Electrochemical Behavior and Differential Pulse Polarographic Determination of Rifampicin in the Pharmaceutical Preparations

  • Differential pulse polarographic(DPP) analytical procedure for the rifampicin antibiotic, which can be applied to monitor its synthetic process from the starting antibiotic of rifamycin B or rifamycin SV has been developed based on the electrochemical reduction of an azomethine group. Rifampicin exhibited a cathodic peak due to the azomethine group in the side chain of 3-[(4-methyl-1-piperazinyl)imino]methyl moiety and another cathodic peak due to the carbonyl group in rifamycin SV by DPP. The experimental peak potential shift of an azomethine reduction was -73 mV/pH in the pH range between 3.0 and 7.5, agreeing with involvement of 4 e-and 5 $H^5$ in its reduction. By the cyclic voltammetric(CV) studies, the azomethine and the carbonyl reductions in rifampicin were processed irreversibly on the mercury electrode. The plot of peak currents vs. concentrations of rifampicin ranging $1.0{\times}10^{-7} M~$1.0{\times}10^{-5} M yielded a straight line with a correlation coefficient of 0.9996. The detection limit was $1.0{\times}10^{-8} M with a modulation amplitude of 50 mV DPP has been successfully applied for the determination of rifampicin in the pharmaceutical preparations.

  • The synthesis and characterization of the monomeric group 13 heterocyclic thiosemicarbazone complexes $Me_2M[SC_4H_3CHNNC(S)SCH_3]$ (M=Al (2), Ga (3)) are described. Compounds 2-3 were prepared using $MMe_3$ (M=Al, Ga) in toluene with 2-thiophenecarboxaldehyde-S-methyldithiocarbazat e under anaerobic conditions. These complexes have been characterized by $^1H\;NMR,\; ^{13}C\; NMR$, elemental analyses, and single-crystal X-ray diffraction. 2 crystallizes in the monoclinic space group $P2_1/c$ with unit cell parameters a=10.2930(5) Å, b=18.564 (1) Å, c=7.3812(6) Å, V=1347.9(2) Å3, $D_{calc}=1.342\; gcm^{-3}$ for Z=4, 9281 reflections with $I_o<3{\sigma}\;(I_o),$ R1=0.0500 and wR2=0.0526. 3 crystalizes in the orthorhombic space group $P_{bca}$ with unit cell parameters a=13.340(3) Å, b=19.9070(5) Å, c=11.3690(2) Å, $V=2673.88(9)\;{\AA}^3$, $D_{calc}=1.511\; gcm^{-3}$ for Z=8, 17004 reflections with $I_o>3{\sigma}\;(I_o),$, R1=0.0480 and wR2=0.0524. Compound 3 is a monomeric gallium compound with a weak interaction between the pendant thiophene and the gallium center.

  • RFSP is a computer program to do fuel management calculations for CANDU reactors. Its main function is to calculate neutron flux and power distributions using two-energy-group, three dimensional neutron diffusion theory. However, up to now the treatment has not been true two-group but actually "one-and-half groups". In other words, the previous (1.5-group) version of RFSP lumps the fast fission term into the thermal fission term. This is based on the POWDERPUFS-V Westcott convention. Also, there is no up-scattering term or bundle power over cell flux (H1 factor) for the fast group. While POWDERPUFS-V provides only 1.5 group properties, true two-group cross sections for the design and analysis of CAUDU reactors can be obtained from WIMS-AECL. To treat the full two-group properties, the previous RFSP version was modified by adding the fast fission, up-scatter terms, and H1 factor. This two-group version of RFSP is a convenient tool to accept lattice properties from any advanced lattice code (e.g. WIMS-AECL DRAGON, HELIOS...) and to apply to advanced fuel cycles. In this study, the modification to implement the true two-group treatment was performed only in the subroutines of the *SIMULATE module of RFSP. This module is the appropriate one to modify first, since it is used for the tracking of reactor operating histories. The modified two-group RFSP was evaluated with true two-group cross sections from WIMS-AECL. Some tests were performed to verify the modified two-group RFSP and to evaluate the effects of fast fission and up-scatter for three core conditions and four cases corresponding to each condition. The comparisons show that the two-group results are quite reasonable and serve as a verification of the modifications made to RFSP. To assess the long-term impact of the full 2-group treatment, it is necessary to simulate a long period (several months) of reactor history. It will also be necessary to implement the full two-group treatment of reactivity devices and assess the reactivity-device worths.ce worths.

