The Effect of Bottom ash in Reducing Cadmium Phytoavailability in Cadmium-contaminated Soil (중금속 오염 농경지 토양에서 바닥재 시용에 의한 카드뮴 식물이용성 저감효과)

  • BACKGROUND: Since bottom ash (BA) contains considerable amounts of CaO and MgO, it could be a useful amendment to increase soil pH and to immobilize cadmium (Cd). This study was conducted to evaluate effect of BA application in reducing Cd phytoavailability.METHODS AND RESULTS: Bottom ash was applied at the rate of 0, 20, 40, and 80 Mg/ha to Cd contaminated soil, and then lettuce was cultivated under field condition. soil pH and net negative charge increased slightly with increasing BA application; however, there was no statistical difference among the rates. Water soluble, exchangeable+acidic, reducible, and oxidizable fraction of Cd decreased with increasing bottom ash application rate, whereas residual fraction of Cd increased with increasing bottom ash application rate. Lettuce yield increased with rate of bottom ash up to 40 kg/ha. Visual evidences of cadmium toxicity and growth inhibition were not found during lettuce cultivation.CONCLUSION: Bottom ash was effective to reduce phytoextractability of Cd and to increase lettuce yield. Conclusively, BA could be a good soil amendment to reduce Cd phytoavailability in contaminated arable soil.

Influence of Pressure Toasting on Starch Ruminal Degradative Kinetics and Fermentation Characteristics and Gelatinization of Whole Horse Beans (Vicia faba) in Lactating Dairy Cows

  • Whole horse beans (Vicia faba cv. Alfred) (WHB) were pressure toasted at different temperatures of 100, 118 and $136^{\circ}C$ for 3, 7, 15 and 30 minutes in order to determine an optimal heating conditions to increase bypass starch (BPSt) as glucose source which is usually limiting nutrient in highly producing dairy cows in the Netherlands. Starch (St) Ruminal Degradative Kinetics and Fermentation Characteristics of (SRDC) of WHB were determined using in sacco technique in 4 lactating dairy cows fed 47% hay and 53% concentrate according to Dutch dairy cow requirements. Measured characteristics of St were soluble fraction (S), potentially degradable fraction (D) and rate of degradation (Kd) of insoluble but degradable fraction. Based on measured characteristics, percentage bypass starch (BPSt) was calculated according to the Dutch new feed evaluation system: the DVE/OEB system. Pressure toasting temperatures significantly affected starch gelatinization (p<0.01). Degradability of Starch in the rumen was highly reduced by pressure toasting (p<0.01). S varied from 58.2% in the raw WHB (RWHB as a control) to 19.6% in $136^{\circ}C/15min$. S was reduced rapidly with increasing time and temperature (p<0.01). D varied from 41.8% in RWHB to 80.5% in $136^{\circ}C/15min$. D fraction was enormously increased with increasing time and temperature (p<0.01). Kd varied from 4.9%h in RWHB to 3.4%/h in $136^{\circ}C/15min$. All these effects resulted in increasing %BPSt from 29.0% in RWHB to 53.1% in $136^{\circ}C/15min$. Therefore BPSt increased from 93.5 g/kg in RWHB to 173.5 g/kg in $136^{\circ}C/15min$. The effects of pressure toasting on %BPSt and BPSt seemed to be linear up to the highest values tested. Therefore no optimal pressure toasting conditions could be determined at this stage. But among 10 treatments, The treatment of $136^{\circ}C/15min$was the best with the highest BPSt content. It was concluded that pressure toasting was effective in shifting starch degradation from rumen to small intestine to increase bypass starch.

