Understanding of the relationships between seeding rate and planting date is important for production of spring oats during the fall season. Therefore, this experiment was conducted to investigate seeding rate and planting date effects on forage performance and quality of spring oats(Avena sativa L.) at Suweon in 1997. The experiment was arranged in a split plot design with three replications. The main plots consisted of different seeding rates such as 150, 200, and 250kg/ha. The subplots consisted of different planting dates of spring oats such as 15, 22, and 29 August and 5 September. Dry matter content of oats was significantly decreased(P<0.01) as the planting date was delayed except the planting date of 5 September(24.8%). Dry matter content of forage oats planted on 15, 22, and 29 August was 23.0, 21.5, and 20.5%, respectively. A seeding $rate\;{\times}\;planting$ date interaction for dry matter content was highly significant(P<0.01). Crude protein content of oats was significantly increased(P<0.01) from 15.6 to 27.9% as the planting was delayed, while a significant seeding $rate\;{\times}\;planting$ date interaction for crude protein content of oats was observed. Acid detergent fiber(ADF) and neutral detergent fiber(NDF) contents of oats increased with early planting(P<0.01), but there was no significant difference among the seeding rates. ln vitro dry matter digestibility(IVDMD) of oats was significantly increased(P<0.01) as the seeding rate was increased together with late planting. Average dry matter yield of oats decreased(P<0.01) from 2,647 to 615kg/ha as the planting was delayed, but there was no significant difference among the seeding rates. A seeding $rate\;{\times}\;planting$ date interaction was observed for average dry matter yield of oats that the seeding rate increase in this experiment showed better at early planting dates. Crude protein yield of oats increased from 171 to 410kg/ha as the planting was delayed, and a significant increases(P<0.01) for crude protein yield was found among the seeding rates. ln vitro digestible dry matter(IVDDM) yield of oats decreased(P<0.01) as the planting was delayed, but there was no significant difference among the seeding rates. A significant seeding $rate\;{\times}\;planting$ date interaction for lVDDM of oats was found. The results of this study indicate that both the seeding rate of 200kg/ha and early planting before 29 August would be recommendable for forage yield and quality of oats in Korea.
A new demand for vegetable peanut (Arachis hypogaea L.) in Korea has increased farmers interest in growing vegetable peanut. Compared to grain peanut production, vegetable peanut production enables the growth period to be shortened by 20 or 30 days and farmers to adopt various cropping systems and to return crop residues in the soil. With the purpose of establishing desirable cropping systems for sustainable vegetable peanut production, three field experiments were conducted from 2000 to 2001 at Milyang, the southeastern part of Korea. Main focuses were given into the effect of cropping systems for vegetable peanut production on each crop's yield and soil sustainability. The cropping systems investigated were single vegetable peanut, peanut-radish-green barley, peanut-barley, and peanut-garlic cropping system, with or without crop residue incorporation in the soil. Among the cropping systems investigated for sustainable vegetable peanut production, peanut-only and peanut-radish-green barley cropping systems showed vulnerable to diseases and lodging while peanut-barley and peanut-garlic cropping systems showed higher stability in response to diseases and lodging, consequently leading to higher yield potential of vegetable peanut production. In the peanut-barley cropping system, both barley and peanut residues returned to the soil played an important role in soil improvement as well as in significantly increased grain yield of peanut and barley. A particular notice was taken to the pronounced increase in Trichoderma population and the amount of nitrogen mineralization induced by the returned barley residue. Soil structure, compactness, pH, and fertility were positively influenced by the returned crop residues, which apparently increased sustainability in vegetable peanut production systems.
This experiment was conducted to investigate the effects of compost leachate and concentrated slurry on growth of tomato in hydroponic culture. In process of composting, compost leachate was produced water was through a saturated compost heap. The concentrated slurry was produced by filtration and concentration by membrane process. Filtration of pig slurry was necessary to prevent the hose clogging in hydroponics culture. The treatments of this experiment were consisted of seven different liquid fertilizers; compost leachate(CL), concentrated pig slurry (CS), compost leachate+byproduct(CL+BP), concentrated pig slurry+byproduct(CS+BP), compost leachate 50%+nutrient solution50%(CL+NS), concentrated pig slurry 50%+nutrient solution50%(CS+NS) and nutrient solution(NS) for tomato based on nitrogen content. The chemical nutrient solution was the solution of National Horticulture Research Station for the growth of tomato. The concentration of nutrient solution was adjusted a range of $1.6{\sim}2.0 mS/cm$ in EC. 1. The compost leachate and concentrated pig slurry were low in phosphorus(P), calcium(Ca), magnesium(Mg), but rich in potassium(K). 2. Plant height, SPAD value of tomato was highest in the plot of CS+NS, intermediate in CL, CS+BP, and lowest in 100% concentrated pig slurry. 3. The tomato yield of compost leachate plot was 91% compared with inorganic nutrient solution. The compost leachate solution could be used as a nutrition solution of tomato in organic hydroponics. 4. The growth including plant height, SPAD value, fruit number, fruit weight and yield of tomato in the CL 50%+NS 50% was similar in the control. In conclusion, the mixture solution of 50% pig slurry and 50% nutrient solution could be used as a nutrition solution of tomato hydroponic culture.
