control. The severity of arthritis within the knee joints, which was evaluated by histological assessment of cartilage destruction and pannus formation, was also lowered by GHS. The production of TNF-and IL-6 in serum was significantly suppressed. The levels of IFN-g in the culture supernatant of splenocytes stimulated with CD3/CD28 or collagen were dramatically decreased, while those of IL-4 was increased. The levels of IgG and IgM RA factor were also decreased in the serum. FACS analysis indicated that B cells (in DLN), CD3+ T cells (in spleen, and paw joint), CD11b+Gr-1+ cells (in paw joint), CD3+CD49b(DX5) (in PBMC) were decreased and there was increased proportion of CD3+, CD4+, CD8+, CD4+CD25+ T cells in DLN. In conclusion, our results demonstrates that GHS significantly suppressed the progression of CIA and this action was characterized by the decreased production of TNF-a, IL-6, and rheumatoid factors, and modulations of immune cell populations.

  • We have grown D $y_{x}$B $i_{3-x}$F $e_{5}$ $O_{12}$ (x = 0.5,1.0, 1.5,2.0) magnetic garnet thin films upon $Al_2$O3i and GGG substrate using Pechini process. The annealing temperature to get single phase D $y_{x}$B $i_{3-x}$F $e_{5}$ $O_{12}$ garnet is dependent on substrate, i.e. the annealing temperature for GGG substrate il 5$0^{\circ}C$ lower than that for $Al_2$ $O_3$ substrate. The grains of garnet thin film grown on GGG (111) plane align along [111] direction, and in this case the hysteresis curve does not saturate up to H : 5000 Oe. We attribute this phenomenon to rotation magnetization process. The maximum amount of Bi substitution in polycrystalline D $y_{x}$B $i_{3-x}$F $e_{5}$ $O_{12}$ thin film prepared by Pechini process is restricted to 2.0 Bi atom/unit cell, and this value is less than that in single garnet crystall grown by LPE method.own by LPE method.ethod.

The Suppressive Effect on Th2 Cytokines Expression and the Signal Transduction Mechanism in MC/9 Mast Cells by PRAL (MC/9 비만세포에서 행인(杏仁) 추출물의 Th2 cytokine 발현 억제 효과 및 신호전달 기전 연구)

  • Objectives PRAL (Prunus armniaca Linne Var) is a herbal formula in Oriental Medicine, known for its anti-inflammatory and anti-allergenic properties. However, its mechanism of action and the cellular targets have not yet been found enough. The purpose of this study is to investigate the effects of PRAL on Th2 cytokines expression in MC/9 mast cells. Methods The effect of PRAL was analyzed by ELISA, Real-time PCR, Western blot in MC/9 mast cells. mRNA levels of GM-CSF, IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$ were analyzed with Real-time PCR. Levels of IL-13, MIP-$1{\alpha}$ were measured using enzyme-linked immunosorbent assays (ELISA). NFAT, AP-1 and NF-${\kappa}B$ p65 were examined by Western blot analysis. Results PRAL inhibited GM-CSF, IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$ mRNA expression in a dose dependent manner. GM-CSF, IL-4, IL-5 mRNA expression were inhibited significantly in comparison to DNP-IgE control group at concentration of 100 ${\mu}g/ml$ and IL-6, IL-13, TNF-${\alpha}$ mRNA expression were inhibited at concentration of 50 ${\mu}g/ml$, 100 ${\mu}g/ml$. PRAL also inhibited the IL-13, MIP-$1{\alpha}$ production significantly in comparison to DNP-IgE control group in a dose dependent manner. IL-13 production was inhibited at a concentration of 200 ${\mu}g/ml$, 400 ${\mu}g/ml$ and MIP-$1{\alpha}$ was inhibited at a concentration of 100 ${\mu}g/ml$, 200 ${\mu}g/ml$, 400 ${\mu}g/ml$. Western blot analysis of transcription factors involving Th2 cytokines expression revealed prominent decrease of the mast cell specific transcription factors including NFAT-1, c-Jun as well as NF-${\kappa}B$ p65 but not NFAT-2 and c-Fos. Conclusion These results indicate that PRAL has the effect of suppressing Th2 cytokines production in the MC/9 mast cells. These data represent that PRAL potentiates therapeutic activities to the allergic disease by regulating Th2 cytokines in the MC/9 mast cells.

