Ultrasonic Nondestructive Evaluation of Microstructural Degradation in Artificially Aging Heat Treated 2.25CrMo Steel (인공 열화 열처리된 2.25CrMo 강의 미세조직 변화에 대한 초음파 비파괴평가)

  • Artificial aging was performed to simulate the microstructural degradation in 2.25CrMo steel arising from long time exposure at $540^{\circ}C$. It was found that the carbides became coarser and spheroidized as aging time increased. An attempt was made to evaluate the microstructural degradation in artificially aging heat treated 2.25CrMo steel by the ultrasonic attenuation and velocity measurements. Ultrasonic velocity was found essentially insensitive to the microstructural changes resulting from aging heat treatment. However, the ultrasonic attenuation was observed to increase with increasing aging time. Also, it was noticed that the change of ultrasonic attenuation with aging time was more sensitive at high frequency regions.

Effect of tensile strain on $I_{c}$ degradation characteristics in Bi-2223 superconducting tapes under bending (Bi-2223 초전도테이프 임계전류의 굽힘하에서 인장변형률 특성)

  • The influences of mixed mode of bending-tension on the Ic degradation and their interaction on the strain effect were evaluated in this study. A test fixture which applies a mixed deformation mode of bending-tension to HTS tapes has been newly devised. When the strain induced in the tape due to the mixed deformation mode was expressed as a total tensile strain, the irreversible strain to the critical current degradation of Bi-2223 tapes increased when compared with the case of simple bending mode, and the value at both ends were larger than that at the central region of the bend part. The Ic degradation behavior at the region exceeding the irreversible strain showed quiet a rapid drop of the Ic when compared with the simple bending cases. As the applied bending strain increased namely as the diameter of mandrel adopted decreased, the apparent irreversible strain of Bi-2223 tapes increased However, the increment decreased as the mandrel diameter decreased. As a result, it could be found that the tension to be applied to the Bi-2223 tapes during cabling of HTS tapes should be smaller, as the mandrel diameter becomes smaller.

  • Studying the effects of gamma radiation on the instrumentation and control (I&C) system of a nuclear power plant is critical to the successful and reliable operation of the plant. In the accidental scenario, the adverse environment of ionizing radiation affects the performance of the I&C system and it leads to inaccurate and incomprehensible results. This paper reports the effects of gamma radiation on the AT89C51RD2, a commercial-off-the-shelf 8-bit high-performance flash microcontroller. The microcontroller, selected for the device under test for this study is used in the remote terminal unit for a nuclear power plant. The custom circuits were made to test the microcontroller under different gamma doses using a 60Co gamma source in both ex-situ and in-situ modes. The device was exposed to a maximum dose of 1.5 kGy. Under this hostile environment, the performance of the microcontroller was studied in terms of device current and voltage changes. It was observed that the microcontroller device can operate up to a total absorbed dose of approximately 0.6 kGy without any failure or degradation in its performance.

Protein Kinase $C-{\alpha}$ Regulates Toll-like Receptor 4-Mediated Inducible Nitric Oxide Synthase Expression

  • Purpose: The nitric oxide (NO) release by inducible nitric oxide synthase (iNOS) is the key events in macrophage response to lipopolysaccharide (LPS) which is suggested to be a crucial mediator for inflammatory and innate immune responses. NO is an important mediator involved in many host defense action and may also lead to a harmful host response to bacterial infection. However, given the importance of iNOS in a variety of pathophysiological conditions, control of its expression and signaling events in response to LPS has been the subject of considerable investigation. Materials and Methods: The Raw264.7 macrophage cell line was used to observe LPS-stimulated iNOS expression. The expression of iNOS is observed by Western blot analysis and real-time RT-PCR. Protein kinase C $(PKC)-{\alpha}$ overexpressing Raw264.7 cells are established to determine the involvement of $PKC-{\alpha}$ in LPS-mediated iNOS expression. $NF-{\kappa}B$ activity is measured by $I{\kappa}B{\alpha}$ degradation and $NF-{\kappa}B$ luciferase activity assay. Results: We found that various PKC isozymes regulate LPS-induced iNOS expression at the transcriptional and translational levels. The involvement of $PKC-{\alpha}$ in LPS-mediated iNOS induction was further confirmed by increased iNOS expression in $PKC-{\alpha}$ overexpressing cells. $NF-{\kappa}B$ dependent transactivation by LPS was observed and $PKC-{\alpha}$ specific inhibitory peptide abolished this activation, indicating that $NF-{\kappa}B$ activation is dependent on $PKC-{\alpha}$. Conclusion: Our data suggests that $PKC-{\alpha}$ is involved in LPS-mediated iNOS expression and that its downstream target is $NF-{\kappa}B$. Although $PKC-{\alpha}$ is a crucial mediator in the iNOS regulation, other PKC isozymes may contribute LPS-stimulated iNOS expression. This finding is needed to be elucidated in further study.