A 2-year field experiment was conducted to determine the influence of early and late soybean (Clycine max Merr) seeding times on competitive relationships between soybeans and annual weeds. Soybeans were planted on May 20 and June 20. Durations of weed competition and weed control were 0, 3, 6, 9, and 12 weeks after planting sad for full growing season. Dominant weeds were Portulaca oleracia, Centipeda minima, Rorippa cantonienais, Chenopodium album, Acalypha australis, Echinochloa crusgalli, and Digitaria sanguinalis. The dry weight of weeds increased with extended competition, especially at early seeding time of soybeans. Soybean yields were decreased as duration of weed competition was extended. More severe yield reduction occurred at late-planted soybeans with early stage weed competition but at early-planted soybeans when weeds competed with soybeans for a 2 weeds and full growing season. The dry weight of weeds emerged after weeding was decreased at early and late seeding times of soybeans as duration of weed control was prolonged. Soybean yields decreased with shortening duration of weed control end this trend of yield reduction was slightly more remarkable at late-planted soybeans than at early-planted soybeans. Late-planted soybeans were required longer than 6 weeks of weed control period to achieve maximum yield as compared to early-planted soybeans.

For the maximum production of pullulan from glucose as a carbon source, the effects of glucose concentration, pH and dissolved oxygen concentration on the cell growth and mass production of high-molecular weight pullulan by A. pullulans ATCC 42023 were evaluated. A. pullulans showed optimum pullulan productivity when glucose concentration was 0.3M (54g/L). And inhibitory effects on the cell growth and the pullulan production were observed at the glucose concentration higher than 0.3M (54g/L). The influence of pH control and dissolved oxygen on the pullulan production and growth of A. pullulans was studied. In shake-flasks, maximum pullulan production was obtained with $11.98g/{\ell}$ when initial pH was 6.5. In the batch fermentation, the maximum pullulan production of $13.31g/{\ell}$ was obtained with constant pH 4.5. And it was found that pullulan yield and synthesis rate increased with oxygen availability. For the production of commercially useful pullulan with high-molecular weight, a mixed carbon source, which was a mixture of glucose and glucosamine, was used for the pullulan fermentation with A. pullulans. On the basis of 5% mixed carbon source, culture with 3% glucosamine with 2% glucose was optimum condition for the production of high (M.W.> 1,000,000) and medium (M.W.> 200,000) molecular weight pullulan with considerable yields of cell mass and product. And the influence of pH control on the molecular weight of pullulan was studied in batch fermentation. It was found that the productivity of high-molecular weight pullulan with pH control at 6.5 was higher than that with no pH control.

The human complement receptor type 1 (CR 1, C3 b/C4b receptor) is a polymorphic membrane glycoprotein expressed on human erythrocytes, peripheral leukocytes, plasma and renal glomerular podocytes, which consists of transmembrane and cytoplasmic domains with 30 repeating homologous protein domains known as short consensus repeats (SCR). CR1 has been used as an inhibitor for inflammatory and immune system for the past several years. Recently; it is reported that CRl was found to suppress the hyper-acute rejection in xeno-transplantation and can be used to cure autoimmune diseases. A soluble form of CRl, called sCRl, is a recombinant CRl by cleaving the transmembrane domain at C-terminus and has been expressed in Chinese Hamster Ovary (CHO) cells. Several purification methods for sCR1 from CHO cells have been reported, but most of them require complicated steps at high cost. Moreover, such methods are mostly performed under the pH condition apt to denaturing sCR1 and causes sCRl losing its activity. We here report a rapid and efficient method to purify sCR1 from CHO cell. The new method consists of a two-stage of cell culture by cultivating cells in serum medium followed by serum-free medium, and a two-stage of column purification by means of heparin and gel filtration column chromatography. By using this novel method, sCR1 can be purified in a simple and effective way with high yield and purity, furthermore, the purified sCR1 was confirmed to retain its activity to suppress the complement activation in vivo and ex vivo.