Effect of Hypothermia on the Prevention of Ventilator-Induced Lung Injury in Rats (백서에서 저체온 전처치가 환기기유발폐손상 억제에 미치는 효과)

  • Backgrounds : Because ventilator-induced lung injury is partly dependent on the intensity of vascular flow, we hypothesized that hypothermia may attenuate the degree of such an injury through a reduced cardiac output. Methods : Twenty-seven male Sprague-Dawley rats were randomly assigned to normothermia ($37{\pm}1^{\circ}C$)-injurious ventilation (NT-V) group (n=10), hypothermia ($27{\pm}1^{\circ}C$)-injurious ventilation (HT-V) group (n=10), or nonventilated control group (n=7). The two thermal groups were subjected to injurious mechanical ventilation for 20 min with peak airway pressure 30 cm $H_2O$ at zero positive end-expiratory pressure, which was translated to tidal volume $54{\pm}6\;ml$ in the NT-V group and $53{\pm}4\;ml$ in the HT-V group (p>0.05). Results : Pressure-volume (P-V) curve after the injurious ventilation was almost identical to the baseline P-V curve in the HT-V group, whereas it was shifted rightward in the NT-V group. On gross inspection, the lungs of the HT-V group appeared smaller in size, and showed less hemorrhage especially at the dependent regions, than the lungs of the NT-V group. [Wet lung weight (g)/body weight (kg)] ($1.6{\pm}0.1$ vs $2.4{\pm}1.2$ ; p=0.014) and [wet lung weight/dry lung weight] ($5.0{\pm}0.1$ vs $6.1{\pm}0.8$ ; p=0.046) of the HT-V group were both lower than those of the NT-V group, while not different from those of the control group($1.4{\pm}0.4$, $4.8{\pm}0.4$, respectively). Protein concentration of the BAL fluid of the HT-V group was lower than that of the NT-V group($1,374{\pm}726\;ug/ml$ vs $3,471{\pm}1,985\;ug/ml$;p=0.003). Lactic dehydrogenase level of the BAL fluid of the HT-V group was lower than that of the NT-V group ($0.18{\pm}0.10\;unit/ml$ vs $0.43{\pm}0.22\;unit/ml$;p=0.046). Conclusions : Hypothermia attenuated pulmonary hemorrhage, permeability pulmonary edema, and alveolar cellular injuries associated with injurious mechanical ventilation, and preserved normal P-V characteristics of the lung in rats.

  • The impressions for odors are subjective and have individual differences. In this study, the Impressions of odors were investigated by covariance structure analysis. 46 subjects (men in their twenty) recorded their reactions to ten odorants by grading them on a seven-point scale in terms of twelve adjective pairs. Their reactions were quantified by using factor analysis and covariance structure analysis. The factors were extracted as "preference", "arousal" and "persistency". The subjects were classified into three groups according to the most suitable causal models (structural equation models). Each group had different causal relationship and different impression structure for odors. It was suggested that there is a possibility to evaluate the subjective impression of odor using covariance structure analysis.

COMPARATIVE STUDY ON PERCENTAGE OF CANAL ENLARGEMENT AND CANAL ANGULATION CHANGE BY PRECURVED ENDOSONIC K-FILE (Precurved Endosonic K-file에 의한 근관확대율 및 만곡도 변화에 대한 연구)

  • The purpose of this study was to evaluate canal shaping ability and canal angulation change of K-file, straight endosonic K-file and pre curved endosonic K-file. Twenty staight canals and thirty curved canals were selected and divided into five groups according to canals curvature and canal instrumentation method. VI group was prepared by straight endosonic K-file and H1 group by K-file in straigt canals. V2 group was instrumented by straight endosonic K-file, V3 group by pre curved endosonic K-file and H2 group by precurved K-file in curved canals. Radiographs of canals were obtained before and after canal shaping. And postoperative radiographs were compared with preoperative radiographs using superimposition method. The results obtained were as follows ; l. In straight canals, K-file group demonstrated lager percentage of canal enlargement than endosonic K-file group on facial view, but reverse results exhibit on mesial view. 2. In curved canals, precurved K-file group showed largest percentage of canal enlargement, followed by precurved endosonic K-fine group and straight endosonic K-file group was smallest. 3. Percantage of canal enlargement at convex side was greater than at concave side in apical portion of each group. Especially in straight endosonic K-file group, percantage of canal enlargement at convex side and concave side showed sharply difference. 4. In angulation change, the straight endosonic K-file group exhibited the greatest its change, followed by precurved endosonic K-file group and precurved K-file group was the least. Above results suggest that K-file is more effective endodontic instrument than endosonic K-file, and that precurved file is effective for canal shaping in curved canal.

Combination effects of nitrite from fermented spinach and sodium nitrite on quality characteristics of cured pork loin

  • Objective: The purpose of this study was to investigate the effect of fermented spinach derived nitrite and sodium nitrite on cured pork loin. Methods: The following treatments were prepared using brine (8% [w/v] salt): Control (-), no nitrite added; Control (+), 0.08% (w/v) sodium nitrite brine; T1, 0.04% (w/v) nitrite fermented spinach juice in 0.04% (w/v) sodium nitrite brine; T2, spinach juice in 0.04% (w/v) sodium nitrite brine; T3, 0.04% (w/v) nitrite fermented spinach juice used as sodium nitrite free brine; and T4, spinach juice used as sodium nitrite free brine. T2 and T4 were incubated to allow to reduce nitrate to nitrite. Results: Spinach juice did not affect cooking loss and pH but negatively influenced flavor and overall acceptability (p<0.05). T1 samples containing synthetic and natural nitrites showed the highest redness values. Spinach juice negatively affected volatile basic nitrogen; however, thiobarbituric acid reactive substance values of T1 and T3 were similar to those of controls (+) (p>0.05). Residual nitrite content decreased with decreasing synthetic nitrite levels. T1 and control samples showed no significant differences in overall acceptability (p>0.05). Conclusion: Thus, combined synthetic and natural nitrites improved the quality of cured pork loin.

