대식세포에서 산화질소 생성에 대한 당귀(當歸) 에탄올 추출물의 억제효과 (Inhibitory Effect of Angelica gigas Nakai Extract on Nitric Oxide Production in RAW 264.7 Cells)

  • Objective : The Angelica gigas Nakai ethanol extract (AGE) was investigated to compare nitric oxide (NO) production and $NF-{\kappa}B$ activity from RAW 264.7 cells, since NO and nuclear $factor-{\kappa}B$ $(NF-{\kappa}B)$ have been shown to be factors implicated in inflammatory disease. Method : AGE was prepared by extracting medicinal herb with 70% (v/v) ethanol solution. We investigated production of nitric oxide (NO) and inducible nitric oxide synthase (iNOS) gene expression by ARE in LPS-stimulated RAW 264.7 macrophage cells. We also investigated inhibition of LPS-induced activation of $NF-{\kappa}B$ on western blot. Result : LPS-induced RAW 264.7 cells increased NO production and iNOS expression. Upon treatment with AGE, nitrite production was significantly inhibited in a concentration-dependent manner compared to the untreated control. AGE inhibited this LPS-induced iNOS mRNA and protein in a dose-dependent manner. AGE markedly inhibited the expression of iNOS mRNA and protein at a concentration of 100 ${\mu}g/ml$. LPS-induced RAW 264.7 cells with AGE blocked inhibitory $factor-{\kappa}B{\alpha}$ degradation. Conclusion :This study shows that AGE seems to attenuate inflammation through inhibition of NO production and iNOS expression by blockade of $NF-{\kappa}B$ activation in LPS-stimulated RAW 264.7 cells.

목단피((牧丹皮).도인(桃仁) 배합(配合)이 항염증(抗炎症) 작용(作用)에 미치는 영향 (Effect of Water Extract of Peonia Suffruticosa and Prunus Percica on Anti-inflammation)

  • Paeonia Suffruticosa and Prunus Persica have been used as oriental medicine for removal of fever, alleviation of pain, an anti-phlogistic effect and removal of extravasated blood. However, it has been never shown the effects of these herbal medicines on anti-inflammatory processes. This experiment was performed to show how these herbs could act as anti-inflammatory medicines at cellular level. Anti-inflammation effects of water extracts from Paeonia Suffruticosa and Prunus Persica as well as their mixture have been investigated, and the results were follows; 1) each extract slightly suppressed the expression and production of inflammatory mediators and enzymes such as NO, iNOS, IL-$1{\beta}$, and TNF-$\alpha$ in lipopolysaccharid(LPS)-stimulated RAW264.7 cells and mouse primary peritoneal macrophages in a dose-dependent manner. These suppressive effects, however, were synergistically increased by their mixture. 2) Each extract of Paeonia Suffruticosa and Prunus Persica insignificantly suppressed the activation and activity of NF-${\kappa}B$ in LPS-stimulated RAW264.7 cells, which controls the expression of inflammatory mediators such as NO, iNOS, IL-$1{\beta}$, and TNF-$\alpha$. However, extract mixture of Paeonia Suffruticosa and Prunus Persica suppressed effectively the activation and activity of NF-${\kappa}B$. 3) Each of Paeonia Suffruticosa and Prunus Persica induced translocation of NF-${\kappa}B$ to the nucleus from the cytosol and DNA-binding activity of nuclear NF-${\kappa}B$ in LPS-activated RAW264.7 cells. The extract mixture of Paeonia Suffruticosa and Prunus Persica showed more significant suppression of the NF-${\kappa}B$ translocation and its DNA-binding activity, as compared to those of the each extract. These results suggest that the extract mixture of Paeonia Suffruticosa and Prunus Persica may affect different control mechanisms for NF-${\kappa}B$ activation and the expression and production of NF-${\kappa}B$-dependent inflammatory mediators, indicating that this extract mixture may be useful for treatment of inflammatory diseases.

