Detection of Fungi Associated with Ear Blighting of Rice Plants and Their Effect on Seed Discoloration and Yield (벼 이삭마름병해(病害)에 관여(關與)하는 균류(菌類)의 검출(檢出)과 검출(檢出)된 균류(菌類)가 종자(種子) 변색(變色) 및 수량(收量)에 미치는 영향(影響))

  • 1. Alternaria tenuis, Cladosporium cladosporioides, Curvularia lunata Fusarium semitectum, Gerlachia oryzae, Phoma sp. Trichoconiella padwickii were found associated with ear blighting of rice plants. The frequency of isolation of the fungi was found to be higher from the severely discolored rice grains than that from the brownish spotted grains, but H. oryzae was encounted highly from the latter. 2. Higher counts of Alternaria tenuis, Helminthosporium oryzae, Trichoconiella padwickii, Fusarium moniliforme, Fusarium semitectum were observed under alternating cycles of 12hrs of light and darkness than under complete darkness but G. oryzae was encountered highly under darkness in the blotter. While no difference in the occurrence of the other fungi was recorded under l2hr NUV light-12hr darkness cycles and darkness. 3. A. tenuis and C. cladosporioides were encountered highly at the lower temp. $(15^{\circ}C)$ but Phoma sp. was occurred highly at the higher temp. $(30^{\circ}C)$ when the seeds were incubated in the blotter. 4. Artificial inoculation of the fungi in the ears of rice plants caused severe discoloration of rice grains and reduced 1,000 grain weights and grain ripening percentage.

Studies on the life history of green peach aphid, Myzus persicae Sulzer (Homoptera) (복숭아흑진딧물(Myzus persicae Sulzer)의 생활사에 관한 연구)

  • The green peach aphid Myzus persicae(Sulzer), is known as the most important vector of potato leaf-roll virus and PVY. Yield of potato plants infested with these viruses are remarkably lower than non-infested plants. A study was conducted to investigate the life history of the green peach aphid at Suweon, Korea (Lat. $37^{\circ}$'N., Long. $126^{\circ}59$'E). The following were obtained: 1. Overwintering eggs hatched from late March to early April, with a hatching rate averaging ninety-five percent. 2. The fundatrigeniae leave the primary host(Punus persica) in early to mid May and migrate to the secondary hosts. 3. From mid to late Oct., the gymnoparae migrate from the secondary hosts to the primary hosts. 4. From early to mid Nov., gymnoparae lay fertilized eggs around buds, in bark crevices, or between bifurcated twigs of the primary hosts. 5. The early-born progeny have 23 generations and the late-born progeny have 9 generations on peach trees, potatoes and raddish from Apr. to Oct. 6. The average length of life was about 28.5 days, with a developmental period of approximate 10.8 days and a reproductive period of 15.8 days. 7. The average number of nymphs produced by a female was fifty, with a maximum number of 118. 8. The average number of nymphs produced by a female per day was about 3.2, with a maximum number of 13.

Changes in Physical and Chemical Properties of Coir Used as the Bag Culture Substrate of Greenhouse Tomatoes for Three Years

  • To identify causes for drastic decrease in yield of tomato with repeated culturing number of the bag culture substrate of greenhouse tomatoes we investigated the physical and chemical properties of a coir used as the bag culture substrate to grow tomato at the Booyeo tomato experimental institute located in Booyeo, Chungnam Province for three years from 2012 to 2014. The results showed that total porosity ranged from 65.4 to 73.1% for the bulk densities of coir ranging from 0.12 to $0.14g\;cm^{-3}$. The volumetric water contents measured at 0.01 bar as air entry point were 25% (before), 33% ($1^{st}yr$), 45% ($2^{nd}yr$), and 37% ($3^{rd}yr$). Organic matter contents ranged from 82.0 to 96.2% (highest in $1^{st}yr$). pH and EC ranged from 4.47 to 6.47 (highest in $2^{nd}yr$), and from 22.2 to $53.5dS\;m^{-1}$ (highest in $1^{st}yr$) and cation exchange capacity ranged from 71.0 to $191.7cmol\;kg^{-1}$ (highest in $3^{rd}yr$). The surface structure observed with electrical microscope showed that the number of large pores decreased with increasing cultivating time while the proportion of smaller pores increased, indicating that the coir was consistently decomposed. Therefore, we could conclude that these changes of all physical and chemical properties of the coir may influence the holding capacities of water and nutrients, resulting in deterioration of quality of culture substrate of greenhouse tomatoes.

