Conditions for Soluble Phosphate Production by Environment-Friendly Biofertilizer Resources, Pseudomonas fluorescens (환경친화적 미생물비료 자원 Pseudomonas fluorescens RAF15에 의한 가용성 인산 생산에 영향을 미치는 조건)

  • The effects of inorganic salts, inoculum concentration, aeration rate and shaking speed on insoluble phosphate solubilization by Pseudomonas fluorescens RAF15 were investigated. Soluble phosphate production was dependent on the presence of $MgCl_2{\cdot}6H_2O$ and $MgSO_4{\cdot}7H_2O$ in the medium. Supplementation of medium with 0.01% $CaCl_2{\cdot}2H_2O$ and 0.01% NaCl slightly increased soluble phosphate production. The optimal medium compositions for the solubilization of insoluble phosphate by P. fluorescens RAF15 were 1.5% glucose, 0.005% urea, 0.3% $MgCl_2{\cdot}6H_2O$, 0.01% $MgSO_4{\cdot}7H_2O$, 0.01% $CaCl_2{\cdot}2H_2O$ and 0.01% NaCl, respectively. Optimal inoculum concentration was 2.0%(v/v). Maximum soluble phosphate production was obtained with 20-50 ml/250-ml flask and 200 rpm of shaking speed, respectively. The addition of EDTA decreased cell growth and soluble phosphate production.

Protective Effects of Omija-tang on $H_2O_2$-induced apoptotic death of H9c2 cardiomyoblast cells (오미자탕(五味子湯)이 심근세포에 미치는 영향)

  • The water extract of Omija-tang(OMJT) has been traditionally used for treatment of ischemic heart and brain damage in oriental medicine. However, little is known about the mechanism by which the water extract of OMJT protects cells from such damage. Therefore, this study was conducted to investigate the protective mechanisms of OMJT on $H_2O_2$-induced toxicity in H9c2 cardiomyoblast cells. Treatment of $H_2O_2$ markedly induced death of H9c2 cardiomyoblast cells in a dose-dependent manner. The characteristics of $H_2O_2$-induced death of H9c2 showed apparent apoptotic features, such as DNA fragmentation. However, OMJT significantly reduced both $H_2O_2$-induced cell death and chromatin fragmentation. The decrease of Bcl-XL expression by $H_2O_2$ was inhibited by OMJT. In addition, the increase of Bcl-XS and Bax expression were also inhibited by OMJT. In particular, Fas expression, which is generally recognized as cell death inducing signal by Fas/FasL interaction, was markedly increased by $H_2O_2$ in a time-dependent manner, whereas this increase was completely prevented by OMJT. The combined treatment of OMJT and $H_2O_2$ in H9c2 cells also reduced activation of caspase-9 and caspase-3 like protease. Taken together, this study indicates that the protective effects of the water extract of OMJT against oxidative damage may be mediated by the modulation of BcI-XL/S and Bax expression by way of the regulation of mitochondrial membrane potential and caspase cascades.

A Study of Efficient Removal of 2-Methylisoborneol and Geosmin by Pulsed Ultraviolet and Ultrasound (효율적인 2-Methylisoborneol, Geosmin의 제거를 위한 Pulsed UV 공정과 Ultrasound 공정의 비교 연구)

  • The degradation of off-flavors which is 2-Methylisoborneol (2-MIB) and geosmin by means of ultrasound (US) and pulsed ultraviolet (PUV) irradiation and its combination with catalyst (wire mesh, wire mesh coated TiO2, and TiO2) and additive (H2O2) were investigated via water system. A combination treatment of TiO2 and H2O2 heterogeneity with US (24 kHz) and PUV (6000 W) has shown improved results in destroying 2-MIB and geosmin, which may be attributed to chain reactions by the enhanced formation of hydroxyl radicals (·OH) through H2O2 dissociation and reactive oxide ions of TiO2 addition. Rapid degradation of off-flavors occurred within 2 min under PUV process with H2O2 100 mg/L (81.5% for 2- MIB; 79.3% for geosmin) and TiO2 100 mg/L (83.7% for 2-MIB; 79.8% for geosmin), while compared with H2O2 100 mg/L (58.4% for 2-MIB; 58.0% for geosmin) and TiO2 100 mg/L (59.2% for 2-MIB; 38.5% for geosmin) within 5 min under US process. Surprisingly, the emphasis was given on the comparison with the same injected energies between PUV and US on degradation efficiency. Based on the injected energy comparison, the US provided better degradation performance under equal input power of 200 kJ with H2O2 100 mg/L, while compared with H2O2 100 mg/L under PUV process. Our findings suggest that US can be more effective compared to PUV for the degradation of off-flavors in aspect of energy consumptions.