탈명산(奪命散)의 항산화 및 항염증효과에 관한 연구 (Anti-oxidative and anti-inflammatory experiments of Talmyung-san in RAW264.7 cells)

  • Objectives : The aim of this study was identification of the anti-oxidative and anti-inflammatory effects of Talmyung-san (TMS) in mouse macrophage, RAW264.7 cells. Methods : To identify the anti-oxidative effect of TMS, scavenging activities of DPPH radical, nitric oxide and peroxynitrite were measured in vitro. In RAW264.7 cells, DCFH-DA assay was conducted to examine the inhibitory effect of TMS on ROS production in response to lipopolysaccharide. And the productions of nitric oxide (NO), $PGE_2$ and pro-inflammatory cytokines were measured. The levels of COX-2, iNOS, nuclear NF-${\kappa}B$ p65 expression and phosphorylation of $I{\kappa}B-{\alpha}$ in cytosol were detected by western blotting analyses. Results : TMS couldn't scavenged DPPH radical, but nitric oxide and peroxynitrite were decreased. TMS decreased intracellular ROS, NO, and IL-$1{\beta}$ production effectively. However, TMS inhibited $PGE_2$ levels only in high concentration ($300{\mu}g/m{\ell}$) and TMS failed to suppress the production of IL-6 and TNF-${\alpha}$. Results from immunoblot analyses revealed that TMS decreased activation of COX-2, iNOS, phosphorylation of $I{\kappa}B-{\alpha}$ and nuclear translocation of p65. Conclusions : TMS has anti-RNS and anti-inflammatory effects via NF-${\kappa}B$ pathway and more intensive studies will be required to evaluate therapeutic potential of TMS.

Artemisia fukudo essential oil attenuates LPS-induced inflammation by suppressing NF-${\kappa}B$ and MAPK activation in RAW 264.7 cells

  • In the present study, the chemical constituents of Artemisia fukudo essential oil (AFE) were investigated using GC-MS. The major constituents were ${\alpha}$-thujone (40.28%), ${\beta}$-thujone (12.69%), camphor (6.95%) and caryophyllene (6.01%). We also examined the effects of AFE on the production of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-IL-$1{\beta}$ (IL-$1{\beta}$), and IL-6 in lipopolysaccharide (LPS)-activated RAW 264.7 cells. Western blotting and RT-PCR analyses indicated that AFE has potent dose-dependent inhibitory effects on pro-inflammatory cytokines and mediators. We investigated the mechanism by which AFE inhibits NO and $PGE_2$ by examining the level of nuclear factor-${\kappa}B$ (NF-${\kappa}B$: p50 and p65) activation within the mitogen-activated protein kinase (MAPK: ERK, JNK and p38) pathway, which is an inflammation induced signal pathway in RAW 264.7 cells. AFE inhibited LPS-induced ERK, JNK and p38 phosphorylation. Furthermore, AFE inhibited the LPS-induced phosphorylation and degradation of $I{\kappa}B-{\alpha}$, which is required for the nuclear translocations of the p50 and p65 NF-${\kappa}B$ subunits in RAW 264.7 cells. Our results suggest that AFE might exert an anti-inflammatory effect by inhibiting the expression of pro-inflammatory cytokines. Such an effect is mediated by a blocking of NF-${\kappa}B$ activation which consequently inhibits the generation of inflammatory mediators in RAW 264.7 cells. AFE may be useful for treating inflammatory diseases.

  • 비지는 대두 가공 시 생산되는 부산물로 대부분 폐기되고 있는 실정이지만, 최근 비지를 유용한 자원으로 이용하기 위한 기능성 연구가 진행되고 있다. 그러나 비지의 항염증에 대한 연구가 미비하여 본 연구진은 비지추출물이 마우스 대식세포인 RAW264.7에 LPS에 의한 염증 반응에 미치는 영향을 평가하였다. 본 연구에서 비지추출물은 NF-${\kappa}B$와 p38의 활성 억제를 통해 만성염증 유발인자인 NO, iNOS, $PGE_2$, COX-2, TNF-${\alpha}$ 및 IL-$1{\beta}$의 발현을 억제하는 것으로 확인되었다. 따라서 비지는 독성과 부작용이 적은 항염증 관련 식의약 소재로 활용될 수 있을 것으로 사료된다.