  • These studies were carried out to select somatic hybrid using selectable marker genes of Nicotiana glauca transformed by NPTII gene and Solanum tuberosum transformed by T- DNA, and to study characteristics of transformant. The results are summarized as follows. 1. Crown gall tumors and hairy roots were formed on potato tuber disc infected by A. tumefaciens Ach5 and A. rhizogenes ATCC15834. These tumors and roots could be grown on the phytohormone free media. 2. Callus formation from hairy root was prompted on the medium containing 2, 4 D 2mg/I with casein hydrolysate lg/l. 3. The survival ratio of crown gall tumor callus derived from potato increased on the medium containing the activated charcoal 0. 5-2. 0mg/I because of the preventions on the other hand, hairy roots were necrosis on the same medium. 4. Callus derived from hairy root were excellently grown for a short time by suspension culture on liquid medium containing 2, 4-D 2mg/I and casein hydrolysate lg/l. 5. The binary vector pGA643 was mobilized from E. coli MC1000 into wild type Agrobacteriurn tumefaciens Ach5, A. tumefaciens $A_4T$ and disarmed A. tuniefaciens LBA4404 using a triparental mating method with E. ccli HB1O1/pRK2013. Transconjugants were obtained on the minimal media containing tetracycline and kanamycin. pGA643 vectors were confirmed by electrophoresis on 0.7% agarose gel. 6. Kanamycin resistant calli were selected on the media supplemented with 2, 4-D 0.5mg/1 and kanamycin $100\mug$/ml after co- cultivating with tobacco stem explants and A. tumefaciens LBA4404/pGA643, and selected calli propagated on the same medium. 7. The multiple shoots were regenerated from kanamycin resistant calli on the MS medium containing BA 2mg/l. 8. Leaf segments of transformed shoot were able to grow vigorusly on the medium supplemented with high concentration of kanamycin $1000\mug$/ml. 9. Kanamycin resistant shoots were rooting and elongated on medium containing kanamycin $100\mug$/ml, but normal shoot were not. 10. For the production of protoplast from potato calli transformed by T-DNA and mesophyll tissue transformed by NPTII gene, the former was isolated in the enzyme mixture of 2.0% celluase Onozuka R-10, 1.0% dricelase, 1.0% macerozyme. and 0.5M mannitol, the latter was isolated in the enzyme mixture 1.0% Celluase Onozuka R-10, 0.3% macerozyme, and 0.7M mannitol. 11. The optimal concentrationn of mannitol in the enzyme mixture for high protoplast yield was 0.8M at both transformed tobacco mesophyll and potato callus. The viabilities of protoplast were shown above 90%, respectively. 12. Both tobacco mesophyll and potato callus protoplasts were fused by using PEG solution. Cell walls were regenerated on hormone free media supplemented with kanamycin after 5 days, and colonies were observed after 4 weeks culture.

Root-zone Application of Insecticides in Gelatin Capsules for the Control of Rice Insect Pests (살충제(gelatin capsule 입)의 수도근부처리에 의한 수도해충 방제효과)

  • During 1973 a field experiment was carried out to evaluate effectiveness of the root zone application of insecticides in medical gelatin capsules for control of several rice insect pests and dwarf virus disease. At three days after transplanting the capsules were pushed by hand about 2.5cm into the soil, near roots of Tongil rice plants. At the given day intervals number of leaf-and plant-hoppers on the hills were recorded by direct count, and dead hearts and white heads by stem borers and dwarf virus infected hills were observed in the experiment plots. Finally grain yields were measured. The percentages of dead hearts and white heads in all the plots were too low for evaluating the effectiveness of insecticides against the striped rice borers. Carbofuran of the insecticides tested was relatively effective against green rice leafhopper (Nephotettixcincticeps) and small brown planthopper (Laodelphax striatellus), although small number of insects on the hills were recorded. Relatively small number of white-backed planthopper (Sogatella furcifera) was occured in the BPMC and Diazinon treated plots. BPMC and Carbofuran were highly effective against the brown planthopper (Nilaparvata lugens) to 80 days after treatment. Incidence of dwarf virus disease was least in the plots of Carbofuran treatment. The highest grain yield was recorded in the plots treated with Carbofuran, and it seemed to be related with good protection from the dwarf virus disease and leaf-and plant-hoppers.

