Effect of Butyric Acid on Performance, Gastrointestinal Tract Health and Carcass Characteristics in Broiler Chickens

  • An experiment was conducted to study the effect of graded levels of butyric acid (butyrate) on performance, gastrointestinal tract health and carcass characteristics in young broiler chickens. Control starter (0-3 wk) and finisher (4-5 wk) diets were formulated to contain 2,900 kcal ME/kg and 22% CP, and 3,000 kcal ME/kg and 20% CP, respectively. Subsequently, four other experimental diets were formulated to contain 0.05% antibiotic (furazolidone) or 0.2, 0.4 and 0.6% butyric acid. Each diet was fed at random to 8 replicates of 6 chicks each throughout the experimental period (0-5 wk). The results showed that 0.4% butyrate in the diet was similar to antibiotic in maintaining body weight gain and reducing E. coli numbers but superior for feed conversion ratio. No added advantage on these parameters was obtained by enhancing the concentration of butyrate from 0.4 to 0.6% in the diet. Feed intake and mortality were not influenced by the dietary treatments. A reduction in pH of the upper GI tract (crop, proventiculus and gizzard) was observed by inclusion of butyrate in the diets of broilers compared to either control or antibiotic-fed group. Butyrate at 0.4% was more effective in reducing the pH than 0.2% butyrate. Within the lower GI tract, 0.4 and 0.6% butyrate was effective in lowering pH in the duodenum, but no effect was found in either the jejunum or ileum. The villus length and crypt depth in the duodenum increased significantly in all the butyrate treated diets irrespective of the level tested. Carcass yield was higher and abdominal fat content was lower significantly in all the butyrate treatment groups compared to the control or antibiotic group. From these findings, it is concluded that 0.4% butyric acid supplementation maintained performance, intestinal tract health, and villi development and carcass quality in broiler chickens.

  • Microalgae do not require much land and make a higher efficient oil production. However, it costs still much higher than other biodiesel resources, such as crops. Sugars charge 80% of culture media when microalgae are massively cultured in the fermenter. This study aims to develop a cost-efficient process for sugar production from Chinese cabbage byproducts. Pre-treatment with 0.25% H2SO4 was most effective when chopped cabbage was incubated 50℃/130 rpm for 24 hours. To hydrolyze cabbage cellulose, we used cellulases secreted from Trichoderma. harzianum. T. harzianum was cultured at 28℃/pH 7/130 rpm for five days. Optimal enzymatic activity of cellulase was obtained by incubating at 0.24 FPU/ml/45℃/pH 5/130 rpm for three days. In comparison to other agricultural waste, such as rice straw, green tea leaves, and palm residue, Chinese cabbage produced the highest sugar yield. We found the optimal conditions to produce sugar from Chinese cabbage byproducts as a carbon source to culture biodiesel-producing microalgae. The efficient process developed in this study helps microalgae as a sustainable alternative energy source by cost-down.

Development of Yeast Strains as Feed for Aquaculture: Possible Yeast Strains (양식을 위한 먹이사료로서의 Yeast 균주의 개발: 가능성 있는 효모 균주)

  • Possible yeast strains that could be used as feed for aquaculture were studied. It was shown that the maximum specific growth rate and the biomass yield of Kluyveromyces fragilis yeast and Candida utilis yeast under optimum pH and temperature were much higher than those of Saccharomyces cervisiae yeast which had been as established yeast diet for rotifer culture. Hence, this work was focussed on the growth characteristics of the two yeasts through flask dultures for mass production. With 5% inoculum dosage, the best values of $\mu$$_{max}$ and OD$_{max}$ were obtained with on 2.5% fructose medium and 2% YE medium for K. fragilis and C. utilis, respectively, where the values of $\mu$$_{max}$ and OD$_{max}$ were found to be 0.73 hr$^{-1}$ and 3.00 for K. fragilis and 0.59 hr$^{-1}$ and 2.80 for C. utilis. It was also found that the lag phase of the growth incresed with increasing initial zinc and NaCl concentrations and decreased with increasing inoculum dosage. Both yeasts could survive relatively well at 3.5% NaCl concentration, and only C. utilis yeast could utilize zinc.