Mouse의 신장상피세포에서 패장(敗醬)추출물이 산화 스트레스 및 NF-${\kappa}B$ signaling에 미치는 영향 (The Effect of Patriniae Radix on the Oxidative Stress and the NF-${\kappa}B$ Signaling in Mouse LLC-$PK_1$ Cell)

  • Objectives : The aims of this study were to investigate the cytoprotective, antioxidative and inflammation genes inhibitory effects of Patriniae Radix on the mouse LLC-$PK_1$ cells (renal epithelial cells). Methods : The cytoprotective effect of Patriniae Radix was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The antioxidative effect was measured in terms of generation amount of superoxide anion radical (${\cdot}{O_2}^-$) by 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), nitric oxide (NO) by 4,5-diaminofluorescein (DAF-2), peroxynitrite ($ONOO^-$) by dihyldrorhodamine 123 (DHR 123) and prostaglandin $E_2$ ($PGE_2$) by $PGE_2$ immunoassay on $H_2O_2$-treated LLC-$PK_1$ cells. For measuring of inflammation genes inhibitory effects, western blot was performed to detect IKK-$\alpha$, phospho-$I{\kappa}B-\alpha$, NF-${\kappa}B$ (p50, p65), COX-2, iNOS, IL-$1{\beta}$ and VCAM-1 protein level in cytosol fractions from LLC-$PK_1$ cells. Results : Patriniae Radix extract reduced the $H_2O_2$-induced cell death and inhibited the amount of $H_2O_2$-induced ${\cdot}{O_2}^-$, NO, $ONOO^-$, $PGE_2$ generation dose-dependently on the mouse LLC-$PK_1$ cells in vitro. Also Patriniae Radix extract inhibited the expression of IKK-$\alpha$, phospho-$I{\kappa}B-\alpha$, COX-2, iNOS, IL-$1\beta$ and VCAM-1 genes dose-dependently by means of decreasing activation of NF-${\kappa}B$. Conclusions : According to above results, it was identified that Patriniae Radix had the cytoprotective, antioxidative and inflammation genes inhibitory effects. So it was suggested that Patriniae Radix would be effective to the treatment for the inflammatory process and inflammation-related diseases.

녹용약침이 백서의 제2형 Collagen 유발 관절염에서 iNOS 발현과 NO 생성 억제에 미치는 영향 (Effect of Cervi Pantotrichum Cornu Pharmacopuncture on Suppressing the Expression of iNOS and Production of NO in Type II Collagen-induced Arthritis Mice)

  • Objectives : The objective of this study is to investigate the suppressing effect of the cervi pantotrichum cornu pharmacopuncture on the expression of iNOS mRNA and production of NO in synoviocytes from artificially arthritis-induced mice. Methods : In vitro test, synoviocytes extracted from a knee joint of a mouse were cultivated, and the herbal extract of cervi pantotrichum cornu($0.4mg/m{\ell}$, $0.6mg/m{\ell}$, $0.8mg/m{\ell}$, and $1.0mg/m{\ell}$) was added into the wells of synoviocytes to suppress the expression of iNOS mRNA and production of NO. In vivo test, each ten mice were allocated into three groups; Normal group, CIA-elicitated group(CIA), and group treated with cervi pantotrichum cornu pharmacopuncture after CIA elicitation(CCA). The extract of cervi pantotrichum cornu was injected into the acupoint of $SP_{10}$ to observe the changes of foot thickness in mice and the suppression of MIF, TNF-$\alpha$, NF-${\kappa}B$ p65, and iNOS. Results : In vitro test, the expression of iNOS mRNA and production of NO were dose-dependently decreased in the wells of synoviocytes treated with PMA. In vivo test, the suppression of MIF, TNF-$\alpha$, NF-${\kappa}B$ p65, and iNOS was clearly shown in the pieces of the synovial joint treated with the extract of cervi pantotrichum cornu. The foot thickness also decreased dose-dependently. Conclusions : It is speculated that the cervi pantotrichum cornu pharmacopuncture can be applicable to the therapy of rheumatoid arthritis by suppressing the expression of iNOS mRNA and production of NO.