  • Column leaching tests were conducted using fresh and weathered pulverised fuel ash of some 17 and 40 years old from two major British power plants, with deionised water and simulated synthetic industrial leachate. The former was to see the leaching behaviour of weathered ash and the latter was to see if the formation of secondary products from water and PFA interaction and ameliorating effect in removing metals from industrial leachates. Fresh PFA liberates elevated concentrations of surface-enriched inorganics, including Ca, Na, K, B, $Cr_{total}$, Li Mo, Se and $SO^{2-}_4$. This might indicate their association with the surface of PFA particles. In the column leaching tests using weathered ash and deionised water, elements are not readily leached but are released more slowly, showing relatively constant concentrations. For the case of weathered ash, some readily soluble surface-enriched elements appears to have been liberated in their early stage of leaching and the liberation of glass associated elements are thought to be more important function in controlling the element concentration. The result from column leaching tests exceed for a number of elements when compared with various Water Standards and suggests the leachate from PFA disposal mound needs dilution to achieve target concentrations. PF A shows element retention effect for many elements, including B, Fe, Zn, Hg, Ni, Li and Mo, in the order of fresh Drax ash > weathered Drax ash > Weathered Meaford ash in retaining capacity. Geochemical modelling using a computer program WATEQ4F reveals some solubility controlling secondary solid products. These include $CaSO_4{\cdot}2H_2O$ for Ca, $Al(OH)_3$ for Al and $Fe(OH)_3$ for Fe.

Antioxidative and Antibacterial Activities of Artemisia princeps Pampanini Extracts (사자발쑥 추출물의 항산화 및 항균 활성)

  • In the present study, the antioxidative and antibacterial activities of Artemisia princeps Pampanini (A. princeps Pamp.) extract were investigated. The ethyl acetate fraction of A. princeps Pamp. showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}=12.27{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of A. princeps Pamp. extract on $Fe^{3+}-EDTA/H_2O_2$ systems were investigated using a luminol-dependent chemiluminescence assay. The ethyl acetate fraction of the extract ($OSC_{50}=0.33{\mu}g/mL$) had a 5 times greater ROS scavenging activity than L-ascorbic acid ($1.50{\mu}g/mL$), known as a water soluble antioxidant. The cellular protective effects of fractions of A. princeps Pamp. on the rose-bengal sensitized photohemolysis of human erythrocytes were examined. The aglycone fraction of extracts suppressed photohemolysis in a concentration dependent manner. The inhibitory effects of A. princeps Pamp. extract on tyrosinase were investigated to assess their whitening efficiency. The ethyl acetate fraction demonstrated a 7 times higher tyrosinase inhibitory effect ($IC_{50}=29.20{\mu}g/mL$) than albutin, known as a whitening agent. The antibacterial activity of ethyl acetate fractions against various normal skin flora were measured. The results showed that the antibacterial activity of the fraction was the highest on Staphylococcus aureus, Bacillus subtilis, and Propionibacterium acnes. Antioxidant substances were isolated and purified from the ethyl acetate fractions. Eupatilin and jaceosidin were identified. These results indicate that the extract/fractions of A. princeps Pamp. can function as antioxidant and/or antibacterial agents for the skin.

Calcium Deficiency Causes Pithiness in Japanese Pear (Pyrus pyrifolia cv. Niitaka) Fruit (칼슘 결핍에 의한 '신고' 배 (Pyrus pyrifolia cv. Niitaka) 과실에서의 바람들이)

  • BACKGROUND: Pithy pear fruit are not distinguished externally from sound fruit and thus often cause unexpected economic losses. To find out the cause of pithiness, the pithiness incidence and characteristics of Japanese pear (Pyrus pyrifolia cv. Niitaka) fruit picked from a spot frequently produced pithy fruit in an orchard were compared with those of fruit picked from another spot produced sound fruit every year. And the soil chemical properties of the two spots and mineral contents in fruit, shoots, and leaves of Japanese pear trees cultivated in the two spots were also examined. METHODS AND RESULTS: The pithiness incidence was 0, 8.8, and 11.3% at 7 days before and 0 and 7 days after optimal harvest date, respectively, in the spot frequently produced pithy fruit. Flesh firmness was significantly lower in pithy fruit than in sound fruit, while soluble solids content was slightly higher in pithy fruit than in sound fruit. Unlike other mineral contents, Ca content was significantly lower in pithy fruit than in sound fruit. These results indicate that Ca deficiency in fruit is closely associated with decrease in flesh firmness and thus pithiness development. Ca content in soil of the spot frequently produced pithy fruit was also significantly lower than that in soil of the spot produced sound fruit. However, shoots or leaves did not exhibit significant difference in Ca and/or other mineral contents between the two spots, indicating that Ca deficiency in fruit is dependent on the translocation of Ca within a plant rather than soil Ca status. Although total-N, available $P_2O_5$, K, and Ca contents were significantly lower in soil of the spot frequently produced pithy fruit than in soil of the spot produced sound fruit, Mg and Na contents and pH were not different between the soil conditions. CONCLUSION(S): Fruit maturity and Ca level in fruit are closely related to the incidence of pithiness in 'Niitaka' Japanese pear.

