The scFv antibody towards the Burkholderia pseudomallei exotoxin was previously constructed by phage display and exhibited good specificity towards the exotoxin. We report here the optimization of the scFv expression in an E. coli expression system. Four different E. coli strains (ER2537, TG1, HB2151, and XL1-Blue) were examined for optimal expression of the scFv protein. Two types of carbon source (i.e. 0.2% glucose and 0.2% glycerol) were also tested for their ability to induce the scFv expression. Cells that carried the scFv construct were grown at $30^{\circ}C$ and induced with 0.05 mM IPTG. The expression was then monitored by SDS-PAGE, Western blotting, and indirect ELISA. The Western blot profile showed different levels of the scFv expression among the host strains; XL1-Blue exhibited the highest level of the scFv protein expression. Glycerol at a concentration of 0.2% (v/v) significantly increased the scFv protein expression level when compared to 0.2% (w/v) glucose. Further optimization demonstrated that the scFv protein expression in XL1-Blue was the most optimal with a glycerol concentration as low as 0.05%. However, by indirect ELISA, only the scFv protein that was expressed in 0.2% (v/v) glycerol exhibited high specificity towards the Burkholderia pseudomallei exotoxin.
Purpose: This study was attempted to estimate the effects of diabetic education fortified with individual practice on plasma glucose, self-care, and self reported physical symptoms in type 2 diabetes patients Method: The subjects consisted of 46 patients with type 2 diabetes patients, who took the hospitalization diabetes education program from July 2003 until February 2004 at Seoul C university hospital. The experimental group was assessed at preand post intervention. The diabetes education was provided for one week. The education consisted of diabetes education videos for the diabetic introduction, group education for medication therapy, dietetic treatment and diabetes complication education. Also individual education for nurses examination of glycemia and insulin injection practice. Results: The HbA1c values significantly decreased from 9.6% on the time of hospitalization to 7.4% 3 months after discharge. In respect to the number of days of self-care, medication, diet, exercise, cleansing feet, and carrying sweets to prepare for hypoglycemia all significantly increased 3 months after discharge compared to the values at the time of hospitalization. Self reported physical symptoms were also significantly increased 3 months after discharge compared to the time of hospitalization. Conclusion: The diabetic education fortified with individual practice can be practically used as a plan for managing glycemia, self-care, and self reported physical symptom of diabetes patients.
This study examined the effects of plums produced in Gimcheon area on the glucose and lipid metabolism in streptozotocin-induced diabetic rats. Male Sprague-Dawley diabetic rats were divided into four groups: control, diabetic control, Diabetes-low plum (containing 10% plum powder), and Diabetes-high plum (containing 20% plum powder). The animals were fed isocaloric experimental diets based on an AIN-76 diet for 6 weeks. Feed efficiency ratio (FER) of the diabetic groups were significantly lower than that of the control. On the other hand, among the diabetic groups, the FER of the high plum intake group was higher than that of the diabetic control. The liver weight per 100 g body weight of each group was similar but the liver weights tended to decrease as the amount of plum intake was increased. Kidney weight per 100 g body weight of the plum intake groups were significantly different compared to that of the diabetic control. The supplementation of plums lowered the fasting blood glucose level of the diabetic groups and improved the glucose tolerance, thereby lowering the glycosylated hemoglobin index. In addition, the supplementation of plum was lowered the blood total cholesterol concentration and increased the HDL-C/TC (%) significantly, thereby lowering the atherosclerotic index (AI) and hepatic peroxide level. A steady diet of plums produced in Gimcheon may be effective in controlling the blood glucose level and preventing chronic diabetes mellitus.