전도산(顚倒散)이 여드름 유발균과 염증에 미치는 영향 (The Effects of Jeondo-san on Anti-Inflammation and Anti-Propionibacterium acnes)

  • Objectives : This study was carried out to investigate the effects of Jeondo-San(JDS) on anti-Inflammation and anti-Propionibacterium acnes. Methods : The effects of JDS on anti-Inflammation and anti-Propionibacterium acnes were measured by the cytotoxicity of Raw 264.7 cell, the inhibition for NO, $TNF-{\alpha}$, $PGE_2$, iNOS and COX-2, the blocking $NF-{\kappa}B$ into nucleus and the sterilizing power for Propionibacterium acnes. Results : 1. All concentrations of JDS has no cytotoxicity in Raw 264.7 cell. 2. All concentrations of JDS inhibited the production of NO in the Raw 264.7 cell stimulated with LPS. 3. All concentrations of JDS did not significantly inhibit the production of $TNF-{\alpha}$ in the Raw 264.7 cell stimulated with LPS. 4. All concentrations of JDS inhibited the production of $PGE_2$ in the Raw 264.7 cell stimulated with LPS. 5. All concentrations of JDS did not inhibit the expression of COX-2 but concentrations of 50\;{\mu}g/ml$, 100\;{\mu}g/ml$ JDS inhibited iNOS expression in the Raw 264.7 cell stimulated with LPS. 6. Concentrations of 50\;{\mu}g/ml$, 100\;{\mu}g/ml$ JDS has the effect of blocking $NF-{\kappa}B$ into nucleus in LPS-induced macrophage Raw 264.7 cell. 7. All concentrations of IDS did not have the inhibitory effect of Propionibactrium acnes. Conclusions : The present date suggest that JDS has a effect on the stage of inflammation of acne.

  • HIV 감염 후 AIDS로의 진행과정에서 cytokines의 변화는 필수적인 결과로서 나타나게 된다. 본 연구에서는 AIDS 동물모델을 사용하여 Pyc Cytokines발현에 미치는 영향과 조절 매개체로서 $NF-{\kappa}B$의 관련성 및 작용 기작 확인에 초점을 두었다. LP-BM5 retrovirus에 감염된 C57BL/6 생쥐를 이용하여12주간에 걸친 실험에서Pyc 투여는 Th1과 Th2 cytokines의 혈중농도를 조절하여 murine AIDS로의 진행을 억제하는 데 역할을 하는 것으로 사료된다. 특히 Th2 cytokines의 mRNA 발현을 억제함에 따라 Th1 cytokines의 혈중농도는 상대적으로 증가한 것으로 사료된다. 이러한 결과는 Pyc가 Th2 cytokines을 선택적으로 transcription level에서 조절하고 있음을 의미하고 있다. 또한 Th2 cytokines mRNA 발현 조절은 $NF-{\kappa}B$의 활성화에 의한 것으로 추정된다. 본 연구는 AIDS 동물모델에서 $NF-{\kappa}B$의 조절을 통한 cytokines의 발현 조절의 가능성을 제시함과 동시에 Pyc의 역할에 대해 자료를 제시하고 있다. 또한 향후 murine AIDS 진행 억제제로서의 Pyc의 기작에 대한 일면을 보인 것에 의의를 두고자 한다.

꽃송이버섯 추출물의 항염활성 효과 (Anti-inflammatory effects of Sparassis crispa extracts)

  • 꽃송이버섯 추출물은 염증반응 시 유도되는 NO 생성을 저해하는 활성이 있는 것으로 나타났으며, 200 ${\mu}g/ml$의 꽃송이버섯 추출물을 처리하였을 때 NO 저해능이 최대 효과를 보였고 RAW264.7 cell에서는 대조군과 유사한 수준으로 NO 생성을 억제하였다. 이러한 NO 생성의 저해는 iNOS의 발현이 감소한 것에 의한 결과임을 단백질과 mRNA의 발현량 변화를 통하여 확인하였으며, mRNA 발현 변화는 iNOS 유전자의 전사를 담당하는 전사인자인 $NF-{\kappa}B$와 STAT-1의 활성감소에 의한 결과임을 western blot을 통하여 확인하였다. 특히, $NF-{\kappa}B$의 활성감소는 $I{\kappa}B{\alpha}$의 활성증가에 의한 $NF-{\kappa}B$의 억제능 향상에 의한 것임을 확인하였고, 활성억제된 $NF-{\kappa}B$가 핵 내부로의 이동이 저해되면서 iNOS 유전자 발현에 영향을 미친 것임을 확인하였다. 그러므로, 꽃송이버섯 추출물은 NO 저해능을 이용한 항염증소재로서 염증성질환의 완하에 도움이 될 것으로 사료된다.