A Kinetic Study on the Solvolysis of Benzyl Chloride under High Pressure (고압하에서 염화벤질의 가용매분해반응에 대한 속도론적 연구)

  • Rates of solvblysis of benzyl chloride in ethanol-water mixtures have been measured at 30 and $40^{\circ}C$ under various pressures up 1.6 kbar. The plots of 1n k as a function of pressure are fitted to a second order function in p, and values of ${\Delta}V^{\neq}$and ${\Delta}{\beta}^{\neq}$ are obtained from the results. Also the values of various pseudo thermodynamic quantities were evaluated from the rate constants. The relationships of the 1n k to $Q_w$ or 1n $C_w$ indicate that the reaction proceeds through $S_N1$ mechanism. A comparison between the present and the previous results gives that the increasing order of ${\mid}{{\Delta}V_0}^{\neq}{\mid}$ and n-values are $p-Cl>p-H>p-CH_3$ and $p-CH_3>p-H.p-Cl$, respectively. From these results, it is believed substituent such as the $p-CH_3$group favors the $S_N1(1)$ character, while the p-Cl group leads to the $S_N1(2)$ character.

Influence of Human Follicular Fluid and Gonadotropins in the Culture Medium on the In Vitro Maturation, Fertilization and Development of Human Immature Oocytes (배양액 내 인간 난포액 및 성선자극호르몬 첨가가 인간 미성숙 난자의 체외성숙, 수정 및 체외 배발달에 미치는 영향)

  • This study was conducted to examine the effects of human follicular fluid and gonadotropin (FSH+HCG+rhEGF) on in vitro maturation, fertilization and development of human immature oocytes. Cumulus-oocyte complexes (COCs) were collected following for in vitro fertilization and embryo transfer (IVF-ET) cycles of the patients. At the time of oocytes collection, oocytes were classified into MII, MI and GV in accordance with their appearance (MII: Fully mature oocyte at metaphase II of meiosis; MI: Nearly mature oocytes at metaphase I of meiosis; GV: Immature oocytes at prophase I of meiosis). After controlled ovarian stimulation using gonadotropin(FSH) and human chorionic gonadotropin (HCG) in 70 ICSI cycles, 158 MI to MII matured oocytes were intracytoplasmic sperm injection (ICSI) ${\sim}4$ h after in vitro culture and 553 MII oocytes were ICSI after denudation. The aspirated MI and GV oocytes were cultured in culture medium containing 10% (v/v) serum protein substitute (SPS), 10% (v/v) human follicular fluid (hFF) and 10% (v/v) serum protein substitute (SPS)+1 IU/ml FSH+10 IU/ml HCG+10 ng/ml recombinant human epidermal growth factor (rhEGF). The maturation rate of immature oocytes was similar among the three group. When maturation medium was supplemented with 10% SPS, 10% hFF or gonadotropins, the fertilization rate of in vitro matured oocytes was higher in 10% SPS (80.0%), but there was no statistical significance (78.2%; hFF, 76.9%; gonadotropin, p>0.05). The development rate of human embryos developed to $6{\sim}8$ cells were not significant difference in the medium containing SPS, hFF and gonadotropins (65.6%, 65.9% and 66.7%). The results of these study suggest that human follicular fluid and gonadotropins supplemented in the culture medium was not effected on the in vitro maturation, fertilization and development of human immature oocytes.

Effects of Bromelain and Double Emulsion on the Physicochemical Properties of Pork Loin

  • Our aim was to investigate the effects of bromelain embedded in double emulsion (DE) on physicochemical properties of pork loin. We evaluated DE characteristics such as size, zeta potential, and microscopy after fabrication. We marinated meat with distilled water (DW), 1% (w/v) bromelain solution, blank DE, and 1% (w/v) bromelain loaded in double emulsion (DE E) for 0, 24, 48, and 72 h at 4℃, and prepared raw meat for control. The marinated samples were assessed for color, water holding capacity, cooking loss, moisture content, pH, protein solubility, Warner-Bratzler shear force (WBSF) and gel electrophoresis. The droplet size of 1% (w/v) bromelain embedded in DE was increased compared with blank DE (p<0.05) and values of zeta potential decreased. The increase in lightness and color difference range of the DE-treated group was lower than that of the DW-treated group (p<0.05). Moreover, treatment by immersion in 1% (w/v) DE E resulted in the highest water holding capacity values (p<0.05) and lower cooking loss values than water base treatment (p<0.05). Results for myofibrillar protein solubility and WBSF showed a similar trend. 1% (w/v) DE E showed degradation of myosin heavy chain after 48 h in sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Thus, bromelain-loaded DE is useful for controlling and handling enzyme activity in food industry.
