Different Effects of Acidic pH Shock on the Prodiginine Production in Streptomyces coelicolor M511 and SJM1 Mutants

  • The changes in prodiginines productions caused by pH shock culture of Streptomyces coelicolor strains were estimated. In Streptomyces coelicolor M511, undecylprodiginine and streptorubin B productions increased 1.8-fold (37.22 mg/g) and 2.5-fold (18.61 mg/g), respectively, by pH shock (from 7.2 to 4.0). In contrast, this resulted in the significantly decreased prodigignines production in the redP deletion mutant SJM1; 3.7-fold for undecylprodiginine, 4.4-fold for streptorubin B, 5.2-fold for methylundecylprodiginine, and 6.4-fold for methyldodecylundecylprodiginine, respectively. RT-PCR analyses showed that, during pH shock, expression of redD, the transcription activator gene, was increased while the expression of fabH, the decarboxylative condensation enzyme gene in fatty acid biosynthesis, was decreased in both strains. The enhanced redD expression was in good accordance with the increased total prodiginines production of M511. However, for SJM1 mutant, the decrease of fabH expression occurred more strikingly, such that it became almost completely turned off during acidic pH shock culture. Therefore, a down-regulation of fabH was considered to be the cause of decreased amount of total prodiginines produced, although redD expression was high in SJM1 mutant.

Isolation and Characterization of Hydrogen Producing Bacterium (수소생산균 Enterobacter cloacae YJ -1의 분리 및 특성)

  • The hydrogen-producing bacterium was isolated from fresh water and identified as Enterobacter cloacae. The isolated was named Enterobacter cloacae YJ-1. In batch culture, The optimum cultivation temperature and pH of strain YJ-1 was 35℃ and 7.5, respectively. All of the added glucose was consumed completely during fermentation even though pH was not controlled. Amount of hydrogen produced on each condition of 2% glucose, 4% sucrose and 5% fructose was 950, 1000 and 948 mL/L, respectively and resulted in increasing hydrogen production approximately 2.5-times more than controlled condition. The maximum hydrogen production was obtained when 50 mM phosphate was added. In repeated-batch culture, hydrogen gas of 1920 mL/L was totally produced for 48. The maximum hydrogen was produced on the condition of 0.5% yeast extract, but the production amount was not changed on the condition of over 0.5%. Most of the organic acids produced during the fermentation were formic and acetic acid, and propionic acid was moiety also generated.

Anti-allergic Effects of Chungshimbohyeoltang in RBL-2H3 Mast Cells and OVA/alum Sensitized Mice (RBL-2H3 비만세포와 OVA/alum 감작 생쥐에서 청심보혈탕(淸心補血湯)의 항알레르기 효과)

  • Objectives The suppressive effect of CSBHT has been mysterious. Thus, the present study is designed to investigate the suppressive effect and its mechanism. Methods To investigate the anti-allergy effect from ChungShimBoHyeolTang(CSBHT), RBL-2H3 cell was used and examined by Real-Time PCR, and IL-4 and IL-13 from RBL-2H3 was examined by ELIS. In addition, GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos, c-Jun, NF-${\kappa}B$ p65 transcription factors of RBL-2H3 mast cell were examined by Western Blotting. Also, OVA/alum-sensitized mice were orally administrated CSBHT and serum OVA-specific IgE production, IL-4, and IL-13 production in splenocytes supernatant were examined. Results As a result of treating with CSBHT extract, RBL-2H3 mast cells significantly suppressed the IL-4 and IL-13 mRNA expression and IL-4 and IL-13 production. Western blot analysis of transcription factors involving IL-4 and IL-13 expression also revealed a prominent decreases of mast cell's specific transcription factors including GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos, and NF-${\kappa}B$ p65. Also, examining the mice, administration of CSBHT suppressed the amount of OVA-specific IgE in OVA/alum-sensitized mice and IL-4 and IL-13 production in splenocytes supernatant. Conclusions The study suggested that the anti-allergic activities of CSBHT suppresses IL-4 and IL-13 production from the Th2 cytokines by suppressing transcription factors as GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos and NF-${\kappa}B$ p65 in mast cells.