Microstructural Observation of Multi-coated YBCO Films Prepared by TFA-MOD (TFA-MOD법으로 제조된 다층 YBCO 박막의 미세구조 관찰)

  • We fabricated $YBa_2Cu_3O_{7-x}$(YBCO) films on (00l) $LaAlO_3$ substrates prepared by metal organic deposition(MOD) method using trifluoroacetate(TFA) solution. The films with various thicknesses were prepared by repeating the dip-coating and calcining processes. The effects of film thickness on phase formation, microstructures, and critical properties were evaluated by X-ray diffraction, scanning electron microscopy, and transmission electron microscopy. The microstructure and resultant critical current($I_C$) and critical current density($J_C$) varied remarkably with film thickness: The ($I_C$) value increased from 39 to 160 A/cm-width as the number of coatings increased from one to four, while the corresponding $J_C$ was measured to be in the range of $0.84-1.21\;MA/cm^2$. Both the $I_C$ and $J_C$ decreased when an additional coating was applied due to microstructural degradation, indicating that the optimum thickness is in the range of $1.1-1.8\;{\mu}m$. The possible cause for the decrease in the $I_C$ and $J_C$ value for film thicker than $1.8\;{\mu}m$ include non-uniform thickness, increased surface roughness, and the poor formability of the YBCO phase and texture arising from the insufficient heat treatment time with respect to the increased thickness.

  • Nonlinear acoustic effect has been considered as an effective tool for the evaluation of material degradation. In this paper, the applicability of nonlinear acoustic effect to the evaluation of degraded 2.25Cr-1Mo steel is investigated. Firstly, artificial aging was performed to simulate the microstructural degradation in 2.25Cr-1Mo steel arising from long time exposure at $540^{\circ}C$. Secondly, ultrasonic nonlinear parameter was quantitatively measured by bi-spectrum and power spectrum. Nonlinear acoustic parameter from bi-spectrum was found to be clearly sensitive to the aging time.

Comparison of In situ Dry Matter Degradation with In vitro Gas Production of Oak Leaves Supplemented with or without Polyethylene Glycol (PEG)

  • Dry matter (DM) degradation of leaves from Quercus cercis, Quercus libari, Quercus branti, and Quercus coccifera was determined using two different techniques: (i) in vitro gas production and (ii) the nylon bag degradability technique. In vitro gas production in the presence or absence of PEG and in situ DM disappearance were measured at 3, 6, 12, 24, 48, 72 and 96 h. In situ and in vitro DM degradation kinetics were described using the equation y = a+b ($1-e^{-ct}$). At all incubation times leaves from Quercus branti incubated with or without PEG gave significantly higher gas production than the other oak leaves except for 3 and 6 h incubation when leaves from Quercus branti without PEG supplementation only gave higher gas production than Quercus cercis and Quercus coccifera. At all incubation times except at 3, 6 and 12 h the DM disappearance from Quercus branti was significantly higher than the other species. Generally, PEG supplementation considerably increased the gas production at all incubation times and estimated parameters such as gas production rate ($c_{gas}$), gas production (ml) from the quickly soluble fraction ($a_{gas}$), gas production (b) from the insoluble fraction, potential gas production (a+b). However, all oak leaves did not give the same response to the PEG supplementation. Although the increase in gas production at 96 h incubation time was 8.9 ml for Quercus libari the increase was 5.5 ml for Quercus coccifera. It was concluded that except at early incubation times the relationships between the two methodologies seem to be sufficiently strong to predict degradability parameters from gas production parameters obtained in the presence or absence of PEG.