The growth kinetics of Streptomyces noursei NRRL 5126 was investigated under different aeration and agitation combinations in a 5.0 l stirred tank fermenter. Poly-${\varepsilon}$-lysine biosynthesis, cell mass formation, and glycerol utilization rates were affected markedly by both aeration and agitation. An agitation speed of 300 rpm and aeration rate at 2.0 vvm supported better yields of 1,622.81 mg/l with highest specific productivity of 15 mg/l.h. Fermentation kinetics performed under different aeration and agitation conditions showed poly- ${\varepsilon}$-lysine fermentation to be a growth-associated production. A constant DO at 40% in the growth phase and 20% in the production phase increased the poly-${\varepsilon}$-lysine yield as well as cell mass to their maximum values of 1,992.35 mg/l and 20.73 g/l, respectively. The oxygen transfer rate (OTR), oxygen utilization rate (OUR), and specific oxygen uptake rates ($qO_2$) in the fermentation broth increased in the growth phase and remained unchanged in the stationary phase.
We established an efficient plant regeneration system for Catharanthus roseus L. (G.) Don through somatic embryogenesis. Embryogenic callus was induced from hypocotyl of seed germinated in vitro. Somatic embryogenesis in Catharanthus has been categorized into three distinct stages: (1) initiation and proliferation of embryo; (2) maturation, and; (3) germination or plantlet conversion. Beside plant growth regulators, various stages of embryogenesis were screened for their response to a wide variety of factors (pH, gelrite, light, sugar alcohols, polyethyleneglycol and amino acids), which affect embryogenesis. All of the tested factors had a small to marked influence on embryogeny and eventual conversion to plantlets. The plantlets were acclimatized successfully in a greenhouse. To our knowledge, this is the first report describing a detailed study of various cultural factors which regulate embryogenesis in C. roseus. The results discussed in this paper may be used in mass propagation to produce medicinal raw material, and the embryo precursor cells could be used in genetic modification programmes that aim to improve the alkaloid yield as well.
A study was conducted to determine the dietary effects of a clay mineral (sodium bentonite, NaB) on manure gas emission, health, production, and meat characteristics of Hanwoo steers. Two diets fed to steers included a control diet (concentrate mix and rice straw) and a treatment diet (control diet+1.0% clay mineral/concentrate mix). Dietary NaB addition considerably reduced concentrations of gases ($H_2S$, $SO_2$ and $NH_3$) in the manure of Hanwoo steers. Growing steers fed NaB had similar blood profiles with the exception of lower (p<0.05) concentrations of blood alkaline phosphatase and lactate dehydrogenase. Dietary NaB addition tended to increase (p = 0.10) live weight by 30 kg at slaughtering and did not affect carcass yield and quality traits. Dietary NaB addition increased concentrations of P (p<0.01), Mg (p<0.01), Na (p<0.01), Zn (p<0.005), K (p = 0.08), Fe (p = 0.08) and Cu (p = 0.07) in the longissimus muscle compared to the control but did not affect (p>0.05) fatty acid composition. The study demonstrated that the dietary addition of a clay mineral could be effective in improving mineral bioavailability to Hanwoo steers, which could be one of the reasons for their improved performance.
The assessment of reproductive potentials of female gayal under semi-intensive management was conducted with 18 animals in Bandarban hilly area of Bangladesh. Age at first estrous (days), age at first conception (days), length of estrous cycle (days), number of service per conception (natural), gestation period (days), age at first calving (days), and calving interval (natural service) (days) of gayal were $598.2{\pm}168.44$, $723{\pm}169.94$, $21.86{\pm}2.93$, 1.41, $296.05{\pm}3.87$, $1014.42{\pm}260.32$ and $465{\pm}80.48$ respectively. Daily milk yield and lactation length of gayal were $304.98{\pm}30.46ml/day$ and $116.67{\pm}8.08days$. Gayal male calves had higher birth weight (19.67 kg) than female calves (15.58 kg) in first lactation. Birth weight increases in second and third lactation than first lactation in both male and female calves. Highest conception rate observed in winter season and 70.60% successful conception occur when service was given within 21-30 h of estrous. Maximum number of female gayal use to come in estrous in winter and calved in monsoon and autumn. Cervicitis and irregular heat were the most reproductive problems followed by metritis, case of abortion, anestrous and repeat breeding. From this study it was observed that the reproductive performance of gayal is very close with the domestic cattle. It may use as beef cattle for the hilly regions of Bangladesh.