  • A field experiment was conducted to evaluate the effectiveness of an organic fertilizer (Glutamic acid fermentation by product ameded with nitrogen) on the growth and yield of radish and chinese cabbage on a relatively fertile soil. Additional laboratory studies were also made to find out the influence of mentioned fertilizer on the phisical and chemical characteristic of soils. The results are summarized as following. 1. Effect of fertilizer on the growth and yield. a. On a relatively fertile soil with high organic matter content (3%), the organic fertilizer was not effective for the growth and yield of radish and yield of radish and chinese cabbage. b. The application of organic fertilizer increased the K contents in plants both in radish and chinese cabbage. It was observed that the high content of K in radish leave lowered the leaf/root ratio and the leaf/root ratio showed a positive correlation with root weight. 2. Effect on soil physical and chemical properties. a. The application of organic fertilizer resulted in the improvement of soil physical property by lowering bulk density and increasing porosity. b. It was found that application of organic fertilizer increased the water stable soil aggregates. c. The application of organic fertilizer did not result in the significant increase in soil organic matter. d. Soil pH was lowered by the application of organic matter. It was speculated that the oxidation of ammonical nitrogen in the fertilizer is responsible for the lowering of soil pH.

Isolation and Characterization of a Novel Lactic Acid Bacterium for the Production of Lactic Acid

  • We isolated a novel lactic acid bacterium from a Korean traditional fermented food, soybean paste. The newly isolated strain, dubbed RKY2, grew well on glucose, sucrose, galactose, and fructose, but it could not utilize xylose, starch, or glycerol. When the partially amplified 16S rDNA sequence (772 bp) of the strain RKY2 was compared with 10 reference strains, it was found to be most similar to Lactobacillus pentosus JCM $1588^T$, with 99.74% similarity. There-fore, the strain RKY2 was renamed Lactobacillus sp. RKY2, which has been deposited in the Korean Collection for Type Cultures as KCTC 10353BP. Lactobacillus sp. RKY2 was found to be a homofermentative lactic acid bacterium, because its end-product from glucose metabolism was found to be mainly lactic acid. It could produce more than 90 g/L of lactic acid from MRS medium supplemented with 100 g/L of glucose, with 5.2 g $L^-1$ $h^-1$ of productivity and 0.95 g/g of lactic acid yield.

  • Polyvoltine silkworm breeds/hybrids play an important role in tropical sericulture. In the process of synthesizing more potential polyvoltine hybrids(polyvoltine${\times}$bivoltine) of superior quantity and quality, the Andhra Pradesh State Sericulture Research and Development Institute (APSSRDI) has developed 8 promising polyvoltine breeding lines (SDMG2, APM16, APM15, APM14, APM5, RM2, APM3, APM13). In order to study their hybrid vigour and performance, these breeds were crossed with 4 potential bivoltine testers (SDD1, SDD2, SDD3, APS8), and 32 different hybrid combinations were prepared and evaluated for their mid parent heterosis (MPH) and better parent heterosis (BPH) of silk productivity. Since silk is the ultimate product required for commercial purpose, based on silk productivity and its heterosis, three superior polyvoltine hybrid combinations namely RM2${\times}$APS8 (24.3%), APM3${\times}$APS8 (12.4%) and APM15${\times}$SDD2 (10.8%) were adjudicated as potential heterotic hybrid combinations of superior silk yield and hence recommended for further large scale field trails and commercial exploitation.

Kinetics of Hydrogen Rich Ethanol as Reductant for HC-SCR over $Al_2O_3$ Supported Ag Catalyst (Ag/$Al_2O_3$ 촉매하의 HC-SCR에서 수소 풍부 에탄올의 반응 특성)

  • Ethanol was used as reductant to remove $NO_x$ over Ag/$Al_2O_3$ catalyst via SCR from stationary emission source. Among the tested hydrocarbon reductants, ethanol showed highest de-$NO_x$ performance over the Ag/$Al_2O_3$ catalyst. De-$NO_x$ efficiency of about 83% was obtained in the condition of GHSV 20,000 $hr^{-1}$, $NO_x$ 200 ppm, CO 200 ppm, $O_2$ 13%, $H_2O$ 5% and mole ratio of ethanol/$NO_x$ = 2 between temperature of $300^{\circ}C$ and $400^{\circ}C$. While $SO_2$ presence in the $NO_x$ exhaust suppressed the catalytic activity, catalyst with acid (0.7% $H_2SO_4$) treatment of catalyst showed higher catalytic activity, where In-Situ DRIFT showed S presence over catalyst surface was increased after acid treatment of catalyst. From in-situ DRIFT and SCR results, it was concluded that sulfur presence over the surface of Ag/$Al_2O_3$ catalyst was the dominant factor to control the de-$NO_x$ reaction yield via HC-SCR from the exhausted gas including $SO_2$.