Objectives : JiaoTeng-Yuan(교등원) is said to be a prescription for preservation of health in $\ulcorner$HuaTuo.ZhongZangJing (화타.중장경)$\lrcorner$. It is known to have the effect of Bu-Shen (보신: strengthening kidney) and Yi-Shou (익수: prolonging the span of one's life). This study investigates whether JTY is effective on inhibition of oxidation stress. Methods : Sprague-Dawley rats (12-week-old, weight $300{\pm}20g$) were divided into 3 groups. The normal group (n=8) was injected with PBS (1ml/body, s.c) subcutaneously behind the neck. The control group (n=8) was injected with D-galactose (50mg/kg, 1ml PBS/body, s.c) to induce pathological animals. The JTY group was injected with the same treatment as the control group, and fed containing JTY (10%). All groups were treated 1 time per day for 6 weeks. After the rats were sacrificed anti-oxidant enzyme (SOD, CAT, G-px) activity, GSH quantity of RBC and tissue (heart, liver and kidney), and plasma Vit-C quantity were examined. Also, the MDA levels of liver and kidney, lipofuscin of heart and endurance of erythrocyte membrane were measured. Results : In the JTY group, RBC's SOD activity decline was halted by 21 % of the normal level, compared to the control group, G-px activity (unit/g of Hb) increased significantly, compared to the normal group, and the level of Vit-C in plasma increased by 16%. Heart's SOD activity was kept at the same level as that of the normal group; and CAT activity decline was halted by 26%. Kidney's CAT and G-px activities were kept at the same level as that shown in the normal group, implying the existence of a halting effect. Liver also showed a slight halting effect against the decline of anti-oxidant ability, but the effect was not significant (${\alpha}=0.05$). A comparison between the levels of peroxide in SD rats showed that the level of TBARS in plasma increased significantly in the control group and that it was normal in the JTY group. The livers in the JTY group, compared to those in the control group, showed 36% halting effect of the normal level while their kidney's indicated the level significantly lower than the normal level. Heart's lipofuscin increased significantly in the control group, but was alike in both the JTY and the normal groups. Endurance of erythrocyte membrane (%) decreased significantly in the control group while it was kept at the similar level in both the JTY and the normal groups, indicating the halting effect. Conclusions : This study suggests that JTY is effective to defend oxidation stress caused by D-galactose in the animals. It showed that the anti-oxidant ability was maintained and strengthened. On the other hand, it reduced the level of peroxide in animals. In sum, JTY appeared to have the equilibrium normal physiological function in SD rat.
Objectives : We implemented the Artemisia Extract Moxibustion Method and had the clinical tests for the diabetes with it. Methods : We implemented Artemisia extract made by extracting the vasodilator and antioxidant compounds from Artemisia-CH2C12 fraction and the moxibustion method constructed with DC Power supply, controller, Artemisia pad. single and multiple heating terminal with PTC(Positive Temperature Coefficient) thermistor. And we performed to estimate the efficiency on the questionnaire and the clinical tests with 23 cases of the diabetics. Results : We have estimated the improvement over 60% the symptoms that were the upper and lower limbs pain, frequent urination, spontaneous perspiration, thirst, decrease of body weight, and malaise after the moxibustion treatment on 5 cases among 23 cases. And the 19 cases took the biochemical check-up after the moxibustion treatment. From the results of biochemical check-up, the average HbAlc of before treatment was 8.400%, and after treatment 7.632%. The average HbAlc was decreased significantly after treatment (P<0.001). And the average urinary blood of before treatment was 0.73 and after treatment 0.27. The average urinary blood was decreased significantly after treatment (P<0.001). In addition the average FBS before treatment was 182.64 mg/dl, after treatment 161.77 mg/dl. Conclusions : We could estimate that our proposed moxibustion method was a significant treatment method for the diabetes.
These studies were performed to investigate the effect of antioxidants, melatonin (MLT) and ascorbic acid (AA), on the diabetic complications. In the experiment, twenty-four Sprague Dawley rats were divided into four groups; Group 1: no treatment (control) (n=6), Group 2: Streptozotocin (STZ; 65 mg/kg b.w. ip) treatment (n=6), Group 3: MLT (10 mg/kg b.w. ip) treatment (n=6), Group 4: ascorbic acid (AA: 40 mg/kg b.w.ip) treatment (n=6). Rats were sacrificed and collected designated samples (blood, kidney, pancreas) at the 21 days after antioxidant treatment (the 8th week after STZ injection). There was no significant change of body weight during the experiment period in the all groups. The concentration of blood glucose was decreased below 200 mg/dl in MLT. The concentration of HbA$_1$c level was decreased significantly in MLT (7.3$\pm$0.66%) compared with STZ (15.1$\pm$1.58%) (p<0.01). No significant change of SOD activity in the RBC and kidney was found in the all groups. In the pancreas, MLT (1.42$\pm$0.17 U/ml) and AA(1.65$\pm$0.16 U/ml) decreased SOD activity compared with STZ (4.10$\pm$0.27 U/ml). However, there was no significant difference among MLT and AA. The level of malondialdehyde(MDA) as an indicator of lipid peroxydation was decreased in MLT compared with STZ in the plasma, kidney and pancreas(p<0.01). These findings suggest that melatonin could prevent the complications of diabetes mellitus.