Hydrothermal Synthesis and Mechanical Characterization of 3mol%Y2O3-ZrO2 by Urea Contents (우레아 첨가량 변화에 따라 수열합성법으로 제조 된 3mol%Y2O3-ZrO2 분말의 합성 및 기계적 특성 평가)

  • The industrial manufacturing of YSZ products can be summarized as a three step process: a) hydrolysis of zirconyl chloride and mixing of other solutions, b) precipitation, and c) calcination. The addition of ammonia or OH- is essential in the precipitation process. However, a strong agglomeration was observed in the results of an ammonia or OH- addition. Thus, it is necessary to disperse the powders smoothly in order to improve the mechanical strength of YSZ. In this study, YSZ was synthesized using the urea stabilizer and hydrothermal method. YSZ powders were synthesized using a hydrothermal method with Teflon Vessels at $180^{\circ}C$ for 24 h. The mole ratio of urea to Zr was 0, 0.5, 1, and 2. The crystal phase, particle size, and morphologies were analyzed. Rectangular specimens ($33\;mm{\times}8\;mm{\times}1{\pm}0.5\;mm$) for three-point bend tests were used in the mechanical properties evaluation. The crystalline of YSZ powders observed a tetragonal phase in the sample with a ratio of Zr:urea = 1:2 addition and a hydrothermal reaction time of 24 h. The average primary particle size of YSZ was measured to be 9 nm to 11 nm. The agglomerated particle size was measured from 15 nm to 30 nm. The three-point bending strength of the YSZ samples was 142.47 MPa, which is the highest value obtained for the Zr:urea = 1:2 ratio addition YSZ sample.

  • plasma-sprayed Cr$_2$O$_3$-based coatings containing MoO$_3$ were studied to gain a better understanding of the influence of MoO$_3$ composition in the coatings on their tribological behaviour. A reciprocal type tribo-tester was employed to examine friction and wear behavior of the specimens at high temperature(450。C). The physical characteristics of worn surfaces were investigated by scanning electron microscopy and chemical composition of the coating surfaces was analyzed using a X-ray photoelectron spectrometer. The results showed that friction coefficient of the MoO$_3$-added coatings were lower than those without MoO$_3$ addition. The larger protecting layers were observed at the worn surface of plasma spray coated specimens with MoO$_3$ addition. XPS analysis of the protecting layer indicated that MoO$_3$ composition was dominantly formed at the surface. MoO$_3$ composition in the protecting layer appears to be more favorable in reducing the friction.

Effects of hydrogen peroxide on voltage-dependent K+ currents in human cardiac fibroblasts through protein kinase pathways

  • Human cardiac fibroblasts (HCFs) have various voltage-dependent $K^+$ channels (VDKCs) that can induce apoptosis. Hydrogen peroxide ($H_2O_2$) modulates VDKCs and induces oxidative stress, which is the main contributor to cardiac injury and cardiac remodeling. We investigated whether $H_2O_2$ could modulate VDKCs in HCFs and induce cell injury through this process. In whole-cell mode patch-clamp recordings, application of $H_2O_2$ stimulated $Ca^{2+}-activated$ $K^+$ ($K_{Ca}$) currents but not delayed rectifier $K^+$ or transient outward $K^+$ currents, all of which are VDKCs. $H_2O_2-stimulated$ $K_{Ca}$ currents were blocked by iberiotoxin (IbTX, a large conductance $K_{Ca}$ blocker). The $H_2O_2-stimulating$ effect on large-conductance $K_{Ca}$ ($BK_{Ca}$) currents was also blocked by KT5823 (a protein kinase G inhibitor) and 1 H-[1, 2, 4] oxadiazolo-[4, 3-a] quinoxalin-1-one (ODQ, a soluble guanylate cyclase inhibitor). In addition, 8-bromo-cyclic guanosine 3', 5'-monophosphate (8-Br-cGMP) stimulated $BK_{Ca}$ currents. In contrast, KT5720 and H-89 (protein kinase A inhibitors) did not block the $H_2O_2-stimulating$ effect on $BK_{Ca}$ currents. Using RT-PCR and western blot analysis, three subtypes of $K_{Ca}$ channels were detected in HCFs: $BK_{Ca}$ channels, small-conductance $K_{Ca}$ ($SK_{Ca}$) channels, and intermediate-conductance $K_{Ca}$ ($IK_{Ca}$) channels. In the annexin V/propidium iodide assay, apoptotic changes in HCFs increased in response to $H_2O_2$, but IbTX decreased $H_2O_2$-induced apoptosis. These data suggest that among the VDKCs of HCFs, $H_2O_2$ only enhances $BK_{Ca}$ currents through the protein kinase G pathway but not the protein kinase A pathway, and is involved in cell injury through $BK_{Ca}$ channels.