Partial Oxidation of Methane to Syngas over M(10)-Ni(5)/SBA-15(M=Ce, Nd, Sm) Catalysts (M(10)-Ni(5)/SBA-15(M=Ce, Nd, Sm) 촉매상에서 합성가스 제조를 위한 메탄의 부분산화반응)

  • M(10)-Ni(5)/SBA-15(M=Ce, Nd, Sm) catalysts were prepared for the partial oxidation of methane (POM) to syngas. The catalysts were characterized by BET, TEM, and XPS. The BET-specific surface area and average pore size for M(10)-Ni(5)/SBA-15(M=Ce, Nd, Sm) were 538.8, 504.3, and $447.3m^2/g$ and 6.4, 6.8, and 7.1 nm, respectively. TEM results showed that the mesoporous hexagonol structure was formed for SBA-15, while the homogeneous dispersion of Ni and Ce particles on the surface was formed for Ce(10)-Ni(5)/SBA-15 caused by the confinment effect of SBA-15. XPS data confirmed that $Ce^{4+}$ and $Ce^{3+}$ on the surface catalyst have two oxidation states due to the lattice oxygen species ($O^{2-}$, $O^-$). The yields of POM to syngas over Ce(10)-Ni(5)/SBA-15 were 52.9% $H_2$ and 21.7% CO at 1 atm, 973 K, $CH_4/O_2=2$, $GHSV=1.08{\times}10^5mL/g_{cat.}{\cdot}h$, and these values were kept constant even after 75 h on streams. The same tendency of syngas yields was observed for M(10)-Ni(5)/SBA-15(M=Ce, Nd, Sm). These results confirm that the redox reaction of promoters including Ce, Nd, and Sm enhanced the stability and yield of catalysts.

Study on Growth Curves of Longissimus dorsi Muscle Area, Backfat Thickness and Body Conformation for Hanwoo (Korean Native) Cows

  • The objective of this study was to estimate the parameters of Gompertz growth curves with the measurements of body conformation, real-time ultrasound longissimus dorsi muscle area (LMA) and backfat thickness (BFT) in Hanwoo cows. The Hanwoo cows (n = 3,373) were born in 97 Hanwoo commercial farms in the 17 cities or counties of Gyeongbuk province, Korea, between 2000 and 2007. A total of 5,504 ultrasound measurements were collected for the cows at the age of 13 to 165 months in 2007 and 2008. Wither height (HW), rump height (HR), the horizontal distance between the top of the hips (WH), and girth of chest (GC) were also measured. Analysis of variance was conducted to investigate variables affecting LMA and BFT. The effect of farm nested in location was included in the statistical model, as well as the effects of HW, HR, WH, and GC as covariates. All of the effects were significant in the analysis of variance for LMA and BFT (p<0.01), except for the HR effect for LMA. The two ultrasound measures and the four body conformation traits were fitted to a Gompertz growth curve function to estimate parameters. Upper asymptotic weights were estimated as $54.0cm^2$, 7.67 mm, 125.6 cm, 126.4 cm, 29.3 cm, and 184.1 cm, for LMA, BFT, HW, HR, WH, and GC, respectively. Results of ultrasound measurements showed that Hanwoo cows had smaller LMA and greater BFT than other western cattle breeds, suggesting that care must be taken to select for thick BFT rather than an increase of only beef yield. More ultrasound records per cow are needed to get accurate estimates of growth curve, which, thus, helps producers select animals with high accuracy.

Synthesis and Characterization of Poly(ethylene oxide) Derivatives Containing Carbonate Linkages (고분자 주사슬에 카보네이트 관능기가 도입된 새로운 폴리에틸렌 옥사이드 유도체의 합성 및 분석)

  • In order to obtain PEO with reduced crystallinity, novel PEO derivatives containing carbonate linkages in the main chain have been synthesized by the reaction of various molecular weight poly (ethylene glycol)s (PEGs) with dimethyl carbonate (DMC) in the presence of $H_2SO_4$ to yield methyl carbonate terminated PEGs, followed by condensation reaction under vacuum in the presence of titanium isopropoxide (TiP) catalyst. The number average molecular weight of PEGs used was in the range of 200 and 600 g/mol. The structure and compositions of the resulting polymers were characterized by $^1$H-NMR and $^{13}C-NMR$. Their thermal behavior and molecular weight were characterized by DSC/TGA and GPC, respectively.