Purpose: Insulin is involved in carbohydrate metabolism and also it's very important because it increases storage of glycogen, synthesis of fatty acids, absorption of amino acid, synthesis of protein. Insulin is clinically useful when we evaluate fasting patients in hypoglycemia, classify and predict diabetes, assess the activity of ${\beta}$-cell, research insulin resistance. We are going to increase usability of insulin assay by establishing normal reference value according to statistical analysis. Material & Method: We selected 6,648 patients who visited asan health medical center from May to August in 2008. We set exclusion criteria as family of diabetes, diabetes medication, the past history of blood glucose rise, more than 100 mg/dL in normal fasting blood glucose, outside the scope of BMI 18.5~22.9 $kg/m^2$, and more than HbA1c 6.5%. We determine whether the subgroup is portioned as sex and age or not and establish normal reference value by conducting statistical analysis as Bayesian's method and Hoffman's method. Result: Portioning of subgroup as sex and age is not needed. By statistical analysis of Bayesian method, results 1.5-11.0 uIU/mL. By statistical analysis of Hoffman method, results 1.8~12.8 uIU/mL. Conclusion: We established 1.8~12.8 uIU/mL as Insulin normal reference value by Hoffman method. This is a similar value with reporting reference value 1.7~11.8 uIU/mL in kit. This will enhance the usability of insulin assay by establishing normal reference value.
Purpose: This study was done to survey health behaviors in people at risk for a Cerebrovascular Accident(CVA). Method: From November 21 to December 29, 2005, a questionnaire survey was conducted with 171 people at risk for a Cerebrovascular Accident(LDL of above 130mg/dl & homocysteine of above $15.0{\mu}mol/L$). Their physical composition was measured and blood was collected. Results: 1. Of the subjects, 34.5% were smokers, 61.4% were drinkers, 56.7% did not exercise regularly, 57.3% did not control their weight, 26.9% preferred eating meat, and 32.2% preferred salty food. 2. The gender was different between smoking status($X^2=10.734$, p= .001), and drinking status($X^2=7.185$, p= .007), and the age was different between smoking status($X^2=6.656$, p=.010), and drinking status($X^2=10.722$, p= .001). The $HbA_1C$ level was different for regular exercise($X^2=4.824$, p=.028) and the HDL-cholesterol was different for meat-eating preference($X^2=7.928$, p= .005). The observance of troubling signs was different for a salty food preference($X^2=4.313$, p= .038). Conclusion: It is necessary to develop programs for taking care of people at risk for a Cerebrovascular Accident and test the effects of the programs in order to reduce the risk factors of CVA and enhance health behavior promotion.

The purpose of this study was to investigate the relationship between diabetic neuropathy, and glycemic control, self-management in type 2 diabetes mellitus(DM) patients. This was a cross sectional descriptive study and data were collected between May 10 and 31, 2011 using a questionnaire and medical record. The participants were 108 with DM who were treated at the endocrine medical outpatient department. The data were analyzed Pearson's correlation with SPSS WIN program. The mean scores of neuropathic pain and self-management were $9.3{\pm}1.4$(Range 0-14.64) and $5.40{\pm}0.76$(Range 1-7), respectively. There were 33.3% of the patients whose HbA1c levels are higher than 7.5%. Neuropathic pain was positively correlated with glycemic control(r=.18, p=.035), and was negatively correlated with self-management(r=-.19, p=.023).
The present study was conducted to investigate the effect of whole wheat bread with added Lindera obtusiloba (LO) and Capsosiphon fulvescens (CF) ethanol extracts on serum glucose and lipid levels in Sprague Dawley rats. Rats were divided into five groups depending on the diet administered: normal bread (NC), whole wheat bread (W), whole wheat bread with LO leaves extract (WL), whole wheat bread with CF extract (WC), and whole wheat bread with freeze-drying CF (WDC). After 4 weeks of consuming the experimental diet, the blood glucose level and hemoglobin A1c contents were found to be significantly lower in the W, WL, WC, and WDC groups than in the NC group. The high-density lipoprotein-cholesterol levels increased in the WL group when compared to those in the NC group and triglycerides levels decreased in all wheat groups compared to those in the NC group. These results suggest that wheat breads containing LO and CF extracts are effective for preventing hypercholesterolemia and obesity.