Study on Two Step Plasma Treatment for Electroless Cu Plating of Fluoropolymer (불소수지의 무전해 동도금을 위한 단계적 플라즈마 전처리법에 관한 연구)

  • Low temperature plasma treatment with different gases and rf powers were performed to improve the adhesion strength between polytetrafluoroethylene(PTFE) and electroless deposited copper. According to the research, $H_2$ plasma having hydrogen radical was more effective in surface polarity modification than $O_2$ plasma due to the defluorination reaction. However, surface roughness of PTFE was more increased with $O_2$ than $H_2$ plasma. PTFE treated with $120W-O_2$ plasma and $250w-H_2$ plasma, consecutively showed rougher surface than single step $250w-H_2$ plasma treated one and more hydrophilic than single step $120W-O_2$ plasma treated one. And it showed 5B tape test grade, which is better adhesion property than 1B or 3B obtained by single step plasma treatment. In addition, adhesion strength between PTFE and Cu deposit is also deeply affected by residual water on its interface.

Removal of Total Organic Carbon and Micropollutants in Tertiary Treated Sewage by Medium Pressure UV/H2O2 (중압 자외선과 과산화수소 공정을 이용한 하수 3차 처리수중 총유기탄소와 미량오염물질 제거)

  • This study evaluated the applicability of UV-AOP process using medium-pressure UV lamp and H2O2 to remove TOC and emerging micropollutants in the effluent from a sewage treatment plant. The UV lamp with higher output(1.6~8.0 kW) showed slightly higher amount of power in removing TOC of 1 mg/L(0.09 kWh/mg/L~0.11 kWh/mg/L), however it was found that there was no significant difference for each cases. In addition, under the condition that the H2O2 concentration is sufficient, as the power consumption of the UV lamp increases, the unit TOC removal concentration per unit H2O2 decomposition concentration also increases, resulting in effective removal of TOC. The removal rate of 7 new trace contaminants, such as antibiotics by the UV-AOP tested, was at least 89.4%, and the ability to remove the emerging micro pollutants in the process was very effective. But, it was judged that it could not be excluded that the probablity of transforming to oxidated by-product in the case of a low TOC removal efficiency. Depending on the operating conditions of the UV and H2O2 processes, a higher BOD concentration is found in the treated water than in the influent, and it is necessary to review the UV power and proper injection conditions of H2O2 to maintain the BOD concentration increase below a certain level.

  • Dense $0.95(Na_{0.5}K_{0.5})NbO_3-0.05LiTaO_3$ (NKN-LT) ceramics were developed by conventional sintering process. Sintering temperature was lowered by adding $Na_2O$ as a sintering aid. The electrical properties of NKN-LT ceramics were investigated as a function of $Na_2O$ concentration. When the sample sintered at $1000^{\circ}C$ for 4h with the addition of 1 mol% $Na_2O$, electromechanical coupling factor ($k_p$) and piezoelectric coefficient ($d_{33}$) of NKN-LT ceramics were found to reach the highest values of 0.43 and 190 pC/N, respectively.

Pre-Irradiation Grafting of Acrylic Acid onto Polyethylene Film (전조사법에 의한 아크릴산의 폴리에틸렌 필름에 대한 그라프트 반응)

  • Acrylic acid was graft-copolymerized on polyethylene film in the presence of additives such as acid and $FeSO_4(NH_4)_2SO_4{\cdot}6H_2O$ using peroxide grafting technique by ${\gamma}$-ray and electron beam, and the effect of $FeSO_4(NH_4)_2SO_4{\cdot}6H_2O$ and acid on the grafting yield was evaluated. The grafting mechanism and the physical property of grafted films were also examined. The results showed that the inclusion of $FeSO_4(NH_4)_2SO_4{\cdot}6H_2O$ in acidified acrylic acid grafting solution was extremely beneficial and led to a most unusual enhancement effect in the radiation grafting. In the other hand, inclusion of mineral acid in the grafting solution in the absence of $FeSO_4(NH_4)_2SO_4{\cdot}6H_2O$ could not lead to he suitable grafting reaction by the severe homopolymerization of acrylic acid. The addition of $H_2SO_4$, and HCl led to much higher grafting yield than $HNO_3$and $CH_3COOH$. It was shown that grafting yield of ${\gamma}$-ray irradiated-polyethylene was higher than that of electron beam irradiated polyethylene.