Versatile Catabolic Properties of Tn4371-encoded bph Pathway in Comamonas testosteroni (Formerly Pseudomonas sp.) NCIMB 10643

  • Comamonas testosteroni (formerly Pseudomonas sp.) NCIMB 10643 can grow on biphenyl and alkylbenzenes $(C_2-C_7)$ via 3-substituted catechols. Thus, to identify the genes encoding the degradation, transposon-mutagenesis was carried out using pAG408, a promoter-probe mini-transposon with a green fluorescent protein (GFP), as a reporter. A mutant, NT-1, which was unable to grow on alkylbenzenes and biphenyl, accumulated catechols and exhibited an enhanced expression of GFP upon exposure to these substrates, indicating that the gfp had been inserted in a gene encoding a broad substrate range catechol 2,3-dioxygenase. The genes (2,826 bp) flanking the gfp cloned from an SphI-digested fragment contained three complete open reading frames that were designated bphCDorfl. The deduced amino acid sequences of bphCDorfl were identical to 2,3-dihydroxybiphenyl 1,2-dioxygenase (BphC), 2-hydroxy-6-oxo-6-phenylhexa-2,4-dienoate hydrolase (BphD), and OrfI, respectively, that are all involved in the degradation of biphenyl/4-chlorobiphenyl (bph) by Ralstonia oxalatica A5. The deduced amino acid sequence of the orfl revealed a similarity to those of outer membrane proteins belonging to the OmpW family. The introduction of the bphCDorfl genes enabled the NT-l mutant to grow on aromatic hydrocarbons. In addition, PCR analysis indicated that the DNA sequence and gene organization of the bph operon were closely related to those in the bph operon from Tn4371 identified in strain A5. Furthermore, strain A5 was also able to grow on a similar set of alkylbenzenes as strain NCIMB 10643, demonstrating that, among the identified aromatic hydrocarbon degradation pathways, the bph degradation pathway related to Tn4371 was the most versatile in catabolizing a variety of aromatic hydrocarbons of mono- and bicyclic benzenes.

Studies on Improvement of Schottky Characteristics for GaN Devices (GaN 소자의 쇼트키 특성 향상에 관한 연구)

  • In this paper, I have fabricated and measured the gallium nitride(GaN) based Schottky diodes, and have carried out analyses of degradation of Schottky barrier characteristics. To improve of degraded Schottky barrier characteristics, I have carried out several experiments such as N$_2$ plasma exposure, annealing in N$_2$ ambient and annealing after N$_2$ plasma exposure. In the results of these experiments, I have achieved that only annealing in N$_2$ ambient is enough to improve the Schottky barrier characteristics, are temperature of 700$\^{C}$ and time of 90 sec in N$_2$ ambient furnace. for the analysis of these experiments, I have carried out the measurement of electric characteristics and quantitative analysis of etching damage using AES(Aguger Electron Spectroscopy).

Effect of Ultrasound on the Acid Hydrolysis of Inulin (Inulin의 산 가수분해 반응에서 초음파가 미치는 영향)

  • The effect of ultrasound on the acid hydrolysis of inulin was studied under significantly mild reaction conditions, at which sugar degradation products were not detected. Reaction conditions were i the range of 50~$60^{\circ}C$ and 0.1~0.3%(w/w) of HCl concentrations. The effects of reactor position inside water bath and mechanical agitation under ultrasound were investigated. The production rates of fructose with/without ultrasound irradiation were compared. The activation energies for both control and ultrasound reaction were the same, i. e., 25kca1/mo1, and ultrasound enhancement was average 22%.