Flavor Compounds of Cholesterol-Reduced Cheddar Cheese Slurries

  • This study was carried out to find the difference in flavor compounds between cholesterol-reduced Cheddar cheese slurries and control sample. The cheeses were made by 3 different treatments as followings: 1) Control (no homogenization, no ${\beta}$-CD), 2) Trt A (1,000 psi milk homogenization, 1% ${\beta}$-CD) and 3) Trt B (cream separation following by 10% ${\beta}$-CD, mixed with skim milk at 1,000 psi homogenization). The cholesterol removals of the cheeses were 79.30% (Trt A) and 91.22% (Trt B). The cheese slurries made by the cheeses were aged at $32^{\circ}C$ for 3 wk. The production of short-chain fatty acids (SCFA) was significantly increased with storage time in all treatments. Total amount of SCFA was dramatically increased at 2 wk and maintained thereafter in control group. The amounts of acetone and acetaldehyde were slightly increased in control at 3 wk, however, no difference was found in others. Ethanol production was dramatically increased at 1 wk and decreased thereafter in all treatments. Based on our results, cheese slurries for Trt B showed a highest cholesterol removal rate. Although little difference was found in flavor production, lower amount of SCFA was found in Trts A and B in 2 and 3 wk. It may indicate that a certain amount of SCFA is decreased during ${\beta}$-CD treatment.

  • In order to treat leachate from aged landfill site effectively, removal of biologically recalcitrant organic matter and denitrification efficiency were evaluated through the combination of $H_2O_2/O_3$ AOP pretreatment process and UASB process. The results can be summarized as follows. In case of leachate having low COD/N ratio from aged landfill site, it is possible to increase available COD for denitrification in nitrate utilizing denitrification and nitrite utilizing denitrification both by enhancing biodegradability of recalcitrant organic matter as applying $H_2O_2/O_3$ AOP to pretreatment process. In this experiment, it is found that available COD for denitrification can be increased to 1.0 and 0.4 g/day, respectively. Comparison has been made between requiring COD and available COD for denitrification in each experimental stages. It is expected that high rate of denitrification can be achieved with leachate from young landfill site because higher amount of available COD for denotrification is present in the leachate than the amount of requiring COD for denitrification. Especially, In leachate from aged landfill site with low COD/N ratio, it can be concluded that denitrification using nitrite nitrogen can enhance overall denitrification performance efficiently because denitrification using nitrite nitrogen requires less amount of carbon source than denitrification using nitrate nitrogen. Comparing the biogas production rate and nitrogen content of biogas under the condition of same amount of nitrate and nitrite addition, biogas production and nitrogen content of biogas are increased during denitrification after $H_2O_2/O_3$ AOP pretreatment process. Therefore, it can be confirmed that COD/N ratio in the leachate is increased. Applying $H_2O_2/O_3$ AOP as pretreatment system of landfill leachate seems to have little economic benefit because it requires additional carbon source to denitrify ammonia nitrogen in leachate coming from aged landfill site. However, it is possible to apply this pretreatment process to leachate from old landfill site in view of AOP process can achieve removal of biologically recalcitrant organic matter and increase of available COD for denitrification simultaneously.

Enhanced of Bio-Hydrogen Production from Microalgae by Thermal Pre-Treatment (열처리를 통한 미세조류로부터 바이오수소 생산 향상)

  • This study was conducted to increase the amount of bio-hydrogen production from microalgae(Chlorella vulgaris) in batch reactors by thermal pre-treatment. The optimization of thermal pre-treatment was conducted using statistic experimental design of response surface methodology. Two experimental parameters of temperature and reaction time were considered. The optimization condition was founded at the coded variables of <0.52, -0.07> corresponding to the experimental of heating temperature of $95.6^{\circ}C$ and reaction time of 57.9 min, respectively. Under the optimal condition, the maximum hydrogen production was predicted to 25.3mL $H_2/g$ dry cell weight (dcw), which was 9.1 times higher value of control(2.8mL $H_2/g$ dcw).