Relationships of Plasma Insulin-like Growth Factor (IGF)-I and IGF-II Concentrations to Litter Size (Landrace와 Yorkshire 돼지에서 혈장의 Insulin-like Growth Factor (IGF)-I과 IGF-II 농도와 산자수 및 비유성적과의 관계)

  • The present study was undertaken to find relationships of plasma insulin-like growth factor (IGF)-I and IGF-II concentrations to litter size and lactation performance. Sixty pure-bred Landrace and Yorkshire pigs having similar farrowing weeks which had been selected from a large number of pregnant gilts and sows were divided into low- (<${\mu}$-0.5SD) and high-litter size (>${\mu}$+0.5 SD) lines under a 2 (breed)${\times}$2 (line) factorial arrange of treatments. After adjusting the litter size to nine piglets per sow at farrowing, total litter weight was measured at three weeks postpartum at weaning as an index of milk yield. Blood samples were obtained from the jugular vein at day (d)-90 pregnancy (Px) and at d-15 postpartum. The litter size or the number of piglets born during the present experiment and the average litter size during the entire parities up to the present one were greater in the high-line than in the low-line by 3.7 and 2.4 piglets, respectively (P<0.01); effect of the breed on litter size was not significant. Plasma IGF-II concentration at d-90 Px was greater in the high-line than in the low-line. Litter size and d-90 Px IGF-I concentration were negatively correlated in Landrace (r=-0.46; P<0.05) and tended to be negatively correlated in Yorkshire (r=-0.31; P=0.09), which resulted in a significant negative correlation between these two variables in total animals (r=-0.35; P<0.01). Litter weight at weaning was not different between the two breeds or lines. Relationships between the litter weight and IGF concentration were not consistent across the breed ${\times}$ physiological stage combinations. Results suggest that d-90 Px IGF concentrations may be indicative of the litter size at impending farrowing.

Environmental Evaluation of the Productivity and Functional Fructo-oligosaccharides of Yacon Cultivated in Various Regions of Korea (우리나라 재배지역별 야콘 생산성 및 기능성 프락토올리고당 평가)

  • The purpose of this study was to investigate the effects of envrionmental factors on the ecological responses, yield, and quality properties and physicochemical characteristics of yacon cultivated in various regions of Korea. Experiments were carried out in eight regions from 2010 to 2013. The temperature range in Jinbu, Bonghwa, Cheolwon, and Gangneung during the growth period of yacon cultivation was $17.5-24.6^{\circ}C$. The total yield and marketable yield of tuberous root in Jinbu were 4,065 and 3,196 kg/10a, respectively. The sugar content of yacon tuberous roots comprised 0.11-0.20% fructose, 0.11-0.37% glucose, 0.39-0.68% sucrose, 0.07-0.37% reducing sugar, and 7.03-9.62% fructo-oligosaccharides. The content of fructo-oligosaccharides, which is a functional substance, was the highest in yacon cultivated in Jinbu. Based on the productivity and functional fructo-oligosaccharides, the optimum areas to cultivate yacon are Jinbu and Bonghwa, which are located in the middle-highland zone (500-560 m) and have a suitable growth temperature of $18-25^{\circ}C$. The results of the present study indicate that temperature could be the factor with the greatest influence on the root growth of yacon.

Fertilizing Effects of Swine Compost Fermented with Sawdust on Mixed Pastures (혼파초지에 대한 톱밥발효돈분의 시용효과)

  • Experiment was carried out to find the fertilizing effects of 8 different application rates of swine compost fermented with sawdust(SCS) including Chemical fertilizer(CF) on forage yield and soil chemical characteristics of mixed pastures sown in Sep. 1993 at National Livestock Research Institute, RDA., in Suwon during low years. It was arranged in a randomized complete block design with three replicates. Dry matter yield were shown at similar among treatments except Control and $50\%$ SCS of standard amount plot. In botanical composition, the legume and weeds percentages of each treatment were increased as advancing year. The final year's legume percentage were high in line with SCS fertilizing plots($39\%{\sim}43\%$), SCS + CF plots($30\%{\sim}41\%$) and CF plot($32\%$). In productions of TDN, NE and crude protein yield, SCS or SCS($75\%$) + CF($25\%$) were nearly same comparing those of CF, respectively. Phosphate, potassium, magnesium contents and K/(Ca + Mg) except calcium contents of those SCS fertilizing plots in plant were generally high with comparing CF. Those contents were proportional according to the fertilizing amount These result indicate the possibility to substitute chemical fertilizer for SCS($75\%$, 25ton/ha) + CF, $25\%$) as manure-N 210 kg/ha, but might be considered accumulation phosphate in the soil.