  • This experiment was conducted to investigate the influence of amounts of lime dressed on the growth of rice by the treatment of percolation and nonpercolation in the acid sulphate soil. And also analysis of soil chemical components after treatment was carried out. The results obtained were summarized as follows: 1. In the initial stage of growth, number of tillers and plant length showed no distinct differences between the treatments of percolation and nonpercolation, but after August the effect of lime appeared and the percolation treatment was more effective than the nonpercolation. 2. Lime dressing affected good influence on the panicles, grain per panicles and the rate of grain formation, and the treatment of percolation showed better results than nonpercolation. 3. If the yield of rough rice in the control (nonpercolation and lime dressing) was 100, it was 194 in the treatment of nonpercolation 12me/100gr of lime dressed, 268 in the treatment of percolation-4me/100gr of lime and 315 in the 8me/100gr-percolation. 4. Lime dressing affected good influence on the control of Helminthosporium leaf spots. 5. In the case of lime dressing, amounts of available phosphate and soluble silicon dioxide were increased, but ferrous ion ($Fe^{{+}{+}}$) were decreased.

  • A pot experiment was conducted to find out the effects of application of slacked lime and fused super-phosphate on the lead uptake of upland crops in a lead added soil. Lead concentration of the soils were adjusted to 0, 150, 300mg/kg respectively. The slacked lime was applied at the equivalent amount of lime requirement with extra 150kg/10a, and 2 times for the fused superphosphate. The results obtained were as follows: 1. Lead contents in crops increased in the order: sesame > maize > potato > sweet potato > soybean > green perilla > peanut > red bean. 2. Lead contents in parts of crops were increased in the order; root > stem > leaf > grain. 3. Increasing lead concentration in soils, lead content in the plant was increased and crops yield were decreased. 4. Lead contents in soybean and green perlilla were decreased in slacked lime application treatment. 5. The lead contents in leaf and grain of soybean and green perllila decreased with decreasing in the ratio of Pb/Ca+Mg equivalent in soil. 6. Grain yield were increased in slacked lime, but were decreased in fused superphosphate application treatment. 7. With increasing the soil Pb contents, calcium and phosphate contents were increased in leaf and stem, but calcium was decreased in roots. 8. $1N-NH_4$ OAC soluble Pb contents in soil were 26-50 ppm and 42-70 ppm, respectively, for 150mg/kg and 300mg/kg lead treatments. 9. The soil pH was increased in the order of slacked lime, fused superphosphate and nontreatment.

Studies on the Microflora and Enzymes Influencing on Korea Native Kochuzang (Red Pepper Soybean Paste) Aging (재래식(在來式) 고추장 숙성(熟成)에 미치는 미생물(微生物) 및 그 효소(酵素)에 관(關)한 연구(硏究))