Inhibitory Effect of Ceongryulsaseuptang-kami on Arthritis occurrence in Collagen Induced Arthritis Mouse (CIA 생쥐의 관절염 유발에 대한 청열사습탕가미방의 억제 효과)

  • To evaluate effect of CRSST on inhibiting the occurrence of arthritis, we performed the experiments including production of inflammatory cytokine and immunoglobin in collagen induced arthritis model. The results were obtained as follows. CRSST extract shows any cytotoxicity effect on mouse lung fibroblast cells at dose of 400 ㎍/㎖. CRSST group shows inhibitory effect on arthritis incidence than control group for six weeks. Arthritis index of CRSST group reduces from 4 weeks (75±17.4%) to 6 weeks (33.3±10.0%) compared with control group. In CRSST group, production of cytokines which shows suppressive effect on inflammation (IL-4, IL-10 ) are increased and which promotes inflammation (TNF-α, INF-γ) are decreased in blood. In CRSST group, production of immunogloblin (IgG2b, IgG3 and IgM) is reduced compared with control group, and rate of CD4+ and CD3+ T cell is lower in joint and higher in lymph node compared with control group. From above results it could be accepted that CRSST shows anti-arthritis effect via immune system especially through the controlling the inflammatory cytokines and immunoglobins. CRSST could be usefully applied for the prevention and treatment of RA. And also is expected to be clinically helpful on the treatment of RA through modification.

Effects of Sopoongsangagambang Administration along with External Spray Treatment on Atopic Dermatitis Development in NC/Nga Mice (소풍산가감방(消風散加減方)과 외치방(外治方) 병용이 NC/Nga 아토피 생쥐에 미치는 영향)

  • Although the parallel prescription of Sopoongsangagambang (SG) administration along with external treatment such as spraying or ointment application on the skin is clinically used for the treatment of atopic dermatitis (AD), molecular mechanism underlying its effectiveness is unknown. Thus in the present study, diverse immune responses in terms of chemical mediators related to AD were investigated using an atopic mouse model NC/Nga after SG administration and external treatment (ET), and major findings are summarized as follows. 1. The clinical severities in 16 and 20 week old NC/Nga mice with SG and ET treatment were decreased to 72.2% and 62.3% respectively compared to the control NC/Nga mice with no drug treatment. 2. IgE, IL-4, IL-5, IL-6, IL-13, IgM, IgG1 and IgG2a levels in the serum of SG and ET treated NC/Nga mouse group were significantly decreased compared to the untreated control mice. In contrast, $IFN-{\gamma}$ showed a significant increase in the experimental group compared to the untreated control group. 3. The spleen weight of SG and ET treated NC/Nga mice was significantly decreased compared to the untreated control group. 4. The B/T ratio in the lymph node of SG and ET treated NC/Nga mice was increased compared to the untreated control group. $CD4^+\;and\;CD8^+$ cell numbers in the lymph node of SG and ET treated NC/Nga mice were significantly increased compared to the untreated control group, but $CD69^+\;and\;CD11a^+$ cells were significantly decreased. 5. mRNA expression levels of IL-4, IL-5, and CCR3 in the skin tissues of SG and ET treated NC/Nga mice were significantly decreased, and expression levels of IL-6, IL-13, $CD69^+/CD3{\varepsilon}^+\;and\;CD19^+/CD44^+$ in the skin tissues of SG and ET treated NC/Mga mice were significantly decreased compared to the untreated control group. $IFN-{\gamma}$ mRNA expression levels were increased compared to the untreated control group. 6. Histological observation of the ear and neck skin tissues showed that the extents of inflammation and infiltrated immune cells in the epidermis and dermis of SG and ET treated NC/Nga mice were highly reduced compared to the untreated control group. 7. Lymphokine assay showed a significant decrease in IL-4 levels in SG and ET treated NC/Nga mice compared to the untreated control group, but the levels of $IFN-{\gamma}$ secretion were significantly increased drug treated NC/Nga mice.

The morphological and functional studies of lymphoid organs in Apodemus agrarius pallas (등줄쥐에서 면역관련 장기의 특성연구)

  • Apodemus agrarius Pallas has been known to be a reservoir of hanta virus. But there was no report of immune organs. So this study was performed to clarify the relation between Structure and function of immune organ. In the present investigation an immunocytochemistry procedure was used to examine the distribution of T and B cell and macrophage populations in the thymus spleen and lymph nodes. Most of lymphoid organ showed decreased pattern of Ly1 positive T and IgM positive B cells. Macrophage positive cells were deceased in vivo. The present study gives an overview on the immune organ structure and function of Apodemus agrarius Pallas.