  • An International Cooperative Biodiversity Group (ICBG) program based at the University of Illinois at Chicago initiated its activities in 1998, with the following specific objectives: (a) inventory and conservation of of plants of Cuc Phuong National Park in Vietnam and of medicinal plants of Laos; (b) drug discovery (and development) based on plants of Vietnam and Laos; and (c) economic development of communities participating in the ICBG project both in Vietnam and Laos. Member-institutions and an industrial partner of this ICBG are bound by a Memorandum of Agreement that recognizes property and intellectual property rights, prior informed consent for access to genetic resources and to indigenous knowledge, the sharing of benefits that may arise from the drug discovery effort, and the provision of short-term and long-term benefits to host country institutions and communities. The drug discovery effort is targeted to the search for agents for therapies against malaria (antimalarial assay of plant extracts, using Plasmodium falciparum clones), AIDS (anti-HIV-l activity using HOG.R5 reporter cell line (through transactivation of the green fluorescent protein/GFP gene), cancer (screening of plant extracts in 6 human tumor cell lines - KB, Col-2, LU-l, LNCaP, HUVEC, hTert-RPEl), tuberculosis (screening of extracts in the microplate Alamar Blue assay against Mycobacterium tuberculosis $H_{37}Ra\;and\;H_{37}Rv),$ all performed at UIC, and CNS-related diseases (with special focus on Alzheimer's disease, pain and rheumatoid arthritis, and asthma), peformed at Glaxo Smith Kline (UK). Source plants were selected based on two approaches: biodiversity-based (plants of Cuc Phuong National Park) and ethnobotany-based (medicinal plants of Cuc Phuong National Park in Vietnam and medicinal plants of Laos). At mc, as of July, 2001, active leads had been identified in the anti-HIV, anticancer, antimalarial, and anti- TB assay, after the screening of more than 800 extracts. At least 25 biologically active compounds have been isolated, 13 of which are new with anti-HIV activity, and 3 also new with antimalarial activity. At GSK of 21 plant samples with a history of use to treat CNS-related diseases tested to date, a number showed activity against one or more of the CNS assay targets used, but no new compounds have been isolated. The results of the drug discovery effort to date indicate that tropical plant diversity of Vietnam and Laos unquestionably harbors biologically active chemical entities, which, through further research, may eventually yield candidates for drug development. Although the substantial monetary benefit of the drug discovery process (royalties) is a long way off, the UIC ICBG program provides direct and real-term benefits to host country institutions and communities.

Effect of Nitrate Concentration in Culture Solution on the Growth and the Uptake of Inorganic Elements of Tomato Plants(Lycopersicum esculentum Mill) (배양액(培養液)의 질산태질소농도가 토마토(Lycopersicum esculentum Mill)의 생육 및 무기원소(無機元素) 흡수에 미치는 영향)

  • This experiment was carried out to investigate the effects of nitrate concentration in culture solution on the growth and the uptake of inorganic elements in Tomato plant in the greenhouse. Tomato plants(cv. TVR-2) were grown with nitrate concentrations 8, 16, 24, 32cmol/l, based on Japan ENSI standard solution. Dry weights of lamina and petiole increased with the nitrate concentration. However, the dry weight of fruit was the highest in the treatment of nitrate concentration of 16cmol/l. The proportion of dry weights of vegitative organ to reproductive organ was the lowest in the treatments of nitrate concentrations of 16cmol/l and it increased with the nitrate concentration. The fruit yield was the highest at the treatment of nitrate concentration of 16cmol/l. With the increase of nitrate level the concentrations of N, $NO_3-N$, Ca and Na increased in lamina and petioles. The concentrations of K, P, S and Cl tended to decline in the nitrate concentration of 16 and 32cmol/l. These results indicate that optimum nitrate concentrations in a tomato grown by hydroponics change with growth stage, and the optimum concentrations for vegitative and reproductive stage were 8 and 16cmol/l, respectively. It also was proved that the nitrate concentrations in the culture solution affected antagonistically the uptake of inorganic anion in tomato : In low nitrate level $Cl^-$ uptake was affected much, while $SO_4{^{2-}}$ and $H_2PO_4{^-}$ uptake were affected in high nitrate level.