  • The study was carried out to investigate the changes of the various chemical components and the microflora during the aging period of Korean navive Kochuzang. (Red pepper soybean paste) Korean native maeju loaves were separated into surface and inner parts. Three kinds of Korean native Kochuzang were prepared from surface part, inner part, and ordinary of maeju. The selection and the indentification of the high enzyme producing strains from the microflora and characteristics of their enzymes were studied. I. The changes of the various chemical components during the aging period of Kochuzang. 1) The changes of pH in the 3 kinds of Kochuzang displayed rapid decrease for the first 10 days after preparing and gradual curve of decrease until 60 days, but slight increase for the next 30 days. The pH of the surface part Kochuzang was lower than that of inner part or ordinary Kochuzang. 2) The total acid contents in the 3 kinds of Kochuzang showed gradual increase until the 60 days but it slowly reduced after this time. 3) The total nitrogen contents in the 3 kind of Kochuzang showed gradual inerease up to the 60 days, but slight decrease after this time. 4) The changes of trichloroacetic acid soluble nitrogen in the 3 kinds of Kochuzang showed a remarkable increase for the first 10 days, however gradual increase after this time. 5) The increase of amino nitrogen contents in the 3 kinds of Kochuzang seemed to be remarkable until the first 30 days, however to be less remarkable after this time. 6) The contents of reducing sugar in the 3 kinds of Kochuzang showed remarkable increase until the first 50 days and it slowly reduced after this time. II. The changes of microflora during the aging period of Kochuzang. 1) Aerobic, anaerobic bacteria and mold in the 3 kinds of Kochuzang were increased until the first 30 to 40 days, but they were reduced after this time. 2) No yeast in the three kinds of Kochuzang appeared until the first 20 days. Yeast were proved to grow, when the pH value was decreased below 5.4 after the 30 days. Yeasts in the surface part and ordinary Kochuzang were gradually increased and those in the inner part Kochuzang were decreased as aging. III. The selection and identification of high amylase and protease producing strains from the microflora during the aging period of Kochuzang. 1) The amylase and protease highly producing strains from microflora were identified as Bacillus subtilis-P, Bacillus subtilis-G, Bacillus licheniformis-K, Aspergillus oryzae-B. 2) Amylase activity of Aspergillus oryzae-B was highest among the strains and the strains in order of the higher activity to the lower one were Bacillus subtilis-P Bacillus licheniformis-K, Bacillus subtilis-G. Protease activities of Aspergillus oryzae-B and Bacillus subtilis-P were about the same and the strains in order of the higher activity to the lower one were Bacillus licheniformis-K, Bacillus subtilis-G. 3) Amylase activity was inhibited more than protease activity was with NaCl concentration. Amylase activity was inhibited by 45 to 65 percent and protease activity by 40 to 46 percent at the concentration of 15 percent NaCl, which was the average concentration of NaCl in Kochuzang.

  • A pot experiment was conducted to find out the effect of water management on the growth and uptake of arsenic and inorganic nutrients of rice plant at As added soil. The arsenic were added to soil at the levels of As 0, 10, 50, 100 and 150 ppm, respectively. Water management was done with two ways: intermittent irrigation from ten days after transplanting, and continuous submersion until harvest. Higher soil As levels increased As content in plant but reduced growth rate. Aresenic content in plant was considerably reduced with intermittent irrigation compared to continuous submersion. Rice growth showed also same trend. With increasing As levels in soil, N content in plant was increased but P, K, Ca, Mg, $SiO_2$, Fe and Mn content in plant were tend to be decreased. These inorganic nutrients in plant were also much absorbed in continuous submersion compared to intermittent irrigation. Soil pH was slightly increased with increasing As levels in soil while soil Eh has no relationship with soil As levels. On the other hand, soil pH was higher in the treatment of continuous submersion than that of intermittent irrigation but soil Eh showed reverse trend. With increasing As levels in soil, water soluble-As and Ca-As fractions in soil tend to be increased with continuous submersion, but these fractions has no tendency with intermittant irrigation.

Antioxidant Activity of Enzymatic Extracts from Sargassum coreanum (큰잎모자반 효소적 추출물의 항산화 활성)

  • In this study, Sargassum coreanum was enzymatically hydrolyzed to prepare water-soluble extracts by using five carbohydrates (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavozyme and Alcalase) and their potential antioxidant activity were evaluated. The Celluclast and Neutrase extracts of Sargassum coreanum exhibited better DPPH radical scavenging activities (92.42% and 92.78%, respectively) and hydrogen peroxide ($H_2O_2$) scavenging activities (58.28% and 57.97%, respectively) compared to those of other enzymatic extracts. These results suggest that Sargassum coreanum would be a good raw materials for antioxidant and enzymatic hydrolysis would be a good strategy to prepare antioxidant extracts from seaweeds.