Process Balance of Natural Indigo Production based on Traditional Niram Method

  • In this work, the natural indigo production process from Polygonum tinctorium was balanced based on the traditional Niram method in Korea. A standard procedure was determined considering the conditions of indican extraction from plant material, the amount of alkali for precipitation, storage of extract, etc. The effect of experimental conditions on the yield of crude dye was investigated. The contents of indigo and indirubin of the crude dyes were analyzed by HPLC. Increase of the amount of crude dye was observed within 1-2.5 days of extraction time. Longer extraction beyond 2.5 days resulted in a slight decrease in the amount of crude dye. There was no consistency in terms of indigo content depending on extraction pH. We found that the storage of extract or harvested plants affected adversely to dye yield and dye quality. Based on the lab scale extraction, large scale extraction was performed for 2-2.5 days in water and 2.0-2.5 g/L of $Ca(OH)_2$ was applied for precipitation of indigo dye. We obtained natural indigo dye containing about 15% of pure indigo in scale-up production using whole plant except root.

  • A strain of alkalophilic Bacillus sp. C-21 has been Isolated from sold. The strain was capable of producing large amount of cyclodextrin glycosyltransferase (CGTase) in the high alkaline pH medium. The preferable medium composition was determined to be as follows : 1.0% soluble starch, 1.0% peptone, 0.5% yeast extract, 0.1% K2HP04, 0.02% MgSO4.7H2O and 1.0% Na2CO3(pH 10.0) The highest enzyme production was observed after 30hours of cultivation at 33$^{\circ}C$. The optimum temperature and pH for the activity of crude enzyme were 6$0^{\circ}C$ and 6.0, respectively. The enzyme was stable between pH 6.0 and 9.6, and up to 55$^{\circ}C$.

Influence of hydraulic retention time on biogas production during leachate treatment

  • The main objective of this study is to investigate the influence of hydraulic retention time (HRT) on biogas production during leachate treatment using an anaerobic reactor type Upflow Anaerobic Sludge Blanket. For this purpose, four HRTs ranging from 12 to 48 h were experienced. The obtained results showed that higher amount of biogas could be produced during longer HRTs. However, HRTs longer than 48 h could not affect clearly the biogas generation and considered as unnecessary given the small additional amount of biogas produced during the degradation process. A volume of $0.434L/L_{leachate}/d$ was achieved during the treatment with a HRT of 48 h. The higher biogas production, the smaller chemical oxygen demand (COD) values achieved. Besides, COD removal and biogas production positively correlate, showing that the active biomass has degraded effectively the organic load to produce biogas. Moreover, all the analyzed physicochemical parameters have experienced a decrease after 48 h except for the pH, which increased to approximately neutral value. More precisely, a decrease of 93.8%, 89.7%, 95%, 70%, 77%, and 84.4% was recorded for COD, electrical conductivity, total suspended solid, turbidity, $NH_4{^+}-N$, and $NO_3{^-}-N$, respectively.

  • This study was undertaken in order to examine the effect of ginger, gerlic and red pepper, usually used as the ingredients of Kimchi, on the growth and acid production of Lact. plantarum ATCC 8014 and Lact. fermenti ATCC 9338 during incubation in the medium containing different amount of each extract. The results obtained are as follows: 1. The effects of ginger extract a. The growth of Lact. plantarum ATCC 8014 and Lact. fermenti ATCC 9338 tended to be stimulated as the amount of extract added into the medium increased in a certain amount (3.64mg soluble solid/ml). b. The pH of the culture medium of Lact. plantarum ATCC 8014 became lower to some degree and acid production tended to be stimulated but acid production of Lact. fermenti ATCC 9338 tended to be suppressed as the amount of extract added in a certain amount (3.64mg soluble solid/ml) increased. 2. The effect of garlic extract a. The growth of Lact. plantarum ATCC 8014 and Lact. fermenti ATCC 9338 tended to be stim ulated as the amount of extract added in a certain amount (31.7 mg soluble solid/ml) increased. b. The acid production of Lact plantarum ATCC 8014 was suppressed (p<0.01) and drop in pH was suppressed (p<0.05) as the amount of extract added in a certain amount increased. In case of Lact. fermenti ATCC 9338, the acid production t ended to be suppressed also. 3. The effects of red pepper extract a. The growth of Lact. plantarum ATCC 8014 and Lact. fermenti ATCC 9338 tended to be stimulated as the amount of extract added in a certain amount (14.5 mg soluble solid/ml) increased.
